    TITLE: The options of prosthetic equipment for people with unilateral transfemoral amputation and its impact on their physical capability OBJECTIVES: The aim of the work is to evaluate the effect of the used components of the prosthesis on the activity level of adults with unilateral transfemoral amputation, which are currently available on the domestic market covered by health insurance with deductibles. METHODS: The basis of the analytical part is literary research and data collection of technical data sheets. The second part of the work is qualitative research and contains case studies of five probands. The research group consisted of 5 persons with unilateral transfemoral amputation aged 49-78 years. Unstructured interview and observation methods were used for data collection. RESULTS: The result of the work is a summary of information related to the assembly of different types of TF prostheses for adults and evaluation of the relation between the level of technology used and the quality of the user's life. Druhá část práce je vedena jako kvalitativní výzkum a obsahuje případové studie pěti probandů. Výzkumný soubor tvořilo 5 osob s unilaterální transfemorální amputací ve věku 49-78 let. Pro sběr dat byly použity metody nestrukturovaný rozhovor a pozorování. VÝSLEDKY: Výsledkem práce je sumarizace informací vztahující se k sestavení různých typů TF protéz pro dospělého člověka a vyhodnocení souvislosti mezi úrovní použité techniky a kvalitou života uživatele.     Import 07/02/2011Bakalářská práce na téma Venkovská turistika je rozdělena na část teoretickou, praktickou a dotazník s doporučením. V teoretické části se práce zabývá obecným cestovním ruchem, jeho formami a také se zaměřuje na část, která se vymezuje na venkově. Zde práce vyčleňuje možnosti venkovské turistiky s důrazem na cykloturistiku, její historii a rozvoj, druhy, význam, možnosti a také druhy cyklistických tras a kol. Praktická část je zaměřena na vymezení regionu Slovácka, zobrazení jeho atraktivit jak už přírodních tak i historických. Také zde uvádí turistickou infrastrukturu, do které patří ubytování, lázně, gastronomie a v neposlední řadě vinařské stezky. V závěru bakalářské práce je provedeno vyhodnocení dotazníků, které je graficky zpracováno a jsou zde navržena doporučení, která by měla přispět ke zvýšení kvality a množství nabízených výrobků, jejich propagace a služeb. Cílem této práce je zachytit problematiku venkovské turistiky, obzvláště cykloturistiky ve spojitosti s vinařstvím, která je v dnešní době velmi oblíbená a zdůraznit hlavní klady, které vinařské cyklostezky v regionu Slovácko přináší.The bachelor thesis is divided into three parts - theoretical, practical and questionnaire with recommendations. In the theoretical part forms of general tourism are described with more detail on countryside tourism and especially the cycle track tourism, its history, forms and potentials. Practical part focuses in describing Slovácko region depicting its natural and historical eye-appeals. Moreover describing its tourist infrastructure including accommodation, spas, gastronomy and wine tracks. Conclusion evaluates the questionnaires in graphs with recommendations to increase its quality, amount of goods and services together with their promotion.     Plasma Concentrations of Extracellular DNA in Acute Kidney Injury

    Current diagnostic methods of acute kidney injury (AKI) have limited sensitivity and specificity. Tissue injury has been linked to an increase in the concentrations of extracellular DNA (ecDNA) in plasma. A rapid turnover of ecDNA in the circulation makes it a potential marker with high sensitivity. This study aimed to analyze the concentration of ecDNA in plasma in animal models of AKI. Three different fractions of ecDNA were measured—total ecDNA was assessed fluorometrically, while nuclear ecDNA (ncDNA) and mitochondrial DNA (mtDNA) were analyzed using quantitative real-time PCR. AKI was induced using four different murine models of AKI-bilateral ureteral obstruction (BUO), glycerol-induced AKI (GLY), ischemia–reperfusion injury (IRI) and bilateral nephrectomy (BNx). Total ecDNA was significantly higher in BUO (p < 0.05) and GLY (p < 0.05) compared to the respective control groups. ncDNA was significantly higher in BUO (p < 0.05) compared to SHAM. No significant differences in the concentrations of mtDNA were found between the groups. The plasma concentrations of different fractions of ecDNA are dependent on the mechanism of induction of AKI and warrant further investigation as potential surrogate markers of AKI.publishedVersio

    Sex differences in long-term effects of collagen-induced arthritis in middle-aged mice

    Introduction: Rheumatoid arthritis (RA) is a chronic inflammatory disorder with high prevalence among middle-aged women. Collagen-induced arthritis (CIA) is the most widely used animal model of RA, however, sex differences and long-term effects of CIA in mice are poorly described in the literature.Aim: Therefore, the present study aimed to analyze the long-term effects of CIA on the joints of middle-aged mice of both sexes and to describe potential sex differences.Materials and methods: CIA was induced in middle-aged DBA/1J mice by immunization with bovine type II collagen and complete Freund’s adjuvant. Saline was administered to control mice. Arthritis score assessment, plethysmometry, and thermal imaging of the joints were performed weekly for 15 weeks. Locomotor activity, micro-computed tomography, joint histology and biochemical analyses were performed at the end of the experiment.Results: Our results indicate a similar prevalence of arthritis in both sexes of mice—67% (8/12) of females and 89% (8/9) males with an earlier onset in males (day 14 vs. day 35). After the arthritis scores peaked on day 56 for males and day 63 for females, they steadily declined until the end of the experiment on day 105. A similar dynamics was observed in paw volume and temperature analyzing different aspects of joint inflammation. Long-term consequences including higher proteinuria (by 116%), loss of bone density (by 33.5%) and joint damage in terms of synovial hyperplasia as well as bone and cartilage erosions were more severe in CIA males compared to CIA females. There were no significant differences in locomotor activity between CIA mice and CTRL mice of any sex.Conclusion: This is the first study to describe the long-term effects of the CIA model in terms of sex differences in DBA/1J mice. Our results indicate sex differences in the dynamics, but not in the extent of arthritis. An earlier onset of arthritis and more severe consequences on joints, bones and kidneys were found in males. The underlying immune pathomechanisms responsible for the limited duration of the arthritis symptoms and the opposite sex difference in comparison to RA patients require further investigation

    The diploma thesis " The transformation of the press releases into the media texts" deals with criteria according to what journalists make decisions, what press release should be included into the news and what should not. Since the present media studies perceive gatekeeping as a broader process of information control involving all aspects of message encoding, therefore not just a selection but also a compilation, a transmition, a formation and the timing of information transmition from a supplier to a recipient, the diploma thesis also pursues the transformation process of a press release into a media text itself. The diploma thesis is divided into a theoretical and a practical part. The theoretical part includes the gatekeeping process analysis, the pseudoevent problems analysis and news production routinization analysis. This chapter summarizes the knowledge to the topic from scholarly literature. The second and the principal part of the diploma thesis is the practical and application part. This part argues a personal and a creative approach to a chosen problem solution. The premediate defined hypothesis are being disproved or approved on the basis of a practical research.
