Os resultados parecem confirmar as hipóteses aí postas, uma vez que apresentam certa zona da serra da Arrábida como centro emissor do mesmo material.     Degradation of Fluoroquinolone Antibiotics and Identification of Metabolites/Transformation Products by LC-MS/MS

    Antibiotics are a therapeutic class widely found inenvironmental matrices and extensively studied due to its persistence and implications for multi-resistant bacteria development. Degradation of four fluoroquinolone antibiotics, namely Ofloxacin (OFL), Norfloxacin (NOR), Ciprofloxacin (CPF) and Moxifloxacin (MOX), at 10 mg L-1 using a mixed bacterial culture, was assessed for 60 days. The assays were followed by a developed and validated analytical method of HPLC with Fluorescence Detection using a Luna PFP (2) 3µm column. The optimized conditions allowed picturing metabolites/transformation products formation and accumulation during the process, stating an incomplete mineralization, also shown byfluoride release. OFL and MOX presented the highest (98.3%) and the lowest (80.5%) extent of degradation after 19 days of assay, respectively. Some of these intermediate compounds were identified by LCMS/MS in selected degradation samples. Most of the intermediates were already described as biodegradation and/or photodegradationproducts in different conditions, but new and/or unknown metabolites were also present

    Impact of anthropogenic noise on the survival and development of meagre (Argyrosomus regius) early life stages

    The growth of human populations has been driving an unprecedent and widespread increase in marine traffic, posing a real threat to marine biodiversity. Even though we are now aware of the negative effects of shipping noise exposure on fish, information about the impact on their early life stages continues to lack. Meagre (Argyrosomus regius) is a vocal fish that uses estuaries with high levels of anthropogenic noise pollution as both breeding areas and nurseries. Here, the effects of boat noise exposure on the development and survival of meagre larvae were studied. Embryos and larvae were exposed to either noise (boat noise playback) or control treatments (coils producing a similar electric field to the speakers) and hatching rate, survival rate, morphometric traits and stress-related biomarkers, at hatching and at 2 days-post-hatching (dph) were analyzed. Results showed no conclusive effects of the impact of boat noise playback, even though there was an increased lipid droplet consumption and a decrease in body depth at 2dph larvae under this stressor. The assessment of oxidative stress and energy metabolism-related biomarkers at hatching showed a marginal decrease in superoxide dismutase (SOD) activity and no changes in DNA damage or electron transport system activity (ETS), although it cannot be disregarded that those effects could only be visible at later stages of larval development. Whether these morphological and developmental results have implications in later stages remains to be investigated.     Boat noise impacts Lusitanian toadfish breeding males and reproductive outcome

    Anthropogenic noise is a growing threat to marine organisms, including fish. Yet very few studies have addressed the impact of anthropogenic noise on fish reproduction, especially in situ. In this study, we investigated the impacts of boat noise exposure in the reproductive success of wild Lusitanian toadfish (Halobatrachus didactylus), a species that relies on advertisement calls for mate attraction, using behavioural, physiological and reproductive endpoints. Two sets of artificial nests were deployed in the Tagus estuary and exposed to either ambient sound or boat noise during their breeding season. Toadfish males spontaneously used these nests to breed. We inspected nests for occupation and the presence of eggs in six spring low tides (in two years) and assessed male vocal activity and stress responses. Boat noise did not affect nest occupation by males but impacted reproductive success by decreasing the likelihood of receiving eggs, decreasing the number of live eggs and increasing the number of dead eggs, compared to control males. Treatment males also showed depressed vocal activity and slightly higher cortisol levels. The assessment of oxidative stress and energy metabolism-related biomarkers revealed no oxidative damage in noise exposed males despite having lower antioxidant responses and pointed towards a decrease in the activity levels of energy metabolism-related biomarkers. These results suggest that males exposed to boat noise depressed their metabolism and their activity (such as parental care and mate attraction) to cope with an acoustic stressor, consistent with a freezing defensive response/behaviour. Together, our study demonstrates that boat noise has severe impacts on reproductive fitness in Lusitanian toadfish. We argue that, at least fishes that cannot easily avoid noise sources due to their dependence on specific spawning sites, may incur in significant direct fitness costs due to chronic noise exposure.Fundação para a Ciência e Tecnologia - FCTinfo:eu-repo/semantics/publishedVersio

