    Objectifying fitness: A content and thematic analysis of #Fitspiration images on social media.

    Research suggests that exposure to #fitspiration content can result in increased body dissatisfaction. Employing a data-driven approach, the present study examines the nature of images and text contained within #fitspiration posts on social media. First, a content analysis of images labelled as #fitspiration on popular site Instagram (N = 1,000) was performed. People featured in 52% of images, of which nearly 90% of individuals were coded as having low body fat and 55% were coded as muscular. Individuals were typically presented in sexually objectified ways, which varied as a function of gender. Second, a thematic analysis of text from the same set of images (N = 400) was conducted to identify common themes and underlying meaning embedded within the messages. Six themes were developed: (1) Fit is sexy, (2) A fit physique requires commitment and self-regulation, (3) Your choices define you, (4) Pleasure and perseverance through pain, (5) Battle of the selves: You vs. You, and (6) Here’s to us! A celebration of a community. In combination, the analyses demonstrate how text and images found in #fitspiration posts perpetuate pervading sociocultural appearance ideals for men and women, positioning exercise as means to achieving these ideals. Furthermore, #fitspiration sexually objectifies the fit body, with text and images encouraging self-objectification and the distancing of the self from internal bodily functions in physical activity settings. Future research should focus on how #fitspiration content influences individuals in relation to how they think and feel about their body and physical activity

    “It’s Better Saying I Look Fat Instead of Saying You Look Fat”: A Qualitative Study of U.K. Adolescents’ Understanding of Appearance-Related Interactions on Social Media

    Appearance-related interactions with peers, both positive and negative, are commonplace on social media. Using qualitative methods, this study explores U.K. adolescents’ shared understandings and experiences of these interactions. Sixty-four adolescents (Age M = 12.56; SD = 0.97; Girls = 33) from a secondary school in Northern England participated in semi-structured focus groups. Using thematic analysis, three themes were developed that encapsulate their shared understandings of appearance-related interactions: (a) positive appearance commentary is the norm, especially if you are popular and attractive; (b) comments to others should be positive, but comments about the self should be modest and self-deprecating; and (c) negative appearance comments are problematic but not always intentionally harmful. Overall, our findings suggest that, to adolescents, the boundaries between positive and negative interactions are blurred, as content, intention, gender, and social rules intersect with social media platform design. Further research is needed to better understand how social media site design alters adolescents’ appearance interactions, as well as the role of these interactions in the development and maintenance of peer relationships and body image concerns

    ‘When you think of exercising, you don’t really want to think of puking, tears, and pain’: Young adolescents’ understanding of fitness and #fitspiration

    Adolescents access information about fitness, including content labelled as #fitspiration, through social media. A total of 77 adolescents (mean age = 12.49; standard deviation = 0.55; girls = 27) participated in semi-structured focus groups to explore their perspectives on #fitspiration and fitness more broadly. Through inductive thematic analysis, four themes were developed: (1) fitness enhances physical function and appearance, but these are not always linked; (2) fitness is transformative but requires hard work; (3) fitness should be an intrinsically motivated personal choice; and (4) pain in the pursuit of fitness. Findings highlight young adolescents’ complex understandings of fitness negotiated through their critical interpretation of #fitspiration

    "Just Shave It Off"

    Introduction Head hair comprises a critical part of the male appearance ideal, which itself is a crucial signifier of a man’s masculinity. However, difficulties in recruitment have meant that research has not yet fully explored how men construct the loss of head hair (baldness), perhaps because it is considered “feminine” to disclose body dissatisfaction experiences to a researcher or other people. Methods and Design Online forums provide an opportunity for the anonymous discussion of body dissatisfaction that may overcome this obstacle. The first 260 forums posts from the two most popular baldness forums were thematically analysed. Ethics Statement Institutional ethics approval was granted. Results and Discussion We identified three themes titled: (1) Baldness is an ugly and demasculinising condition, (2) Baldness is stigmatised by a superficial society and superficial women and (3) Resistance to baldness despair. Our findings show baldness distress, and stigma exist though so does resistance, which can be comforting to men experiencing baldness or any form of body dissatisfaction. Conclusion and Implications Online forums are a salient resource to enhance our understanding of men’s balding concerns and disclosure barriers. Independent, professional and effective baldness support that unpacks baldness masculinised and medicalised framing is recommended

