    OcorrĂȘncia de Leptospira spp. e fatores associados Ă  infecção em equinos de contingente militar no estado do Rio Grande do Sul, Brasil

    The seroprevalence of Leptospira spp. and infection risk factors were investigated in horses from a military contingent in the Rio Grande do Sul State, Brazil. A total of 446 horses were evaluated and categorized into 309 mares, 11 stallions, and 126 horses with an average age of three years. To determine seroprevalence, serum samples from all horses were submitted to the microscopic agglutination test against 12 serovars belonging to nine serogroups, usually circulating in equine populations. To investigate the possible risk factors of infection by Leptospira spp., questionnaires were applied in that military unit. The seroprevalence in the horses with an average age of 3 years, mares, and stallions were 57.94% (73/126), 54.05% (165/309), and 45.45% (5/11), respectively. The main risk factors identified were contact with different water sources, the presence of rodents, and contact with wild and domestic animals. The high seroprevalence of anti‑Leptospira spp. antibodies may be associated with the presence of these risk factors. Therefore, the exposure of horses to possible reservoirs of Leptospira spp. should be minimized. Also, the immunoprophylaxis protocol should be reviewed and a shorter interval between vaccinations adopted to control leptospirosis in this herd. A soroprevalĂȘncia de Leptospira spp. e os fatores de risco para infecção foram investigados em cavalos de um contingente militar no Rio Grande do Sul, Brasil. Para determinação da soroprevalĂȘncia, amostras de soro de todos os equinos foram submetidas ao teste de aglutinação microscĂłpica contra 12 sorovares pertencentes a nove sorogrupos, normalmente circulantes em populaçÔes equinas. Para investigar os possĂ­veis fatores de risco da infecção por Leptospira spp., foi aplicado um questionĂĄrio naquela unidade militar. A soroprevalĂȘncia nos cavalos com idade mĂ©dia de trĂȘs anos, Ă©guas e garanhĂ”es foi de 57,94% (73/126), 54,05% (165/309) e 45,45% (5/11), respectivamente. Os principais fatores de risco identificados foram o contato com diferentes fontes de ĂĄgua, a presença de roedores e o contato com animais silvestres e domĂ©sticos. A alta soroprevalĂȘncia de anticorpos anti-Leptospira spp. pode estar associada Ă  presença desses fatores de risco. Portanto, a exposição destes equinos aos possĂ­veis reservatĂłrios de Leptospira spp. deve ser minimizada.     Inert Coats of Magnetic Nanoparticles Prevent Formation of Occlusive Intravascular Co-aggregates With Neutrophil Extracellular Traps

    If foreign particles enter the human body, the immune system offers several mechanisms of response. Neutrophils forming the first line of the immune defense either remove pathogens by phagocytosis, inactivate them by degranulation or release of reactive oxygen species or immobilize them by the release of chromatin decorated with the granular proteins from cytoplasm as neutrophil extracellular traps (NETs). Besides viable microbes like fungi, bacteria or viruses, also several sterile inorganic particles including nanoparticles reportedly activate NET formation. The physicochemical nanoparticle characteristics fostering NET formation are still elusive. Here we show that agglomerations of non-stabilized superparamagnetic iron oxide nanoparticles (SPIONs) induce NET formation by isolated human neutrophils, in whole blood experiments under static and dynamic conditions as well as in vivo. Stabilization of nanoparticles with biocompatible layers of either human serum albumin or dextran reduced agglomeration and NET formation by neutrophils. Importantly, this passivation of the SPIONs prevented vascular occlusions in vivo even when magnetically accumulated. We conclude that higher order structures formed during nanoparticle agglomeration primarily trigger NET formation and the formation of SPION-aggregated NET-co-aggregates, whereas colloid-disperse nanoparticles behave inert and are alternatively cleared by phagocytosis

    TBX2 is a neuroblastoma core regulatory circuitry component enhancing MYCN/FOXM1 reactivation of DREAM targets

    Chromosome 17q gains are almost invariably present in high-risk neuroblastoma cases. Here, we perform an integrative epigenomics search for dosage-sensitive transcription factors on 17q marked by H3K27ac defined super-enhancers and identify TBX2 as top candidate gene. We show that TBX2 is a constituent of the recently established core regulatory circuitry in neuroblastoma with features of a cell identity transcription factor, driving proliferation through activation of p21-DREAM repressed FOXM1 target genes. Combined MYCN/TBX2 knockdown enforces cell growth arrest suggesting that TBX2 enhances MYCN sustained activation of FOXM1 targets. Targeting transcriptional addiction by combined CDK7 and BET bromodomain inhibition shows synergistic effects on cell viability with strong repressive effects on CRC gene expression and p53 pathway response as well as several genes implicated in transcriptional regulation. In conclusion, we provide insight into the role of the TBX2 CRC gene in transcriptional dependency of neuroblastoma cells warranting clinical trials using BET and CDK7 inhibitors

    Joint effects of climate, tree size, and year on annual tree growth derived from tree-ring records of ten globally distributed forests

