    Biosignatures in chimney structures and sediment from the Loki's Castle low-temperature hydrothermal vent field at the Arctic Mid-Ocean Ridge

    We investigated microbial life preserved in a hydrothermally inactive silica-barite chimney in comparison with an active barite chimney and sediment from the Loki's Castle low-temperature venting area at the Arctic Mid-Ocean Ridge (AMOR) using lipid biomarkers. Carbon and sulfur isotopes were used to constrain possible metabolic pathways. Multiple sulfur (δ34S, ∆33S) isotopes on barite over a cross section of the extinct chimney range between 21.1 and 22.5‰ in δ34S, and between 0.020 and 0.034‰ in Δ33S, indicating direct precipitation from seawater. Biomarker distributions within two discrete zones of this silica-barite chimney indicate a considerable difference in abundance and diversity of microorganisms from the chimney exterior to the interior. Lipids in the active and inactive chimney barite and sediment were dominated by a range of 13C-depleted unsaturated and branched fatty acids with δ13C values between −39.7 and −26.7‰, indicating the presence of sulfur-oxidizing and sulfate-reducing bacteria. The majority of lipids (99.5%) in the extinct chimney interior that experienced high temperatures were of archaeal origin. Unusual glycerol monoalkyl glycerol tetraethers (GMGT) with 0-4 rings were the dominant compounds suggesting the presence of mainly (hyper-) thermophilic archaea. Isoprenoid hydrocarbons with δ13C values as low as −46‰ also indicated the presence of methanogens and possibly methanotrophs

    Upper ocean temperature characteristics in the subantarctic Southeast Pacific based on biomarker reconstructions

    Alkenones and Glycerol Dialkyl Glycerol Tetraether lipids (GDGT) as remnants of living organisms are widely used biomarkers for determining past oceans&rsquo; water temperatures. The organisms these proxy carriers stem from, are influenced by a number of environmental parameters, such as water depth, nutrient availability, light conditions or seasonality, which all may significantly bias the calibration to ambient water temperatures. Reliable temperature determinations remain thus challenging, especially in higher latitudes and for under-sampled regions. We analyzed 33 sediment surface samples from the Southern Chilean continental margin and the Drake Passage for alkenones and GDGTs and compared the results with gridded instrumental reference data from the World Ocean Atlas 2005 (WOA05), as well as previously published data from an extended study area covering the Central and Western South Pacific towards the New Zealand continental margin. We show that for alkenone-derived SSTs, the widely-used global core-top calibration of M&uuml;ller et al. (1998) yields the smallest residuals, whereas the calibration of Sikes et al. (1997), adapted to higher latitudes and supposed to show summer SSTs, overestimates modern WOA05-based (summer and annual mean) SSTs. Our alkenone SSTs show a slight seasonal shift of ~1 &deg;C at the Southern Chilean Margin and up to ~2 &deg;C in the Drake Passage towards austral summer SSTs, whereas samples in the Central South Pacific reflect an annual mean signal. We show that for GDGT-based temperatures, a more complex pattern emerges. In areas north of the Subantarctic Front (SAF) the subsurface calibration of Kim et al. (2012a) best reflects temperatures from the WOA05, largely within the margin error of &plusmn;2.2 &deg;C. Temperatures south of the SAF instead are significantly overestimated by up to 14 &deg;C, irrespective of the applied calibration. Based on a qualitative assessment of the GDGT [2] / [3]-ratios, which likely indicate water depth of origin, our samples reflect a subsurface (0 to 200 m water depth) rather than a surface (0&ndash;50 m water depth) signal. The overestimation of surface and subsurface temperatures south of the SAF highlights the need for a re-assessment of existing calibrations in the polar Southern Ocean, and leads to limitations in reliably both obtaining absolute values and assessing relative changes. Therefore, we suggest a modified Southern Ocean calibration for surface and subsurface GDGT-based temperatures, which shows a lower temperature sensitivity of the TEXL86 and yields principally lower absolute temperatures, which align more closely with WOA05-derived values.</p

    Microbial methane cycling in sediments of Arctic thermokarst lagoons

    Thermokarst lagoons represent the transition state from a freshwater lacustrine to a marine environment, and receive little attention regarding their role for greenhouse gas production and release in Arctic permafrost landscapes. We studied the fate of methane (CH4) in sediments of a thermokarst lagoon in comparison to two thermokarst lakes on the Bykovsky Peninsula in northeastern Siberia through the analysis of sediment CH4 concentrations and isotopic signature, methane-cycling microbial taxa, sediment geochemistry, lipid biomarkers, and network analysis. We assessed how differences in geochemistry between thermokarst lakes and thermokarst lagoons, caused by the infiltration of sulfate-rich marine water, altered the microbial methane-cycling community. Anaerobic sulfate-reducing ANME-2a/2b methanotrophs dominated the sulfate-rich sediments of the lagoon despite its known seasonal alternation between brackish and freshwater inflow and low sulfate concentrations compared to the usual marine ANME habitat. Non-competitive methylotrophic methanogens dominated the methanogenic community of the lakes and the lagoon, independent of differences in porewater chemistry and depth. This potentially contributed to the high CH4 concentrations observed in all sulfate-poor sediments. CH4 concentrations in the freshwater-influenced sediments averaged 1.34 ± 0.98 μmol g−1, with highly depleted δ13C-CH4 values ranging from −89‰ to −70‰. In contrast, the sulfate-affected upper 300 cm of the lagoon exhibited low average CH4 concentrations of 0.011 ± 0.005 μmol g−1 with comparatively enriched δ13C-CH4 values of −54‰ to −37‰ pointing to substantial methane oxidation. Our study shows that lagoon formation specifically supports methane oxidizers and methane oxidation through changes in pore water chemistry, especially sulfate, while methanogens are similar to lake conditions

