    How virtuous is humor? Evidence from everyday behavior

    Historical as well as contemporary writers across many disciplines have referred to humor as a virtue. However, in psychological research it is not clear in which ways humor can serve (as) a virtue, and for which virtues this holds. The current study addresses this issue from the perspective of lay people's everyday lives. The aims of the study are to investigate (1) how often people achieved each of six core virtues identified by Dahlsgaard et al. (Review of General Psychology 9: 203-213, 2005) by means of humor, also in relation to how important the respective virtue was for the participants, (2) to collect reports of situations where participants actually used humor to achieve any of the six virtues, and (3) to study the use of eight comic styles (Schmidt-Hidding, Europäische Schlüsselwörter. Band I: Humor und Witz, Huber, 1963) within the reported situations. Whereas justice and humanity were the virtues considered most important, the virtues most compatible with humor seemed to be humanity and wisdom. However, it was possible to report situations for each of the virtues. More benevolent comic styles were used more frequently to achieve virtue than more malevolent styles.     Titze (Humor and Health Journal 5:1-11, 1996) concluded from individual case studies that gelotophobes do not experience humor and laughter as a shared enjoyment but rather as a threat. Two studies examined whether gelotophobes are less humorous in general or whether this is true only for certain components of humor. In study I, three samples (N = 120 and 70 students; N = 169 adults) filled in the GELOPH46 along with several humor instruments (i.e., 3 WD, CHS, HBQD, HSQ, HUWO, STCI-T60). Results showed that gelotophobes are less cheerful and characterize their humor style as inept, socially cold, and mean-spirited. They report less frequent use of humor as a means for coping and indulge less often in self-enhancing and social humor. Appreciation of incongruity-resolution humor and nonsense humor (but not sexual humor) was lower than for non-gelotophobes. Study II (N = 131 adults) focused on the relation between gelotophobia, gelotophilia, and katagelasticism and the ability to create humor (i.e., the CPPT). The ability to create humor is unrelated to gelotophobia, and tends to be positively correlated with gelotophilia and katagelasticism.     Next generation of antiretroviral agents targeting the RNA binding site of the HIV-1 cellular cofactor DDX3: an innovative therapeutic approach

    Introduction: Efficacy of currently approved anti-HIV drugs is hampered by mutations of the viral enzymes, leading invariably to drug resistance and chemotherapy failure. Recent data suggest that cellular co-factors also represent useful targets for anti-HIV therapy. We have recently provided evidence for the possibility to block HIV-1 replication by targeting its cellular cofactor DDX3. Material and methods: Molecular modeling and in silico technologies were applied to rationally design small molecules specifically targeting the RNA binding site of human DDX3. Biochemical studies of mutated DDX3 enzymes were also used to identify additional potential drug binding sites. Results Optimization of compounds identified by application of a high-throughput docking approach afforded a promising lead compound which proved to inhibit both the helicase and ATPase activity of DDX3 and to reduce the viral load of peripheral blood mononuclear cells (PBMC) infected with HIV-1. A novel interaction site has been also identified in DDX3, which, when blocked, can reduce viral replication, representing an additional target for small molecules inhibitors. Conclusions: We have identified the first inhibitors of HIV-1 replication targeting the RNA binding site of the cellular cofactor human DDX3. These compounds may offer superior selectivity over the ATP-competitive inhibitors previously developed. In addition, a novel RNA interacting motif specific to DDX3 has been identified, opening new venues for HIV-1 drug development

    Short alleles, bigger smiles? The effect of 5-HTTLPR on positive emotional expressions.

    The present research examined the effect of the 5-HTTLPR polymorphism in the serotonin transporter gene on objectively coded positive emotional expressions (i.e., laughing and smiling behavior objectively coded using the Facial Action Coding System). Three studies with independent samples of participants were conducted. Study 1 examined young adults watching still cartoons. Study 2 examined young, middle-aged, and older adults watching a thematically ambiguous yet subtly amusing film clip. Study 3 examined middle-aged and older spouses discussing an area of marital conflict (that typically produces both positive and negative emotion). Aggregating data across studies, results showed that the short allele of 5-HTTLPR predicted heightened positive emotional expressions. Results remained stable when controlling for age, gender, ethnicity, and depressive symptoms. These findings are consistent with the notion that the short allele of 5-HTTLPR functions as an emotion amplifier, which may confer heightened susceptibility to environmental conditions

