    Near-infrared evolution of the equatorial ring of SN 1987A

    We use adaptive-optics imaging and integral field spectroscopy from the Very Large Telescope, together with images from the \emph{Hubble Space Telescope}, to study the near-infrared (NIR) evolution of the equatorial ring (ER) of SN~1987A. We study the NIR line and continuum flux and morphology over time in order to lay the groundwork for \emph{James Webb Space Telescope} observations of the system. We also study the differences in the interacting ring structure and flux between optical, NIR and other wavelengths, and between line and continuum emission, to constrain the underlying physical processes. Mostly the evolution is similar in the NIR and optical. The morphology of the ER has been skewed toward the west side (with roughly 2/3 of the NIR emission originating there) since around 2010. A steady decline in the ER flux, broadly similar to the MIR and the optical, is ongoing since roughly this time as well. The expansion velocity of the ER hotspots in the NIR is fully consistent with the optical. However, continuum emission forms roughly 70 per cent of the NIR luminosity, and is relatively stronger outside the hotspot-defined extent of the ER than the optical emission or NIR line emission since 2012--2013, suggesting a faster-expanding continuum component. We find that this outer NIR emission can have a significant synchrotron contribution. Even if emission from hot (\sim2000~K) dust is dominant within the ER, the mass of this dust must be vanishingly small (a few ×1012\times10^{-12}~M_\odot) compared to the total dust mass in the ER (105\gtrsim10^{-5}~M_\odot) to account for the observed HKsHKs flux. The NIR continuum emission, however, expands slower than the more diffuse 180-K dust emission that dominates in the MIR, indicating a different source, and the same hot dust component cannot account for the JJ-band emission.Comment: 17 pages, 17 figures.     Tiivistelmä. Tämän pro gradu -tutkielman tarkoituksena on tutkia yhden suuren yhtenäisperuskoulun pelastussuunnitelman vastaavuuta suhteessa lakiin kirjattuihin sisältövaatimuksiin. Lisäksi tarkoituksena on selvittää saman tutkimukseen osallistuneen koulun turvallisuusvastaavan ja opettajien suhtautumista oman koulun turvallisuuskulttuuriin. Turvallisuuskulttuuria tarkastellaan pelastussuunnitelmaan perehtymisen, turvallisuusjohtamisen, turvallisuuskasvatuksen ja turvallisuuskulttuurin ylläpidon näkökulmista. Tutkimuksessa on käytetty fenomenografista tutkimusotetta. Turvallisuusvastaavan ja opettajien suhtautumista, asenteita ja mielipiteitä kuvaava aineisto on kerätty sähköistä kyselylomaketta apuna käyttäen. Kyselylomake sisälsi neljätoista avointa kysymystä sekä kyllä/ei-väittämiä. Lomake lähetettiin turvallisuusvastaavan lisäksi 55 opettajalle. Opettajien vastauksia kertyi yksitoista. Tutkimusvastaukset analysoitiin käyttäen fenomenografiseen tutkimusotteeseen soveltuvaa aineistolähtöistä analyysimenetelmää. Vastauksista tehtiin tulkintoja opettajien suhtautumisesta turvallisuuskulttuuriin asteikolla positiivinen-neutraali-negatiivinen. Tutkimustulosten mukaan tutkimukseen osallistuneen koulun pelastussuunnitelma on sisällöllisestä laadukkuudesta huolimatta enemmän linjassa säädösten kanssa, jotka on laadittu ennen uusinta pelastuslakia. Tuloksista käy myös ilmi, että opettajien suhtautuminen koulun turvallisuuskulttuuriin on pääosin positiivista ja neutraalia. Positiiviset vastaukset kuvastavat vahvaa luottamusta koulun turvallisuuteen ja turvallisuutta edistäviin toimiin. Neutraaleista vastauksista puolestaan heijastuu opettajien välinpitämättömyys ja tietämättömyys turvallisuusasioita koskien. Negatiivista suhtautumista sisältävien vastausten mukaan turvallisuuskasvatus tulisi huomioida opettajankoulutuksessa nykyistä paremmin. Tutkimuksen luotettavuuteen on pyritty kuvaamalla avoimesti ja tarkasti tutkimuksen eri vaiheet, menetelmät ja tulokset. Luotettavuutta lisää myös se, että vastaajat olivat entuudestaan tuntemattomia, eikä heitä ole voitu johdatella vastaamaan tietyllä tavalla. Tutkimustulosten todenmukaisuutta ajatellen sekä koululle että kaikille vastaajille on taattu täysi anonymiteetti. Tutkimuksen tarkoituksena ei ole perinteisesti ajateltuna yleistettävän tiedon luominen koulujen turvallisuuskulttuurista. Tutkimustuloksilla voi olla sosiokulttuurisesti laajempaa merkitystä, koska koululaitoksissa useiden lakien ja asetusten määrittämä turvallisuus on opetuksen järjestämisessä kaiken lähtökohta

