Determining scoring values in cases of UGIB is important to do so it can be used as a basis to determine the mortality rate of UGIB patients. Despite the dependency of most risk scoring systems for this disorder on endoscopic findings, the Glasgow-Blatchford Bleeding Score (GBS) and Incomplete Rockall Score (IRS) are based on simple variables. Our purpose to differentiate about sensitivity and spesificity between IRS and GBS to predict the severity of patient with non variceal upper GI bleeding without the endoscopic score. Our study was undertaken in emergency department Saiful Anwar hospital, Malang. The sample consist of 102 patients with non variceal upper GI bleeding from November 2012 to April 2013. With receiving-operator characteristics (ROC), we compare the significance between IRS and GBS in predicting the severity of patient with non variceal upper GI bleeding. In next step, we compare both of them with the diagnostic test. ROC curve shows that GBS is slightly better than IRS (0,955 vs 0,744, p<0,05). IRS has better specificity (79,55% vs 70,45%) than GBS. But, GBS has better sensitivity (96,55% vs 65,52%). The final results is GBS as an alternative diagnostic score has better results than IRS in understanding the severity of patient with non variceal upper GI bleeding in Saiful Anwar hospital. Therefore, it can be used as a simple predictor replacing the endoscopic score

One  kind  of  fishing  gear  that  use  to  captured  phototaxis  positif  fish  is  Lift  Net.  Lamp  was  no longer  used  as  fish  aggregating  device  for  lift  net  because  it  was  uneffective  and  unefficient  to gathered  fish  target.  If  the  problem  still  continued  it  can  raised  fuels  consumption  for  lamp attractor. Another fish aggregating device “rumpon” was a solution for lift net operation. Research objectives were to increase effectivity, efficiency and fishing production, and hopefully it can raise fisheries income.  Keyword : Liftnet, Rumpon, Demak Seawater

Hydrodynamics of domain growth in nematic liquid crystals

We study the growth of aligned domains in nematic liquid crystals. Results are obtained solving the Beris-Edwards equations of motion using the lattice Boltzmann approach. Spatial anisotropy in the domain growth is shown to be a consequence of the flow induced by the changing order parameter field (backflow). The generalization of the results to the growth of a cylindrical domain, which involves the dynamics of a defect ring, is discussed.Comment: 12 revtex-style pages, including 12 figures; small changes before publicatio

Dominant Mutations in GRHL3 Cause Van der Woude Syndrome and Disrupt Oral Periderm Development

Mutations in interferon regulatory factor 6 (IRF6) account for ∌70% of cases of Van der Woude syndrome (VWS), the most common syndromic form of cleft lip and palate. In 8 of 45 VWS-affected families lacking a mutation in IRF6, we found coding mutations in grainyhead-like 3 (GRHL3). According to a zebrafish-based assay, the disease-associated GRHL3 mutations abrogated periderm development and were consistent with a dominant-negative effect, in contrast to haploinsufficiency seen in most VWS cases caused by IRF6 mutations. In mouse, all embryos lacking Grhl3 exhibited abnormal oral periderm and 17% developed a cleft palate. Analysis of the oral phenotype of double heterozygote (Irf6+/−;Grhl3+/−) murine embryos failed to detect epistasis between the two genes, suggesting that they function in separate but convergent pathways during palatogenesis. Taken together, our data demonstrated that mutations in two genes, IRF6 and GRHL3, can lead to nearly identical phenotypes of orofacial cleft. They supported the hypotheses that both genes are essential for the presence of a functional oral periderm and that failure of this process contributes to VWS

Protein kinase C activation disrupts epithelial apical junctions via ROCK-II dependent stimulation of actomyosin contractility

<p>Abstract</p> <p>Background</p> <p>Disruption of epithelial cell-cell adhesions represents an early and important stage in tumor metastasis. This process can be modeled <it>in vitro </it>by exposing cells to chemical tumor promoters, phorbol esters and octylindolactam-V (OI-V), known to activate protein kinase C (PKC). However, molecular events mediating PKC-dependent disruption of epithelial cell-cell contact remain poorly understood. In the present study we investigate mechanisms by which PKC activation induces disassembly of tight junctions (TJs) and adherens junctions (AJs) in a model pancreatic epithelium.</p> <p>Results</p> <p>Exposure of HPAF-II human pancreatic adenocarcinoma cell monolayers to either OI-V or 12-O-tetradecanoylphorbol-13-acetate caused rapid disruption and internalization of AJs and TJs. Activity of classical PKC isoenzymes was responsible for the loss of cell-cell contacts which was accompanied by cell rounding, phosphorylation and relocalization of the F-actin motor nonmuscle myosin (NM) II. The OI-V-induced disruption of AJs and TJs was prevented by either pharmacological inhibition of NM II with blebbistatin or by siRNA-mediated downregulation of NM IIA. Furthermore, AJ/TJ disassembly was attenuated by inhibition of Rho-associated kinase (ROCK) II, but was insensitive to blockage of MLCK, calmodulin, ERK1/2, caspases and RhoA GTPase.</p> <p>Conclusion</p> <p>Our data suggest that stimulation of PKC disrupts epithelial apical junctions via ROCK-II dependent activation of NM II, which increases contractility of perijunctional actin filaments. This mechanism is likely to be important for cancer cell dissociation and tumor metastasis.</p
