    Quality assurance of diatom counts in Europe: towards harmonized datasets

    Investigations on organism ecology, biodiversity and biogeography often use large compiled datasets to extract information on species ecological preferences, which then can be used in environmental assessment. Freshwater benthic diatoms are commonly used in this context. However, it is important that the taxonomic information of the separate diatom datasets is compatible. At present, inconsistencies between diatom datasets, mainly due to differences and uncertainties in diatom identification, may misinform diatom taxon-specific ecological preferences, geographical distribution and water quality assessment. It is our opinion that these inconsistencies in diatom datasets can be reduced with quality assurance (QA), such as identification exercises. However, the results of these exercises must be well documented and well communicated; otherwise, gained knowledge may not spread inter-regionally or internationally. As a first step to reach greater consistency in QA/harmonization studies, this article (1) presents and compares information of existing diatom identification and counting QA from published and grey (non-peer reviewed) European literature to identify advantages and drawbacks of each approach; (2) summarizes taxa that can easily be misidentified according to European identification exercises; and (3) suggests a consistent design of identification exercises for diatom dataset QA

    On the identity and distribution in northern Portugal of three gomphonema species currently misidentified as gomphonema clevei

    Gomphonema specimens with wide axial areas collected from streams in Northern Portugal were examined by light and scanning electron microscopy. Three species were found and identified as G. rhombicum, G. uniserhombicum and G. ibericum. No specimens were found with the features of G. clevei sensu strict, and previous records from the region with the latter name are regarded as representing one or a mixture of the three species examined herein. The three species differed in the fine details of the striae; double alternating poroids in G. rhombicum, slit-like with irregular outlines in G. uniserhombicum and single or divided slit-like poroids in G. ibericum. Differences in size and shape of the valve and axial area allow discrimination of at least medium-sized to large specimens of the three species. Gomphonema rhombicum was the most common species, sometimes occurring as dominant in the epilithon. Both G. uniserhombicum and G. ibericum were found more sparsely and always in communities containing G. rhombicum. All species were found in slightly acidic to circumneutral, well-oxygenated waters with low conductivity, poor in nutrients and with low organic content.Instituto da Água (Portugal

    Predictive models based on invertebrate assemblages are a powerful tool for assessing water quality. Yet, few attempts have been made to build predictive models with diatoms although they are widely used in freshwater assessment and are among the mandatory biological monitoring elements indicated in the European Water Framework Directive. We built two predictive models for central Portugal, one for macroinvertebrates and the other for diatoms, using data sampled simultaneously from 43 sites in the Spring of 2004 and 2005. Our objective was to test the performance of models based on the diatom community and compare the assessments made by the two indicator communities for the same sites. Each model was tested with 17 sites. Both models showed good performance in predicting reference sites to the correct group: 76% for the macroinvertebrate model using the discriminant variables longitude, distance to source, alkalinity, depth, number of riffles and channel substrate diversity; 85% for the diatoms model using latitude, runoff, mineralization, distance to source, slope, discharge and hardness. The water quality assessments of test sites produced by the two models were similar in 76% of the cases. According to the predictive models, invertebrates were more sensitive to morphological alterations of the channel and habitat conditions (presence of dams and artificial walls, destruction of riparian corridor) while diatoms were more sensitive to changes in water chemistry. Moreover, in lowland streams, where the habitat and substrate heterogeneity are naturally low, assessment by diatoms was more useful than evaluations using invertebrates.     Biosilica from diatoms is formed at ambient conditions under the control of biological and physicochemical processes. The changes in growth and biosilica formation through uptake of different concentrations of Cd2+ by the diatom Nitzschia palea (Kutzing) W. Smith was investigated, correlating Cd2+ effects to changes in the biosilica nanostructure and the relative content of the encapsulated biomolecules. Diatom growth rates at different Cd2+ concentrations (as 1, 2, 3, 4, and 5 X 10-1 mg L-1 CdCl2) were studied in order to determine the concentrations at which sublethal effects were visible, allowing the harvest of sufficient diatom cells for further experiments. We found a clear correlation between the Cd2+ concentrations and both the nanostructure of the biosilica and content of encapsulated peptides. Cd2+ induced biosilica deformation was assessed by scanning electron microscopy and attenuated total reflectance-Fourier Transformed Infrared Spectroscopy (FTIR), revealing that micromorphological changes in frustule features (striae, costae, pores) and nanostructural modifications (structure of the silica and conformation of the encapsulated peptides) occurred at applied Cd2+ concentrations of 2 and 3 X 10-1 mg L-1. In particular the FTIR contribution of peptides decreased at elevated Cd2+ concentrations, whereas shifts in wave number of several relevant organic bonds as C = O stretching (1765 cm-1) and possibly hydrated sulfate (1160, 1110 and 980 cm-1) were assigned. Additional analysis of the amide I band showed a relative increase in beta-sheet structure (16801620 cm-1) when Cd2+ concentration increased. Cadmium uptake clearly affected the molecular ordering of the biosilica in Nitzschia palea, most probably by interfering in biological or physicochemical processes involved in diatom biosilicification