    Tree rings provide an invaluable long-term record for understanding how climate and other drivers shape tree growth and forest productivity. However, conventional tree-ring analysis methods were not designed to simultaneously test effects of climate, tree size, and other drivers on individual growth. This has limited the potential to test ecologically relevant hypotheses on tree growth sensitivity to environmental drivers and their interactions with tree size. Here, we develop and apply a new method to simultaneously model nonlinear effects of primary climate drivers, reconstructed tree diameter at breast height (DBH), and calendar year in generalized least squares models that account for the temporal autocorrelation inherent to each individual tree\u27s growth. We analyze data from 3811 trees representing 40 species at 10 globally distributed sites, showing that precipitation, temperature, DBH, and calendar year have additively, and often interactively, influenced annual growth over the past 120 years. Growth responses were predominantly positive to precipitation (usually over ≄3-month seasonal windows) and negative to temperature (usually maximum temperature, over ≀3-month seasonal windows), with concave-down responses in 63% of relationships. Climate sensitivity commonly varied with DBH (45% of cases tested), with larger trees usually more sensitive. Trends in ring width at small DBH were linked to the light environment under which trees established, but basal area or biomass increments consistently reached maxima at intermediate DBH. Accounting for climate and DBH, growth rate declined over time for 92% of species in secondary or disturbed stands, whereas growth trends were mixed in older forests. These trends were largely attributable to stand dynamics as cohorts and stands age, which remain challenging to disentangle from global change drivers. By providing a parsimonious approach for characterizing multiple interacting drivers of tree growth, our method reveals a more complete picture of the factors influencing growth than has previously been possible

    Reproducibility of quantitative F-18-3'-deoxy-3'-fluorothymidine measurements using positron emission tomography

    Positron emission tomography (PET) using F-18-3'-deoxy-3'-fluorothymidine ([F-18]FLT) allows noninvasive monitoring of tumour proliferation. For serial imaging in individual patients, good reproducibility is essential. The purpose of the present study was to evaluate the reproducibility of quantitative [F-18]FLT measurements. Nine patients with non-small-cell lung cancer (NSCLC) and six with head-and-neck cancer (HNC) underwent [F-18]FLT PET twice within 7 days prior to therapy. The maximum pixel value (SUVmax) and a threshold defined volume (SUV41%) were defined for all delineated lesions. The plasma to tumour transfer constant (K-i) was estimated using both Patlak graphical analysis and nonlinear regression (NLR). NLR was also used to estimate k(3), which, at least in theory, selectively reflects thymidine kinase 1 activity. The level of agreement between test and retest values was assessed using the intraclass correlation coefficient (ICC) and Bland-Altman analysis. All primary tumours and > 90% of clinically suspected locoregional metastases could be delineated. In total, 24 lesions were defined. NLR-derived K-i, Patlak-derived K-i, SUV41% and SUVmax showed excellent reproducibility with ICCs of 0.92, 0.95, 0.98 and 0.93, and SDs of 16%, 12%, 7% and 11%, respectively. Reproducibility was poor for k(3) with an ICC of 0.43 and SD of 38%. Quantitative [F-18]FLT measurements are reproducible in both NSCLC and HNC patients. When monitoring response in individual patients, changes of more than 15% in SUV41%, 20-25% in SUVmax and Patlak-derived K-i, and 32% in NLR3k-derived K-i are likely to represent treatment effect

    Alcohol Consumption, Genetic Variants in Alcohol Deydrogenases, and Risk of Cardiovascular Diseases: A Prospective Study and Meta-Analysis

    OBJECTIVE: First, to investigate and compare associations between alcohol consumption and variants in alcohol dehydrogenase (ADH) genes with incidence of cardiovascular diseases (CVD) in a large German cohort. Second, to quantitatively summarize available evidence of prospective studies on polymorphisms in ADH1B and ADH1C and CVD-risk. METHODS: We conducted a case-cohort study nested within the European Prospective Investigation into Cancer and Nutrition (EPIC)-Potsdam cohort including a randomly drawn subcohort (n = 2175) and incident cases of myocardial infarction (MI; n = 230) or stroke (n = 208). Mean follow-up time was 8.2±2.2 years. The association between alcohol consumption, ADH1B or ADH1C genotypes, and CVD-risk was assessed using Cox proportional hazards regression. Additionally, we report results on associations of variants in ADH1B and ADH1C with ischemic heart disease and stroke in the context of a meta-analysis of previously published prospective studies published up to November 2011. RESULTS: Compared to individuals who drank >0 to 6 g alcohol/d, we observed a reduced risk of MI among females consuming >12 g alcohol/d (HR = 0.31; 95% CI: 0.10-0.97) and among males consuming >24 to 60 g/d (HR = 0.57; 95% CI: 0.33-0.98) or >60 g alcohol/d (HR = 0.30; 95% CI: 0.12-0.78). Stroke risk was not significantly related to alcohol consumption >6 g/d, but we observed an increased risk of stroke in men reporting no alcohol consumption. Individuals with the slow-coding ADH1B*1/1 genotype reported higher median alcohol consumption. Yet, polymorphisms in ADH1B or ADH1C were not significantly associated with risk of CVD in our data and after pooling results of eligible prospective studies [ADH1B*1/1: RR = 1.35 (95% CI: 0.98-1.88; p for heterogeneity: 0.364); ADH1C*2/2: RR = 1.07 (95% CI: 0.90-1.27; p for heterogeneity: 0.098)]. CONCLUSION: The well described association between alcohol consumption and CVD-risk is not reflected by ADH polymorphisms, which modify the rate of ethanol oxidation

