    Dissertation presented to obtain the Ph.D degree in Developmental BiologyProtein N-terminal acetylation is a highly conserved and widespread modification that occurs on approximately 80% of all soluble, cytoplasmic human proteins. Nevertheless, with few exceptions, little is known about the biological function of protein N- acetylation. Recently, it was suggested to act as a general protein destabilization signal in yeast (Hwang et al., 2010). Yet, other reports suggest it might act as stabilizer, for instance by blocking protein degradation (Ciechanover and Ben-Saadon, 2004). This protein modification is catalyzed by N-terminal acetyltransferases (NATs), which are highly conserved from yeast to humans both in subunit composition and substrate specificity (Starheim et al., 2009). Since NATs enzymatic activity and function has been mostly studied in yeast and tissue culture cells, our current understanding of the role of these enzymes during development of multicellular organisms is extremely poor, being our main goal to understand the role of this ubiquitous protein modification during development.(...)This dissertation was sponsored by Fundação para a Ciência e Tecnologia.     Satisfação profissional: um estudo com enfermeiros especialistas em Enfermagem de Reabilitação

    oai:ojs2.rper.aper.pt:article/7The goal of this study was to analyze the professional satisfaction of rehabilitation nurses, as well as to verify whetherthe practice of specialized care or the practice of general care influences your professional satisfaction.The study involved 306 rehabilitation nurses and their professional satisfaction was evaluated through their responsesto the “Satisfaction in Nurses’ Work Scale”. We used an analytics transverse methodology.For statistical evaluation, we used factorial analysis, Spearman’s correlation coefficient and parametric test t-studentfor independent samples.Our results show that the rehabilitation nurses are slightly dissatisfied.We found that Nurses who practice general care are more dissatisfied than those who practice specialty care.Este estudio pretendió analizar la satisfacción profesional de los enfermeros especialistas en Enfermería deRehabilitación y también averiguar si el ejercicio de cuidados de especialidad o el ejercicio de cuidados generalesinfluye en su satisfacción profesional.La satisfacción profesional fue evaluada a través de la aplicación de la “Escala de Evaluación de la Satisfacción en eltrabajo de los Enfermeros” a 306 profesionales de Enfermería, especialistas en Enfermería de Rehabilitación. Serealizó un estudio transverso analítico.Como instrumentos de análisis estadística de los datos, se recorrió al análisis factorial, al coeficiente de correlaciónde Spearman y a un teste paramétrico t-student para muestras independientes.Los resultados muestran que los enfermeros especialistas en Enfermería de Rehabilitación se encuentran ligeramenteinsatisfechos.Verificamos que los enfermeros que ejercen cuidados generales se encuentran más insatisfechos que los que trabajancomo especialistas.Este estudo pretendeu analisar a satisfação profissional dos Enfermeiros Especialistas em Enfermagem de Reabilitação (EEER) e também averiguar se o exercício de cuidados de especialidade ou o exercício de cuidados gerais influencia a sua satisfação profissional.A satisfação profissional foi avaliada através da aplicação da “Escala de Avaliação da Satisfação no Trabalho dos Enfermeiros” a 306 profissionais de Enfermagem, especialistas  Enfermagem de Reabilitação.     Maintaining centrosomes and cilia

There is no public supplementary material available for this publication.Centrosomes and cilia are present in organisms from all branches of the eukaryotic tree of life. These structures are composed of microtubules and various other proteins, and are required for a plethora of cell processes such as structuring the cytoskeleton, sensing the environment, and motility. Deregulation of centrosome and cilium components leads to a wide range of diseases, some of which are incompatible with life. Centrosomes and cilia are thought to be very stable and can persist over long periods of time. However, these structures can disappear in certain developmental stages and diseases. Moreover, some centrosome and cilia components are quite dynamic. While a large body of knowledge has been produced regarding the biogenesis of these structures, little is known about how they are maintained.     Is Redshirting Beneficial for Reading Acquisition Success?

