    O presente trabalho tem como tema as técnicas de Reprodução Humana Assistida, analisando a responsabilidade civil frente às lacunas do ordenamento jurídico brasileiro e tem por objetivo principal promover reflexões sobre os problemas bioéticos e jurídicos proveniente de tão controverso tema. A problemática se verifica principalmente na concepção do filho através das técnicas de reprodução humana assistida, levando em consideração o direito ao planejamento familiar previsto na Constituição e a carência de legislação que regulamente tais procedimentos de forma ética e juridicamente corretas. Assim, a presente pesquisa se justifica por tratar de problemática relacionada a uma das esferas mais importantes do ser humano, o direito à vida e consequentemente a procriação. Por conseguinte, é necessário que os operadores do Direito conheçam os tipos de técnicas de Reprodução Humana Assistida, bem como sua classificação no mundo científico, para que possam conduzir e litigar de forma justa e sempre relacionada à bioética. Na abordagem do tema proposto, serão observadas e conceituadas as técnicas de reprodução humana assistida, de forma que se esclareça e não deixe dúvidas sobre tais procedimentos. Apresentaremos ainda, os princípios norteadores do direito relacionados ao planejamento familiar em uma análise conceitual e interdisciplinar do assunto, para tentar minimizar os questionamentos relacionados ao tema

    Evaluation of the supramolecular interaction of Congo red with cucurbiturils using mass spectrometry and spectroscopic methods

    The ability of cucurbit[n]urils (CB[n]) to decolourise aqueous solutions of the azo dye Congo red (CR) was described more than a century ago alongside the first synthesis of CB[n]. No subsequent studies of the nature of the physical interactions have been reported despite the interest in using CB[n] as adsorbents for the removal of CR and related organic dyes from wastewaters. In the present work the supramolecular interaction between CB[n] (n = 7, 8) and CR was studied by electrospray ionisation mass spectrometry (ESI-MS), 1H NMR, and solid-state characterisation of isolated complexes. Under positive ESI, the formation of host–guest complexes in the gas phase was not observed, suggesting that CR anions do not interact with the portals and the nonpolar inner cavity of the CB[n] molecules. Conversely, under negative ESI, 1 : 1 and higher order (1 : 2, 2 : 1, 3 : 1 and 2 : 2) CR: CB[7] and CR: CB[8] adducts were detected, which is attributed to interaction between CR and the outer surface hydrogens of CB[n]. Solid-state supramolecular adducts between CB[n] and CR were isolated from aqueous media under either ambient conditions, giving structures denoted as CR@CB[n](RT), or hydrothermal (100 ºC) conditions, giving structures denoted as CR@CB[n](100). The adducts were characterised by elemental and thermogravimetric analyses (TGA), powder X-ray diffraction (PXRD), and spectroscopic methods (FT-IR, FT-Raman, 13C{1H} CP MAS NMR, UV/vis and near-IR absorption, fluorescence excitation and emission). The interaction conditions and acid content of the starting CB[n] influenced the protonation state of CR molecules. CR@CB[7](RT) contained only unprotonated CR, while CR@CB[8](100) contained exclusively protonated CR (ammonium and azonium/quinoid structures).     In vitro binding and survival assays of Leishmania parasites to peripherical blood monocytes and monocyte-derived macrophages isolated from dogs naturally and experimentally infected with Leishmania (Leishmania) chagasi

    <p>Abstract</p> <p>Background</p> <p>There are a few works considering the characterization of canine monocyte-derived macrophages as well as a standardized procedure for isolation, culture, and infection of these cells with <it>Leishmania</it>. We have performed several modifications in order to improve the canine monocyte-derived macrophage cultures. In addition, we have done a comparative study between monocytes and monocyte-derived macrophages from dogs naturally and experimentally infected with <it>L. chagasi</it>.</p> <p>Results</p> <p>In the presence of exogenous serum, opsonized <it>Leishmania </it>promastigotes binds better to monocytes/macrophages than without serum. Otherwise, this binding occurs due to the strict correlation between the opsonized biologic particles with the third receptor of the complement (CR3-CD11b/CD18). In fact, our assays with CD11b confirmed the importance of this receptor for canine cells and the <it>L. chagasi </it>experimental system. Moreover, monocytes obtained from naturally infected dogs have shown a higher number of monocytes bounded to promastigotes. The experimental results regarding survival have shown that promastigote forms of opsonized <it>L. chagasi </it>were more infective, because we found higher numbers of promastigotes bound to the different cells. As a consequence, after forty-eight hours of binding, higher numbers of amastigotes appeared inside monocyte-macrophages.</p> <p>Conclusion</p> <p>These studies have given support to continue comparative studies involving canine monocytes, monocyte-derived macrophages and peritoneal macrophages. Since we have standardized the canine cell culture, we are looking forward to determining the phenotypic properties of these cells before and after <it>L. chagasi </it>infection using flow cytometry.</p

    Supramolecular organizations in the aerobic respiratory chain of Escherichia coli

