    Modeling the effect of copper availability on bacterial denitrification

    When denitrifying bacteria such as Paracoccus denitrificans respire anaerobically they convert nitrate to dinitrogen gas via a pathway which includes the potent greenhouse gas, nitrous oxide (NO). The copper-dependent enzyme Nitrous Oxide reductase (Nos) catalyzes the reduction of NO to dinitrogen. In low-copper conditions, recent experiments in chemostats have demonstrated that Nos efficiency decreases resulting in significant NO emissions. For the first time, a chemostat-based mathematical model is developed that describes the anaerobic denitrification pathway based on Michaelis-Menten kinetics and published kinetic parameters. The model predicts steady-state enzyme levels from experimental data. For low copper concentrations, the predicted Nos level is significantly reduced, whereas the levels for the non copper-dependent reductases in the pathway remain relatively unaffected. The model provides time courses for the pathway metabolites that accurately reflect previously published experimental data. In the absence of experimental data purely predictive analyses can also be readily performed by calculating the relative Nos level directly from the copper concentration. Here, the model quantitatively estimates the increasing level of emitted NO as the copper level decreases. We have developed a mathematical model for the denitrification pathway based on existing experimental results, Michaelis-Menten kinetics and experimentally obtained kinetic constants. This is the first such model to incorporate the copper concentration in order to predict emissions of the potent greenhouse gas, nitrous oxide (NO), as well as the other nitrogenous compounds in the pathway. The model predicts increasing NO emissions as the copper level is lowered, in agreement with experimental observations in chemostats. © 2013 The Authors.     Evaluation of work-related psychosocial factors and regional musculoskeletal pain: results from a EULAR Task Force

    Objectives: to establish whether review articles provide consistent conclusions on associations between workplace psychosocial factors and musculoskeletal pain and, if differences exist, to explore whether this is related to the methods used.Methods: reviews, reported up to February 2007, that included consideration of workplace psychosocial factors and upper limb, back or knee pain were identified through searches of multiple databases. The specific work-related psychosocial factors considered were job demands, support, job autonomy and job satisfaction. The conclusions of each review on one or more of the psychosocial/musculoskeletal pain associations were extracted.Results: 15 review articles were identified that considered one or more of the regional pain syndromes included in the study. For back pain, the most consistent conclusions (four reviews positive out of six) were with high job demands and low job satisfaction. The studies of upper limb pain were exclusively related to shoulder and/or neck pain, and the most consistent positive conclusions were with high and low job demands (four reviews positive out of six and two reviews positive out of three, respectively). For knee pain, only a single review was identified. For individual reviews of back and upper limb pain, there were marked differences in the number of associations concluded to be positive between reviews.Conclusions: the reasons for reviews coming to different conclusions included that they were often evaluating different bodies of evidence (according to their search criteria, the year when the review was conducted, the role that quality assessment played in whether studies contributed to evidence, and the combination of risk factors addressed in individual studies), but more important was whether the review specified explicit criteria for making conclusions on strength of evidence. These conclusions emphasise the importance of developing standardised methods for conducting such evaluations of existing evidence and the importance of new longitudinal studies for clarifying the temporal relationship between psychosocial factors and musculoskeletal pain in the workplac

    Investigating the palaeoenvironmental context of Late Pleistocene human dispersals into Southeast Asia: a review of stable isotope applications

    We review palaeoenvironmental applications of stable isotope analysis to Late Pleistocene archaeological sites across Southeast Asia (SEA), a region critical to understanding the evolution of Homo sapiens and other co-existing Late Pleistocene (124–11.7 ka) hominins. Stable isotope techniques applied to archaeological deposits offer the potential to develop robust palaeoenvironmental reconstructions, to contextualise the occupational and non-occupational history of a site. By evaluating the published research in this field, we show that sediments, guano, tooth enamel, speleothem and biomolecular material such as leaf waxes have great potential to provide site-specific palaeoenvironmental records and local and catchment-scale landscape context to hominin dispersal in the region. However, stable isotope techniques used in these contexts are in their infancy in SEA, and the diagenetic controls associated with hot and humid environments that typify the region are not yet fully understood. Additionally, availability of sources of stable isotopes varies between sites. Nonetheless, even the limited research currently available shows that stable isotope analyses can aid in developing a better understanding of the role of the environment on the nature and timing of dispersals of our species eastwards into SEA and beyond.Meghan S.     Spatial regulation of the glycocalyx component podocalyxin is a switch for prometastatic function

    The glycocalyx component and sialomucin podocalyxin (PODXL) is required for normal tissue development by promoting apical membranes to form between cells, triggering lumen formation. Elevated PODXL expression is also associated with metastasis and poor clinical outcome in multiple tumor types. How PODXL presents this duality in effect remains unknown. We identify an unexpected function of PODXL as a decoy receptor for galectin-3 (GAL3), whereby the PODXL-GAL3 interaction releases GAL3 repression of integrin-based invasion. Differential cortical targeting of PODXL, regulated by ubiquitination, is the molecular mechanism controlling alternate fates. Both PODXL high and low surface levels occur in parallel subpopulations within cancer cells. Orthotopic intraprostatic xenograft of PODXL-manipulated cells or those with different surface levels of PODXL define that this axis controls metastasis in vivo. Clinically, interplay between PODXL-GAL3 stratifies prostate cancer patients with poor outcome.     Measurements of exclusive B_s^0 decays at the Y(5S) resonance