    The controversy around the effect of academic redshirting on reading acquisition continues receiving attention in the international literature. However, few studies are known with non-English speaking children. In this study we intend to understand this phenomenon with 698 Portuguese speaking first graders, 360 girls (51.6%), aged between 5 years old and 8 months and 7 years old and 6 months (M = 6.3 months, SD = 3.9 months). Reading acquisition precursors were assessed namely phonemic awareness and letter-sound knowledge. Results reveal that 5.9% of first graders are redshirted. Clusters analysis indicated two clusters per variable. Cluster 1 with low phonemic awareness and letter-sound knowledge and low socioeconomic status, cluster 2 with high phonemic awareness and letter-sound knowledge and medium-high socioeconomic status. The cluster results suggest a prevalence of 24.5% children at risk of having learning difficulties. The MANOVA indicated that only socioeconomic status has an effect on phonemic awareness and letter-sound knowledge, with children from medium-high level presenting higher results. It is concluded that redshirting did not bring additional advantages for reading acquisition success. Implications about the importance of education in order to lessen those differences, as well as prevent difficulties are presented.info:eu-repo/semantics/publishedVersio

    Usos da memória como recurso de contextualização no jornalismo digital

    This article aims to discuss aspects of the production of the so-called contextual journalism linked to the notion of memory, in order to understand how it is triggered in the construction of journalistic contents that seek to offer, in a perspective of contextualization, better circumstances for understanding the facts and information reported. To this purpose, reports from the Nexo Jornal and UOL Tab websites are taken as objects of analysis. A first understanding provided by such approach is that memory can be present in journalism in different ways, highlighting here how it, in the activation of facts and circumstances of the past, can shed light on the present of everyday life. Journalism, so to speak, uses memory at the same time that it contributes to shaping it.O presente artigo tem o objetivo de discutir aspectos da produção do chamado jornalismo contextual articulado à noção de memória, a fim de compreender como esta é acionada na construção dos conteúdos jornalísticos que buscam oferecer, em uma perspectiva de contextualização, melhores circunstâncias de compreensão dos fatos e informações abordados. Para tal, toma-se como objetos de análise reportagens dos portais Nexo Jornal e do UOL Tab.     Lower airway flow influences peak nasal inspiratory flow in school-aged children*

    Background: Rhinitis and asthma frequently coexist. Peak nasal inspiratory flow (PNIF) objectively evaluates nasal obstruction. Lower airway flow’s impact on PNIF has seldom been analysed in children. We aimed to study the associations between PNIF and: (1)forced expiratory volume in one second (FEV1) and peak expiratory flow (PEF) in children with allergic rhinitis and asthma and healthy controls; (2)allergic rhinitis and asthma control subjective evaluation. Methods: Sequential assessments of PNIF before and after nasal decongestion and spirometry with bronchodilation test were performed in 65 children (6-12 years) with allergic rhinitis and asthma, and 24 gender, age-matched healthy controls. The Control of Allergic Rhinitis and Asthma Test in children (CARATkids) was used for control assessment. Associations were investigated by multiple linear regression models. Results: Baseline and decongested PNIF correlated with baseline and post-bronchodilation FEV1 and PEF, observed independently of rhinitis and asthma diagnosis. The best model for PNIF included PEF, age and gender. No association was found between PNIF and CARATkids scores, except for nasal obstruction self-report. Conclusion: In school-aged children, besides age and gender, PEF values should ideally be known to interpret PNIF values.     Distribuição dos casos de cardiopatias congênitas em um hospital do Oeste Paulista