    Waist‑height ratio and waist are the best estimators of visceral fat in type 1 diabetes

    Visceral fat is associated with cardiovascular and kidney disease. However, the relationship between body composition and anthropometric measures in type 1 diabetes is unknown. Using z-statistics, we ranked the ability of body mass index (BMI), waist circumference (WC), waist-hip ratio (WHR), waist-height ratio (WHtR) and a body shape index (ABSI) to capture measures of body composition from 603 Dual-energy-X-Ray-Absorptiometry scans of adults with type 1 diabetes. Albuminuria was defined as urinary albumin excretion rate of at least 30 mg/24 h. Women with albuminuria had higher visceral fat mass % (VFM%) (0.9 vs. 0.5%, p = 0.0017) and lower appendicular lean mass % (AppLM%) (25.4 vs 26.4%, p = 0.03) than those without. Men with albuminuria had higher VFM% (1.5 vs. 1.0%, p = 0.0013) and lower AppLM% (30.0 vs 32.3, p < 0.0001) than those without. In men, WHtR estimated VFM% best (z-statistics = 21.1), followed by WC (z = 19.6), BMI (z = 15.1), WHR (z = 14.6) and ABSI (z = 10.1). In women, the ranking was WC (z = 28.9), WHtR (z = 27.3), BMI (z = 20.5), WHR (z = 12.7) and ABSI (z = 10.5). Overall, the ranking was independent of albuminuria. Adults with type 1 diabetes and albuminuria have greater VFM% and lower AppLM% than those without. WHtR and WC best estimate the VFM% in this population, independently of albuminuria and sex.Peer reviewe

    Obatoclax Inhibits Alphavirus Membrane Fusion by Neutralizing the Acidic Environment of Endocytic Compartments

    As new pathogenic viruses continue to emerge, it is paramount to have intervention strategies that target a common denominator in these pathogens. The fusion of viral and cellular membranes during viral entry is one such process that is used by many pathogenic viruses, including chikungunya virus, West Nile virus, and influenza virus. Obatoclax, a small-molecule antagonist of the Bcl-2 family of proteins, was previously determined to have activity against influenza A virus and also Sindbis virus. Here, we report it to be active against alphaviruses, like chikungunya virus (50% effective concentration [EC50] = 0.03 mu M) and Semliki Forest virus (SFV; EC50 = 0.11 mu M). Obatoclax inhibited viral entry processes in an SFV temperaturesensitive mutant entry assay. A neutral red retention assay revealed that obatoclax induces the rapid neutralization of the acidic environment of endolysosomal vesicles and thereby most likely inhibits viral fusion. Characterization of escape mutants revealed that the L369I mutation in the SFV E1 fusion protein was sufficient to confer partial resistance against obatoclax. Other inhibitors that target the Bcl-2 family of antiapoptotic proteins inhibited neither viral entry nor endolysosomal acidification, suggesting that the antiviral mechanism of obatoclax does not depend on its anticancer targets. Obatoclax inhibited the growth of flaviviruses, like Zika virus, West Nile virus, and yellow fever virus, which require low pH for fusion, but not that of pH-independent picornaviruses, like coxsackievirus A9, echovirus 6, and echovirus 7. In conclusion, obatoclax is a novel inhibitor of endosomal acidification that prevents viral fusion and that could be pursued as a potential broad-spectrum antiviral candidate.Peer reviewe

    CHARACTERISATION AND IN VITRO AND IN VIVO EVALUATION OF SUPERCRITICAL-CO2-FOAMED B-TCP/PLCL COMPOSITES FOR BONE APPLICATIONS

    Most synthetic bone grafts are either hard and brittle ceramics or paste-like materials that differ in applicability from the gold standard autologous bone graft, which restricts their widespread use. Therefore, the aim of the study was to develop an elastic, highly porous and biodegradable beta-tricalciumphosphate/poly(L-lactide-co-epsilon-caprolactone) (beta-TCP/PLCL) composite for bone applications using supercritical CO2 foaming. Ability to support osteogenic differentiation was tested in human adipose stem cell (hASC) culture for 21 d. Biocompatibility was evaluated for 24 weeks in a rabbit femur-defect model. Foamed composites had a high ceramic content (50 wt%) and porosity (65-67 %). After 50 % compression, in an aqueous environment at 37 degrees C, tested samples returned to 95 % of their original height. Hydrolytic degradation of beta-TCP/PLCL composite, during the 24-week follow-up, was very similar to that of porous PLCL scaffold both in vitro and in vivo. Osteogenic differentiation of hASCs was demonstrated by alkaline phosphatase activity analysis, alizarin red staining, soluble collagen analysis, immunocytochemical staining and qRT-PCR. In vitro, hASCs formed a pronounced mineralised collagen matrix. A rabbit femur defect model confirmed biocompatibility of the composite. According to histological Masson-Goldner's trichrome staining and micro-computed tomography, beta-TCP/PLCL composite did not elicit infection, formation of fibrous capsule or cysts. Finally, native bone tissue at 4 weeks was already able to grow on and in the beta-TCP/PLCL composite. The elastic and highly porous beta-TCP/PLCL composite is a promising bone substitute because it is osteoconductive and easy-to-use and mould intraoperatively.Peer reviewe

