    A study was carried out on Met- and Leu-enkephalin, Gastrin / CCK-, SP-, CGRP-, NPY- immunoreactive fibers using paraffin sections of dental pulp taken from 8 apparently normal teeth (wisdom teeth or teeth extracted for orthodontic reasons).Within the limitations of the samples studied, dental pulp is characterized by the presence of sensory (Enkephalin-, Gastrin/CCK- immunoreactive) and pain fibers (SP-immunoreactive) and of fibers with a potent vasodilatory action (CGRP-immunoreactive) and by the absence of fibers with a vasoconstrictor action (NPY-immunoreactive).Notre étude a porté sur la mise en évidence sur des coupes après inclusion à la paraffine des fibres Met- et Leu-enképhaline, Gastrine / CCK-, SP-, CGRP- et NPY-immunoréactives, dans la pulpe de huit dents normales (dents de sagesse ou dents extraites pour des raisons orthodontiques).Dans la limite des échantillons utilisés, la pulpe dentaire est caractérisée par la présence de fibres sensitives (Enképhalin-, Gastrin/CCK-immunoréactives), des fibres de la douleur (SP-immunoréactives) et de fibre à action vasodilatatrice (CGRP-immunoréactives) et par l’absence de fibres à action vasoconstrictive (NPY-immunoréactives)

    Ultrastructural changes of collagen and elastin in human gingiva during orthodontic tooth movement

    After 15 days of mesializing or distalizing orthodontic treatment, 10 permanent premolars of young patients were extracted with the interdental gingiva. The connective tissues of the compressed or stretched interdental papillae were compared to that of untreated samples by light and transmission electron microscope.Large collagen fibres bundles represented by fibrils with a banding pattern of 64 nm and a mean diameter of 75 nm were observed in compressed interdental gingiva. Several elastic fibres with a mean diameter of 950 nm were also present. In some central areas of compressed gingiva collagen fibrils longitudinally split into widely spaced microfibrils were often observed in proximity to the elastic fibres.In stretched and untreated interdental papillae the collagen fibrils presented a mean diameter of 66 nm and 57 nm respectively. In both groups, few elastic fibres ranging in diameter 600 nm were seen. The increased size of the gingival collagen fibrils undergoing pressure and tension is indicative of remodelling of the fibrous collagen system.The fair increase in number and size of elastic fibres in compressed gingiva suggests that the elastic fibre system takes over the place whenever a collapse of the collagenous framework occurs.Après 15 jours de traitement orthodontique par mésialisation ou distalisation, 10 prémolaires permanentes ont été extraites chez de jeunes patients, ainsi que la gencive interdentaire adjacente. Le tissu conjonctif des papilles interdentaires comprimées ou étirées a été comparé avec celui d’échantillons de dents n’ayant pas subi de traitement. L’observation a été faite en microscopie classique et en microscopie électronique à transmission. Dans la gencive interdentaire comprimée, on observe de longs faisceaux de fibres de collagène faits de fibrilles qui présentent une périodicité de 64 nm et un diamètre inférieur à 75 nm. Ont été observées également de fibres élastiques d’une diamètre inférieur à 950 nm. Dans certaines zones centrales de gencive comprimée, on peut voir, à proximité dess fibres élastiques, des fibrilles de collagène réparties longitudinalement en microfibrilles très espacées.Dans la gencive interdentaire étirée et dans celle non soumise au traitement orthodontique, les fibrilles de collagène présentent un diamètre de 66 nm et 57 nm respectivement. Dans ces deux groupes, on note un très petit nombre de fibres élastiques de 600 nm de diamètre. L’augmentation du nombre et de la taille des fibres élastiques dans la gencive comprimée indique que le système de fibres élastiques remplace la trame collagène qui a subi un collapsus

    Novel role for polycystin-1 in modulating cell proliferation through calcium oscillations in kidney cells

