    The RMS survey: Ammonia mapping of the environment of young massive stellar objects - II

    We present the results from NH3 mapping observations towards 34 regions identified by the Red MSX Source (RMS) survey. We have used the Australia Telescope Compact Array to map ammonia (1, 1) and (2, 2) inversion emission spectra at a resolution of 10 arcsec with velocity channel resolution of 0.4 km s−1 towards the positions of embedded massive star formation. Complementary data have been used from the ATLASGAL and GLIMPSE Legacy Surveys in order to improve the understanding of the regions and to estimate physical parameters for the environments. The fields have typical masses of ∼1000 M⊙, radii of ∼0.15 pc, and distances of ∼3.5 kpc. Luminosities range between ∼103 and ∼106 L⊙ and kinetic temperatures between 10 and 40 K. We classify each field into one of two subsets in order to construct an evolutionary system for massive star formation in these regions based on the morphology and relative positions of the NH3 emission, RMS sources, and ATLASGAL thermal dust emission. Differences in morphology between NH3 emission and ATLASGAL clumps are shown to correspond to evolutionary stages of ongoing massive star formation in these regions. The study has been further refined by including the positions of known methanol and water masers in the regions to gain insight into possible protostellar regions and triggered star formation

    ATLASGAL - physical parameters of dust clumps associated with 6.7 GHz methanol masers

    We have constructed the largest sample of dust-associated class II 6.7 GHz methanol masers yet obtained. New measurements from the Methanol Multibeam (MMB) survey were combined with the 870 μm APEX Telescope Large Area Survey of the Galaxy (ATLASGAL) and the 850 μm JCMT Plane Survey (JPS). Together with two previous studies we have now identified the host clumps for 958 methanol masers across the Galactic Plane, covering approximately 99 per cent of the MMB catalogue and increasing the known sample of dust-associated masers by over 30 per cent. We investigate correlations between the physical properties of the clumps and masers using distances and luminosities drawn from the literature. Clumps hosting methanol masers are significantly more compact and have higher volume densities than the general population of clumps. We determine a minimum volume density threshold of n(H2) ≥ 104 cm−3 for the efficient formation of intermediate- and high-mass stars. We find 6.7 GHz methanol masers are associated with a distinct part of the evolutionary process (Lbol/Mfwhm ratios of between 100.6 and 102.2) and have well-defined turning on and termination points. We estimate the lower limit for the mass of embedded objects to be ≥6 M⊙ and the statistical lifetime of the methanol maser stage to be ∼3.3 × 104 yr. This suggests that methanol masers are indeed reliable tracers of high-mass star formation, and indicates that the evolutionary period traced by this marker is relatively rapid

    Inhibiting mevalonate pathway enzymes increases stromal cell resilience to a cholesterol-dependent cytolysin

    Animal health depends on the ability of immune cells to kill invading pathogens, and on the resilience of tissues to tolerate the presence of pathogens. Trueperella pyogenes causes tissue pathology in many mammals by secreting a cholesterol-dependent cytolysin, pyolysin (PLO), which targets stromal cells. Cellular cholesterol is derived from squalene, which is synthesized via the mevalonate pathway enzymes, including HMGCR, FDPS and FDFT1. The present study tested the hypothesis that inhibiting enzymes in the mevalonate pathway to reduce cellular cholesterol increases the resilience of stromal cells to PLO. We first verified that depleting cellular cholesterol with methyl-β-cyclodextrin increased the resilience of stromal cells to PLO. We then used siRNA to deplete mevalonate pathway enzyme gene expression, and used pharmaceutical inhibitors, atorvastatin, alendronate or zaragozic acid to inhibit the activity of HMGCR, FDPS and FDFT1, respectively. These approaches successfully reduced cellular cholesterol abundance, but mevalonate pathway enzymes did not affect cellular resilience equally. Inhibiting FDFT1 was most effective, with zaragozic acid reducing the impact of PLO on cell viability. The present study provides evidence that inhibiting FDFT1 increases stromal cell resilience to a cholesterol-dependent cytolysin

    Identification of novel clostridium perfringens type E strains that carry an iota toxin plasmid with a functional enterotoxin gene

    Clostridium perfringens enterotoxin (CPE) is a major virulence factor for human gastrointestinal diseases, such as food poisoning and antibiotic associated diarrhea. The CPE-encoding gene (cpe) can be chromosomal or plasmid-borne. Recent development of conventional PCR cpe-genotyping assays makes it possible to identify cpe location (chromosomal or plasmid) in type A isolates. Initial studies for developing cpe genotyping assays indicated that all cpe-positive strains isolated from sickened patients were typable by cpe-genotypes, but surveys of C. perfringens environmental strains or strains from feces of healthy people suggested that this assay might not be useful for some cpe-carrying type A isolates. In the current study, a pulsed-field gel electrophoresis Southern blot assay showed that four cpe-genotype untypable isolates carried their cpe gene on a plasmid of ~65 kb. Complete sequence analysis of the ~65 kb variant cpe-carrying plasmid revealed no intact IS elements and a disrupted cytosine methyltransferase (dcm) gene. More importantly, this plasmid contains a conjugative transfer region, a variant cpe gene and variant iota toxin genes. The toxin genes encoded by this plasmid are expressed based upon the results of RT-PCR assays. The ~65 kb plasmid is closely related to the pCPF4969 cpe plasmid of type A isolates. MLST analyses indicated these isolates belong to a unique cluster of C. perfringens. Overall, these isolates carrying a variant functional cpe gene and iota toxin genes represent unique type E strains. © 2011 Miyamoto et al

