    Sdg 6, water management and sustainable practices: insights from the cusio-valsesia taps and valves industrial district in Italy

    Water is a vital resource, but there are only a few studies on how to integrate it into corporate sustainability management. This article analyses the relationship between business and sustainability practices with the aim of integrating the challenges posed by the water-related SDG 6 and all other interlinked Sustainable Development Goals (SDGs) of the 2030 Agenda. Through out qualitative research, the tap and valve district of Cusio-Valsesia in Italy was investigated. Al though participating firms showed little knowledge of the challenges posed by the United Nations, the insights from their practices, even if poorly formalised and structured, made it possible to identify "silent SDGs". Therefore, companies, if duly incentivised, are not far from understanding and engaging with the SDGs in a formalized manner

    Digital transformation in healthcare: Analyzing the current state-of-research

    Abstract Digital transformation in healthcare is of increasing relevance for both scholars and practitioners in the field. Our article attempts to assess the research question how multiple stakeholders implement digital technologies for management and business purposes. To answer this question, we perform a systematic literature review about the state of the art of digital transformation in healthcare. Our findings show that prior research falls into five clusters: operational efficiency by healthcare providers; patient-centered approaches; organizational factors and managerial implications; workforce practices; and socio-economic aspects. These clusters are linked together into a model showing how these various forms of technology implementation lead to operational efficiencies for services providers. Various directions for future research and management implications are offered

    Microbiological characteristics of poultry meats - Results of inspections carried out in the province of Milano, Italy

    Examinations were conducted in terms of microbiological quality/quantity (TMC, Coliforms, E. coli, S. aureus, Sulphate-reducing Clostridia, B. cereus, Salmonella spp. and Lysteria spp. and Campylobacter spp.) on 240 poultry meat samples (chicken, turkey and quail) acquired pursuant to the standards set by the Regional Plan of programming and coordination in the field of operations concerning official inspections of Lombardia animal origin and by a few private companies for self-inspection. The TMC was consistently low and in line with reports in the literature, as was the case with coliforms, E. coli, S. aureus, sulphatereducing Clostrides and B. cereus. In the case of Salmonella spp., only 5 samples tested positive: one for S. typhimurium and one for S. enteritidis (chicken); only one sample from turkey tested positive for S. blokley, and two out of five samples analysed from quail tested positive result for S. typhimurium. About 3% of the samples analyzed tested positive for Listeria monocitogenes, but they were within the legal limits. Research on Campylobacter thermophiles has involved only 50 samples, of which only 5 have tested positive. These results confirm the high quality of hygiene and cleanliness of poultry meat, in accordance with that reported in the national literature and with respect to EU norms

    Transcriptional regulation of lipid metabolism and inflammation in transition dairy goats by fish oil and stearate

    To better understand the interaction between saturated or unsaturated fatty acids and its effect onexpression of genes involved in subcutaneous adipose tissue metabolism, 23 second parity alpine dairygoats were enrolled in the experiment and fed either a non fat-supplemented basal diet (C; n=8), thebasal diet supplemented with stearic acid (ST; n=7) or the basal diet supplemented with fish oil (FO;n=8). 30g/head/d supplemental fatty acids during the dry period and 50g/head/d during lactation weredelivered starting one week before parturition up to 21 days in milk. Subcutaneous adipose tissuesamples were harvested at day -7, 7 and 21 relative to kidding and mRNA levels of genes involved ininflammation were measured via qPCR. Data were analyzed using the MIXED procedure of SAS. Nosignificant effects for treatment were observed, however eight genes were significant for time. HP andSAA3 expression peaked at day 7 postpartum, to then return at prepartum level around 21 d relative tokidding, while IL8, IL10, and IL18 expression constantly increased along the transition period. Vice versa,expression of IL1β, IL6R, and RXRA decreased in response to kidding, with a subsequent increase at day21. The obtained results led us to hypothesize that goats face a postponed lipomobilization afterkidding, probably related to their reduced production. The next step will involve the analysis of miRNArelated to immune cell infiltration, adipocyte inflammation and lipolysis and positive regulation ofadipogenesis to better understand the complex network of lipid metabolism in periparturient goats

    Antidiabetic thiazolidinediones induce ductal differentiation but not apoptosis in pancreatic cancer cells

    AIM: Thiazolidinediones (TZD) are a new class of oral antidiabetic drugs that have been shown to inhibit growth of same epithelial cancer cells. Although TZD were found to be ligands for peroxisome proliferator-activated receptor gamma (PPARgamma), the mechanism by which TZD exert their anticancer effect is presently unclear. In this study, we analyzed the mechanism by which TZD inhibit growth of human pancreatic carcinoma cell lines in order to evaluate the potential therapeutic use of these drugs in pancreatic adenocarcinoma. METHODS: The effects of TZD in pancreatic cancer cells were assessed in anchorage-independent growth assay. Expression of PPARgamma was measured by reverse-transcription polymerase chain reaction and confirmed by Western blot analysis. PPARgamma activity was evaluated by transient reporter gene assay. Flow cytometry and DNA fragmentation assay were used to determine the effect of TZD on cell cycle progression and apoptosis respectively. The effect of TZD on ductal differentiation markers was performed by Western blot. RESULTS: Exposure to TZD inhibited colony formation in a PPARgamma-dependent manner. Growth inhibition was linked to G1 phase cell cycle arrest through induction of the ductal differentiation program without any increase of the apoptotic rate. CONCLUSION: TZD treatment in pancr