    Objectives: Polycystin-1 (PC1), a signalling receptor regulating Ca2+-permeable cation channels, is mutated in autosomal dominant polycystic kidney disease, which is typically characterized by increased cell proliferation. However, the precise mechanisms by which PC1 functions on Ca2+ homeostasis, signalling and cell proliferation remain unclear. Here, we investigated the possible role of PC1 as a modulator of non-capacitative Ca2+ entry (NCCE) and Ca2+ oscillations, with downstream effects on cell proliferation. Results and discussion: By employing RNA interference, we show that depletion of endogenous PC1 in HEK293 cells leads to an increase in serum-induced Ca2+ oscillations, triggering nuclear factor of activated T cell activation and leading to cell cycle progression. Consistently, Ca2+ oscillations and cell proliferation are increased in PC1-mutated kidney cystic cell lines, but both abnormal features are reduced in cells that exogenously express PC1. Notably, blockers of the NCCE pathway, but not of the CCE, blunt abnormal oscillation and cell proliferation. Our study therefore provides the first demonstration that PC1 modulates Ca2+ oscillations and a molecular mechanism to explain the association between abnormal Ca2+ homeostasis and cell proliferation in autosomal dominant polycystic kidney disease

    Are pharmaceuticals removal and membrane fouling in electromembrane bioreactor affected by current density?

    Abstract Pharmaceutical active compounds (PhACs) have been detected at significant concentrations in various natural and artificial aquatic environments. In this study, electro membrane bioreactor (eMBR) technology was used to treat simulated municipal wastewater containing widely-used pharmaceuticals namely amoxicillin (AMX), diclofenac (DCF) and carbamazepine (CBZ). The effects of varying current density on the removal of PhACs (AMX, DCF and CBZ) and conventional pollutants (chemical oxygen demand (COD), dissolved organic carbon (DOC), humic substances, ammonia nitrogen (NH 4 -N), nitrate nitrogen (NO 3 -N) and orthophosphate (PO 4 -P) species) were examined. High COD and DOC removal efficiencies (~100%) were obtained in all the experimental runs regardless of applied current density. In contrast, enhanced removal efficiencies for AMX, DCF and CBZ were achieved at high current densities. Membrane fouling rate in eMBR with respect to conventional MBR was reduced by 24, 44 and 45% at current densities of 0.3, 0.5 and 1.15 mA/cm 2 , respectively. The mechanism for pharmaceutical removal in this study proceeded by: (1) charge neutralization between negatively-charged pharmaceutical compounds and positive electro-generated aluminium coagulants to form larger particles and (2) size exclusion by membrane filtration

    Combination of Electrochemical Processes with Membrane Bioreactors for Wastewater Treatment and Fouling Control: A Review

    This paper provides a critical review about the integration of electrochemical processes into membrane bioreactors (MBR) in order to understand the influence of these processes on wastewater treatment performance and membrane fouling control. The integration can be realized either in an internal or an external configuration. Electrically enhanced membrane bioreactors or electro membrane bioreactors (eMBRs) combine biodegradation, electrochemical and membrane filtration processes into one system providing higher effluent quality as compared to conventional MBRs and activated sludge plants. Furthermore, electrochemical processes, such as electrocoagulation, electrophoresis, and electroosmosis, help to mitigate deposition of foulants into the membrane and enhance sludge dewaterability by controlling the morphological properties and mobility of the colloidal particles and bulk liquid. Intermittent application of minute electric field has proven to reduce energy consumption and operational cost as well as minimize the negative effect of direct current field on microbial activity which are some of the main concerns in eMBR technology. The present review discusses important design considerations of eMBR, its advantages as well as its applications to different types of wastewater. It also presents several challenges that need to be addressed for future development of this hybrid technology which include treatment of high strength industrial wastewater and removal of emerging contaminants, optimization study, cost benefit analysis and the possible combination with microbial electrolysis cell for biohydrogen production

    Psychostimulant effect of the synthetic cannabinoid JWH-018 and AKB48: Behavioral, neurochemical, and dopamine transporter scan imaging studies in mice

