    In an effort to strengthen philanthropic investments among its membership, the Youth Transition Funders Group (YTFG) asked a group of policy experts to provide recommendations on how foundations can work to encourage effective policy solutions on issues affecting youth in transition to adulthood. The primary challenge was to think beyond the systemic silos that so deeply shape the services and expectations of youth and move towards an overall framework that could produce improved outcomes. YTFG's work is based on the Connected by 25 framework, in which all youth reach the following outcomes by age 25:Educational achievement in preparation for career and community participation, including a high school diploma, post-secondary degree, and/or vocational certificate trainingGainful employment and/or access to career training to achieve life-long economic successConnections to a positive support system -- namely, guidance from family members and caring adults, as well as access to health, counseling, and mental health servicesThe ability to be a responsible and nurturing parentThe capacity to be actively engaged in the civic life of one's communityThis issue brief offers a summary of those recommendations, focusing on four primary transition points that often threaten the ability for youth to be connected by age 25 to the institutions and support systems that help them succeed throughout life

    Solid Footing: Reinforcing the Early Care and Education Economy for Infants and Toddlers in DC

    The District of Columbia (DC) is a vibrant, diverse, financially stable city that has become one of the most expensive places to live in the nation. It also ranks among the five major U.S. cities with the greatest income inequality. Because of this economic divide, the District struggles to create equity among its population, particularly in education where the achievement gap between poor and wealthy stubbornly persists. Research has consistently shown that this achievement gap begins not in kindergarten, but in the cradle, with the differences between the early learning environments of children who live in low-income and upper-income households producing cognitive differences before a child even reaches the public school system. Access to high-quality early learning environments can reduce or even eliminate that gap, which is why District policymakers have invested heavily in quality universal preschool and Pre-Kindergarten. But children from low-income households can already be cognitively behind by preschool, so the District must also invest in the early education needs of its infants and toddlers.This report attempts to quantify and qualify what investment need to be made. Until now, no one has assessed how much it costs early care and education (ECE) providers to meet the level of quality that the District requires, or how providers are able to maintain quality while serving families who depend on child care subsidy payments from the government. DC Appleseed and the DC Fiscal Policy Institute have collaborated to produce a study to better understand these realities. The work grew from concern that the District's payment rates to ECE providers for the child care subsidy program are not keeping up with the costs, even though the children receiving subsidized services and the nearly 200 providers who serve them are among the District's most vulnerable and precious resources. The underpaid workforce that cares for and educates infants and toddlers is essentially subsidizing the system through low wages

    Unilateral snow banking in tuberculosis-related intermediate uveitis

    Snow banking is usually a term coined to describe the accumulation of vitreous exudates over the pars plana and the peripheral retina in pars planitis. Snow banking is very rare in tubercular intermediate uveitis. A 32-year-old male was diagnosed to have intermediate uveitis due to tubercular etiology in the right eye. Laboratory investigations include an increased erythrocyte sedimentation rate, positive Mantoux test, and computed tomography thorax showing mediastinal lymphadenopathy. Transbronchial needle aspiration of the lymph nodes showed chronic granulomatous inflammation with caseation. There were no recurrences following antitubercular therapy (ATT). This case report highlights the unique finding of snow banking in tubercular uveitis and course following treatment with ATT

    Screening of GPCR drugs for repurposing in breast cancer

    Drug repurposing can overcome both substantial costs and the lengthy process of new drug discovery and development in cancer treatment. Some Food and Drug Administration (FDA)-approved drugs have been found to have the potential to be repurposed as anti-cancer drugs. However, the progress is slow due to only a handful of strategies employed to identify drugs with repurposing potential. In this study, we evaluated GPCR-targeting drugs by high throughput screening (HTS) for their repurposing potential in triple-negative breast cancer (TNBC) and drug-resistant human epidermal growth factor receptor-2-positive (HER2+) breast cancer (BC), due to the dire need to discover novel targets and drugs in these subtypes. We assessed the efficacy and potency of drugs/compounds targeting different GPCRs for the growth rate inhibition in the following models: two TNBC cell lines (MDA-MB-231 and MDA-MB-468) and two HER2+ BC cell lines (BT474 and SKBR3), sensitive or resistant to lapatinib + trastuzumab, an effective combination of HER2-targeting therapies. We identified six drugs/compounds as potential hits, of which 4 were FDA-approved drugs. We focused on β-adrenergic receptor-targeting nebivolol as a candidate, primarily because of the potential role of these receptors in BC and its excellent long-term safety profile. The effects of nebivolol were validated in an independent assay in all the cell line models. The effects of nebivolol were independent of its activation of β3 receptors and nitric oxide production. Nebivolol reduced invasion and migration potentials which also suggests its inhibitory role in metastasis. Analysis of the Surveillance, Epidemiology and End Results (SEER)-Medicare dataset found numerically but not statistically significant reduced risk of all-cause mortality in the nebivolol group. In-depth future analyses, including detailed in vivo studies and real-world data analysis with more patients, are needed to further investigate the potential of nebivolol as a repurposed therapy for BC

    MUSiC : a model-unspecific search for new physics in proton-proton collisions at root s=13TeV

    Results of the Model Unspecific Search in CMS (MUSiC), using proton-proton collision data recorded at the LHC at a centre-of-mass energy of 13 TeV, corresponding to an integrated luminosity of 35.9 fb(-1), are presented. The MUSiC analysis searches for anomalies that could be signatures of physics beyond the standard model. The analysis is based on the comparison of observed data with the standard model prediction, as determined from simulation, in several hundred final states and multiple kinematic distributions. Events containing at least one electron or muon are classified based on their final state topology, and an automated search algorithm surveys the observed data for deviations from the prediction. The sensitivity of the search is validated using multiple methods. No significant deviations from the predictions have been observed. For a wide range of final state topologies, agreement is found between the data and the standard model simulation. This analysis complements dedicated search analyses by significantly expanding the range of final states covered using a model independent approach with the largest data set to date to probe phase space regions beyond the reach of previous general searches.Peer reviewe
