    Validación y detección automática del transporte dispersivo del emisario submarino de Mar del Plata, Argentina

    The submarine outfall of Mar del Plata city at Camet was projected considering the mean and maximum of forecasted sewage discharges, the inner-shelf depth, coliform concentration and its decay (T90) mainly induced by sunlight effect and costal salinity. In 2016 the outfall was operating with a length of 3,810 m and diffusers in the last 526 m. An economical method to monitor its performance in relation to the surroundings, is remote-sensing techniques, applying either visible or radar images. Tidal currents parallel to the coast are responsible for the transport of the sedimentary plume in the far field, after a primary dilution from a depth of 11 m. Visible images (1.5 to 6 m spatial resolution) are effective in monitoring the plume entrained in the upper portion of the water column. These analyses led to study the interaction between waves and coastal currents. Radar images (30 m resolution X and C bands) permit to survey the slick-alike plume that differs from the environment water by the surface roughness. Comparing both techniques visible images can distinguish the different colours of the plume; instead, the radar images are showing the surface roughness from the slick-alike plume. The main advantage of active sensors is that they can map the plume during a cloudy weather and even during night time.El emisario submarino de Mar del Plata en Camet fue proyectado considerando las descargas cloacales promedio y máximas previstas, la profundidad de la plataforma vecina, la concentración de coliformes y el decaimiento (T90) inducido por la luz solar y la salinidad. En 2016 el emisario operaba con una longitud de 3.810 m con difusores en los últimos 526 m. Un método poco oneroso para analizar su comportamiento en relación a su entorno es la aplicación de técnicas de teledetección tanto en el espectro visible como mediante imágenes de radar. Las corrientes de marea paralelas a la costa son responsables de una pluma sedimentaria en el campo lejano, luego de una dilución primaria desde una profundidad de 11 m. Las imágenes visibles (resolución espacial de 1,5 a 6 m) son efectivas para monitorear la pluma extendida en la capa superior del mar. Estos análisis permiten el estudio de la interacción entre olas y corrientes costeras. Las imágenes de radar (resolución de 30 m en las bandas X y C) permiten relevar plumas superficiales semejantes a derrames de aceites por su rugosidad. Comparando ambas técnicas las imágenes visibles pueden distinguir plumas de diferentes colores del agua; por el contrario, las imágenes de radar están mostrando diferencias en la tensión superficial.     BRCA1 expression modulates chemosensitivity of BRCA1-defective HCC1937 human breast cancer cells

    Germline mutations of the tumour suppressor gene BRCA1 are involved in the predisposition and development of breast cancer and account for 20–45% of all hereditary cases. There is an increasing evidence that these tumours are characterised by a specific phenotype and pattern of gene expression. We have hypothesised that differences in chemosensitivity might parallel molecular heterogeneity of hereditary and sporadic breast tumours. To this end, we have investigated the chemosensitivity of the BRCA1-defective HCC1937 breast cancer cell line, and the BRCA1-competent MCF-7 (hormone-sensitive) and MDA-MB231 (hormone-insensitive) breast cancer cell lines using the MTT assay. The 50% inhibitory concentration (IC50) for the individual compounds were derived by interpolate plot analysis of the logarithmic scalar concentration curve after a 48 h exposure. HCC1937 cells were significantly (P<0.005) more sensitive to cisplatin (CDDP) (IC50 : 30–40 μM) compared with MCF-7 (IC50 : 60–70 μM) and MDA-MB231 (IC50 : 90–100 μM) cells. On the other hand, BRCA1-defective breast cancer cells were significantly less sensitive to doxorubicin (Dox) (IC50 : 45–50 μM) compared with MCF-7 (IC50 : 1–5 μM) and MDA-MB231 (IC50 : 5–10 μM) (P<0.02), as well as to paclitaxel (Tax) (IC50 : >2 μM for HCC1937, 0.1–0.2 μM for MCF-7 and 0.01–0.02 μM for MDA-MB231) (P<0.001). Full-length BRCA1 cDNA transfection of BRCA1-defective HCC1937 cells led to the reconstituted expression of BRCA1 protein in HCC1937/WTBRCA1-derived cell clone, but did not reduce tumour cell growth in soft agar. BRCA1 reconstitution reverted the hypersensitivity to CDDP (P<0.02), and restored the sensitivity to Dox (P<0.05) and Tax (P<0.001), compared with parental HCC1937 cells. Taken together, our findings suggest a specific chemosensitivity profile of BRCA1-defective cells in vitro, which is dependent on BRCA1 protein expression, and suggest prospective preclinical and clinical investigation for the development of tailored therapeutical approaches in this setting

