Quantifying the Coastal Hazard Risk Reduction Benefits of Coral Reef Restoration in the U.S. Virgin Islands
Coastal habitat restoration, especially of coral reef ecosystems, can significantly reduce the exposure of coastal communities to natural hazards and, consequently, the risk of wave-driven flooding. Likewise, reef degradation can increase coastal flood risks to people and property. In this study, the valuation of coral reefs in the United States Virgin Islands (USVI), along the coasts of St. Croix, St. John, and St. Thomas, demonstrated the social and economic benefits provided by these natural defenses. Across the territory, more than 481 people and 31.2 million of infrastructure were estimated to receive protection from coral reefs per year (2010 U.S. dollars). In 2017, Hurricanes Irma and Maria significantly damaged coral reefs throughout the archipelago. By combining engineering, ecological, geospatial, social, and economic data and tools, this study provided a rigorous valuation of where potential coral reef restoration projects could help rebuild these damaged habitats and decrease the risks from coastal hazards faced by USVI’s reef-fronted communities. Multiple restoration scenarios were considered in the analysis, two of which are detailed in this report. These include (1) ‘Ecological’ restoration, where restoration creates a structure that is 0.25 m high and 25-m-wide reef, and (2) ‘Hybrid’ restoration, where restoration creates a structure that is 1.25 m high and 5 m wide. There are many ways that such structures could be developed. In the hydrodynamic analyses, there are no assumptions about how the restoration is developed. Many practitioners of both coral (and oyster reef) restoration consider that a reef height of 0.25 m might be delivered from planting corals alone and that 1.25 m might require a combination of artificial structures and coral planting. In a third scenario, the analysis investigated the reduction of protection benefits that would occur through the reduction of 1 meter of naturally occurring reef height due to reef degradation. The reduction of protection due to the loss of reefs can also be interpreted as the protection value of the existing reefs. In all studied restoration scenarios, it was assumed that the planting of corals would enhance hydrodynamic roughness, effectively dissipating incident wave energy and reducing the potential for coastal flooding. A standardized approach was employed to strategically locate potential restoration projects along the entire linear extent of existing reefs bordering the USVI, and to identify where coral reef restoration could offer valuable benefits in flood reduction. Potential restoration projects were only located within the existing distribution of reefs across the region, even though numerous sites were positioned far offshore (2-3 km), and some were at relatively deep depths (up to 7 m). Risk-based valuation approaches were followed to delineate flood zones at a 10 m2 resolution along the entire region's reef-lined shorelines for all the potential coral reef restoration scenarios. These were subsequently compared to flood zones without coral reef restoration. The potential reduction in coastal flood risk provided by coral reef restoration, and the protection value of existing reefs, were quantified utilizing the latest information available at the time of analysis from the U.S. Census Bureau, Federal Emergency Management Agency (FEMA), and Bureau of Economic Analysis for return-interval storm events. The change in Expected Annual Damages (EAD), a metric indicating the annual protection gained due to coral reef restoration, was calculated based on the damages associated with each storm probability. The findings suggest that the benefits of reef restoration are spatially variable within the USVI. In some areas, the analysis showed limited benefits from reef restoration, which may be attributed to the depth or offshore distances of proposed restoration sites. However, there were a number of key areas where reef restoration could have substantial benefits for flood risk reduction. The annual flood risk reduction attributed to potential ‘ecological’ coral reef restoration in the USVI was 99 people and 6.1 million (2010 U.S. dollars). The Benefit-to-Cost Ratio (BCR) for this restoration approach was found to be larger than 1 (i.e., cost-effective) along 11% of the St. Croix coastline, 4.9% of the St. John coastline, and 8.7% of the St. Thomas coastline. This analysis offers stakeholders and decision-makers a spatially explicit and rigorous evaluation that illustrates how, where, and when potential coral reef restoration efforts in St. Croix, St. John, and St. Thomas could be instrumental to reducing coastal storm-induced flooding. Understanding areas where reef management, recovery, and restoration could effectively reduce climate hazard-related risks is crucial to protect and enhance the resilience of coastal communities in USVI
Cortisol reactivity and suicidal behavior: investigating the role of hypothalamic-pituitary-adrenal axis responses to stress in suicide attempters and ideators
