    Local Lung Targeting of Tumor Associated Macrophages Combined with Cytoreductive Therapy Decrease Tumor Burden in a Secondary Lung Cancer Model

    Backgrounds. The efficacy of a locally administered small molecule colony stimulating factor 1 receptor inhibitor (CSF-1Ri), PLX3397, alone or in combination with cytoreductive therapy (paclitaxel) in reducing the tumor burden of an in vivo model of secondary lung cancer was investigated in this work. Local administration of immunotherapy to the lungs may enhance lung biodistribution of such therapies and reduce potential unwanted off-target toxicity. In addition, combination of such therapy with low dose standard of care chemotherapy may offer improved anti-tumor effects. Methods. Murine breast cancer cells (4T1, known to be highly metastasized to the lungs) were transduced to express the genes luciferase and tdTomato, and cells were injected to female Balb/c mice before being treated with PLX (intratracheally administered), paclitaxel (intravenously given), or the combination therapy. Both ex vivo bioluminescent imaging and lung weights were used to evaluate tumor burden. Western blot was performed using lung tumors to assess the effect of PLX3397 on its molecular target (phosphorylated CSF-1R). Immunofluorescence and Flowcytometry were utilized to examine the impact of treatment on tumorigenic tumor associated macrophages (M2 TAM). Results. Single-agent treatment partially decreased tumor burden, while combination therapy led to a significant reduction in tumor burden. PLX3397 significantly inhibited the expression of phosphorylated CSF-1R and reduced the number of M2 TAM without affecting the total macrophage population, thereby increasing the anti-tumorigenic (M1)/M2 ratio. Conclusion. Tumor burden reduction upon local administration of PLX3397 to the lungs correlates with the marked inhibition of the molecular target and the decrease in M2 TAM.https://scholarscompass.vcu.edu/gradposters/1065/thumbnail.jp

    Self-concept and facial appearance of individuals with an intellectual disability:A scoping review

    This paper looks at how self-concept is related to facial appearance. Very little information can be found on how persons with intellectual disabilities think and feel about their own facial appearance. In this review 19 articles on subject of self-concept in people with intellectual disabilities were included. Results show that self-concept of individuals with intellectual disabilities is affected by the relationships they have (positively and negatively). The perceptions of caregivers, peers and their awareness of stigma effects self-concept. How facial appearance affects the self-concept of persons with intellectual disabilities remains unclear and should be a focus of research in future. Abstract: Background The mental health and well-being of individuals can be positively affected by improving facial appearance through orthodontic treatment. However, research is limited on the relationship between facial appearance and self-concept of individuals with intellectual disabilities. Materials and methods This scoping review includes 19 articles and is aimed to examine the self-concept of individuals with intellectual disabilities and determine how the self-concept might be impacted by facial appearance. Furthermore, this review is exploratory in the way that an attempt was made to extract constructs that contribute to the self-concept of persons with intellectual disabilities. Results Unfortunately, no articles were found on how individuals with intellectual disabilities think about their own facial appearance. However, the articles on self-concept in persons with intellectual disabilities do mention some interesting facts: social comparisons lead to social stigma that can contribute negatively to the self-concept of individuals with intellectual disabilities. Also, the ability to communicate and express oneself can contribute to one's self-esteem. Furthermore, parents/caregivers often influence the concept of oneself. Moreover, an individual's relationship with their social environment defines their self-concept. Conclusion It is currently unknown how individuals with intellectual disabilities view their own facial appearance and whether their self-concept is influenced by their appearance. Questionnaires which address attitudes on facial appearance could provide a deeper understanding of the self-concept of individuals with intellectual disabilities and possibly make way for orthodontic treatment. However, caution is needed concerning changing the physical appearance of persons with intellectual disabilities. One should also battle stigma by encouraging the social inclusion of persons with intellectual disability

    Tumor necrosis factor receptor 1 (TNFRI) for ventilator-associated pneumonia diagnosis by cytokine multiplex analysis

