    Development of new nano-enhanced phase change materials (NEPCM) to improve energy efficiency in buildings: Lab-scale characterization

    Fatty acids are promising organic phase change materials (PCMs) for thermal energy storage (TES) in buildings because of their high storage capacity, non-toxic nature and little subcooling. Their phase change temperatures make them suitable for heating, ventilating and air conditioning (HVAC) applications in the building sector. However, one of their main drawbacks is their poor thermal conductivity which limits their application. In the present study two fatty acids within the building application temperature range, capric acid (CA) and capric-myristic acid (CA-MA) eutectic mixture, were nano-enhanced throughout silicon dioxide nanoparticles (nSiO2) addition (0.5 wt.%, 1.0 wt.% and 1.5 wt.%). Main properties of the nano-enhanced phase change materials (NEPCM) obtained were characterized by means of differential scanning calorimetry (DSC), Hot wire technique, Fourier transformed infrared (FT-IR) spectroscopy, thermogravimetric analyses (TGA), scanning electron microscopy (SEM), and rheological measurements. Furthermore, their long-term performance was evaluated after 2000 cycles by means of cycling stability tests. The NEPCM obtained showed high thermal conductivity and specific heat capacity.     Climate and Nothofagus pumilio establishment at upper treelines in the patagonian andes

    The upper treeline provides a unique opportunity for documenting changes in forest dynamics in the context of current environmental fluctuations. Documenting the response of forests to present environmental changes will be very useful in developing and validating vegetation models that predict plant responses to global changes resulting fromhuman activities. It is generally suggested that the expansion of the upper treeline is limited by cold temperatures, so warmer conditions are expected to induce an advance of forests into alpine vegetation. Significant changes in temperature and precipitation have already been documented in regional studies along the Patagonian Andes during the last decades. Across Patagonia, there are strong relationships between changes in climate and variations in the Southern Annular Mode (SAM). The atmospheric circulation indexes, act as climate packages summarizing climatic information since they combine changes in environmental variables. In order to complement previous studies in the northern Patagonia treeline, we have conducted a this study in the southern Patagonian Andes to: (1) characterize the patterns of Nothofagus pumilio establishment in the upper treeline along a precipitation gradient, (2) to establish the relationship between variations in regional climate and tree establishment and (3) to determine the influence of continental and hemispheric-scale climatic forcing on tree establishment at regional scale. Our studies suggest that the location of the upper treeline of the N. pumilio forest in the Patagonian Andes is sensitive to changes in climate variations. The temperature increment has facilitated the tree recruitment, however, the rate of seedling establishment appears to be more strongly modulated by the interaction between temperature increase and variations in precipitation. The expansion of the upper treeline has been limited to 5–10m from the abrupt treeline. This comparatively reduced advance of the forest in the alpine grassland is regulated by other biotic and abiotic factors rather than the temperature increase. While the recent expansion of the upper treeline in northern Patagonia was regulated by temperature and precipitation variations associated with the different phases of the Pacific Decadal Oscillation, the establishment above the upper treeline in the southern Patagonian Andes was facilitated by climatic variations induced by the positive trend of the SAM during the last decades.Fil: Srur, Ana Marina.     Dkk1 Regulates Ventral Midbrain Dopaminergic Differentiation and Morphogenesis

    Dickkopf1 (Dkk1) is a Wnt/ÎČ-catenin inhibitor that participates in many processes during embryonic development. One of its roles during embryogenesis is to induce head formation, since Dkk1-null mice lack head structures anterior to midbrain. The Wnt/ÎČ-catenin pathway is also known to regulate different aspects of ventral midbrain (VM) dopaminergic (DA) neuron development and, in vitro, Dkk1-mediated inhibition of the Wnt/ÎČ-catenin pathway improves the DA differentiation in mouse embryonic stem cells (mESC). However, the in vivo function of Dkk1 on the development of midbrain DA neurons remains to be elucidated. Here we examined Dkk1+/− embryos and found that Dkk1 is required for the differentiation of DA precursors/neuroblasts into DA neurons at E13.5. This deficit persisted until E17.5, when a defect in the number and distribution of VM DA neurons was detected. Furthermore, analysis of the few Dkk1−/− embryos that survived until E17.5 revealed a more severe loss of midbrain DA neurons and morphogenesis defects. Our results thus show that Dkk1 is required for midbrain DA differentiation and morphogenesis

    An efficient GUI-based clustering software for simulation and Bayesian cluster analysis of single-molecule localization microscopy data

    Ligand binding of membrane proteins triggers many important cellular signaling events by the lateral aggregation of ligand-bound and other membrane proteins in the plane of the plasma membrane. This local clustering can lead to the co-enrichment of molecules that create an intracellular signal or bring sufficient amounts of activity together to shift an existing equilibrium towards the execution of a signaling event. In this way, clustering can serve as a cellular switch. The underlying uneven distribution and local enrichment of the signaling cluster’s constituting membrane proteins can be used as a functional readout. This information is obtained by combining single-molecule fluorescence microscopy with cluster algorithms that can reliably and reproducibly distinguish clusters from fluctuations in the background noise to generate quantitative data on this complex process. Cluster analysis of single-molecule fluorescence microscopy data has emerged as a proliferative field, and several algorithms and software solutions have been put forward. However, in most cases, such cluster algorithms require multiple analysis parameters to be defined by the user, which may lead to biased results. Furthermore, most cluster algorithms neglect the individual localization precision connected to every localized molecule, leading to imprecise results. Bayesian cluster analysis has been put forward to overcome these problems, but so far, it has entailed high computational cost, increasing runtime drastically. Finally, most software is challenging to use as they require advanced technical knowledge to operate. Here we combined three advanced cluster algorithms with the Bayesian approach and parallelization in a user-friendly GUI and achieved up to an order of magnitude faster processing than for previous approaches. Our work will simplify access to a well-controlled analysis of clustering data generated by SMLM and significantly accelerate data processing. The inclusion of a simulation mode aids in the design of well-controlled experimental assays