    The mechanical response of talin

    Talin, a force-bearing cytoplasmic adapter essential for integrin-mediated cell adhesion, links the actin cytoskeleton to integrin-based cell–extracellular matrix adhesions at the plasma membrane. Its C-terminal rod domain, which contains 13 helical bundles, plays important roles in mechanosensing during cell adhesion and spreading. However, how the structural stability and transition kinetics of the 13 helical bundles of talin are utilized in the diverse talin-dependent mechanosensing processes remains poorly understood. Here we report the force-dependent unfolding and refolding kinetics of all talin rod domains. Using experimentally determined kinetics parameters, we determined the dynamics of force fluctuation during stretching of talin under physiologically relevant pulling speeds and experimentally measured extension fluctuation trajectories. Our results reveal that force-dependent stochastic unfolding and refolding of talin rod domains make talin a very effective force buffer that sets a physiological force range of only a few pNs in the talin-mediated force transmission pathway

    Prioritizing Environmental Issues around the World: Opinions from an International Central and Eastern European Environmental Health Conference

    BACKGROUND: As the next generation of scientists enters the field of environmental health, it is imperative that they view their contributions in the context of global environmental stewardship. In this commentary, a group of international graduate students facilitated by three experienced environmental health scientists present their views on what they consider to be the global environmental health concerns of today. This group convened initially in October 2004 at an international health conference in Prague, Czech Republic. OBJECTIVES: In this report we identify perceived environmental health concerns that exist around the world, with a focus on Central and Eastern Europe. Additionally, we address these perceived problems and offers some potential solutions. DISCUSSION: At the meeting, students were invited to participate in two panel discussions. One group of young international scientists identified several significant global environmental health concerns, including air pollution, occupational hazards, and risk factors that may exacerbate current environmental health issues. The second panel determined that communication, education, and regulation were the mechanisms for addressing current environmental challenges. CONCLUSIONS: In this commentary we expand on the views presented at the meeting and represent the concerns of young investigators from nine different countries. We provide ideas about and support the exchange of information between developed and developing countries on how to handle the environmental health challenges that face the world today

    Matrix metalloproteinase-9 activity and a downregulated Hedgehog pathway impair blood-brain barrier function in an <i>in vitro</i> model of CNS tuberculosis

    Central nervous system tuberculosis (CNS TB) has a high mortality and morbidity associated with severe inflammation. The blood-brain barrier (BBB) protects the brain from inflammation but the mechanisms causing BBB damage in CNS TB are uncharacterized. We demonstrate that Mycobacterium tuberculosis (Mtb) causes breakdown of type IV collagen and decreases tight junction protein (TJP) expression in a co-culture model of the BBB. This increases permeability, surface expression of endothelial adhesion molecules and leukocyte transmigration. TJP breakdown was driven by Mtb-dependent secretion of matrix metalloproteinase (MMP)-9. TJP expression is regulated by Sonic hedgehog (Shh) through transcription factor Gli-1. In our model, the hedgehog pathway was downregulated by Mtb-stimulation, but Shh levels in astrocytes were unchanged. However, Scube2, a glycoprotein regulating astrocyte Shh release was decreased, inhibiting Shh delivery to brain endothelial cells. Activation of the hedgehog pathway by addition of a Smoothened agonist or by addition of exogenous Shh, or neutralizing MMP-9 activity, decreased permeability and increased TJP expression in the Mtb-stimulated BBB co-cultures. In summary, the BBB is disrupted by downregulation of the Shh pathway and breakdown of TJPs, secondary to increased MMP-9 activity which suggests that these pathways are potential novel targets for host directed therapy in CNS TB

    Prenatal Famine and Genetic Variation Are Independently and Additively Associated with DNA Methylation at Regulatory Loci within IGF2/H19