    Arcanobacterium haemolyticum has an established role in the etiology of human pharyngitis. There are increasing reports of systemic infections caused by this organism. From India, we report the first case of Arcanobacterium haemolyticum causing pyothorax in an immunocompetent adolescent male patient. The probable mode of infection is also discussed. The role of A. hemolyticum as an animal pathogen needs further study

    Ovine pedomics : the first study of the ovine foot 16S rRNA-based microbiome

    We report the first study of the bacterial microbiome of ovine interdigital skin based on 16S rRNA by pyrosequencing and conventional cloning with Sanger-sequencing. Three flocks were selected, one a flock with no signs of footrot or interdigital dermatitis, a second flock with interdigital dermatitis alone and a third flock with both interdigital dermatitis and footrot. The sheep were classified as having either healthy interdigital skin (H), interdigital dermatitis (ID) or virulent footrot (VFR). The ovine interdigital skin bacterial community varied significantly by flock and clinical condition. The diversity and richness of operational taxonomic units was greater in tissue from sheep with ID than H or VFR affected sheep. Actinobacteria, Bacteriodetes, Firmicutes and Proteobacteria were the most abundant phyla comprising 25 genera. Peptostreptococcus, Corynebacterium and Staphylococcus were associated with H, ID and VFR respectively. Sequences of Dichelobacter nodosus, the causal agent of ovine footrot, were not amplified due to mismatches in the 16S rRNA universal forward primer (27F). A specific real time PCR assay was used to demonstrate the presence of D. nodosus which was detected in all samples including the flock with no signs of ID or VFR. Sheep with ID had significantly higher numbers of D. nodosus (104-109 cells/g tissue) than those with H or VFR feet

    OH maser emission in the THOR survey of the northern Milky Way*

    Context. OH masers trace diverse physical processes, from the expanding envelopes around evolved stars to star-forming regions or supernovae remnants. Providing a survey of the ground-state OH maser transitions in the northern hemisphere inner Milky Way facilitates the study of a broad range of scientific topics. Aims. We want to identify the ground-state OH masers at ∼18 cm wavelength in the area covered by The HI/OH/Recombination line survey of the Milky Way (THOR). We will present a catalogue of all OH maser features and their possible associated environments. Methods. The THOR survey covers longitude and latitude ranges of 14. ◦3 < l < 66. ◦8 and b < ± 1.◦25. All OH ground state lines 2Π3/2 (J = 3/2) at 1612 (F = 1−2), 1665 (F = 1−1), 1667 (F = 2−2) and 1720 MHz (F = 2−1) have been observed, employing the Very Large Array (VLA) in its C configuration. The spatial resolution of the data varies between 12.5" and 19", the spectral resolution is 1.5 km s−1, and the rms sensitivity of the data is ∼10 mJy beam−1 per channel. Results. We identify 1585 individual maser spots (corresponding to single spectral features) distributed over 807 maser sites (regions of size ∼103−104 AU). Based on different criteria from spectral profiles to literature comparison, we try to associate the maser sites with astrophysical source types. Approximately 51% of the sites exhibit the double-horned 1612 MHz spectra typically emitted from the expanding shells of evolved stars. The separations of the two main velocity features of the expanding shells typically vary between 22 and 38 km s−1. In addition to this, at least 20% of the maser sites are associated with star-forming regions. While the largest fraction of 1720 MHz maser spots (21 out of 53) is associated with supernova remnants, a significant fraction of the 1720 MHz maser spots (17) are also associated with star-forming regions. We present comparisons to the thermal 13CO(1–0) emission as well as to other surveys of class II CH3OH and H2O maser emission. The catalogue attempts to present associations to astrophysical sources where available, and the full catalogue is available in electronic form. Conclusions. This OH maser catalogue presents a unique resource of stellar and interstellar masers in the northern hemisphere. It provides the basis for a diverse range of follow-up studies from envelopes around evolved stars to star-forming regions and Supernova remnants

    Skewed genomic variability in strains of the toxigenic bacterial pathogen, Clostridium perfringens

    Clostridium perfringens is a Gram-positive, anaerobic spore-forming bacterium commonly found in soil, sediments, and the human gastrointestinal tract. C. perfringens is responsible for a wide spectrum of disease, including food poisoning, gas gangrene (clostridial myonecrosis), enteritis necroticans, and non-foodborne gastrointestinal infections. The complete genome sequences of Clostridium perfringens strain ATCC 13124, a gas gangrene isolate and the species type strain, and the enterotoxin-producing food poisoning strain SM101, were determined and compared with the published C. perfringens strain 13 genome. Comparison of the three genomes revealed considerable genomic diversity with >300 unique "genomic islands" identified, with the majority of these islands unusually clustered on one replichore. PCR-based analysis indicated that the large genomic islands are widely variable across a large collection of C. perfringens strains. These islands encode genes that correlate to differences in virulence and phenotypic characteristics of these strains. Significant differences between the strains include numerous novel mobile elements and genes encoding metabolic capabilities, strain-specific extracellular polysaccharide capsule, sporulation factors, toxins, and other secreted enzymes, providing substantial insight into this medically important bacterial pathogen. ©2006 by Cold Spring Harbor Laboratory Press

    Foreign direct investment and institutional reform : evidence and an application to Portugal

    We examine the role of geographic, economic, and institutional factors in attracting Foreign Direct Investment (FDI) in Europe, using a cross-section of inward bilateral investments. We estimate and assess the expected benefits, the required reform efforts, and the efficiency of reform options corresponding to a con- vergence of Portuguese institutions to EU standards. We conclude that improving home institutions is likely to have a quantitatively very significant role in attracting FDI. Geographical and market size factors also play a role. Reforms promoting the independence of financial institutions and a leaner bureaucracy, lowering political risk and corruption, and improving the investment code may significantly affect the amount of bilateral inward FDI that is targeted to Portugal.info:eu-repo/semantics/publishedVersio