    Several exclusive Bs0B_s^0 decays are studied using a 1.86 fb-1 data sample collected at the Y(5S) resonance with the Belle detector at the KEKB asymmetric energy e^+ e^- collider. In the Bs0Dsπ+B_s^0 \to D_s^- \pi^+ decay mode we find 10 Bs0B_s^0 candidates and measure the corresponding branching fraction. Combining the B_s^0 -> D_s^{(*)-} \pi^+, B_s^0 -> D_s^{(*)-} \rho^+, B_s^0 -> J/\psi \phi and B_s^0 -> J/\psi \eta decay modes, a significant Bs0B_s^0 signal is observed. The ratio \sigma (e^+ e^- -> B_s^* \bar{B}_s^*) / \sigma (e^+ e^- -> B_s^{(*)} \bar{B}_s^{(*)}) = (93^{+7}_{-9} \pm 1)% is obtained at the Y(5S) energy, indicating that Bs0B_s^0 meson production proceeds predominantly through the creation of BsBˉsB^*_s \bar{B}^*_s pairs. The Bs0B_s^0 and BsB_s^* meson masses are measured to be M(B_s^0)=(5370 \pm 1 \pm 3)MeV/c^2 and M(B_s^*)=(5418 \pm 1 \pm 3)MeV/c^2. Upper limits on the B_s^0 -> \gamma \gamma, B_s^0 -> \phi \gamma, B_s^0 -> K^+ K^- and B_s^0 -> D_s^{(*)+} D_s^{(*)-} branching fractions are also reported.Comment: 9 pages, 5 figures, published in Phys.     Measurement of the ratio B(D0->pi+pi-pi0)/B(D0->K-pi+pi0) and the time-integrated CP asymmetry in D0->pi+pi-pi0

    We report a high-statistics measurement of the relative branching fraction B(D0->pi+pi-pi0)/B(D0->K-pi+pi0) using a 532 fb^{-1} data sample collected with the Belle detector at the KEKB asymmetric-energy e+e- collider. The measured value of the relative branching fraction is B(D0->pi+pi-pi0)/B(D0->K-pi+pi0) = (10.12 +/- 0.04(stat) +/- 0.18(syst))x10^{-2} which has an accuracy comparable to the world average. We also present a measurement of the time-integrated CP asymmetry in D0->pi+pi-pi0 decay. The result, A_{CP} = (0.43 +/- 1.30)%, shows no significant CP violation.Comment: 15 pages, 11 figures, submitted to Physics Letters

    Observation of B+ -> K+ eta gamma

    We report measurements of radiative B decays with K eta gamma final states, using a data sample of 253 /fb recorded at the Upsilon(4S) resonance with the Belle detector at the KEKB e+e- storage ring. We observe B+ -> K+ eta gamma for the first time with a branching fraction of (8.4 +- 1.5(stat) +1.2 -0.9(syst)) X 10^{-6} for M(Keta) K0 eta gamma.     Time-Dependent CP Violation Effects in Partially Reconstructed B0DπB^0 \to D^* \pi Decays

    We report measurements of time-dependent decay rates for B0Dπ±B^0 \to D^{*\mp} \pi^\pm decays and extraction of CP violation parameters related to ϕ3\phi_3. We use a partial reconstruction technique, whereby signal events are identified using information only from the primary pion and the charged pion from the decay of the DD^{*\mp}. The analysis uses 140fb1140 {\rm fb}^{-1} of data accumulated at the Υ(4S)\Upsilon(4S) resonance with the Belle detector at the KEKB asymmetric-energy e+ee^{+}e^{-} collider. We measure the CP violation parameters S+=0.035±0.041(stat)±0.018(syst)S^+ = 0.035 \pm 0.041 ({\rm stat}) \pm 0.018 ({\rm syst}) and S=0.025±0.041(stat)±0.018(syst)S^- = 0.025 \pm 0.041 ({\rm stat}) \pm 0.018 ({\rm syst}).Comment: 15 pages, 5 figures.     Updated precision measurement of the average lifetime of B hadrons

    The measurement of the average lifetime of B hadrons using inclusively reconstructed secondary vertices has been updated using both an improved processing of previous data and additional statistics from new data. This has reduced the statistical and systematic uncertainties and gives \tau_{\mathrm{B}} = 1.582 \pm 0.011\ \mathrm{(stat.)} \pm 0.027\ \mathrm{(syst.)}\ \mathrm{ps.} Combining this result with the previous result based on charged particle impact parameter distributions yields \tau_{\mathrm{B}} = 1.575 \pm 0.010\ \mathrm{(stat.)} \pm 0.026\ \mathrm{(syst.)}\ \mathrm{ps.
