    Essays on Monotone Comparative Statics for Constrained Optimization Problems with Applications

    My dissertation consists of three chapters. Chapters 1 and 2 both address extensions to existing monotone comparative statics results for constrained optimization problems using lattice programming techniques. Chapter 3 applies monotone comparative statics results to the question how environmental regulation affects investment in innovation in imperfectly competitive markets. Generally we can distinguish between two types of comparative statics problems. The first type of problem considers the change of the optimal solution to a maximization problem as the objective function changes, the other type considers the change due to a change in the constraint set. Lattice-based comparative statics theorems have been developed for both types of problems in the literature. The strengths of these lattice-theoretic comparative statics results are that they don't depend on the usual smoothness, interiority and concavity assumptions as required by the classical approach based on the Implicit Function Theorem, as well as convexity of the constraint set. Moreover, these comparative statics results also apply in the case of non-unique solutions.Quah (2007) expanded existing results by Milgrom and Shannon (1994) by making them applicable to some non-lattice constraint sets. In the first chapter, I extend existing comparative statics theorems to parametrized objective functions and non-lattice constraint sets. This generalization makes it possible to analyze a variety of economic optimization problems that fall into this class of problems which cannot be addressed using existing lattice-based techniques. I provide examples from consumer theory, producer theory and environmental economics that show the result's broad scope of applications. The second chapter studies monotone comparative statics with respect to price changes in the consumer's utility maximization problem. Most attempts to derive this property rely on aspects of the demand curve, and it has been hard to derive this property using assumptions on the primitive utility function. Using new results on the comparative statics of demand in Quah (2007), I provide simple and easy conditions on utility functions that yield the gross substitutes property. Quah (2007) provides conditions on utility functions that yield normal demand. I add an assumption on elasticity of marginal rate of substitution, which combined with Quah's assumptions yields gross substitutes. I apply this assumption to the family of constant elasticity of substitution preferences. My approach is grounded in the standard comparative statics decomposition of a change in demand due to a change in price into a substitution effect and an income effect. Quah's assumptions are helpful to sign the income effect. Combined with the elasticity assumption, we can sign the overall effect. As a by-product, I also present conditions which yield the gross complements property. Chapter 3 is an application of monotone comparative statics results to the question how environmental regulation affects incentives for R&D investment. For decades, there has been debate among economists whether environmental regulation hurts firms by restricting their choices or provides them with a comparative advantage through investment in innovation in more efficient technology. This chapter studies the effect of environmental regulation on firms' investment in R&D in imperfectly competitive output markets using a monotone comparative statics approach. I provide necessary and sufficient conditions on the profit function that guarantee nondecreasing R&D investment as regulation is tightened and find that a form of weak complementarity between environmental R&D investment and the policy variable plays a crucial role. Moreover, I provide properties of such profit functions through assumptions on demand, Cournot output and cost functions

    Highlighting the impact of shear strain on the SiO 2 glass structure: From experiments to atomistic simulations

    International audienceSiO 2 glass structure has been permanently modified by uniaxial compression. Within such a loading, the structure is supposed to be affected both by densification and shear flow. We propose to compare recovered silica samples with similar densities, initially deformed plastically under a hydrostatic compression or under a uniaxial compression. From micro-Raman spectroscopy experiments, the shear strain effects have been highlighted on the structural modifications of the glass and have been confirmed from molecular dynamic simulations. In particular , medium range order depends on the mechanical history in plastically deformed glasses. Indeed, both experiments and simulations demonstrate that small rings are favored when permanent shear strain acts with densification, thus allowing a structural signature identification of the densification process

    Inhibition of melanoma cell-intrinsic Tim-3 stimulates MAPK-dependent tumorigenesis.

    T-cell immunoglobulin and mucin domain 3 (Tim-3) is an immune checkpoint receptor that dampens effector functions and causes terminal exhaustion of cytotoxic T-cells. Tim-3 inhibitors are under investigation in immuno-oncology (IO) trials, because blockade of T-cell-Tim-3 enhances antitumor immunity. Here, we identify an additional role for Tim-3 as a growth-suppressive receptor intrinsic to melanoma cells. Inhibition of melanoma cell-Tim-3 promoted tumor growth in both immunocompetent and immunocompromised mice, while melanoma-specific Tim-3 overexpression attenuated tumorigenesis. Antibody (Ab)-mediated Tim-3 blockade inhibited growth of immunogenic murine melanomas in T-cell-competent hosts, consistent with established antitumor effects of T-cell Tim-3 inhibition. In contrast, Tim-3 Ab administration stimulated tumorigenesis of both highly and lesser immunogenic murine and human melanomas in T-cell-deficient mice, confirming growth-promoting effects of melanoma-Tim-3 antagonism. Melanoma-Tim-3 activation suppressed, while its blockade enhanced, phosphorylation of pro-proliferative downstream mitogen-activated protein kinase (MAPK) signaling mediators. Finally, pharmacologic MAPK inhibition reversed unwanted Tim-3 Ab-mediated tumorigenesis in T-cell-deficient mice and promoted desired antitumor activity of Tim-3 interference in T-cell-competent hosts. These results identify melanoma-Tim-3 blockade as a mechanism that antagonizes T-cell-Tim-3-directed IO therapeutic efficacy. They further reveal MAPK targeting as a combination strategy for circumventing adverse consequences of unintended melanoma-Tim-3 inhibition

