    The defective behaviour of an 8T SRAM cell with open defects is analyzed. Full and resistive open defects have been considered in the electrical characterization of the defective cell. Due to the similarity between the classical 6T SRAM cell and the 8T cell, only defects affecting the read port transistors have been considered. In the work, it is shown how an open in a defective cell may influence the correct operation of a victim cell sharing the same read circuitry. Also, it is shown that the sequence of bits written on the defective cell prior to a read action can mask the presence of the defect.     Safety Profile and Tolerability of Topical Phosphodiesterase 4 Inhibitors for the Treatment of Atopic Dermatitis: A Systematic Review and Meta-Analysis

    Evaluate the safety profile and tolerability of topical phosphodiesterase 4 (PDE4) inhibitors versus vehicle as treatment for atopic dermatitis in published studies.A search was performed in Medline/PubMed, Web of Science, and Cochrane Library databases on September 27, 2021, by 1 evaluator, without restrictions on publication dates or languages. Terms such as atopic dermatitis, phosphodiesterase 4 inhibitors, calcineurin inhibitors, and randomized controlled trials were included. The database searches were carried out by 1 evaluator. The titles and abstracts were reviewed for the identification and evaluation of potentially eligible studies. Study selection was made by two reviewers, so there was no intra-examiner statistic at the study selection step. The full-text articles were reviewed to determine whether or not they would be included in the systematic review. Global analyses, which included studies with both unclear and low risk of bias and subanalyses of studies with a low risk of bias were performed.Out of 237 identified articles, 14 clinical trials were included in the meta-analysis. In global analyses of studies with low and unclear risk of bias, topical treatment with PDE4 inhibitors did not differ from vehicle treatment in global treatment emergent adverse events (relative risk?=?0.99; 95% CI, 0.87-1.14; P?=?0.94) or in serious emergent adverse events appearance (relative risk?=?0.92; 95% CI, 0.39-2.20; P?=?0.86). In subanalyses of studies with a low risk of bias, a reduced rate of atopic dermatitis exacerbation was observed in PDE4 inhibitors compared with the vehicle (relative risk?=?0.62; 95% CI, 0.39-0.98; P?=?0.04) and risk of pain at the application site was confirmed (relative risk?=?2.59; 95% CI, 1.27-5.28; P?=?0.01).PDE4 inhibitors did not show differences from vehicle treatment in treatment emergent adverse events or serious emergent adverse events incidence. In studies with low risk of bias, PDE4 inhibitors had a statistically significant risk of producing pain and reduced occurrence of atopic dermatitis exacerbation.© 2022 The Author(s)

    Variability in the meridional overturning circulation at 32°S in the Pacific Ocean diagnosed by inverse box models

    The meridional circulation and transport at 32°S in the Pacific Ocean in 1992 and 2017 are compared with analogous data from 2003 and 2009 computed by Hernández-Guerra and Talley (2016). The hydrographic data come from the GO-SHIP database and an inverse box model has been applied with similar constraints as in Hernández-Guerra and Talley (2016). In 1992, 2003 and 2017 the pattern of the overturning streamfunction and circulation are similar, but in 2009 the pattern of the circulation changes in the whole water column. The horizontal distribution of mass transports at all depths in 1992 and 2017 resembles the familiar shape of the “classical gyre” also observed in 2003 and is notably different to the “bowed gyre” found in 2009. The hydrographic data have been compared with data obtained from the numerical modelling outputs of ECCO, SOSE, GLORYS, and MOM. Results show that none of these models properly represents the “bowed gyre” circulation in 2009, and this change in circulation pattern was not observed during the entire length of model simulations. Additionally, the East Australian Current in the western boundary presents higher mass transport in the hydrographic data than in any numerical modelling output. Its poleward mass transport ranges from −35.1 ± 2.0 Sv in 1992 to −54.3 ± 2.6 Sv in 2003. Conversely, the Peru-Chile Current is well represented in models and presents an equatorward mass transport from 2.3 ± 0.8 Sv in 2009 to 4.4 ± 1.0 Sv in 1992. Furthermore, the Peru-Chile Undercurrent presents a more intense poleward mass transport in 2009 (−3.8 ± 1.2 Sv). In addition, the temperature and freshwater transports in 1992 (0.42 ± 0.12 PW and 0.26 ± 0.08 Sv), 2003 (0.38 ± 0.12 PW and 0.25 ± 0.02 Sv), and 2017 (0.42 ± 0.12 PW and 0.34 ± 0.08 Sv) are similar, but significantly different from those in 2009 (0.16 ± 0.12 PW and 0.50 ± 0.03 Sv, respectively).     Thymidine Kinase 2 Deficiency-Induced Mitochondrial DNA Depletion Causes Abnormal Development of Adipose Tissues and Adipokine Levels in Mice