    If there is a theme running through the Message Covid-19 special issue, it is one of caring. Of our own and others’ resilience and wellbeing, of friendship and community, of students, practitioners and their futures, of social justice, equality and of doing the right thing. The veins of designing with care run through the edition, wide and deep. It captures, not designers as heroes, but those with humble views, exposing the need to understand a diversity of perspectives when trying to comprehend the complexity that Covid-19 continues to generate. As graphic designers, illustrators and visual communicators, contributors have created, documented, written, visualised, reflected, shared, connected and co-created, designed for good causes and re-defined what it is to be a student, an academic and a designer during the pandemic. This poignant period in time has driven us, through isolation, towards new rules of living, and new ways of working; to see and map the world in a different light. A light that is uncertain, disjointed, and constantly being redefined. This Message issue captures responses from the graphic communication design community in their raw state, to allow contributors to communicate their experiences through both their written and visual voice. Thus, the reader can discern as much from the words as the design and visualisations. Through this issue a substantial number of contributions have focused on personal reflection, isolation, fear, anxiety and wellbeing, as well as reaching out to community, making connections and collaborating. This was not surprising in a world in which connection with others has often been remote, and where ‘normal’ social structures of support and care have been broken down. We also gain insight into those who are using graphic communication design to inspire and capture new ways of teaching and learning, developing themselves as designers, educators, and activists, responding to social justice and to do good; gaining greater insight into society, government actions and conspiracy.     Autecological and molecular research for the characterization of the webbing clothes moth Tineola bisselliella (Lepidoptera) as a reference organism

    Die Kleidermotte Tineola bisselliella, ursprĂŒnglich vermutlich aus Afrika stammend (Zeller, 1852), ist als weltweit verbreiteter MaterialschĂ€dling in Wohnungen, Lagern und Museen bekannt (Key & Common, 1959; Querner, 2014). Um in Form des Integrated Pest Management (IPM) Methoden zur Erkennung, PrĂ€vention und BekĂ€mpfung von Vorrats- und MaterialschĂ€dlingen zu organisieren und zu entwickeln, ist eine genaue Kenntnis der Biologie, Ökologie und des Verhaltens des SchĂ€dlings entscheidend. In dieser Arbeit wird aufbauend auf den Studien zur Entwicklung von T. bisselliella unter verschiedenen Temperaturen und Luftfeuchten von Titschak (1925) das Falterschlupfverhalten quantifiziert und mit dem unter der Konkurrenzsituation mit dem aus der gleichen Nahrungsgilde stammenden PolsterwarenkĂ€fers (Anthrenus flavipes) verglichen. Die KonkurrenzfĂ€higkeit der Kleidermotte ĂŒber das gesamte betrachtete Spektrum an Temperatur und Luftfeuchtigkeiten machen sie als MaterialschĂ€dling so erfolgreich. DarĂŒber hinaus wird das Vorkommen in natĂŒrlichen Reservoiren durch die Untersuchung von FreilandfĂ€ngen an verschiedenen Standorten in Berlin und Umgebung als unwahrscheinlich angesehen. Molekularbiologische Betrachtungen des Genflusses anhand von mitochondrialen Sequenzabschnitten sowie BOX-PCR Mustern zeigen zwar keine Fragmentierung der Populationen in die einzelnen Fangstandorte, jedoch kann auch ein Netzwerk aus passiver Verbreitung durch den Menschen fĂŒr einen kontinuierlichen Genfluss sorgen.The webbing clothes moth Tineola bisselliela, assumed to original derive from Afrika (Zeller, 1852), is a cosmopolitan pest in households, warehouses and on culture heritage (Key & Common, 1959; Querner, 2014). To organize and develop strategies for the identification, prevention and control of stored product and culture heritage pest organism in the manner of Integrated Pest Management (IPM), a precise knowledge of the biology, ecology and physiology of the pest organism is needed. Based on the observations by Titschak (1925) on the development of T. bisselliella under different temperatures and relative humidities, this work shows the quantification of moth eclosion under three conditions of temperature and relative humidity compared in control and interspecific competition with the furniture carpet beetle (Anthrenus flavipes) from the same feeding guild. The overall competition of the webbing clothes moth demonstrates her success as a culture heritage pest organism. Monitoring T. bisselliella at differnt outdoor stations in the broader area of Berlin leads to the absence of an occurrence in natural habitats. Molecular analysis of mitochondrial sequences as well as BOX-PCR patters demonstrate no population fragmentation into the trapping locations.