    Stealth magnetoliposomes based on calcium-substituted magnesium ferrite nanoparticles for curcumin transport and release

    Despite the promising pharmacological properties of curcumin, the transport and effective release of curcumin is still a challenge. The advances in functionalized nanocarriers for curcumin have also been motivated by the anticancer activity of this natural compound, aiming at targeted therapies. Here, stealth (aqueous and solid) magnetoliposomes containing calcium-substituted magnesium ferrite nanoparticles, CaxMg1−xFe2O4 (with x = 0.25, 0.50, 0.75) were developed as nanocarriers for curcumin. The magnetic nanoparticles exhibit superparamagnetic properties and crystalline structure, with sizes below 10 nm. The magnetoliposomes based on these nanoparticles have hydrodynamic diameters around or below 150 nm and a low polydispersity. The influence of an alternating magnetic field (AMF) on drug release over time was evaluated and compared with curcumin release by diffusion.     Effects of fluoxetine schock loadings in a aerobic granular sludge sequencing batch reactor

    Pharmaceuticals have received increasing attention as emerging organic pollutants due to their frequent occurrence in the environment and potential adverse effects on ecossistems and to human health. Pharmaceuticals may not be completely metabolized in the human body and can enter municipal sewage systems as the parent drug and as their “biologically active” metabolites. Some of these compounds cannot be easily removed at wastewater treatment plants (WWTPs) [1]. Fluoxetine (FLX) is a chiral fluorinated pharmaceutical indicated mainly for treatment of depression and is one of the most dispensed drugs in the world. There is a clear evidence of environmental contamination with this drug and its active metabolite norfluoxetine (NFLX) [2]. Granular sludge sequencing batch reactors (SBR) constitute a promising technology for the treatment of effluents containing micropollutants. The main biological processes occurring in wastewater treatment plants - COD, N and P removal - can be inhibited by these pollutants. This study focused on the effect of FLX on the performance of granular sludge SBR and on the diversity of the microbial population under continuous and intermittent feeding of the compound. The COD removal was not markedly affected by FLX shock loads. Ammonium removal was initially affected but after ca. 20 days of FLX feeding, NH4+ was not detected in the treated effluent – maximum of 0.03 mM – indicating that ammonia oxidizing bacteria (AOB) became adapted to the presence of FLX. Nitrite was also practically not detected in the treated effluent - maximum of 0.01 mM - indicating that nitrite oxidizing bacteria (NOB) were not inhibited by the presence of the FLX, whereas nitrate accumulated in the effluent, indicating that denitrification was affected. Phosphate removal was markedly affected in the beginning of FLX feeding showing a gradual adaptation to the presence of FLX, being practically not detected in the treated effluent (maximum of 0.04 mM) after 70 days. There was no evidence of FLX biodegradation. Changes in the bacterial community from aerobic granules were examined using denaturing gradient gel electrophoresis (DGGE) of 16S rRNA. Samples taken before starting the shock loadings with FLX clearly shift from other samples.     In this work, a new method for magnetoliposomes synthesis with improved and adequate structural, physicochemical and magnetic properties was developed. The overall results confirmed the development of a promising new method for the synthesis of cubic-shaped magnetic ferrite nanoparticles and a novel route for drug-loaded SMLs with improved features.     Magnetoliposomes based on shape anisotropic calcium/magnesium ferrite nanoparticles as nanocarriers for doxorubicin

