    The Ramsey method in high-precision mass spectrometry with Penning traps: Experimental results

    The highest precision in direct mass measurements is obtained with Penning trap mass spectrometry. Most experiments use the interconversion of the magnetron and cyclotron motional modes of the stored ion due to excitation by external radiofrequency-quadrupole fields. In this work a new excitation scheme, Ramsey's method of time-separated oscillatory fields, has been successfully tested. It has been shown to reduce significantly the uncertainty in the determination of the cyclotron frequency and thus of the ion mass of interest. The theoretical description of the ion motion excited with Ramsey's method in a Penning trap and subsequently the calculation of the resonance line shapes for different excitation times, pulse structures, and detunings of the quadrupole field has been carried out in a quantum mechanical framework and is discussed in detail in the preceding article in this journal by M. Kretzschmar. Here, the new excitation technique has been applied with the ISOLTRAP mass spectrometer at ISOLDE/CERN for mass measurements on stable as well as short-lived nuclides. The experimental resonances are in agreement with the theoretical predictions and a precision gain close to a factor of four was achieved compared to the use of the conventional excitation technique.Comment: 12 pages, 14 figures, 2 table

    Mutant and chimeric recobinant plasminogen activatorsproduction in eukaryotic cellsand preliminary characterization

    Mutant urokinase-type plasminogen activator (u-PA) genes and hybrid genes between tissue-type plasminogen activator (t-PA) and u-PA have been designed to direct the synthesis of new plasminogen activators and to investigate the structure-function relationship in these molecules. The following classes of constructs were made starting from cDNA encoding human t-PA or u-PA: 1) u-PA mutants in which the Arg156 and Lys158 were substituted with threonine, thus preventing cleavage by thrombin and plasmin; 2) hybrid molecules in which the NH2-terminal regions of t-PA (amino acid residues 1-67, 1-262, or 1-313) were fused with the COOH-terminal region of u-PA (amino acids 136-411, 139-411, or 195-411, respectively); and 3) a hybrid molecule in which the second kringle of t-PA (amino acids 173-262) was inserted between amino acids 130 and 139 of u-PA. In all cases but one, the recombinant proteins, produced by transfected eukaryotic cells, were efficiently secreted in the culture medium. The translation products have been tested for their ability to activate plasminogen after in situ binding to an insolubilized monoclonal antibody directed against urokinase. All recombinant enzymes were shown to be active, except those in which Lys158 of u-PA was substituted with threonine. Recombination of structural regions derived from t-PA, such as the finger, the kringle 2, or most of the A-chain sequences, with the protease part or the complete u-PA molecule did not impair the catalytic activity of the hybrid polypeptides. This observation supports the hypothesis that structural domains in t-PA and u-PA fold independently from one to another

    A framework for power analysis using a structural equation modelling procedure

    BACKGROUND: This paper demonstrates how structural equation modelling (SEM) can be used as a tool to aid in carrying out power analyses. For many complex multivariate designs that are increasingly being employed, power analyses can be difficult to carry out, because the software available lacks sufficient flexibility. Satorra and Saris developed a method for estimating the power of the likelihood ratio test for structural equation models. Whilst the Satorra and Saris approach is familiar to researchers who use the structural equation modelling approach, it is less well known amongst other researchers. The SEM approach can be equivalent to other multivariate statistical tests, and therefore the Satorra and Saris approach to power analysis can be used. METHODS: The covariance matrix, along with a vector of means, relating to the alternative hypothesis is generated. This represents the hypothesised population effects. A model (representing the null hypothesis) is then tested in a structural equation model, using the population parameters as input. An analysis based on the chi-square of this model can provide estimates of the sample size required for different levels of power to reject the null hypothesis. CONCLUSIONS: The SEM based power analysis approach may prove useful for researchers designing research in the health and medical spheres

    A linear radiofrequency ion trap for accumulation, bunching, and emittance improvement of radioactive ion beams

    An ion beam cooler and buncher has been developed for the manipulation of radioactive ion beams. The gas-filled linear radiofrequency ion trap system is installed at the Penning trap mass spectrometer ISOLTRAP at ISOLDE/CERN. Its purpose is to accumulate the 60-keV continuous ISOLDE ion beam with high efficiency and to convert it into low-energy low-emittance ion pulses. The efficiency was found to exceed 10% in agreement with simulations. A more than 10-fold reduction of the ISOLDE beam emittance can be achieved. The system has been used successfully for first on-line experiments. Its principle, setup and performance will be discussed

    Highly specific and sensitive non-radioactive molecular identification of Phytophthora cinnamomi

    In response to the need for a faster, more reliable method for identifying Phytophthora cinnamomi in cork oak soils in Portugal, a simple, fast, sensitive molecular identification method is described. It is based on a colorimetric assay which involves an oligonucleotide capture probe covalently immobilised on microtitration wells, a multi-biotinylated oligonucleotide detection probe and the PCR-amplified target DNA. The target DNA is a 349 bp DNA fragment partially covering the 3'-translated and 3'- untranslated regions of the cinnamomin gene. When the specificity of the PCR reaction was evaluated in vitro using isolates of P. cinnamomi and eight other Phytophthora species, including the related P. cambivora, it was specific to P. cinnamomi. When 30 isolates of P. cinnamomi from oak roots in southern Portugal were assayed, 26 gave a strong positive response. The assay has a sensitivity of about 2±5 genome equivalents of P. cinnamomi. The reason for the negative response of four isolates remains unclear

Some experiments can be carried out under conditions which prevail in hot stellar plasmas. The experimental potential of storage and cooling of exotic nuclei is illustrated with recent experimental results and an outlook to future experiments is presented

    Beating the news using social media: the case study of American Idol

    We present a contribution to the debate on the predictability of social events using big data analytics. We focus on the elimination of contestants in the American Idol TV shows as an example of a well defined electoral phenomenon that each week draws millions of votes in the USA. This event can be considered as basic test in a simplified environment to assess the predictive power of Twitter signals. We provide evidence that Twitter activity during the time span defined by the TV show airing and the voting period following it correlates with the contestants ranking and allows the anticipation of the voting outcome. Twitter data from the show and the voting period of the season finale have been analyzed to attempt the winner prediction ahead of the airing of the official result. We also show that the fraction of tweets that contain geolocation information allows us to map the fanbase of each contestant, both within the US and abroad, showing that strong regional polarizations occur. The geolocalized data are crucial for the correct prediction of the final outcome of the show, pointing out the importance of considering information beyond the aggregated Twitter signal. Although American Idol voting is just a minimal and simplified version of complex societal phenomena such as political elections, this work shows that the volume of information available in online systems permits the real time gathering of quantitative indicators that may be able to anticipate the future unfolding of opinion formation events