    Transmembrane TNF-α: structure, function and interaction with anti-TNF agents

    Transmembrane TNF-α, a precursor of the soluble form of TNF-α, is expressed on activated macrophages and lymphocytes as well as other cell types. After processing by TNF-α-converting enzyme (TACE), the soluble form of TNF-α is cleaved from transmembrane TNF-α and mediates its biological activities through binding to Types 1 and 2 TNF receptors (TNF-R1 and -R2) of remote tissues. Accumulating evidence suggests that not only soluble TNF-α, but also transmembrane TNF-α is involved in the inflammatory response. Transmembrane TNF-α acts as a bipolar molecule that transmits signals both as a ligand and as a receptor in a cell-to-cell contact fashion. Transmembrane TNF-α on TNF-α-producing cells binds to TNF-R1 and -R2, and transmits signals to the target cells as a ligand, whereas transmembrane TNF-α also acts as a receptor that transmits outside-to-inside (reverse) signals back to the cells after binding to its native receptors. Anti-TNF agents infliximab, adalimumab and etanercept bind to and neutralize soluble TNF-α, but exert different effects on transmembrane TNF-α-expressing cells (TNF-α-producing cells). In the clinical settings, these three anti-TNF agents are equally effective for RA, but etanercept is not effective for granulomatous diseases. Moreover, infliximab induces granulomatous infections more frequently than etanercept. Considering the important role of transmembrane TNF-α in granulomatous inflammation, reviewing the biology of transmembrane TNF-α and its interaction with anti-TNF agents will contribute to understanding the bases of differential clinical efficacy of these promising treatment modalities

    Natural History and Outcome of Hepatic Vascular Malformations in a Large Cohort of Patients with Hereditary Hemorrhagic Teleangiectasia

    BACKGROUND: Hereditary hemorrhagic telangiectasia is a genetic disease characterized by teleangiectasias involving virtually every organ. There are limited data in the literature regarding the natural history of liver vascular malformations in hemorrhagic telangiectasia and their associated morbidity and mortality. AIM: This prospective cohort study sought to assess the outcome of liver involvement in hereditary hemorrhagic telangiectasia patients. METHODS: We analyzed 16 years of surveillance data from a tertiary hereditary hemorrhagic telangiectasia referral center in Italy. We considered for inclusion in this study 502 consecutive Italian patients at risk of hereditary hemorrhagic telangiectasia who presented at the hereditary hemorrhagic telangiectasia referral center and underwent a multidisciplinary screening protocol for the diagnosis of hereditary hemorrhagic telangiectasia. Of the 502 individuals assessed in the center, 154 had hepatic vascular malformations and were the subject of the study; 198 patients with hereditary hemorrhagic telangiectasia and without hepatic vascular malformations were the controls. Additionally, we report the response to treatment of patients with complicated hepatic vascular malformations. RESULTS: The 154 patients were included and followed for a median period of 44 months (range 12-181); of these, eight (5.2%) died from VM-related complications and 39 (25.3%) experienced complications. The average incidence rates of death and complications were 1.1 and 3.6 per 100 person-years, respectively. The median overall survival and event-free survival after diagnosis were 175 and 90 months, respectively. The rate of complete response to therapy was 63%. CONCLUSIONS: This study shows that substantial morbidity and mortality are associated with liver vascular malformations in hereditary hemorrhagic telangiectasia patients
