Combined treatment of graft versus host disease using donor regulatory T cells and ruxolitinib
Donor derived regulatory T lymphocytes and the JAK1/2 kinase inhibitor ruxolitinib are currently
being evaluated as therapeutic options in the treatment of chronic graft versus host disease (cGvHD).
In this work, we aimed to determine if the combined use of both agents can exert a synergistic efect
in the treatment of GvHD. For this purpose, we studied the efect of this combination both in vitro
and in a GvHD mouse model. Our results show that ruxolitinib favors the ratio of thymic regulatory T
cells to conventional T cells in culture, without afecting the suppressive capacity of these Treg. The
combination of ruxolitinib with Treg showed a higher efcacy as compared to each single treatment
alone in our GvHD mouse model in terms of GvHD incidence, severity and survival without hampering
graft versus leukemia efect. This benefcial efect correlated with the detection in the bone marrow of
recipient mice of the infused donor allogeneic Treg after the adoptive transfer

Integrated Multi-Omics Analysis for Inferring Molecular Players in Inclusion Body Myositis
Inclusion body myositis (IBM) is an acquired inflammatory myopathy affecting proximal and distal muscles that leads to weakness in patients over 50. It is diagnosed based on clinical and histological findings in muscle related to inflammation, degeneration, and mitochondria. In relation to IBM, a shortage of validated disease models and a lack of biomarkers and effective treatments constitute an unmet medical need. To overcome these hurdles, we performed an omics analysis of multiple samples from IBM patients (saliva, fibroblasts, urine, plasma, and muscle) to gain insight into the pathophysiology of IBM. Degeneration was evident due to the presence of amyloid β peptide 1–42 (Aβ1–42) in the saliva of the analyzed IBM patients. The presence of metabolic disarrangements in IBM was indicated by an imbalanced organic acid profile in fibroblasts and urine. Specifically, abnormal levels of L-pyroglutamic and orotic acid were supported by the abnormal expression of related metabolites in plasma and urine (glutathione and pyrimidines) and the aberrant expression of upstream gene regulators (L2HGDH, IDH2, OPLAH, and ASL) in muscle. Combined levels of L-pyroglutamic and orotic acid displayed an outstanding biomarker signature in urine with 100% sensitivity and specificity. The confirmation of systemic metabolic disarrangements in IBM and the identification of novel biomarkers reported herein unveil novel insights that require validation in larger cohorts

El trabajo obtuvo un Premio Tomás García Verdejo a las buenas prácticas educativas en la Comunidad Autónoma de Extremadura para el curso 2020/2021. Modalidad BSe describe un proyecto llevado a cabo en el IES Bioclimático y el IES San Roque, ambos de Badajoz, cuyo objetivo principal era atender al alumnado de altas capacidades o con talento/alto rendimiento, mediante la creación de una escuela de teatro en inglés. Otros objetivos de la iniciativa fueron: fomentar el interés por la investigación e innovación utilizando las nuevas tecnologías de la información y comunicación; estimular el potencial de aprendizaje del alumnado participante para el enriquecimiento común contribuyendo al fomento de la igualdad entre hombres y mujeres; aprovechar el teatro como una forma innovadora para el aprendizaje del inglés; ofrecer procedimientos para la mejora del rendimiento escolar y el éxito educativo, la integración social y la reducción de la tasa de fracaso escolar y abandono escolar de estos alumnos; fomentar la investigación, la lectura y la escritura en lengua inglesa; facilitar espacios colaborativos; contribuir a la adquisición de valores como la responsabilidad, la solidaridad, etc. y promover la participación de la comunidad educativaExtremaduraES