    Weight Loss Trajectories in Healthy Weight Coaching : Cohort Study

    Background: As global obesity prevalence continues to increase, there is a need for accessible and affordable weight management interventions, such as web-based programs. Objective: This paper aims to assess the outcomes of healthy weight coaching (HWC), a web-based obesity management program integrated into standard Finnish clinical care. Methods: HWC is an ongoing, structured digital 12-month program based on acceptance and commitment therapy. It includes weekly training sessions focused on lifestyle, general health, and psychological factors. Participants received remote one-on-one support from a personal coach. In this real-life, single-arm, prospective cohort study, we examined the total weight loss, weight loss profiles, and variables associated with weight loss success and program retention in 1189 adults (963 women) with a BMI >25 kg/m(2) among participants of the program between October 2016 and March 2019. Absolute (kg) and relative (%) weight loss from the baseline were the primary outcomes. We also examined the weight loss profiles, clustered based on the dynamic time-warping distance, and the possible variables associated with greater weight loss success and program retention. We compared different groups using the Mann-Whitney test or Kruskal-Wallis test for continuous variables and the chi-squared test for categorical variables. We analyzed changes in medication using the McNemar test. Results: Among those having reached the 12-month time point (n=173), the mean weight loss was 4.6% (SE 0.5%), with 43% (n=75) achieving clinically relevant weight loss (>= 5%). Baseline BMI >= 40 kg/m(2) was associated with a greater weight loss than a lower BMI (mean 6.6%, SE 0.9%, vs mean 3.2%, SE 0.6%; P=.02). In addition, more frequent weight reporting was associated with greater weight loss. No significant differences in weight loss were observed according to sex, age, baseline disease, or medication use. The total dropout rate was 29.1%. Dropouts were slightly younger than continuers (47.2, SE 0.6 years vs 49.2, SE 0.4 years; P=.01) and reported their weight less frequently (3.0, SE 0.1 entries per month vs 3.3, SE 0.1 entries per month; P Conclusions: A comprehensive web-based program such as HWC is a potential addition to the repertoire of obesity management in a clinical setting. Heavier patients lost more weight, but weight loss success was otherwise independent of baseline characteristics.Peer reviewe

    Ibuprofen-loaded calcium phosphate granules : combination of innovative characterization methods to relate mechanical strength to drug location

    This paper studies the impact of the location of a drug substance on the physicochemical and mechanical properties of two types of calcium phosphate granules loaded with seven different contents of ibuprofen, ranging from 1.75% to 46%. These implantable agglomerates were produced by either low or high shear granulation. Unloaded Mi-Pro pellets presented higher sphericity and mechanical properties, but were slightly less porous than Kenwood granules (57.7% vs 61.2%). Nevertheless, the whole expected quantity of ibuprofen could be integrated into both types of granules. A combination of surface analysis, using near-infrared (NIR) spectroscopy coupling chemical imaging, and pellet porosity, by mercury intrusion measurements, allowed ibuprofen to be located. It was shown that, from 0% to 22% drug content, ibuprofen deposited simultaneously on the granule surface, as evidenced by the increase in surface NIR signal, and inside the pores, as highlighted by the decrease in pore volume. From 22%, porosity was almost filled, and additional drug substance coated the granule surfaces, leading to a large increase in the surface NIR signal. This coating was more regular for Mi-Pro pellets owing to their higher sphericity and greater surface deposition of drug substance. Unit crush tests using a microindenter revealed that ibuprofen loading enhanced the mechanical strength of granules, especially above 22% drug content, which was favorable to further application of the granules as a bone defect filler

    Adaptive responses of animals to climate change are most likely insufficient

    Biological responses to climate change have been widely documented across taxa and regions, but it remains unclear whether species are maintaining a good match between phenotype and environment, i.e. whether observed trait changes are adaptive. Here we reviewed 10,090 abstracts and extracted data from 71 studies reported in 58 relevant publications, to assess quantitatively whether phenotypic trait changes associated with climate change are adaptive in animals. A meta-analysis focussing on birds, the taxon best represented in our dataset, suggests that global warming has not systematically affected morphological traits, but has advanced phenological traits. We demonstrate that these advances are adaptive for some species, but imperfect as evidenced by the observed consistent selection for earlier timing. Application of a theoretical model indicates that the evolutionary load imposed by incomplete adaptive responses to ongoing climate change may already be threatening the persistence of species
