The role of ATF2 in insulin action

    Get PDF
We have identified ATF2 as a component of the cellular and in vivo insulin signaling systems. Insulin induced ATF2-phosphorylation in A14 fibroblasts, 3T3L1-adipocytes and several mouse tissues in vivo. In cell lines, the insulin-induced ATF2-phosphorylation was dependent on cooperation between two Ras-dependent MAPK-pathways: ERK and p38/JNK. Analysis of several described ATF2-target genes identified insulin-induced expression of Egr1, ATF3, c-jun and SREBP1c as being ATF2-dependent in cell lines. These genes encode transcription factors whose targets have been postulated to be involved in several aspects of normal insulin action, like control of hepatic fat and glucose metabolism and proliferation and differentiation of beta-cells. Quantitative PCR analysis showed increased mRNA expression of these genes in mouse livers correlating with hepatic ATF2-phosphorylation induced by insulin, but also in response to HFD-induced insulin resistance. In addition, the known ATF2 target genes, the inflammatory cytokines IL1-beta and TNF-alpha were also significantly induced by the HFD in liver. Although the elucidation of the exact role of ATF2 activation under these conditions needs further experimentation, the data presented in this thesis suggest a potential dual function for ATF2 as a mediator of insulin action on the one hand and a putative regulator of the development or maintenance of insulin resistance and/or diabetes-associated complications on the other.Dutch Diabetes Research Foundation (DFN), J.E. Acute necrotizing pancreatitis in rats

    Get PDF
The specific aim of the present study was to investigate whether eicosanoids play a role in acute necrotizing pancreatitis. Because of the limited number of patients with acute pancreatitis admitted to the hospital each year, as well as the practical difficulties encountered in studying these patients, we had to turn to an experimental model of acute necrotizing pancreatitis. Experimental pancreatitis has been known since 1856 (9) when Claude Bernard injected a mixture of bile and oliYe oil into the pancreatic duct of a dog. We used a model based on the introduction of a bile salt into the pancreatic ducts of rats

A morphological algorithm for improving radio-frequency interference detection

    Get PDF
A technique is described that is used to improve the detection of radio-frequency interference in astronomical radio observatories. It is applied on a two-dimensional interference mask after regular detection in the time-frequency domain with existing techniques. The scale-invariant rank (SIR) operator is defined, which is a one-dimensional mathematical morphology technique that can be used to find adjacent intervals in the time or frequency domain that are likely to be affected by RFI. The technique might also be applicable in other areas in which morphological scale-invariant behaviour is desired, such as source detection. A new algorithm is described, that is shown to perform quite well, has linear time complexity and is fast enough to be applied in modern high resolution observatories. It is used in the default pipeline of the LOFAR observatory.Comment: Accepted for publication in A&

Photon Orbital Angular Momentum and Mass in a Plasma Vortex

We analyse the Anderson-Higgs mechanism of photon mass acquisition in a plasma and study the contribution to the mass from the orbital angular momentum acquired by a beam of photons when it crosses a spatially structured charge distribution. To this end we apply Proca-Maxwell equations in a static plasma with a particular spatial distribution of free charges, notably a plasma vortex, that is able to impose orbital angular momentum (OAM) onto light. In addition to the mass acquisition of the conventional Anderson-Higgs mechanism, we find that the photon acquires an additional mass from the OAM and that this mass reduces the Proca photon mass.Comment: Four pages, no figures. Transforming towards sustainable health and wellbeing systems:Eight guiding principles based on the experiences of nine Dutch Population Health Management initiatives

Introduction Population Health Management initiatives are increasingly introduced, aiming to develop towards sustainable health and wellbeing systems. Yet, little is known about which strategies to implement during this development. This study provides insights into which strategies are used, why, and when, based on the experiences of nine Dutch Population Health Management initiatives. Methods The realist evaluation approach was used to gain an understanding of the relationships between context, mechanisms and outcomes when Population Health Management strategies were implemented. Data were retrieved from three interview rounds (n = 207) in 2014, 2016 and 2017. Data was clustered into guiding principles, underpinned with strategy-context-mechanism-outcome configurations. Results The Dutch initiatives experienced different developments, varying between immediate large-scale collaborations with eventual relapse, and incremental growth towards cross-sector collaboration. Eight guiding principles for development towards health and wellbeing systems were identified, focusing on: 1. Shared commitment for a Population Health Management-vision; 2. Mutual understanding and trust; 3. Accountability; 4. Aligning politics and policy; 5. Financial incentives; 6. A learning cycle based on a data-infrastructure; 7. Community input and involvement; and 8. Stakeholder representation and leadership. Conclusion Development towards a sustainable health and wellbeing system is complex and time-consuming. Its success not only depends on the implementation of all eight guiding principles, but is also influenced by applying the right strategies at the right moment in the development

Photon orbital angular momentum in a plasma vortex

We study theoretically the exchange of angular momentum between a photon beam and a plasma vortex, and demonstrate the possible excitation of photon angular momentum states in a plasma. This can be relevant to laboratory and space plasma diagnostics; radio astronomy self-calibration; and generating photon angular momentum beams. A static plasma perturbation with helical structure, and a rotating plasma vortex are studied in detail and a comparison between these two cases, and their relevance to the physical nature of photon OAM, is established.Comment: 4 pages, submitted to Phys. Population health management guiding principles to stimulate collaboration and improve pharmaceutical care

    Get PDF
Purpose: A range of strategies to improve pharmaceutical care has been implemented by population health management (PHM) initiatives. However, which strategies generate the desired outcomes is largely unknown. The purpose of this paper is to identify guiding principles underlying collaborative strategies to improve pharmaceutical care and the contextual factors and mechanisms through which these principles operate. Design/methodology/approach: The evaluation was informed by a realist methodology examining the links between PHM strategies, their outcomes and the contexts and mechanisms by which these strategies operate. Guiding principles were identified by grouping context-specific strategies with specific outcomes. Findings: In total, ten guiding principles were identified: create agreement and commitment based on a long-term vision; foster cooperation and representation at the board level; use layered governance structures; create awareness at all levels; enable interpersonal links at all levels; create learning environments; organize shared responsibility; adjust financial strategies to market contexts; organize mutual gains; and align regional agreements with national policies and regulations. Contextual factors such as shared savings influenced the effectiveness of the guiding principles. Mechanisms by which these guiding principles operate were, for instance, fostering trust and creating a shared sense of the problem. Practical implications: The guidin