    A life course examination of the physical environmental determinants of physical activity behaviour: A “Determinants of Diet and Physical Activity” (DEDIPAC) umbrella systematic literature review

    Background: Participation in regular physical activity is associated with a multitude of health benefits across the life course. However, many people fail to meet PA recommendations. Despite a plethora of studies, the evidence regarding the environmental (physical) determinants of physical activity remains inconclusive. Objective: To identify the physical environmental determinants that influence PA across the life course. Methods: An online systematic literature search was conducted using MEDLINE, ISI Web of Science, Scopus and SPORTDiscus. The search was limited to studies published in English (January 2004 to April 2016). Only systematic literature reviews (SLRs) and meta-analyses (MAs) of observational studies, that investigated the association between physical determinants and physical activity outcomes, were eligible for inclusion. The extracted data were assessed on the importance of determinants, strength of evidence and methodological quality. Results: The literature search identified 28 SLRs and 3 MAs on 67 physical environmental characteristics potentially related to physical activity that were eligible for inclusion. Among preschool children, a positive association was reported between availability of backyard space and outdoor toys/equipment in the home and overall physical activity. The availability of physical activity programs and equipment within schools, and neighbourhood features such as pedestrian and cyclist safety structure were positively associated with physical activity in children and adolescents. Negative street characteristics, for example, lack of sidewalks and streetlights, were negatively associated with physical activity in adults. Inconsistent associations were reported for the majority of reviewed determinants in adults. Conclusion: This umbrella SLR provided a comprehensive overview of the physical environment determinants of physical activity across the life course and has highlighted, particularly amongst youth, a number of key determinants that may be associated with overall physical activity. Given the limited evidence drawn mostly from cross-sectional studies, longitudinal studies are needed to further explore these associations. Registration: PROSPERO CRD42015010616

Knowing factors associated with PA might help reducing the disease and economic burden caused by low activity. Studies suggest that socio-cultural factors may affect PA, but systematic overviews of findings across the life course are scarce. This umbrella systematic literature review (SLR) summarizes and evaluates available evidence on socio-cultural determinants of PA in children, adolescents, and adults. Methods: This manuscript was drafted following the recommendations of the \u27Preferred Reporting Items for Systematic reviews and Meta-Analyses\u27 (PRISMA) checklist. The MEDLINE, Web of Science, Scopus, and SPORTDiscus databases were searched for SLRs and meta-analyses (MAs) on observational studies published in English that assessed PA determinants between January 2004 and April 2016. The methodological quality was assessed and relevant information on socio-cultural determinants and any associations with PA was extracted. The available evidence was evaluated based on the importance of potential determinants and the strength of the evidence. Results: Twenty SLRs and three MAs encompassing 657 eligible primary studies investigated potential socio-cultural PA determinants, with predominantly moderate methodological quality. Twenty-nine potential PA determinants were identified that were primarily assessed in children and adolescents and investigated the micro-environmental home/household level. We found probable evidence that receiving encouragement from significant others and having a companion for PA were associated with higher PA in children and adolescents, and that parental marital status (living with partner) and experiencing parental modeling were not associated with PA in children. Evidence for the other potential determinants was limited, suggestive, or non-conclusive. In adults, quantitative and conclusive data were scarce. Conclusions: A substantial number of SLRs and MAs investigating potential socio-cultural determinants of PA were identified. Our data suggest that receiving social support from significant others may increase PA levels in children and adolescents, whereas parental marital status is not a determinant in children. Evidence for other potential determinants was limited. This was mainly due to inconsistencies in results on potential socio-cultural determinants of PA across reviews and studies.     Atmosphere-ocean changes in the Pacific Southern Ocean over the past 1 Million years and implications for global climate