    Advances in the use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems

    Conservation and sustainable management of aquatic ecosystems is a priority in environmental programs worldwide. However, these aims are highly dependent on the efficiency, accuracy and cost of existent methods for the detection of keystone species and monitoring of biological communities. Rapid advances in eDNA, barcoding and metabarcoding promoted by high-throughput sequencing technologies are generating millions of sequences in a fast way, with a promising cost reduction, and overcoming some difficulties of the traditional taxonomic approaches. This paper provides an updated broad perspective of the current developments in this dynamic field presented in the special session (SS) "The use of molecular tools in ecological and biodiversity assessment of aquatic ecosystems" of the XIX Congress of the Iberian Association of Limnology (AIL2018), held in Coimbra, Portugal.Developments presented are mainly focused on the Iberian Peninsula (Portugal and Spain, including Atlantic Macaronesian islands) but include studies in France, Germany, Finland, Russia (Siberia) and South America. The networks within which these researchers are involved are yet even broader, profiting from existing molecular facilities, and traditional taxonomic expertise, which can be viewed as a characteristic of this new research area. It was evident in the SS that the use of molecular tools is widespread, being used to study a diversity of aquatic systems, from rivers' headwaters to estuaries and coastal lagoons, and volcanic, mountain and frozen lakes to hot springs. The organisms targeted are likewise varied and include fish, macroinvertebrates, meiofauna, microalgae such as diatoms and dinoflagellates, other protists, fungi, and bacteria (cyanobacteria and other). Some studies address the whole biodiversity (i.e., all species present independently of the taxonomic group) from environmental samples of water, biofilms and preservative solution from field samples (e.g., ethanol from macroinvertebrate samples).     Prospective observational cohort study on grading the severity of postoperative complications in global surgery research

    Background The Clavien–Dindo classification is perhaps the most widely used approach for reporting postoperative complications in clinical trials. This system classifies complication severity by the treatment provided. However, it is unclear whether the Clavien–Dindo system can be used internationally in studies across differing healthcare systems in high- (HICs) and low- and middle-income countries (LMICs). Methods This was a secondary analysis of the International Surgical Outcomes Study (ISOS), a prospective observational cohort study of elective surgery in adults. Data collection occurred over a 7-day period. Severity of complications was graded using Clavien–Dindo and the simpler ISOS grading (mild, moderate or severe, based on guided investigator judgement). Severity grading was compared using the intraclass correlation coefficient (ICC). Data are presented as frequencies and ICC values (with 95 per cent c.i.). The analysis was stratified by income status of the country, comparing HICs with LMICs. Results A total of 44 814 patients were recruited from 474 hospitals in 27 countries (19 HICs and 8 LMICs). Some 7508 patients (16·8 per cent) experienced at least one postoperative complication, equivalent to 11 664 complications in total. Using the ISOS classification, 5504 of 11 664 complications (47·2 per cent) were graded as mild, 4244 (36·4 per cent) as moderate and 1916 (16·4 per cent) as severe. Using Clavien–Dindo, 6781 of 11 664 complications (58·1 per cent) were graded as I or II, 1740 (14·9 per cent) as III, 2408 (20·6 per cent) as IV and 735 (6·3 per cent) as V. Agreement between classification systems was poor overall (ICC 0·41, 95 per cent c.i. 0·20 to 0·55), and in LMICs (ICC 0·23, 0·05 to 0·38) and HICs (ICC 0·46, 0·25 to 0·59). Conclusion Caution is recommended when using a treatment approach to grade complications in global surgery studies, as this may introduce bias unintentionally

    The surgical safety checklist and patient outcomes after surgery: a prospective observational cohort study, systematic review and meta-analysis

    © 2017 British Journal of Anaesthesia Background: The surgical safety checklist is widely used to improve the quality of perioperative care. However, clinicians continue to debate the clinical effectiveness of this tool. Methods: Prospective analysis of data from the International Surgical Outcomes Study (ISOS), an international observational study of elective in-patient surgery, accompanied by a systematic review and meta-analysis of published literature. The exposure was surgical safety checklist use. The primary outcome was in-hospital mortality and the secondary outcome was postoperative complications. In the ISOS cohort, a multivariable multi-level generalized linear model was used to test associations. To further contextualise these findings, we included the results from the ISOS cohort in a meta-analysis. Results are reported as odds ratios (OR) with 95% confidence intervals. Results: We included 44 814 patients from 497 hospitals in 27 countries in the ISOS analysis. There were 40 245 (89.8%) patients exposed to the checklist, whilst 7508 (16.8%) sustained ≥1 postoperative complications and 207 (0.5%) died before hospital discharge. Checklist exposure was associated with reduced mortality [odds ratio (OR) 0.49 (0.32–0.77); P\u3c0.01], but no difference in complication rates [OR 1.02 (0.88–1.19); P=0.75]. In a systematic review, we screened 3732 records and identified 11 eligible studies of 453 292 patients including the ISOS cohort. Checklist exposure was associated with both reduced postoperative mortality [OR 0.75 (0.62–0.92); P\u3c0.01; I2=87%] and reduced complication rates [OR 0.73 (0.61–0.88); P\u3c0.01; I2=89%). Conclusions: Patients exposed to a surgical safety checklist experience better postoperative outcomes, but this could simply reflect wider quality of care in hospitals where checklist use is routine