    Multifunctional lipid nanocarriers are a promising therapeutic approach for controlled drug release in cancer therapy. Combining the widely used liposome structure with magnetic nanoparticles in magnetoliposomes allies, the advantages of using liposomes include the possibility to magnetically guide, selectively accumulate, and magnetically control the release of drugs on target. The effectiveness of these nanosystems is intrinsically related to the individual characteristics of the two main components-lipid formulation and magnetic nanoparticles-and their physicochemical combination. Herein, shape-anisotropic calcium-substituted magnesium ferrite nanoparticles (Ca0.25Mg0.75Fe2O4) were prepared for the first time, improving the magnetic properties of spherical counterparts. The nanoparticles revealed a superparamagnetic behavior, high saturation magnetization (50.07 emu/g at 300 K), and a large heating capacity. Furthermore, a new method for the synthesis of solid magnetoliposomes (SMLs) was developed to enhance their magnetic response. The manufacturing technicalities were optimized with different lipid compositions (DPPC, DPPC/Ch, and DPPC/DSPE-PEG) originating nanosystems with optimal sizes for biomedical applications (around or below 150 nm) and low polydispersity index. The high encapsulation efficiency of doxorubicin in these magnetoliposomes was proven, as well as the ability of the drug-loaded nanosystems to interact with cell membrane models and release DOX by fusion. SMLs revealed to reduce doxorubicin interaction with human serum albumin, contributing to a prolonged bioavailability of the drug upon systemic administration.     Behavioral alterations and Fos protein immunoreactivity in brain regions of bile duct-ligated cirrhotic rats

    Hepatic encephalopathy (HE) encompasses a variety of neuropsychiatric symptoms, including anxiety and psychomotor dysfunction. Although HE is a frequent complication of liver cirrhosis, the neurobiological substrates responsible for its clinical manifestations are largely unclear. In the present study, male Wistar rats were bile duct-ligated (BDL), a procedure which induces liver cirrhosis, and on the 21st day after surgery tested in the elevated plus-maze (EPM) and in an open field for anxiety and locomotor activity measurements. Analysis of Fos protein immunoreactivity (Fos-ir) was used to better understand the neurobiological alterations present in BDL animals. Plasma levels of ammonia were quantified and histopathological analysis of the livers was performed. BDL rats showed a significant decrease in the percentage of entries and time spent in the open arms of the EPM, an anxiogenic effect. These animals also presented significant decreases in Fos-ir in the lateral septal nucleus and medial amygdalar nucleus. Their ammonia plasma levels were significantly higher when compared to the sham group and the diagnosis of cirrhosis was confirmed by histopathological analysis. These results indicate that the BDL model induces anxiogenic results, possibly related to changes in the activation of anxiety-mediating circuitries and to increases in ammonia plasma levels.A Encefalopatia hepática (HE) engloba uma variedade de sintomas neuropsiquiátricos, incluindo ansiedade e disfunção psicomotora. Embora seja uma complicação frequente da cirrose hepática, os substratos neurobiológicos responsáveis por suas manifestações clínicas são em grande parte desconhecidos. No presente estudo, ratos Wistar machos foram submetidos ao procedimento cirúrgico de ligação e secção do ducto biliar (BDL; bile-duct ligation), para indução da cirrose hepática e, no 21º dia após a cirurgia, submetidos aos testes comportamentais no labirinto em cruz elevado (LCE) e campo aberto para avaliação da ansiedade e atividade locomotora. A análise da imunorreatividade à proteína Fos (Fos-ir) foi utilizada para melhor compreender as alterações neurobiológicas presentes nos animais do grupo BDL. Foi realizada a quantificação da concentração de amônia plasmática e análise histopatológica dos fígados. Os ratos do grupo BDL mostraram diminuição significativa na porcentagem de entradas e tempo gasto nos braços abertos do LCE, caracterizando efeito ansiogênico. Estes animais também apresentaram redução significativa na Fos-ir no núcleo septal lateral e núcleo medial da amígdala. A concentração plasmática de amônia foi significativamente mais elevada que a do grupo sham e o diagnóstico de cirrose foi confirmado por análise histopatológica.