    Mammal adipose tissues require mitochondrial activity for proper development and differentiation. The components of the mitochondrial respiratory chain/oxidative phosphorylation system (OXPHOS) are encoded by both mitochondrial and nuclear genomes. The maintenance of mitochondrial DNA (mtDNA) is a key element for a functional mitochondrial oxidative activity in mammalian cells. To ascertain the role of mtDNA levels in adipose tissue, we have analyzed the alterations in white (WAT) and brown (BAT) adipose tissues in thymidine kinase 2 (Tk2) H126N knockin mice, a model of TK2 deficiency-induced mtDNA depletion. We observed respectively severe and moderate mtDNA depletion in TK2-deficient BAT and WAT, showing both tissues moderate hypotrophy and reduced fat accumulation. Electron microscopy revealed altered mitochondrial morphology in brown but not in white adipocytes from TK2-deficient mice. Although significant reduction in mtDNA-encoded transcripts was observed both in WAT and BAT, protein levels from distinct OXPHOS complexes were significantly reduced only in TK2-deficient BAT. Accordingly, the activity of cytochrome c oxidase was significantly lowered only in BAT from TK2-deficient mice. The analysis of transcripts encoding up to fourteen components of specific adipose tissue functions revealed that, in both TK2-deficient WAT and BAT, there was a consistent reduction of thermogenesis related gene expression and a severe reduction in leptin mRNA. Reduced levels of resistin mRNA were found in BAT from TK2-deficient mice. Analysis of serum indicated a dramatic reduction in circulating levels of leptin and resistin. In summary, our present study establishes that mtDNA depletion leads to a moderate impairment in mitochondrial respiratory function, especially in BAT, causes substantial alterations in WAT and BAT development, and has a profound impact in the endocrine properties of adipose tissues

    CIBERER : Spanish national network for research on rare diseases: A highly productive collaborative initiative

This innovative research structure comprises 11 different specific areas dedicated to the main public health priorities in the National Health System. CIBERER, the thematic area of CIBER focused on rare diseases (RDs) currently consists of 75 research groups belonging to universities, research centers, and hospitals of the entire country. CIBERER's mission is to be a center prioritizing and favoring collaboration and cooperation between biomedical and clinical research groups, with special emphasis on the aspects of genetic, molecular, biochemical, and cellular research of RDs. This research is the basis for providing new tools for the diagnosis and therapy of low-prevalence diseases, in line with the International Rare Diseases Research Consortium (IRDiRC) objectives, thus favoring translational research between the scientific environment of the laboratory and the clinical setting of health centers. In this article, we intend to review CIBERER's 15-year journey and summarize the main results obtained in terms of internationalization, scientific production, contributions toward the discovery of new therapies and novel genes associated to diseases, cooperation with patients' associations and many other topics related to RD research

    Crise de abastecimento de água em São Paulo e falta de planejamento estratégico

    Embora a crise no abastecimento de água na Região Metropolitana de São Paulo (RMSP) tenha se manifestado de maneira mais intensa no verão de 2013-2014, ela revela um problema crônico que vem afetando toda a Região nos últimos dez anos. Esse problema foi gerado pela falta de um planejamento estratégico que considere questões climatológicas que podem indicar, com meses de antecedência, problemas de recomposição dos níveis dos mananciais, permitindo que ações sejam empreendidas com razoável antecedência, reduzindo os impactos para a população. Este estudo mostra como é possível utilizar informações climáticas na gestão estratégica do sistema de abastecimento da RMSP.Though the crisis in the water supplying system in the Metropolitan Region of São Paulo (RMSP) was more intensively felt in the 2013-2014 summer, it reveals a chronic problem that has been affecting the whole RMSP for the past ten years. This problem is originated from the lack of a strategic planning that takes into consideration climate issues that could, months before, foresee problems to restore the levels of water resources, allowing measures to be implemented within a reasonable anticipation, therefore reducing the impacts on the population. This study shows how it is possible to use climate information in the strategic management of the water supply in the RMSP