    A Motif Unique to the Human Dead-Box Protein DDX3 Is Important for Nucleic Acid Binding, ATP Hydrolysis, RNA/DNA Unwinding and HIV-1 Replication

    DEAD-box proteins are enzymes endowed with nucleic acid-dependent ATPase, RNA translocase and unwinding activities. The human DEAD-box protein DDX3 has been shown to play important roles in tumor proliferation and viral infections. In particular, DDX3 has been identified as an essential cofactor for HIV-1 replication. Here we characterized a set of DDX3 mutants biochemically with respect to nucleic acid binding, ATPase and helicase activity. In particular, we addressed the functional role of a unique insertion between motifs I and Ia of DDX3 and provide evidence for its implication in nucleic acid binding and HIV-1 replication. We show that human DDX3 lacking this domain binds HIV-1 RNA with lower affinity. Furthermore, a specific peptide ligand for this insertion selected by phage display interferes with HIV-1 replication after transduction into HelaP4 cells. Besides broadening our understanding of the structure-function relationships of this important protein, our results identify a specific domain of DDX3 which may be suited as target for antiviral drugs designed to inhibit cellular cofactors for HIV-1 replication

    DNA barcode reference libraries for the monitoring of aquatic biota in Europe: Gap-analysis and recommendations for future work

    Effective identification of species using short DNA fragments (DNA barcoding and DNA metabarcoding) requires reliable sequence reference libraries of known taxa. Both taxonomically comprehensive coverage and content quality are important for sufficient accuracy. For aquatic ecosystems in Europe, reliable barcode reference libraries are particularly important if molecular identification tools are to be implemented in biomonitoring and reports in the context of the EU Water Framework Directive (WFD) and the Marine Strategy Framework Directive (MSFD). We analysed gaps in the two most important reference databases, Barcode of Life Data Systems (BOLD) and NCBI GenBank, with a focus on the taxa most frequently used in WFD and MSFD. Our analyses show that coverage varies strongly among taxonomic groups, and among geographic regions. In general, groups that were actively targeted in barcode projects (e.g. fish, true bugs, caddisflies and vascular plants) are well represented in the barcode libraries, while others have fewer records (e.g. marine molluscs, ascidians, and freshwater diatoms). We also found that species monitored in several countries often are represented by barcodes in reference libraries, while species monitored in a single country frequently lack sequence records. A large proportion of species (up to 50%) in several taxonomic groups are only represented by private data in BOLD. Our results have implications for the future strategy to fill existing gaps in barcode libraries, especially if DNA metabarcoding is to be used in the monitoring of European aquatic biota under the WFD and MSFD. For example, missing species relevant to monitoring in multiple countries should be prioritized for future collaborative programs. We also discuss why a strategy for quality control and quality assurance of barcode reference libraries is needed and recommend future steps to ensure full utilisation of metabarcoding in aquatic biomonitoring.This paper is a deliverable of the European Cooperation in Science and Technology (COST) Action DNAqua-Net (CA15219) Working Group 1, led by Torbjørn Ekrem and Fedor Čiampor.     Can people really “laugh at themselves?”—experimental and correlational evidence

    Laughing at oneself is considered a core component of the sense of humor in the theories of several authors. In McGhee’s (1996) eight-step-training program of the sense of humor, laughing at oneself constitutes one of the most difficult levels. However, until now, only little empirical evidence on laughing at oneself exists. Using a multimethod approach, in the current study, 70 psychology students and a total of 126 peers filled in the Sense of Humor Scale (SHS, McGhee, 1996), containing as a subscale “Laughing at oneself”. In addition, the participants answered the Trait and State forms of the State- Trait-Cheerfulness-Inventory (STCI, Ruch, Köhler, & van Thriel, 1996; Ruch, Köhler, & van Thriel, 1997). They then were confronted with six distorted images of themselves. Facial responses of the participants were videotaped and analyzed using the Facial Action Coding System (FACS, Ekman, Friesen, & Hager, 2002). Four indicators of exhilaration were examined: (a) experienced funniness, (b) AU12 smiles, (c) Duchenne displays, and (d) laughter. Furthermore, fake and masking smiles were studied. Results demonstrated that self- and peer reports of “laughing at oneself” converged moderately. All four indicators of exhilaration were shown, but funniness and laughter seemed to be the most strongly related indicators. Trait cheerfulness and (low) seriousness, and a cheerful mood state formed further characteristics of persons who laugh at themselves
