    The Consensus Molecular Subtypes of Colorectal Cancer

To resolve inconsistencies among the reported gene expression-based CRC classifications and facilitate clinical translation, we formed an international consortium dedicated to large-scale data sharing and analytics across expert groups. We show marked interconnectivity between six independent classification systems coalescing into four consensus molecular subtypes (CMS) with distinguishing features: CMS1 (MSI Immune, 14%), hypermutated, microsatellite unstable, strong immune activation; CMS2 (Canonical, 37%), epithelial, chromosomally unstable, marked WNT and MYC signaling activation; CMS3 (Metabolic, 13%), epithelial, evident metabolic dysregulation; and CMS4 (Mesenchymal, 23%), prominent transforming growth factor ÎČ activation, stromal invasion, and angiogenesis. Samples with mixed features (13%) possibly represent a transition phenotype or intra-tumoral heterogeneity. We consider the CMS groups the most robust classification system currently available for CRC - with clear biological interpretability - and the basis for future clinical stratification and subtype-based targeted interventions

    AprĂšs avoir Ă©tĂ© la premiĂšre d’Europe, la marine de Louis XIV a commencĂ© Ă  s’effondrer Ă  partir de 1707, et elle n’était dĂ©jĂ  plus que l’ombre d’elle-mĂȘme lorsque Louis XV a succĂ©dĂ© Ă  son arriĂšre-grand-pĂšre en 1715. SecrĂ©taire d’État de la Marine de 1723 Ă  1749, le comte de Maurepas est traditionnellement considĂ©rĂ© comme le bĂątisseur d’une nouvelle marine, qui aurait fait ses preuves au cours de la guerre de Succession d’Autriche, aprĂšs une longue pĂ©riode de paix avec la Grande-Bretagne. Les archives du Conseil de Marine rĂ©vĂšlent pourtant que c’est dĂšs 1719, que cette reconstruction a Ă©tĂ© entreprise. Avec le soutien du RĂ©gent, les membres de ce conseil dirigĂ© par le comte de Toulouse, Amiral de France, ont alors entrepris de se doter d’un instrument naval puissant, qu’ils destinaient Ă  la guerre d’escadre. Pendant quelques annĂ©es, d’importants efforts ont Ă©tĂ© faits pour mettre en chantier un grand nombre de nouveaux vaisseaux, avant que ce projet ne soit abandonnĂ© en 1725, suite Ă  une forte rĂ©duction des dĂ©penses. Il s’avĂšre donc que le rĂŽle de Maurepas a surtout consistĂ© Ă  entretenir, tant bien que mal, une marine restĂ©e inachevĂ©e. Il s’est Ă©galement efforcĂ©, il est vrai, de la prĂ©parer Ă  la guerre de course qu’il entendait mener en cas de nouveau conflit contre la Grande-Bretagne. Le fait est cependant que les opĂ©rations qui ont suivi l’entrĂ©e en guerre de la France en 1744 ont trĂšs vite rĂ©vĂ©lĂ©, non seulement les limites de cette stratĂ©gie, mais aussi l’impuissance et les fragilitĂ©s de la premiĂšre marine de Louis XV, dont Maurepas prononcera lui-mĂȘme l’acte de dĂ©cĂšs dans ses « RĂ©flexions sur le commerce et sur la marine » de 1745.After being the first in Europe, Louis XIV’s navy began to collapse from 1707, and it was already half-ruined when Louis XV succeeded its great grandfather in 1715. Having been Secretary of State for the Navy from 1723 to 1749, the Comte de Maurepas is traditionally regarded as the founder of a new navy, which would have proved its worth during the War of the Austrian Succession, after a long period of peace with Great Britain. However, the archives of the Navy Council reveal that it was as early as 1719 that this reconstruction was undertaken. With the support of the Regent, the members of this board headed by the Comte de Toulouse, Admiral of France, planned to create the naval instrument they needed for a guerre d’escadre. For a few years, great efforts were made to build a large number of new ships, before this program was abandoned in 1725, following a drastic budget reduction. It turns out, therefore, that Maurepas’s role was mainly to maintain, as best he could, a navy that remained unfinished. Admittedly, he also tried to prepare it for the guerre de course he intended to fight in the event of a new war with Great Britain. The fact is, however, that the naval operations which followed the outbreak of war in 1744 quickly revealed not only the limits of this strategy, but also the inability and the weaknesses of Louis XV's first navy, of which Maurepas himself performs the autopsy in his « Reflec- tions on Trade and Navy » of 1745

    Windowed OFDM versus OFDM/OQAM: A Transmission Capacity Comparison in the HomePlug AV Context

    International audienceWith the HPAV specification, windowed OFDM is now the most commonly used modulation technique in PLC networks. However, another modulation, named OFDM/OQAM, could notably improve the capacity of these networks. Imposing an orthogonality condition only in the real field, this modulation offers a great flexibility in the design of the prototype filter that shapes its symbols. Nevertheless, to take advantage of this degree of freedom, a study needs to be conducted to decide which prototype filter is the most adapted for PLC networks. The prototype filter length, its time and frequency localization and its orthogonality or nearly-orthogonality properties, represent some parameters that will determine the performance of OFDM/OQAM in a particular context. So, this contribution shows the achieved throughputs by OFDM/OQAM in an in-home PLC context, using different prototype filters and following the HPAV specification mask. These throughputs are also compared to the ones provided by the windowed OFDM modulation

    Frequency Division Multiplexing Analysis for Point-to-Multipoint Transmissions in Power Line Networks

    In Power Line Networks, a link adaptation is realized by combining Multi-Carrier modulations techniques and bit-loading algorithms, resulting in the creation of tone maps. A tone map defines, for each subcarrier on a particular transmission path, the appropriate constellation that has to be applied based on a link quality criterion. In this paper, we aim at showing how point-to-multipoint communications could take advantage of the definition of a Frequency Division Multiplexing (FDM) transmission mode, considering the context of IEEE P1901 networks. To do so, this new mode has been integrated in a PLC network simulator and the packet-level performance is assessed, using UDP flows. The first results exhibited in this paper show that, when associated with a recently released allocation algorithm, called the TMS (Tone Maps Splitting) algorithm, this solution could greatly improve data rates in future PLC networks
