    From 4D medical images (CT, MRI, and Ultrasound) to 4D structured mesh models of the left ventricular endocardium for patient-specific simulations

    With cardiovascular disease (CVD) remaining the primary cause of death worldwide, early detection of CVDs becomes essential. The intracardiac flow is an important component of ventricular function, motion kinetics, wash-out of ventricular chambers, and ventricular energetics. Coupling between Computational Fluid Dynamics (CFD) simulations and medical images can play a fundamental role in terms of patient-specific diagnostic tools. From a technical perspective, CFD simulations with moving boundaries could easily lead to negative volumes errors and the sudden failure of the simulation. The generation of high-quality 4D meshes (3D in space + time) with 1-to-l vertex becomes essential to perform a CFD simulation with moving boundaries. In this context, we developed a semiautomatic morphing tool able to create 4D high-quality structured meshes starting from a segmented 4D dataset. To prove the versatility and efficiency, the method was tested on three different 4D datasets (Ultrasound, MRI, and CT) by evaluating the quality and accuracy of the resulting 4D meshes. Furthermore, an estimation of some physiological quantities is accomplished for the 4D CT reconstruction. Future research will aim at extending the region of interest, further automation of the meshing algorithm, and generating structured hexahedral mesh models both for the blood and myocardial volume

    Tricuspid annuloplasty concomitant with mitral valve surgery: Effects on right ventricular remodeling

    ObjectivesTricuspid valve annuloplasty (TVP) has been advocated concomitantly with left-sided cardiac surgery in case of more than moderate tricuspid regurgitation (TR) or tricuspid annular dilation (TAD) (diameter >40 mm or 21 mm/m²) even in the absence of significant TR. Data on postoperative right ventricular (RV) remodeling are lacking in such patients.MethodsPreoperative and postoperative echocardiography data from 45 consecutive TVP procedures, performed in mitral valve surgery in a single tertiary center, were retrospectively analyzed and compared with a propensity-matched control group of 33 procedures without concomitant TVP. RV function and geometry was analyzed by measuring RV size, fractional area change, and end-diastolic sphericity index (RVSI = long-axis length/short-axis width) and compared at baseline versus follow-up.ResultsAt a mean follow-up of 5 months, a favorable change in RV geometry was observed in TVP patients (RVSI increased from 1.99 ± 0.33 to 2.21 ± 0.42; P = .001), whereas the opposite was observed in the control group (RVSI decreased from 2.34 ± 0.52 to 2.17 ± 0.13; P = .05). Only in control patients, indexed RV end-diastolic area increased significantly (P = .003). In TVP patients, when comparing patients with baseline more than moderate TR (n = 13) to patients with isolated TAD (n = 32), there was a significant decrease in RV end-diastolic area only in the group with more than moderate TR (from 12.9 ± 3.5 cm2/m2 to 10.3 ± 1.9 cm2/m2; P = .009).ConclusionsAdding TVP to mitral valve surgery in patients with more than moderate TR or TAD leads to favorable changes in RV geometry and prevents postoperative RV dilation. This is most pronounced in patients with more than moderate TR at baseline

    Site-directed spin labeling is emerging as an essential tool to investigate the structural and dynamical features of RNA. We propose here an enzymatic method, which allows the insertion of a paramagnetic center at a specific position in an RNA molecule. The technique is based on a segmental approach using a ligation protocol with T4 RNA ligase 2. One transcribed acceptor RNA is ligated to a donor RNA in which a thio-modified nucleotide is introduced at its 5'-end by in vitro transcription with T7 RNA polymerase. The paramagnetic thiol-specific reagent is subsequently attached to the RNA ligation product. This novel strategy is demonstrated by introducing a paramagnetic probe into the 55 nucleotides long RNA corresponding to K-turn and Specifier Loop domains from the Bacillus subtilis tyrS T-Box leader RNA. The efficiency of the coupling reaction and the quality of the resulting spin-labeled RNA were assessed by Mass Spectrometry, Electron Paramagnetic Resonance (EPR) and Nuclear Magnetic Resonance (NMR). This method enables various combinations of isotopic segmental labeling and spin labeling schemes, a strategy that will be of particular interest to investigate the structural and dynamical properties of large RNA complexes by NMR and EPR spectroscopies

    Sucesso considerável tem sido obtido no uso do melhoramento clássico para o controle de doenças de plantas economicamente importantes, tais como a ferrugem alaranjada das folhas e a antracnose dos frutos do cafeeiro (CBD). Há um grande consenso de que o uso de plantas geneticamente resistentes é o meio mais apropriado e eficaz em termos de custos do controle das doenças das plantas, sendo também um dos elementos chave do melhoramento da produção agrícola. Tem sido também reconhecido que um melhor conhecimento do agente patogênico e dos mecanismos de defesa das plantas permitirá o desenvolvimento de novas abordagens no sentido de aumentar a durabilidade da resistência. Após uma breve descrição de conceitos na área da resistência das plantas às doenças, nesta revisão tentou-se dar uma idéia do progresso na investigação da ferrugem alaranjada do cafeeiro e do CBD relativamente ao processo de infecção e variabilidade dos agentes patogênicos, melhoramento do cafeeiro para a resistência e mecanismos de resistência do cafeeiro