    The organization of respiratory chain complexes in supercomplexes has been shown in the mitochondria of several eukaryotes and in the cell membranes of some bacteria. These supercomplexes are suggested to be important for oxidative phosphorylation efficiency and to prevent the formation of reactive oxygen species. Here we describe, for the first time, the identification of supramolecular organizations in the aerobic respiratory chain of Escherichia coli, including a trimer of succinate dehydrogenase. Furthermore, two heterooligomerizations have been shown: one resulting from the association of the NADH:quinone oxidoreductases NDH-1 and NDH-2, and another composed by the cytochrome bo3 quinol:oxygen reductase, cytochrome bd quinol:oxygen reductase and formate dehydrogenase (fdo). These results are supported by blue native-electrophoresis, mass spectrometry and kinetic data of wild type and mutant E . coli strains

    Identification of impact aroma compounds in Eugenia uniflora L. (Brazilian Pitanga) leaf essential oil

    The leaf essential oil of Eugenia uniflora L. (Myrtaceae) was extracted by Clevenger apparatus and analysed by gas chromatography-mass spectrometry (GC-MS). The leaves were collected and immediately extracted for five consecutive days at 9:00 am and 2:00 pm. No variance in the oil yields were observed in the period. Furanodiene and its rearrangement product, furanoelemene (or curzerene, 50.2%), beta-elemene (5.9%) and alpha-cadinol (4.7%) were identified as the most abundant compounds. GC-Olfatometry (GC-O) associated to Aroma Extract Dilution Analysis (AEDA) allowed the identification of nine active aroma compounds, where furanodiene (along with furanoelemene, FD 1024), beta-elemene (FD 256) and (E,E)-germacrone (FD 256) were characterized as the main impact aroma compounds in the odor of this essential oil. Those substances were collected through a sniffing port adapted on the GC allowing to obtain a typical essence of pitanga as indicated by comparative olfatometric analysis

    Health status and labor force participation : evidence for urban low and middle income individuals in Colombia

    This paper analyzes the relationship between individual health status and labor force participation using the first wave of the Colombian Longitudinal Survey. The empirical modeling strategy accounts for the presence of potential endogeneity between these two variables. The results show that there is a positive relationship between health and labor force participation in both directions, indicating that better health is likely to lead to a higher probability of participation in the labor market, but also that individuals who participate in the labor market are more likely to report better health. Interesting differences are uncovered when comparing the results by gender and/or age groups. For instance, for younger females, health status and higher education positively affect the probability of labor participation, whereas having children under the age of 5 and being married reduce their probability of participation. Our findings also highlight the importance of public policy to guarantee good health conditions of the population which could also have a positive impact on labor productivity and consequently on long-run economic growth.info:eu-repo/semantics/publishedVersio

    Bacteriophage-receptor binding proteins for multiplex detection of Staphylococcus and Enterococcus in blood

    Health care-associated infections (HCAIs) affect hundreds of millions of patients, representing a significant burden for public health. They are usually associated to multidrug resistant bacteria, which increases their incidence and severity. Bloodstream infections (BSIs) are among the most frequent and life-threatening HCAIs, with Enterococcus and Staphylococcus among the most common isolated pathogens. The correct and fast identification of the etiological agents is crucial for clinical decision-making, allowing to rapidly select the appropriate antimicrobial and to prevent from overuse and misuse of antibiotics and the consequent increase in antimicrobial resistance. Conventional culture methods are still the gold standard to identify these pathogens, however are time-consuming and may lead to erroneous diagnosis, which compromises an efficient treatment. (Bacterio)phage receptor binding proteins (RBPs) are the structures responsible for the high specificity conferred to phages against bacteria and thus are very attractive biorecognition elements with high potential for specific detection and identification of pathogens.     T-Cell Memory Responses Elicited by Yellow Fever Vaccine are Targeted to Overlapping Epitopes Containing Multiple HLA-I and -II Binding Motifs

    The yellow fever vaccines (YF-17D-204 and 17DD) are considered to be among the safest vaccines and the presence of neutralizing antibodies is correlated with protection, although other immune effector mechanisms are known to be involved. T-cell responses are known to play an important role modulating antibody production and the killing of infected cells. However, little is known about the repertoire of T-cell responses elicited by the YF-17DD vaccine in humans. In this report, a library of 653 partially overlapping 15-mer peptides covering the envelope (Env) and nonstructural (NS) proteins 1 to 5 of the vaccine was utilized to perform a comprehensive analysis of the virus-specific CD4+ and CD8+ T-cell responses. The T-cell responses were screened ex-vivo by IFN-γ ELISPOT assays using blood samples from 220 YF-17DD vaccinees collected two months to four years after immunization. Each peptide was tested in 75 to 208 separate individuals of the cohort. The screening identified sixteen immunodominant antigens that elicited activation of circulating memory T-cells in 10% to 33% of the individuals. Biochemical in-vitro binding assays and immunogenetic and immunogenicity studies indicated that each of the sixteen immunogenic 15-mer peptides contained two or more partially overlapping epitopes that could bind with high affinity to molecules of different HLAs. The prevalence of the immunogenicity of a peptide in the cohort was correlated with the diversity of HLA-II alleles that they could bind. These findings suggest that overlapping of HLA binding motifs within a peptide enhances its T-cell immunogenicity and the prevalence of the response in the population. In summary, the results suggests that in addition to factors of the innate immunity, "promiscuous" T-cell antigens might contribute to the high efficacy of the yellow fever vaccines. © 2013 de Melo et al