    Successful Thrombectomy Improves Functional Outcome in Tandem Occlusions with a Large Ischemic Core

    International audienceBackground: Emergent stenting in tandem occlusions and mechanical thrombectomy (MT) of acute ischemic stroke related to large vessel occlusion (LVO-AIS) with a large core are tested independently. We aim to assess the impact of reperfusion with MT in patients with LVO-AIS with a large core and a tandem occlusion and to compare the safety of reperfusion between large core with tandem and nontandem occlusions in current practice. Methods: We analyzed data of all consecutive patients included in the prospective Endovascular Treatment in Ischemic Stroke Registry in France between January 2015 and March 2023 who presented with a pretreatment ASPECTS (Alberta Stroke Program Early CT Score) of 0–5 and angiographically proven tandem occlusion. The primary end point was a favorable outcome defined by a modified Rankin Scale (mRS) score of 0–3 at 90 days. Results: Among 262 included patients with a tandem occlusion and ASPECTS 0–5, 203 patients (77.5%) had a successful reperfusion (modified Thrombolysis in Cerebral Infarction grade 2b-3). Reperfused patients had a favorable shift in the overall mRS score distribution (adjusted odds ratio [aOR], 1.57 [1.22–2.03]; P < 0.001), higher rates of mRS score 0–3 (aOR, 7.03 [2.60–19.01]; P < 0.001) and mRS score 0–2 at 90 days (aOR, 3.85 [1.39–10.68]; P = 0.009) compared with nonreperfused. There was a trend between the occurrence of successful reperfusion and a decreased rate of symptomatic intracranial hemorrhage (aOR, 0.5 [0.22–1.13]; P = 0.096). Similar safety outcomes were observed after large core reperfusion in tandem and nontandem occlusions. Conclusions: Successful reperfusion was associated with a higher rate of favorable outcome in large core LVO-AIS with a tandem occlusion, with a safety profile similar to nontandem occlusion

    Prevalence Estimates of Amyloid Abnormality Across the Alzheimer Disease Clinical Spectrum

    Importance: One characteristic histopathological event in Alzheimer disease (AD) is cerebral amyloid aggregation, which can be detected by biomarkers in cerebrospinal fluid (CSF) and on positron emission tomography (PET) scans. Prevalence estimates of amyloid pathology are important for health care planning and clinical trial design. Objective: To estimate the prevalence of amyloid abnormality in persons with normal cognition, subjective cognitive decline, mild cognitive impairment, or clinical AD dementia and to examine the potential implications of cutoff methods, biomarker modality (CSF or PET), age, sex, APOE genotype, educational level, geographical region, and dementia severity for these estimates. Design, Setting, and Participants: This cross-sectional, individual-participant pooled study included participants from 85 Amyloid Biomarker Study cohorts. Data collection was performed from January 1, 2013, to December 31, 2020. Participants had normal cognition, subjective cognitive decline, mild cognitive impairment, or clinical AD dementia. Normal cognition and subjective cognitive decline were defined by normal scores on cognitive tests, with the presence of cognitive complaints defining subjective cognitive decline. Mild cognitive impairment and clinical AD dementia were diagnosed according to published criteria. Exposures: Alzheimer disease biomarkers detected on PET or in CSF. Main Outcomes and Measures: Amyloid measurements were dichotomized as normal or abnormal using cohort-provided cutoffs for CSF or PET or by visual reading for PET. Adjusted data-driven cutoffs for abnormal amyloid were calculated using gaussian mixture modeling. Prevalence of amyloid abnormality was estimated according to age, sex, cognitive status, biomarker modality, APOE carrier status, educational level, geographical location, and dementia severity using generalized estimating equations. Results: Among the 19 097 participants (mean [SD] age, 69.1 [9.8] years; 10 148 women [53.1%]) included, 10 139 (53.1%) underwent an amyloid PET scan and 8958 (46.9%) had an amyloid CSF measurement. Using cohort-provided cutoffs, amyloid abnormality prevalences were similar to 2015 estimates for individuals without dementia and were similar across PET- and CSF-based estimates (24%; 95% CI, 21%-28%) in participants with normal cognition, 27% (95% CI, 21%-33%) in participants with subjective cognitive decline, and 51% (95% CI, 46%-56%) in participants with mild cognitive impairment, whereas for clinical AD dementia the estimates were higher for PET than CSF (87% vs 79%; mean difference, 8%; 95% CI, 0%-16%; P = .04). Gaussian mixture modeling-based cutoffs for amyloid measures on PET scans were similar to cohort-provided cutoffs and were not adjusted. Adjusted CSF cutoffs resulted in a 10% higher amyloid abnormality prevalence than PET-based estimates in persons with normal cognition (mean difference, 9%; 95% CI, 3%-15%; P = .004), subjective cognitive decline (9%; 95% CI, 3%-15%; P = .005), and mild cognitive impairment (10%; 95% CI, 3%-17%; P = .004), whereas the estimates were comparable in persons with clinical AD dementia (mean difference, 4%; 95% CI, -2% to 9%; P = .18). Conclusions and Relevance: This study found that CSF-based estimates using adjusted data-driven cutoffs were up to 10% higher than PET-based estimates in people without dementia, whereas the results were similar among people with dementia. This finding suggests that preclinical and prodromal AD may be more prevalent than previously estimated, which has important implications for clinical trial recruitment strategies and health care planning policies