    High temperature piezoelectric properties of flux-grown α-GeO 2 single crystal

    International audienceThe temperature-dependence of the piezoelectric properties of trigonal -GeO2 single-crystals obtained by the high-temperature flux method was measured by the resonance technique of the electrical impedance in the 20°C-600°C range. To approach the values of the two independent piezoelectric coefficients d11 and d14, we first measured as a function of temperature the elastic coefficients S11, S14 and S66 and the dielectric permittivity 11 which are involved in the coupling coefficient k of both the thickness shear mode and the transverse mode. A Y-cut plate with a simple +45°-rotation ((YXtwl) +45°/0°/0°) was used to measure the coupling coefficient of the thickness shear mode, and two X-turned plates ((XYtwl) +45°/0°/0° and (XYtwl)-45°/0°/0°) were prepared to characterize the coupling coefficient of two transverse modes. From the whole experimental measurements, the piezoelectric coefficients of -GeO2 were calculated up to 600 °C. They show that this crystal is one of the most efficient in regard of the -quartz-like family at room temperature, and that its thermal comportment retains large piezoelectric properties up to 600°C

    Potential of EPR spin-trapping to investigate in situ free radicals generation from skin allergens in reconstructed human epidermis: cumene hydroperoxide as proof of concept

    The first step in the development of skin sensitisation to a chemical, and in the elicitation offurther allergic contact dermatitis (ACD), is the binding of the allergen to skin proteins after pene-trating into the epidermis. The so-formed antigenic adduct is then recognised by the immunesystem as foreign to the body. Sensitising organic hydroperoxides derived from autoxidation ofnatural terpenes are believed to form antigens through radical-mediated mechanisms, althoughthis has not yet been established. So far,in vitroinvestigations on reactive radical intermediatesderived from these skin sensitisers have been conducted in solution, yet with experimental condi-tions being far away from real-life sensitisation. Herein, we report for the first time, the potentialuse of EPR spin-trapping to study thein situgeneration of free radicals derived from cumenehydroperoxide CumOOH in a 3D reconstructed human epidermis (RHE) model, thus much closerto what may happenin vivo. Among the undesirable effects associated with dermal exposure toCumOOH, it is described to cause allergic and irritant dermatitis, being reported as a significantsensitiser. We considered exploiting the usage of spin-trap DEPMPO as an extensive view of allsort of radicals derived from CumOOH were observed all at once in solution. We showed that inthe EpiskinTMRHE model, both by incubating in the assay medium and by topical application,carbon radicals are mainly formed by redox reactions suggesting the key role of CumOOH-derived carbon radicals in the antigen formation process

    Rapport de sondages et d'analyses, Le Kilian et les carrières anciennes de trachyte dans la Chaîne des Puys (Puy-de-Dôme)

    En 2008, l'existence de carrières souterraines médiévales avait été mise en évidence dans la pente ouest du Bois de Manson qui domine la dépression du Cratère Kilian, au pied sud du puy de Dôme. En 2009-2010, des sondages et prospections ont été étendus à tout l'ensemble du Kilian de façon à préciser l'étendue et, si possible, la chronologie de son exploitation dans le passé. Ces travaux ont permis d'observer, dans le fond du cratère et sur son flanc interne ouest, des amoncellements de déblais d'un volume considérable, témoignant d'une extraction de roche à grande échelle durant le haut Moyen Âge et très probablement aussi à l'époque gallo-romaine. Une nouvelle carrière souterraine a été découverte dans la pente interne ouest du cratère. La base du remplissage de cette carrière a livré des charbons datés entre la fin du IV e siècle et le début du VI e siècle par le radiocarbone, tandis que le sommet du remplissage contenait des tessons de céramique datables, par leur typologie, de la fin du V e siècle au début du VIII e siècle. L'état actuel des investigations conduit à faire l'hypothèse que les gallo-romains ont exploité, au fond du cratère, un trachyte compact dont on ne trouve aujourd'hui que les déchets de taille, et dont les affleurements sont masqués par les déblais, tandis que les artisans du Moyen Âge ont recherché un trachyte plus tendre dans les pentes hautes du cratère. Le Kilian doit donc s'ajouter aux trois sources actuellement connues de trachyte d'oeuvre dans le passé, à savoir les volcans Sarcoui, Aumône (ou petit Suchet) et Cliersou. Dès cette découverte, en 2008, s'est posée la question de savoir quelle part éventuelle le trachyte du Kilian avait pu prendre dans la construction du temple de Mercure au sommet du puy de Dôme et dans l'agglomération gallo-romaine située au col de Ceyssat. Pour y répondre, une campagne d'analyses géochimiques et pétrographiques été engagée pour caractériser, aux fins de comparaison, non seulement les trachytes du Kilian et ceux des ruines gallo-romaines, mais, de plus, les trachytes du Cliersou, de l'Aumône et du Sarcoui. Ces analyses ont finalement montré qu'il est possible de faire une discrimination statistiquement significative entre les différents trachytes étudiés, à l'exception de ceux du couple Cliersou-Aumône dont les laves sont très peu différentes les unes des autres. Le résultat le plus remarquable est que tous les trachytes gallo-romains échantillonnés (dont 10 échantillons distincts au temple de Mercure et 10 au col de Ceyssat) se rattachent sans ambiguïté au Kilian. En toute rigueur, ces nouvelles données ne permettent pas d'exclure sans appel la possibilité d'utilisation à l'époque gallo-romaine, au temple de Mercure et au col de Ceyssat, de trachytes provenant d'autres sources que le Kilian (cas des chaperons de mur du col de Ceyssat, provenant du puy de Dôme). Cependant, il faut ajouter qu'un examen visuel des trachytes d'oeuvre dans ces deux sites, portant sur un nombre de moellons et d'éléments architecturaux bien supérieur au nombre de ceux qui ont été analysés, conduit à conclure que leur source est probablement commune. Cette conclusion est basée sur un faciès minéralogique particulier, observable à l'oeil nu ou à la loupe