    A transmembrane histidine kinase functions as a pH sensor

    The two-component system DesK-DesR regulates the synthesis of unsaturated fatty acids in the soil bacteria Bacillus subtilis. This system is activated at low temperature and maintains membrane lipid fluidity upon temperature variations. Here, we found that DesK—the transmembrane histidine kinase—also responds to pH and studied the mechanism of pH sensing. We propose that a helix linking the transmembrane region with the cytoplasmic catalytic domain is involved in pH sensing. This helix contains several glutamate, lysine, and arginine residues At neutral pH, the linker forms an alpha helix that is stabilized by hydrogen bonds in the i, i + 4 register and thus favors the kinase state. At low pH, protonation of glutamate residues breaks salt bridges, which results in helix destabilization and interruption of signaling.     An Immune-CNS Axis Activates Remote Hippocampal Stem Cells Following Spinal Transection Injury

    External stimuli such as injury, learning, or stress influence the production of neurons by neural stem cells (NSCs) in the adult mammalian brain. These external stimuli directly impact stem cell activity by influencing areas directly connected or in close proximity to the neurogenic niches of the adult brain. However, very little is known on how distant injuries affect NSC activation state. In this study, we demonstrate that a thoracic spinal transection injury activates the distally located hippocampal-NSCs. This activation leads to a transient increase production of neurons that functionally integrate to improve animal’s performance in hippocampal-related memory tasks. We further show that interferon-CD95 signaling is required to promote injury-mediated activation of remote NSCs. Thus, we identify an immune-CNS axis responsible for injury-mediated activation of remotely located NSCs

    Multiproxy summer and winter surface air temperature field reconstructions for southern South America covering the past centuries

    We statistically reconstruct austral summer (winter) surface air temperature fields back to ad 900 (1706) using 22 (20) annually resolved predictors from natural and human archives from southern South America (SSA). This represents the first regional-scale climate field reconstruction for parts of the Southern Hemisphere at this high temporal resolution. We apply three different reconstruction techniques: multivariate principal component regression, composite plus scaling, and regularized expectation maximization. There is generally good agreement between the results of the three methods on interannual and decadal timescales. The field reconstructions allow us to describe differences and similarities in the temperature evolution of different sub-regions of SSA. The reconstructed SSA mean summer temperatures between 900 and 1350 are mostly above the 1901-1995 climatology. After 1350, we reconstruct a sharp transition to colder conditions, which last until approximately 1700. The summers in the eighteenth century are relatively warm with a subsequent cold relapse peaking around 1850. In the twentieth century, summer temperatures reach conditions similar to earlier warm periods. The winter temperatures in the eighteenth and nineteenth centuries were mostly below the twentieth century average. The uncertainties of our reconstructions are generally largest in the eastern lowlands of SSA, where the coverage with proxy data is poorest. Verifications with independent summer temperature proxies and instrumental measurements suggest that the interannual and multi-decadal variations of SSA temperatures are well captured by our reconstructions. This new dataset can be used for data/model comparison and data assimilation as well as for detection and attribution studies at sub-continental scales

    Trends in the epidemiology of catheter-related bloodstream infections; towards a paradigm shift, Spain, 2007 to 2019

Aim: To analyse changes in CRBSI epidemiology observed by the Infection Control Catalan Programme (VINCat). Methods: A cohort study including all hospital-acquired CRBSI episodes diagnosed at 55 hospitals (2007-2019) in Catalonia, Spain, was prospectively conducted. CRBSI incidence rates were adjusted per 1,000patientdays. To assess the CRBSI rate trend per year, negative binomial models were used, with the number of events as the dependent variable, and the year as the main independent variable. From each model, the annual rate of CRBSI diagnosed per 1,000patientdays and the incidence rate ratio (IRR) with its 95% confidence intervals (CI) were reported. Results: During the study, 9,290 CRBSI episodes were diagnosed (mean annual incidence rate:0.20episodes/1,000patientdays). Patients' median age was 64.1years; 36.6% (3,403/9,290) were female. In total, 73.7% (n=6,845) of CRBSI occurred in non-intensive care unit (ICU) wards, 62.7% (n=5,822) were related to central venous catheter (CVC), 24.1% (n=2,236) to peripheral venous catheters (PVC) and 13.3% (n=1,232) to peripherally-inserted central venous catheters (PICVC). Incidence rate fell over the study period (IRR:0.94;95%CI:0.93-0.96), especially in the ICU (IRR:0.88;95%CI:0.87-0.89). As a whole, while episodes of CVC CRBSI fell significantly (IRR:0.88;95%CI:0.87-0.91), peripherally-inserted catheter CRBSI (PVC and PICVC) rose, especially in medical wards (IRR PICVC:1.08;95%CI:1.05-1.11; IRR PVC: 1.03; 95% 1.00-1.05). Conclusions: Over the study, CRBSIs associated with CVC and diagnosed in ICUs decreased while episodes in conventional wards involving peripherally-inserted catheters increased. Hospitals should implement preventive measures in conventional wards