    Differentiating Vogt-Koyanagi-Harada syndrome from recurrent optic neuritis: a case report and review of the literature concerning Hispanic patients

    Abstract Background First recognized at the beginning of twentieth century and named after three authors who independently described some affected patients, Vogt-Koyanagi-Harada syndrome is a rare multisystemic autoimmune disease targeting melanin-containing tissues of the eye, meninges, inner ear and skin. It predominantly affects Asian people, but also people with darker skin pigmentation such as Native Americans and Hispanics (Mestizos), whose ancestors moved from Asia across the Bering strait to North America and further down to Central and South America. Heterogenous presentation is observed, especially among different ethnic groups. Here we describe the case of an Hispanic South American patient presenting with multiple visual relapses and thus mimicking recurrent optic neuritis; we provide insights into the differential diagnosis and a brief review of the literature concerning the epidemiology of Vogt-Koyanagi-Harada syndrome in Hispanic patients compared with other ethnic groups. Case presentation A 34-year-old Ecuadorian woman presented over years with multiple relapses involving the visual system. She was investigated in both neurologic and ophthalmic clinical settings. Brain Magnetic Resonance Imaging, cerebrospinal fluid examination, Spectral Domain Optical Coherence Tomography and Fluorescein Angiography were performed. She was misdiagnosed first as an optic neuritis pointing to a demyelinating disorder, then as a posterior scleritis. Due to the protean manifestations of Vogt-Koyanagi-Harada syndrome and the incomplete clinical presentation at the beginning, the right diagnosis was made only at a later disease stage using retrospective criteria. Conclusions Hispanic patients often present without extraocular symptoms in early phases of the disease and they have globally lower rates of intertegumentary signs compared to Asian patients. The diagnosis of a multisystemic disease such as Vogt-Koyanagi-Harada syndrome is a challenge involving specialists operating in different medical fields; especially in urban multiethnic populations, rare etiologies of common symptoms have to be taken into account when performing a differential diagnosis

    Different ataxin-3 amyloid aggregates induce intracellular Ca2+ deregulation by different mechanisms in cerebellar granule cells

    AbstractThis work aims at elucidating the relation between morphological and physicochemical properties of different ataxin-3 (ATX3) aggregates and their cytotoxicity. We investigated a non-pathological ATX3 form (ATX3Q24), a pathological expanded form (ATX3Q55), and an ATX3 variant truncated at residue 291 lacking the polyQ expansion (ATX3/291Δ). Solubility, morphology and hydrophobic exposure of oligomeric aggregates were characterized. Then we monitored the changes in the intracellular Ca2+ levels and the abnormal Ca2+ signaling resulting from aggregate interaction with cultured rat cerebellar granule cells. ATX3Q55, ATX3/291Δ and, to a lesser extent, ATX3Q24 oligomers displayed similar morphological and physicochemical features and induced qualitatively comparable time-dependent intracellular Ca2+ responses. However, only the pre-fibrillar aggregates of expanded ATX3 (the only variant which forms bundles of mature fibrils) triggered a characteristic Ca2+ response at a later stage that correlated with a larger hydrophobic exposure relative to the two other variants. Cell interaction with early oligomers involved glutamatergic receptors, voltage-gated channels and monosialotetrahexosylganglioside (GM1)-rich membrane domains, whereas cell interaction with more aged ATX3Q55 pre-fibrillar aggregates resulted in membrane disassembly by a mechanism involving only GM1-rich areas. Exposure to ATX3Q55 and ATX3/291Δ aggregates resulted in cell apoptosis, while ATX3Q24 was substantially innocuous. Our findings provide insight into the mechanisms of ATX3 aggregation, aggregate cytotoxicity and calcium level modifications in exposed cerebellar cells

    Toxoplasma gondii in small ruminants in Northern Italy – prevalence and risk factors

    [b]Objective[/b]. The aim of the survey was to evaluate [i]Toxoplasma gondii[/i] seroprevalence in small ruminants and possible risk factors associated with the infection. [b]Materials and methods[/b]. Sera from 474 goats and 502 sheep reared on 42 farms in northern Italy were collected and tested for IgG antibodies to [i]T. gondii[/i] by IFAT (indirect immunofluorescence antibody test). To identify risk factors, a binary logistic regression analysis of the variables was performed. An audit form about farm management was used. [b]Results[/b]. Antibodies to [i]T. gondii [/i]were found in 96.6% of goat farms and in 87.5% of sheep farms; 41.7% goats and 59.3% sheep resulted positive. Seroprevalence was significantly higher in sheep than in goats. Seroprevalence values were similar in goats from eastern and western areas, whereas goats from the southern area were at lower risk of infection. Saanen goats presented the lowest seroprevalence (30.7 %), whereas cross-breed exhibited the highest rate (48.7%). Goats from farms housing both sheep and goats had an infection risk 1.39 times higher than goats from farms that did not house sheep. Animals bred on intensive farms showed lower prevalence (22.1%) in comparison with those from extensive (45.6%) or semi-intensive farms (60%). Sampling area was one of the strongest predictors of [i]T. gondii [/i]infection in sheep flocks. Transhumant flocks showed a higher risk of infection by [i]T. gondii[/i] compared with semi-intensive farms (66.8% vs. 38.4%). [b]Conclusions[/b]. The highest [i]T. gondii[/i] seroprevalence values were registered in transhumant flocks of sheep and in family businesses rearing goats. As these traditional activities represent an important resource for the conservation of the territory and its economy, management practices for a better control of the disease should be improved