Every 40 seconds a person dies by suicide somewhere in the world. The causes of suicidal behavior are not fully understood. Dysregulated hypothalamic-pituitary-adrenal (HPA) axis activity, as measured by cortisol levels, is one potential risk factor. The current study aimed to investigate whether cortisol reactivity to a laboratory stress task differentiated individuals who had previously made a suicide attempt from those who had thought about suicide (suicide ideators) and control participants. One hundred and sixty participants were recruited to a previous attempt, a suicidal ideation or a control group. Participants completed background questionnaires before completing the Maastricht Acute Stress Test (MAST). Cortisol levels were assessed throughout the stress task. Measures of suicide behavior were measured at baseline, 1 month and 6 month follow-up. Participants who had made a previous suicide attempt exhibited significantly lower aggregate cortisol levels during the MAST compared to participants in the control group; suicide ideators were intermediate to both groups. This effect, however, was driven by participants who made an attempt within the past year, and to some degree by those with a family history of attempt. Participants who had made a suicide attempt and had a family history of suicide exhibited the lowest levels of cortisol in response to stress. Finally, lower levels of cortisol in response to the MAST were associated with higher levels of suicidal ideation at 1-month follow-up in the suicide attempter group. These results are consistent with other findings indicating that blunted HPA axis activity is associated with some forms of suicidal behavior. Characteristics and sources of fluorescent aerosols in the central Arctic Ocean
The Arctic is sensitive to cloud radiative forcing. Due to the limited number of aerosols present throughout much of the year, cloud formation is susceptible to the presence of cloud condensation nuclei and ice nucleating particles (INPs). Primary biological aerosol particles (PBAP) contribute to INPs and can impact cloud phase, lifetime, and radiative properties. We present yearlong observations of hyperfluorescent aerosols (HFA), tracers for PBAP, conducted with a Wideband Integrated Bioaerosol Sensor, New Electronics Option during the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition (October 2019–September 2020) in the central Arctic. We investigate the influence of potential anthropogenic and natural sources on the characteristics of the HFA and relate our measurements to INP observations during MOSAiC. Anthropogenic sources influenced HFA during the Arctic haze period. But surprisingly, we also found sporadic “bursts” of HFA with the characteristics of PBAP during this time, albeit with unclear origin. The characteristics of HFA between May and August 2020 and in October 2019 indicate a strong contribution of PBAP to HFA. Notably from May to August, PBAP coincided with the presence of INPs nucleating at elevated temperatures, that is, &gt;−9°C, suggesting that HFA contributed to the “warm INP” concentration. The air mass residence time and area between May and August and in October were dominated by the open ocean and sea ice, pointing toward PBAP sources from within the Arctic Ocean. As the central Arctic changes drastically due to climate warming with expected implications on aerosol–cloud interactions, we recommend targeted observations of PBAP that reveal their nature (e.g., bacteria, diatoms, fungal spores) in the atmosphere and in relevant surface sources, such as the sea ice, snow on sea ice, melt ponds, leads, and open water, to gain further insights into the relevant source processes and how they might change in the future.</jats:p
Neoamphimedine Circumvents Metnase-Enhanced DNA Topoisomerase IIα Activity Through ATP-Competitive Inhibition
Type IIα DNA topoisomerase (TopoIIα) is among the most important clinical drug targets for the treatment of cancer. Recently, the DNA repair protein Metnase was shown to enhance TopoIIα activity and increase resistance to TopoIIα poisons. Using in vitro DNA decatenation assays we show that neoamphimedine potently inhibits TopoIIα-dependent DNA decatenation in the presence of Metnase. Cell proliferation assays demonstrate that neoamphimedine can inhibit Metnase-enhanced cell growth with an IC50 of 0.5 μM. Additionally, we find that the apparent Km of TopoIIα for ATP increases linearly with higher concentrations of neoamphimedine, indicating ATP-competitive inhibition, which is substantiated by molecular modeling. These findings support the continued development of neoamphimedine as an anticancer agent, particularly in solid tumors that over-express Metnase
Interaction between the microbiome and TP53 in human lung cancer.
BACKGROUND: Lung cancer is the leading cancer diagnosis worldwide and the number one cause of cancer deaths. Exposure to cigarette smoke, the primary risk factor in lung cancer, reduces epithelial barrier integrity and increases susceptibility to infections. Herein, we hypothesize that somatic mutations together with cigarette smoke generate a dysbiotic microbiota that is associated with lung carcinogenesis. Using lung tissue from 33 controls and 143 cancer cases, we conduct 16S ribosomal RNA (rRNA) bacterial gene sequencing, with RNA-sequencing data from lung cancer cases in The Cancer Genome Atlas serving as the validation cohort.