    O objetivo deste trabalho foi analisar a distribuição espaço-temporal dos pacientes com cardiopatias congênitas atendidos no Ambulatório de Cardiologia Pediátrica do Hospital de referência do Oeste Paulista. Realizamos um estudo retrospectivo com análise de dados de base eletrônica e prontuários dos pacientes diagnosticados com cardiopatiacongênita entre os períodos de julho de 2013 a julho de 2018. Foram selecionados 298 prontuários para análise das variáveis de CID-10, gênero, distribuição espacial e série temporal. Foi possível observar que os defeitos septais foram as cardiopatias mais prevalentes, não houve diferença entre os gêneros. Notou-se aumento do diagnóstico a partir de 2014, com implementação do teste do coraçãozinho e 51% dos casos eram da cidade de Presidente Prudente,com maior concentração de casos na região do parque industrial. Há uma relação na incidência das malformações cardíacas com o meio ambiente desfavorável. Os resultados encontrados podem guiar políticas de saúde pública, visando reduzir a exposição da população mais vulnerável, na busca da melhora nos índices de saúde.The aim of this research was to conduct a comprehensive spatial-temporal analysis of the population affected by congenital heart anomalies assisted at the Pediatric Cardiology Outpatient Department at the distinguished Western Paulista reference hospital. We conducted a retrospective study involving the analysis of electronic databaserecords and patient medical charts for individuals diagnosed with congenital heart disease during the period from July 2013 to July 2018. A total of 298 medical records were selected for the analysis of variables encompassing the ICD-10 codes, gender, spatial distribution, and temporal trends. It was possible to observe that septal defects were the most prevalent congenital heart abnormalities, and there was no gender-based difference. An increase in diagnoses was noted from 2014, coinciding with the implementation of the “heart test,” and 51% of the cases were from Presidente Prudente, with a higher concentration of cases in the industrial park area. There is an association between cardiac congenital malformations and an adverse environmental context. The findings can inform public health policies aimed at reducing the exposure of the most vulnerable population in pursuit of improvinghealth indicators

    In post-modern society, changes in people's lifestyle have triggered both professional and personal problems. In this context, encouraging the biopsychosocial look at the patient, there is a concern to research more about Takotsubo Cardiomyopathy. The objective of this research is to identify studies that address the factors that can trigger Takotsubo myocardiopathy. This is an integrative literature review. The evidence found through the retrieved articles indicates the association of CMT with stressors, this being the most likely variable, it is an underdiagnosed pathology because its findings are confused with Acute Coronary Syndrome. The results of the present study demonstrate the association of stressors with the development of Takotsubo Cardiomyopathy, clarifying possible relationships between them.Na sociedade pós-moderna, as mudanças no estilo de vida das pessoas desencadearam problemas tanto profissionais, quanto pessoais. Nesse contexto, incentivando o olhar biopsicossocial para o paciente, surge a inquietação de se pesquisar mais sobre a Cardiomiopatia de Takotsubo. O objetivo dessa pesquisa é identificar estudos que abordem os fatores que podem desencadear a mio cardiopatia de Takotsubo.     Regulation of Autophosphorylation Controls PLK4 Self-Destruction and Centriole Number

The SCF-Slimb/βTrCP-E3 ubiquitin ligase controls PLK4 levels through recognition of a conserved phosphodegron. SCF-Slimb/βTrCP substrate binding and targeting for degradation is normally regulated by phosphorylation cascades, controlling complex processes, such as circadian clocks and morphogenesis. Here, we show that PLK4 is a suicide kinase, autophosphorylating in residues that are critical for SCF-Slimb/βTrCP binding. We demonstrate a multisite trans-autophosphorylation mechanism, likely to ensure that both a threshold of PLK4 concentration is attained and a sequence of events is observed before PLK4 can autodestruct. First, we show that PLK4 trans-autophosphorylates other PLK4 molecules on both Ser293 and Thr297 within the degron and that these residues contribute differently for PLK4 degradation, the first being critical and the second maximizing auto-destruction. Second, PLK4 trans-autophosphorylates a phospho-cluster outside the degron, which regulates Thr297 phosphorylation, PLK4 degradation, and centriole number. Finally, we show the importance of PLK4-Slimb/βTrCP regulation as it operates in both soma and germline.
