This study aimed to investigate the therapeutic effects of intranasal administration of human endometrium-derived stem cells (HEDSCs) in the mouse model of Parkinsons disease (PD). Thirty days after intrastriatal injection of 6-OHDA, HEDSCs were administrated intranasally in three doses (104, 5 104 and 105 cells µl1). During 120 days after stem cell administration, behavioral tests were examined. Then the mice were sacrificed and the fresh section of the substantia nigra pars compacta (SNpc) was used for detection of HEDSCs-GFP labeled by fluorescence microscopy method. In addition, immunohistochemistry was used to assay GFP, human neural Nestin, and tyrosine hydroxylase (TH) markers in the fixed brain tissue at the SNpc. Our data revealed that behavioral parameters were significantly improved after cell therapy. Fluorescence microscopy assay in fresh tissue and GFP analysis in fixed tissue were showed that the HEDSCs-GFP labeled migrated to SNpc. The data from immunohistochemistry revealed that the Nestin as a differential neuronal biomarker was expressed in SNpc. Also, TH as a dopaminergic neuron marker significantly increased after HEDSCs therapy in an optimized dose 5 104 cells µl1. Our results suggest that intranasal administration of HEDSCs improve the PD symptoms in the mouse model of PD dose-dependent manner as a noninvasive method. © 2019, Springer Nature B.V

Introduction: Progress in nanotechnology in the past decayed has created various opportunities for evaluation of biological effects such as anti-bacterial effects of nanoparticles. This study was aimed to examine synthesis and the antibacterial effect of Nano-Polyamidoamine-G5 (NPAMAM-G5) dendrimer on Klebsiella Pneumoniae, Pseudomonas Aeruginosa, Shigella Dysenteriae and Bacillus Subtilis. Materials and Methods: NPAMAM-G5 dendrimers was synthesized by Tomalias divergent growth approach. The antibacterial effects of NPAMAM-G5 dendrimer were studied by disc diffusion and micro-dilution method. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against gram-positive and gram-negative bacteria were determined according to Clinical and Laboratory Standards Institute (CLSI) guideline. Transmission electron microscopy (TEM) was used to analyze morphology and size of NPAMAM-G5. Results: Zone of inhibition in concentration 25μg/ml of NPAMAM-G5 dendrimers for Klebsiella Pneumoniae, Pseudomonas Aeruginosa, Shigella Dysenteriae and Bacillus Subtilis were 27, 13, 30 and 18 mm, respectively. There was a significant difference regarding the zone of inhibition between gram-negative and gram-positive bacteria (p<0.05). Remarkably, the MIC for Klebsiella Pneumoniae, Pseudomonas Aeruginosa and Shigella Dysenteriae was 2.5μg/ml and for Bacillus Subtilis was 25μg/ml. The MBC for Shigella Dysenteriae and Pseudomonas aeruginosa were 50 and 200 μg/ml, respectively and for Klebsiella Oxytoca and Bacillus Subtilis was100 μg/ml. It was found that NPAMAM-G5 particles had a spherical shape with a mean diameter size of 10 nm. Conclusion: According to the results, the NPAMAM-G5 dendrimer with end amine groups displayed a positive effect on the removal of standard strains of gram-positive and gram-negative bacteria. © 2019, Semnan University of Medical Sciences. Preparation and optimization of macroalgae-derived solid acid catalysts

    Get PDF
Solid acid catalysts were synthesized from macroalgae Sargassum horneri via hydrothermal carbonization followed by sulfuric acid sulfonation. A three-variable Box-Behnken design and optimization was used to maximize surface acidity. The optimal preparation conditions were found to be at the carbonization temperature of 217 °C, the carbonization time of 4.6 h and the sulfonation temperature of 108.5 °C. Under these conditions, the highest surface acidity achieved was 1.62 mmol g−1. Physical and chemical properties of prepared solid acid catalyst were characterized by powder X-ray diffraction (PXRD), Fourier transform infrared (FTIR) spectroscopy, and elemental analysis. The results proved the grafting of –SO3H groups on an amorphous carbon structure. The catalyst activity was evaluated by the esterification of oleic acid with methanol. The sample prepared achieved 96.6% esterification yield, which was higher than the 86.7% yield achieved by commercial Ambersyst-15 under the same reaction conditions

A Novel Mouse Fgfr2 Mutant, Hobbyhorse (hob), Exhibits Complete XY Gonadal Sex Reversal

The secreted molecule fibroblast growth factor 9 (FGF9) plays a critical role in testis determination in the mouse. In embryonic gonadal somatic cells it is required for maintenance of SOX9 expression, a key determinant of Sertoli cell fate. Conditional gene targeting studies have identified FGFR2 as the main gonadal receptor for FGF9 during sex determination. However, such studies can be complicated by inefficient and variable deletion of floxed alleles, depending on the choice of Cre deleter strain. Here, we report a novel, constitutive allele of Fgfr2, hobbyhorse (hob), which was identified in an ENU-based forward genetic screen for novel testis-determining loci. Fgr2hob is caused by a C to T mutation in the invariant exon 7, resulting in a polypeptide with a mis-sense mutation at position 263 (Pro263Ser) in the third extracellular immunoglobulin-like domain of FGFR2. Mutant homozygous embryos show severe limb and lung defects and, when on the sensitised C57BL/6J (B6) genetic background, undergo complete XY gonadal sex reversal associated with failure to maintain expression of Sox9. Genetic crosses employing a null mutant of Fgfr2 suggest that Fgr2hob is a hypomorphic allele, affecting both the FGFR2b and FGFR2c splice isoforms of the receptor. We exploited the consistent phenotype of this constitutive mutant by analysing MAPK signalling at the sex-determining stage of gonad development, but no significant abnormalities in mutant embryos were detected

A method for the preconcentration of trace heavy metal ions in environmental samples has been reported. The presented method is based on the sorption of Cu(II), Cd(II), Ni(II) and Cr(III) ions with salicylic acid as respective chelate on silica-coated magnetite nanoparticles. Prepared adsorbent was characterized by XRD, SEM, BET and FT-IR measurements. The metals content of the sorbed complexes are eluted using 4.0 mL of 1.0 mol L-1 nitric acid. The influences of the analytical parameters including pH, amount of solid phase and condition of eluting solution, the effects of matrix ions on the retention of the analytes were examined. The accuracy and precision of suggested method were tested by analyzing of certified reference materials. The detection limits (3Sb/m, N = 8) for Cu(II), Cd(II), Ni(II) and Cr(III) ions are 0.22, 0.11, 0.27 and 0.15 μg L-1, respectively, and the maximum preconcentration factor is 200. The method was successfully applied to the evaluation of these trace and toxic metals in various waters, foods and other samples

    Get PDF
This chapter surveys the state-of-the-art in forecasting cryptocurrency value by Sentiment Analysis. Key compounding perspectives of current challenges are addressed, including blockchains, data collection, annotation, and filtering, and sentiment analysis metrics using data streams and cloud platforms. We have explored the domain based on this problem-solving metric perspective, i.e., as technical analysis, forecasting, and estimation using a standardized ledger-based technology. The envisioned tools based on forecasting are then suggested, i.e., ranking Initial Coin Offering (ICO) values for incoming cryptocurrencies, trading strategies employing the new Sentiment Analysis metrics, and risk aversion in cryptocurrencies trading through a multi-objective portfolio selection. Our perspective is rationalized on the perspective on elastic demand of computational resources for cloud infrastructures