    Both the early environment and genetic variation may affect DNA methylation, which is one of the major molecular marks of the epigenome. The combined effect of these factors on a well-defined locus has not been studied to date. We evaluated the association of periconceptional exposure to the Dutch Famine of 1944–45, as an example of an early environmental exposure, and single nucleotide polymorphisms covering the genetic variation (tagging SNPs) with DNA methylation at the imprinted IGF2/H19 region, a model for an epigenetically regulated genomic region. DNA methylation was measured at five differentially methylated regions (DMRs) that regulate the imprinted status of the IGF2/H19 region. Small but consistent differences in DNA methylation were observed comparing 60 individuals with periconceptional famine exposure with unexposed same-sex siblings at all IGF2 DMRs (PBH<0.05 after adjustment for multiple testing), but not at the H19 DMR. IGF2 DMR0 methylation was associated with IGF2 SNP rs2239681 (PBH = 0.027) and INS promoter methylation with INS SNPs, including rs689, which tags the INS VNTR, suggesting a mechanism for the reported effect of the VNTR on INS expression (PBH = 3.4×10−3). Prenatal famine and genetic variation showed similar associations with IGF2/H19 methylation and their contributions were additive. They were small in absolute terms (<3%), but on average 0.5 standard deviations relative to the variation in the population. Our analyses suggest that environmental and genetic factors could have independent and additive similarly sized effects on DNA methylation at the same regulatory site

    A content analysis of thinspiration, fitspiration, and bonespiration imagery on social media

    Background On social media, images such as thinspiration, fitspiration, and bonespiration, are shared to inspire certain body ideals. Previous research has demonstrated that exposure to these groups of content is associated with increased body dissatisfaction and decreased self-esteem. It is therefore important that the bodies featured within these groups of content are more fully understood so that effective interventions and preventative measures can be informed, developed, and implemented. Method A content analysis was conducted on a sample of body-focussed images with the hashtags thinspiration, fitspiration, and bonespiration from three social media platforms. Results The analyses showed that thinspiration and bonespiration content contained more thin and objectified bodies, compared to fitspiration which featured a greater prevalence of muscles and muscular bodies. In addition, bonespiration content contained more bone protrusions and fewer muscles than thinspiration content. Conclusions The findings suggest fitspiration may be a less unhealthy type of content; however, a subgroup of imagery was identified which idealised the extremely thin body type and as such this content should also be approached with caution. Future research should utilise qualitative methods to further develop understandings of the body ideals that are constructed within these groups of content and the motivations behind posting this content

    Quantitative assessment of microbicide-induced injury in the ovine vaginal epithelium using confocal microendoscopy

    <p>Abstract</p> <p>Background</p> <p>The development of safe topical microbicides that can preserve the integrity of cervicovaginal tract epithelial barrier is of great interest as this may minimize the potential for increased susceptibility to STI infections. High resolution imaging to assess epithelial integrity in a noninvasive manner could be a valuable tool for preclinical testing of candidate topical agents.</p> <p>Methods</p> <p>A quantitative approach using confocal fluorescence microendoscopy (CFM) for assessment of microbicide-induced injury to the vaginal epithelium was developed. Sheep were treated intravaginally with one of five agents in solution (PBS; 0.02% benzalkonium chloride (BZK); 0.2% BZK) or gel formulation (hydroxyethyl cellulose (HEC); Gynol II nonoxynol-9 gel (N-9)). After 24 hours the vaginal tract was removed, labeled with propidium iodide (PI), imaged, then fixed for histology. An automated image scoring algorithm was developed for quantitative assessment of injury and applied to the data set. Image-based findings were validated with histological visual gradings that describe degree of injury and measurement of epithelial thickness.</p> <p>Results</p> <p>Distinct differences in PI staining were detected following BZK and N-9 treatment. Images from controls had uniformly distributed nuclei with defined borders, while those after BZK or N-9 showed heavily stained and disrupted nuclei, which increased in proportion to injury detected on histology. The confocal scoring system revealed statistically significant scores for each agent versus PBS controls with the exception of HEC and were consistent with histology scores of injury.</p> <p>Conclusions</p> <p>Confocal microendoscopy provides a sensitive, objective, and quantitative approach for non-invasive assessment of vaginal epithelial integrity and could serve as a tool for real-time safety evaluation of emerging intravaginal topical agents.</p