    Atmosphere-ocean interactions play an important role for understanding processes and feedbacks in the Southern Ocean (SO) and are relevant for changes in Antarctic ice-sheets and atmospheric CO2 concentrations. The most important atmospheric forcing at high and mid-latitudes of the Southern Hemisphere is the westerly wind belt (SWW), which strongly affects the strength and extension of the Antarctic Circumpolar Current (ACC), upwelling of deep-water masses, and controls the back-flow of intermediate waters to the tropics. In order to address orbital and millennial-scale changes of the SWW and the ACC, we present sediment proxy records from the Pacific SO including the Chilean Margin and the Drake Passage. The Drake Passage (DP) represents the most important oceanic gateway along the ACC. Based on grain-size and geochemical properties of sediment records from the southernmost continental margin of South America, we reconstruct changes in DP throughflow dynamics over the past 65,000 years. In combination with published sediment records from the Scotia Sea and preliminary sediment records from the central Drake Passage (Polarstern cruise PS97, 2016), we argue for a considerable total reduction of DP transport and reveal an up to ~40% decrease in flow speed along the northernmost ACC pathway entering the DP during glacial times. Superimposed on this long-term decrease are high-amplitude millennial-scale variations, which parallel Southern Ocean and Antarctic temperature patterns. The glacial intervals of strong weakening of the ACC entering the DP imply a reduced Pacific-Atlantic exchange via the DP (“cold-water route”). The reduced Drake Passage glacial throughflow was accompanied by a pronounced northward extension of the Antarctic cold-water sphere in the Southeast Pacific sector and stronger export of northern ACC water into the South Pacific gyre. These oceanographic changes are consistent with reduced SWW within the modern maximum wind strength zone over the subantarctic ACC and reduced wind forcing due to extended sea-ice further south. Despite this reduction in winds in the core of the westerlies, we observe 3-fold higher dust deposition during glacial periods in Past Antarctic Ice Sheet Dynamics (PAIS) Conference September 10-15th 2017, Trieste - Italy the Pacific Southern Ocean (SO). This observation may be explained by a combination of factors including more expanded arid dust source areas in Australia and a northward extent or enhancement of the SWW over Southeast Australia during glacials that would plausibly increase the dust uptake and export into the Pacific SO. Such scenario would imply stronger SWW at the present northernmost margin of the wind belt coeval with weaker core westerlies in the south and reduced ACC strength, including Drake Passage throughflow during glacials. We conclude that changes in DP throughflow play a critical role for the global meridional overturning circulation and interbasin exchange in the Southern Ocean, most likely regulated by variations in the westerly wind field and changes in Antarctic sea-ice extent.     Importance of TLR2 on Hepatic Immune and Non-Immune Cells to Attenuate the Strong Inflammatory Liver Response During Trypanosoma cruzi Acute Infection

    Trypanosoma cruzi, an obligate intracellular protozoan, is the etiological agent of Chagas Disease that represents an important public health burden in Latin America. The infection with this parasite can lead to severe complications in cardiac, liver and gastrointestinal tissue depending on the strain of parasite and host genetics. Recently, we reported a fatal liver injury in T. cruzi infected B6 mice. However, the local immune response against this parasite is poorly understood. This work highlights some of the molecular and cellular mechanisms involved in liver pathology during the acute phase of infection. Using two mouse strains with different genetic backgrounds and responses to infection, B6 and BALB/c, we found that infected B6 mice develop a strong pro-inflammatory environment associated with high TLR9 expression. Conversely, infected BALB/c mice showed a more balanced inflammatory response in liver. Moreover, higher TLR2 and TLR4 expression were found only in hepatocytes from BALB/c. These data emphasize the importance of an adequate integration of signalling between immune and non-immune cells to define the outcome of infection. In addition, the pre-treatment with TLR2-agonist reverts the strong pro-inflammatory environment in T. cruzi infected B6 mice. These results could be useful in the understanding and design of novel immune strategies in controlling liver pathologies

    Behavioural determinants of physical activity across the life course: a "Determinants of Diet and Physical Activity" (DEDIPAC) umbrella literature review

    Background Low levels of physical activity (PA) are a global concern and increasing PA engagement is becoming a priority in current public health policies. Despite the large number of studies and reviews available, the evidence regarding the behavioral determinants of PA is still inconclusive. Thus, the aim of this umbrella systematic literature review (SLR) was to summarize the evidence on the behavioral determinants of PA across the life course. Methods A systematic online search was conducted on MEDLINE, ISI Web of Science, Scopus, and SPORTDiscus databases. The search was limited to studies published in English from January, 2004 to April, 2016. SLRs and meta-analyses (MAs) of observational studies that investigated the behavioral determinants of PA were considered eligible. The extracted data were assessed based on the importance of the determinants, the strength of evidence, and the methodological quality. The full protocol is available from PROSPERO (PROSPERO 2014:CRD42015010616). Results Seventeen reviews on 35 behavioral determinants of PA were eligible for this umbrella SLR. Regardless of age, the most investigated determinants were those related with ‘screen use’ and ‘smoking’. For youth, probable positive evidence emerged for ‘previous PA’ and ‘independent mobility and active transport’ among children and adolescents. For the adult population, ‘transition to university’ and ‘pregnancy/having a child’ showed probable negative associations. Conclusions Although the majority of the evidence was limited and most of the determinants were not associated with PA, this umbrella SLR provided a comprehensive overview of the associations between behavioral determinants and PA. Youth should be physically active in the early years and increase active transportation to/from school, independent mobility, and ‘free-range activities’ without adult supervision, whilst adult PA behaviors are mostly influenced by the life events. Finally, more research is needed that incorporates prospective study designs, standardized definitions of PA, objective measurement methods of PA assessment, and the use of interactionist and mediational approaches for the evaluation of different behavioral determinants influencing PA behaviors