RESULTS: Overall, we demonstrate a lower alpha diversity in normal lung as compared to non-tumor adjacent or tumor tissue. In squamous cell carcinoma specifically, a separate group of taxa are identified, in which Acidovorax is enriched in smokers. Acidovorax temporans is identified within tumor sections by fluorescent in situ hybridization and confirmed by two separate 16S rRNA strategies. Further, these taxa, including Acidovorax, exhibit higher abundance among the subset of squamous cell carcinoma cases with TP53 mutations, an association not seen in adenocarcinomas.
CONCLUSIONS: The results of this comprehensive study show both microbiome-gene and microbiome-exposure interactions in squamous cell carcinoma lung cancer tissue. Specifically, tumors harboring TP53 mutations, which can impair epithelial function, have a unique bacterial consortium that is higher in relative abundance in smoking-associated tumors of this type. Given the significant need for clinical diagnostic tools in lung cancer, this study may provide novel biomarkers for early detection
Long-Distance Signals Are Required for Morphogenesis of the Regenerating Xenopus Tadpole Tail, as Shown by Femtosecond-Laser Ablation
tadpoles has recently emerged as an important model for these studies; we explored the role of the spinal cord during tadpole tail regeneration.Using ultrafast lasers to ablate cells, and Geometric Morphometrics to quantitatively analyze regenerate morphology, we explored the influence of different cell populations. For at least twenty-four hours after amputation (hpa), laser-induced damage to the dorsal midline affected the morphology of the regenerated tail; damage induced 48 hpa or later did not. Targeting different positions along the anterior-posterior (AP) axis caused different shape changes in the regenerate. Interestingly, damaging two positions affected regenerate morphology in a qualitatively different way than did damaging either position alone. Quantitative comparison of regenerate shapes provided strong evidence against a gradient and for the existence of position-specific morphogenetic information along the entire AP axis.We infer that there is a conduit of morphology-influencing information that requires a continuous dorsal midline, particularly an undamaged spinal cord. Contrary to expectation, this information is not in a gradient and it is not localized to the regeneration bud. We present a model of morphogenetic information flow from tissue undamaged by amputation and conclude that studies of information coming from far outside the amputation plane and regeneration bud will be critical for understanding regeneration and for translating fundamental understanding into biomedical approaches
EML4-ALK induces cellular senescence in mortal normal human cells and promotes anchorage-independent growth in hTERT-transduced normal human cells
Background: Chromosomal inversions involving anaplastic lymphoma kinase (ALK) and echinoderm microtubule associated protein like 4 (EML4) generate a fusion protein EML4-ALK in non-small cell lung cancer (NSCLC). The understanding of EML4-ALK function can be improved by a functional study using normal human cells.
Methods: Here we for the first time conduct such study to examine the effects of EML4-ALK on cell proliferation, cellular senescence, DNA damage, gene expression profiles and transformed phenotypes.
Results: The lentiviral expression of EML4-ALK in mortal, normal human fibroblasts caused, through its constitutive ALK kinase activity, an early induction of cellular senescence with accumulated DNA damage, upregulation of p16INK4A and p21WAF1, and senescence-associated β-galactosidase (SA-β-gal) activity. In contrast, when EML4-ALK was expressed in normal human fibroblasts transduced with telomerase reverse transcriptase (hTERT), which is activated in the vast majority of NSCLC, the cells showed accelerated proliferation and acquired anchorage-independent growth ability in soft-agar medium, without accumulated DNA damage, chromosome aberration, nor p53 mutation. EML4-ALK induced the phosphorylation of STAT3 in both mortal and hTERT-transduced cells, but RNA sequencing analysis suggested that the different signaling pathways contributed to the different phenotypic outcomes in these cells. While EML4-ALK also induced anchorage-independent growth in hTERT-immortalized human bronchial epithelial cells in vitro, the expression of EML4-ALK alone did not cause detectable in vivo tumorigenicity in immunodeficient mice.
Conclusions: Our data indicate that the expression of hTERT is critical for EML4-ALK to manifest its in vitro transforming activity in human cells. This study provides the isogenic pairs of human cells with and without EML4-ALK expression