    JWH-018 and AKB48 are two synthetic cannabinoids (SCBs) belonging to different structural classes and illegally marketed as incense, herbal preparations, or chemical supply for theirs psychoactive cannabis-like effects. Clinical reports from emergency room reported psychomotor agitation as one of the most frequent effects in people assuming SCBs. This study aimed to investigate the psychostimulant properties of JWH-018 and AKB48 in male CD-1 mice and to compare their behavioral and biochemical effects with those caused by cocaine and amphetamine. In vivo studies showed that JWH-018 and AKB48, as cocaine and amphetamine, facilitated spontaneous locomotion in mice. These effects were prevented by CB1 receptor blockade and dopamine (DA) D1/5 and D2/3 receptors inhibition. SPECT-CT studies on dopamine transporter (DAT) revealed that, as cocaine and amphetamine, JWH-018 and AKB48 decreased the [123I]-FP-CIT binding in the mouse striatum. Conversely, in vitro competition binding studies revealed that, unlike cocaine and amphetamine, JWH-018 and AKB48 did not bind to mouse or human DAT. Moreover, microdialysis studies showed that the systemic administration of JWH-018, AKB48, cocaine, and amphetamine stimulated DA release in the nucleus accumbens (NAc) shell of freely moving mice. Finally, unlike amphetamine and cocaine, JWH-018 and AKB48 did not induce any changes on spontaneous [3H]-DA efflux from murine striatal synaptosomes. The present results suggest that SCBs facilitate striatal DA release possibly with different mechanisms than cocaine and amphetamine. Furthermore, they demonstrate, for the first time, that JWH-018 and AKB48 induce a psychostimulant effect in mice possibly by increasing NAc DA release. These data, according to clinical reports, outline the potential psychostimulant action of SCBs highlighting their possible danger to human health

    The pandemic within the pandemic: the surge of neuropsychological disorders in Italian children during the COVID-19 era

    Background: Quarantine and isolation measures during COVID-19 pandemic may have caused additional stress and challenged the mental health of the youth. Aim of the study is to investigate the COVID-19 pandemic impact on neuropsychological disorders (NPD) of Italian children and adolescents to provide general pediatric recommendations. Material and methods: A retrospective multicenter observational study was planned by the Italian Pediatric Society (SIP) to explore the impact of COVID-19 on the access of children to pediatric Emergency Departments (pED) for the evaluation of neuropsychological symptoms, collecting the classification codes of diagnoses between March 1, 2019 and March 2, 2021. The period study was split into two sub-periods: a pre COVID-19 period (from March 1 2019 to March 1, 2020) and a COVID-19 period (from March 2, 2020 to March 2, 2021). As additional information, data on NPD hospitalizations in any pediatric department of the involved centers were recorded. Results: During the study period, a total of 533,318 children were admitted to the pED involved in the study. Despite a 48.2% decline of pED admissions, there was a significant increase (83.1%) in patient admissions for NPD. The most frequent NPD conditions which increased during the COVID-19 pandemic were suicidal ideation (+ 147%), depression (+ 115%), eating disorder (+ 78.4%), and psychosis (+ 17.2%). During the pandemic period, a 39.5% increase in NPD hospitalizations was observed as well. The NPD disorders that mostly required hospitalizations were suicidal ideation (+ 134%), depression (+ 41.4%), eating disorder (+ 31.4%), and drug abuse (+ 26.7%). COVID-19 pandemic had a major impact on children's health, mainly on their NPD development. Neuropsychological assessment should be required at the primary level, in the pediatrician's office, to facilitate early capture of the sign of impairment and provide an adequate treatment. Conclusion: SIP underlines the psychological consequences of COVID 19 pandemic on the youngest and recommends an early identification of NPD in the pediatric population to avoid other serious consequences for children's physical and mental health

    Odontostomatologic management of patients receiving oral anticoagulant therapy: a retrospective multicentric study

    Introduction: Today, we frequently find patients taking oral anticoagulant therapy (OAT), a prophylaxis against the occurrence of thromboembolic events. An oral surgeon needs to know how to better manage such patients, in order to avoid hemorrhagic and thromboembolic complications. Materials and methods: A group of 193 patients (119 men aged between 46 and 82 and 74 women aged between 54 and 76) undergoing OAT for more than 5 years were managed with a standardized management protocol and a 2-months follow-up. The aim of the present study was to apply a protocol, which could provide a safe intra- and postoperative management of patients on OAT. Results: Among the 193 patients, only 2 had postoperative complications. Conclusions: We think that the protocol used in the present study can be used for complete safety in the treatment of this type of patients.
