    Pharmaceuticals in industrial wastewater and their removal using photo-Fenton’s oxidation

    Active pharmaceutical ingredients are known contaminants of surface and ground water. In some cases these are persistent organic chemicals which are only partially eliminated during conventional wastewater treatment. They have been detected in the effluent of various European wastewater treatment plants. However, there is a growing body of evidence to suggest that discharges from pharmaceutical plants themselves can contribute to the levels of pharmaceuticals in the environment. This project is based on both the detection and removal of pharmaceuticals from industrial sources. A SPE-LC-MS/MS method for the detection of famotidine, tamsulosin hydrochloride and solifenacin succinate in wastewater at a pharmaceutical production facility was developed and validated. The results of a six month sampling programme showed substantial concentrations of the analytes in both influent and effluent. Famotidine was detected at an average concentration of 1.6 mg/L and 2.6mg/L, tamsulosin hydrochloride at 5 μg/L and 4 μg/L and solifenacin succinate at 39 μg/L and 28 μg/L in influent and effluent respectively. Photo-Fenton’s oxidation conditions were optimised for the removal of each of the three APIs from aqueous solutions. Intermediates and final products of the degradation have been identified via LC-MS

    Alleviating health risks associated with rainwater harvesting

    Perceived and real public health risks associated with the quality of water from alternative water sources and supply systems, such as rainwater harvesting (RWH) and grey water reuse, continue to restrict their uptake in many countries. One option to alleviate these health risks is to treat alternative water to potable standard at the point of use (POU) as opposed to the point of supply, as undertaken in centralised systems. This paper presents the results of three international empirical field trials of a novel POU RWH treatment device. The results indicate that where the harvested rainwater did not contain elevated levels of pesticides or physico-chemical determinands, the POU device was able to reduce levels in outlet water to meet UK, EU and World Health Organization potable standards. Regarding microbiological determinands, such as total viable counts and coliforms, and microbial pathogens, such as Pseudomonas aeruginosa and Legionella spp., the device achieved reduction to potable standard and full pathogen removal, respectively. Thus, while it is possible to treat harvested rainwater to potable standard with a POU device, whether it is desirable to do so to alleviate risks for all end uses remains a question for further debate

    The availability of public genomic resources can greatly assist biodiversity assessment, conservation, and restoration efforts by providing evidence for scientifically informed management decisions. Here we survey the main approaches and applications in biodiversity and conservation genomics, considering practical factors, such as cost, time, prerequisite skills, and current shortcomings of applications. Most approaches perform best in combination with reference genomes from the target species or closely related species. We review case studies to illustrate how reference genomes can facilitate biodiversity research and conservation across the tree of life.     Bi-allelic Loss-of-Function CACNA1B Mutations in Progressive Epilepsy-Dyskinesia.

    The occurrence of non-epileptic hyperkinetic movements in the context of developmental epileptic encephalopathies is an increasingly recognized phenomenon. Identification of causative mutations provides an important insight into common pathogenic mechanisms that cause both seizures and abnormal motor control. We report bi-allelic loss-of-function CACNA1B variants in six children from three unrelated families whose affected members present with a complex and progressive neurological syndrome. All affected individuals presented with epileptic encephalopathy, severe neurodevelopmental delay (often with regression), and a hyperkinetic movement disorder. Additional neurological features included postnatal microcephaly and hypotonia. Five children died in childhood or adolescence (mean age of death: 9 years), mainly as a result of secondary respiratory complications. CACNA1B encodes the pore-forming subunit of the pre-synaptic neuronal voltage-gated calcium channel Cav2.2/N-type, crucial for SNARE-mediated neurotransmission, particularly in the early postnatal period. Bi-allelic loss-of-function variants in CACNA1B are predicted to cause disruption of Ca2+ influx, leading to impaired synaptic neurotransmission. The resultant effect on neuronal function is likely to be important in the development of involuntary movements and epilepsy.     A SPE-LC-MS/MS method for the detection of low concentrations of pharmaceuticals in industrial waste streams

    A SPE-LC-MS/MS method was developed and validated for the determination of three active pharmaceutical ingredients in the wastewater of a chemical synthesis production facility. The SPE-LC-MS/MS method was validated in actual influent and effluent samples. Linearity, LOD, LOQ, repeatability, intermediate precision, and recovery were determined. The LC-MS/MS method was used for an on-going monitoring program for these pharmaceuticals in wastewater. The method development techniques, validation procedures, and results from real wastewater samples are presented in this paper