    The diagnosis of ventilator-associated pneumonia (VAP) is challenging. An important aspect to improve outcome is early recognition of VAP and the initiation of the appropriate empirical treatment. We hypothesized that biological markers in plasma can rule out VAP at the moment of clinical suspicion and could rule in VAP before the diagnosis can be made clinically. In this prospective study, patients with VAP (n = 24, microbiology confirmed) were compared to controls (n = 19) with a similar duration of mechanical ventilation. Blood samples from the day of VAP diagnosis and 1 and 3 days before were analyzed with a multiplex array for markers of inflammation, coagulation, and apoptosis. The best biomarker combination was selected and the diagnostic accuracy was given by the area under the receiver operating characteristic curve (ROC-AUC). TNF-receptor 1 (TNFRI) and granulocyte colony-stimulating factor (GCSF) were selected as optimal biomarkers at the day of VAP diagnosis, which resulted in a ROC-AUC of 0.96, with excellent sensitivity. Three days before the diagnosis TNFRI and plasminogen activator inhibitor-1 (PAI-1) levels in plasma predicted VAP with a ROC-AUC of 0.79. The slope of IL-10 and PAI-1 resulted in a ROC-AUC of 0.77. These biomarkers improved the classification of the clinical pulmonary infection score when combined. Concentration of TNFRI and PAI-1 and the slope of PAI-1 and IL-10 may be used to predict the development of VAP as early as 3 days before the diagnosis made clinically. TNFRI and GCSF may be used to exclude VAP at the moment of clinical suspicion. Especially TNFRI seems to be a promising marker for the prediction and diagnosis of VA

    Frost Multidimensional Perfectionism Scale: the portuguese version

    CONTEXTO: A Escala Multidimensional de Perfeccionismo de Frost (FMPS) é uma das escalas mais usadas em todo o mundo para avaliar o perfeccionismo. OBJETIVO: Analisar as características psicométricas da versão portuguesa da FMPS. MÉTODOS: A amostra foi constituída por 217 estudantes do ensino superior (178 mulheres). Um subgrupo (n = 166) completou o reteste após quatro semanas. RESULTADOS: A consistência interna da escala mostrou ser elevada (alfa de Cronbach = ,857). As correlações item-total corrigido variaram entre ,019 e ,548. Os resultados também sugeriram uma boa estabilidade temporal da escala, sendo a correlação teste-reteste de ,765. Foi realizada a análise das componentes principais com rotação Varimax e com base no Scree plot foram extraídas duas soluções fatoriais robustas (quatro e seis fatores). A análise paralela (Monte Carlo PCA) confirmou a solução de seis fatores. A validade concorrente com a escala MPS de Hewitt e Flett foi elevada, assim como a sua capacidade discriminante dos afetos positivos e negativos (Perfil de Estados de Humor - POMS). CONCLUSÃO: As duas estruturas fatoriais (quatro e seis fatores) encontradas na versão portuguesa da Escala Multidimensional de Perfeccionismo de Frost replicam os resultados obtidos por diferentes autores, em diferentes amostras e culturas. Esse fato sugere que essa escala é um instrumento robusto para a avaliação do perfeccionismo em vários contextos, clínicos e de investigação, bem como em estudos transculturais.BACKGROUND: The Frost Multidimensional Perfectionism Scale is one of the most world widely used measures of perfectionism. OBJECTIVE: To analyze the psychometric properties of the Portuguese version of the Frost Multidimensional Perfectionism Scale. METHODS: Two hundred and seventeen (178 females) students from two Portuguese Universities filled in the scale, and a subgroup (n = 166) completed a retest with a four weeks interval. RESULTS: The scale reliability was good (Cronbach alpha = .857). Corrected item-total correlations ranged from .019 to .548. The scale test-retest reliability suggested a good temporal stability with a test-retest correlation of .765. A principal component analysis with Varimax rotation was performed and based on the Scree plot, two robust factorial structures were found (four and six factors). The principal component analyses, using Monte Carlo PCA for parallel analyses confirmed the six factor solution. The concurrent validity with Hewitt and Flett MPS was high, as well as the discriminant validity of positive and negative affect (Profile of Mood Stats-POMS). DISCUSSION: The two factorial structures (of four and six dimensions) of the Portuguese version of Frost Multidimensional Perfectionism Scale replicate the results from different authors, with different samples and cultures. This suggests this scale is a robust instrument to assess perfectionism, in several clinical and research settings as well as in transcultural studies

    A partir da evolução do ensino EAD no Brasil pode-se afirmar que tal prática se encontra consolidada no curso superior. Apesar desta afirmativa, pouco se relata na estrutura básica de ensino bem como sua aplicaçao. Logo, o trabalho objetiva realizar o estudo de caso na aplicação do ensino EAD, de forma complementar ao ensino básico tradicional. A pesquisa abrange as dificuldades que envolvem a sua aplicabilidade ao corpo docente, demonstrando que a inserção da forma de ensino híbrida complementar na educação, aplicada de forma gradual, não obrigatória e com suporte conciso da área técnica, propiciando uma forte ligação entre a instituição, o professor e aluno por meio do ambiente virtual de aprendizagem. O método de aplicação e construção do Moodle apresentado demostrou ser eficaz como complemento do ensino tradicional, com aceitação junto ao corpo de alunos e professores, tornando o professor mais presente na vida do estudante.
