    Discursive design thinking: the role of explicit knowledge in creative architectural design reasoning

    The main hypothesis investigated in this paper is based upon the suggestion that the discursive reasoning in architecture supported by an explicit knowledge of spatial configurations can enhance both design productivity and the intelligibility of design solutions. The study consists of an examination of an architect’s performance while solving intuitively a well-defined problem followed by an analysis of the spatial structure of their design solutions. One group of architects will attempt to solve the design problem logically, rationalizing their design decisions by implementing their explicit knowledge of spatial configurations. The other group will use an implicit form of such knowledge arising from their architectural education to reason about their design acts. An integrated model of protocol analysis combining linkography and macroscopic coding is used to analyze the design processes. The resulting design outcomes will be evaluated quantitatively in terms of their spatial configurations. The analysis appears to show that an explicit knowledge of the rules of spatial configurations, as possessed by the first group of architects can partially enhance their function-driven judgment producing permeable and well-structured spaces. These findings are particularly significant as they imply that an explicit rather than an implicit knowledge of the fundamental rules that make a layout possible can lead to a considerable improvement in both the design process and product.     Silicon photonics offers tremendous potential for inexpensive high-yield photonic-electronic integration. Besides conventional dielectric waveguides, plasmonic structures can also be efficiently realized on the silicon photonic platform, reducing device footprint by more than an order of magnitude.     Volumetric modulated arc therapy for spine SBRT patients to reduce treatment time and intrafractional motion

    Volumetric modulated arc therapy (VMAT) is an efficient technique to reduce the treatment time and intrafractional motion to treat spine patients presented with severe back pain. Five patients treated with spine stereotactic body radiation therapy (SBRT) using 9 beams intensity modulated radiation therapy (IMRT) were retrospectively selected for this study. The patients were replanned using two arcs VMAT technique. The average mean dose was 104% ± 1.2% and 104.1% ± 1.0% in IMRT and VMAT, respectively (p = 0.9). Accordingly, the average conformal index (CI) was 1.3 ± 0.1 and 1.5 ± 0.3, respectively (p = 0.5). The average dose gradient (DG) distance was 1.5 ± 0.1 cm and 1.4 ± 0.1 cm, respectively (p = 0.3). The average spinal cord maximum dose was 11.6 ± 1.0 Gy and 11.8 ± 1.1 Gy (p = 0.8) and V10Gy was 7.4 ± 1.4 cc and 8.6 ± 1.7 cc (p = 0.4) for IMRT and VMAT, respectively. Accordingly, the average number of monitor units (MUs) was 6771.7 ± 1323.3 MU and 3978 ± 576.7 MU respectively (p = 0.02).     Automatic recognition of feeding and foraging behaviour in pigs using deep learning

    Highlights• An automated detection method of pig feeding and foraging behaviour was developed.• The automated method is based on convolutional deep neural networks.• The automated method does not rely on pig tracking to estimate behaviours.• Detection of feeding behaviour is highly accurate (99.4%) and fast (0.02 sec/image).• The robust method can be applied under different husbandry/ management conditions.Automated, vision-based early warning systems have been developed to detect behavioural changes in groups of pigs to monitor their health and welfare status. In commercial settings, automatic recording of feeding behaviour remains a challenge due to problems of variation in illumination, occlusions and similar appearance of different pigs. Additionally, such systems, which rely on pig tracking, often overestimate the actual time spent feeding, due to the inability to identify and/or exclude non-nutritive visits (NNV) to the feeding area. To tackle these problems, we have developed a robust, deep learning-based feeding detection method that (a) does not rely on pig tracking and (b) is capable of distinguishing between feeding and NNV for a group of pigs. We first validated our method using video footage from a commercial pig farm, under a variety of settings. We demonstrate the ability of this automated method to identify feeding and NNV behaviour with high accuracy (99.4% ± 0.6%). We then tested the method's ability to detect changes in feeding and NNV behaviours during a planned period of food restriction. We found that the method was able to automatically quantify the expected changes in both feeding and NNV behaviours. Our method is capable of monitoring robustly and accurately the feeding behaviour of groups of commercially housed pigs, without the need for additional sensors or individual marking. This has great potential for application in the early detection of health and welfare challenges of commercial pigs

    Renal neuroendocrine control of desiccation and cold tolerance by Drosophila suzukii

    Background: Neuropeptides are central to the regulation of physiological, and behavioural processes in insects, directly impacting cold and desiccation survival. However, little is known about the control mechanisms governing these responses in D. suzukii. The close phylogenetic relationship of D. suzukii with D. melanogaster allows, through genomic and functional studies, an insight into the mechanisms directing stress tolerance in D. suzukii. Results: Capa, Leucokinin, DH44 and DH31 neuropeptides demonstrate a high level of conservation between D. suzukii and D. melanogaster with respect to peptide sequences, neuronal expression, receptor localisation, and diuretic function in the Malpighian tubules. Despite D. suzukii’s ability to populate cold environments, they proved sensitive to both cold and desiccation. Furthermore, in D. suzukii, Capa acts as a desiccation-and cold stress-responsive gene, while DH44 gene expression is increased only after desiccation exposure, and the LK gene after nonlethal cold stress recovery. Conclusion: This study provides a comparative investigation into stress tolerance mediation by neuroendocrine signalling in two Drosophila species, providing evidence that similar signalling pathways control fluid secretion in the Malpighian tubules. Identifying processes governing specific environmental stresses affecting D. suzukii could lead to the development of targeted integrated management strategies to control insect pest populations

    The Importance of pH in Regulating the Function of the Fasciola hepatica Cathepsin L1 Cysteine Protease

    The helminth parasite Fasciola hepatica secretes cathepsin L cysteine proteases to invade its host, migrate through tissues and digest haemoglobin, its main source of amino acids. Here we investigated the importance of pH in regulating the activity and functions of the major cathepsin L protease FheCL1. The slightly acidic pH of the parasite gut facilitates the auto-catalytic activation of FheCL1 from its inactive proFheCL1 zymogen; this process was ∼40-fold faster at pH 4.5 than at pH 7.0. Active mature FheCL1 is very stable at acidic and neutral conditions (the enzyme retained ∼45% activity when incubated at 37°C and pH 4.5 for 10 days) and displayed a broad pH range for activity peptide substrates and the protein ovalbumin, peaking between pH 5.5 and pH 7.0. This pH profile likely reflects the need for FheCL1 to function both in the parasite gut and in the host tissues. FheCL1, however, could not cleave its natural substrate Hb in the pH range pH 5.5 and pH 7.0; digestion occurred only at pH≤4.5, which coincided with pH-induced dissociation of the Hb tetramer. Our studies indicate that the acidic pH of the parasite relaxes the Hb structure, making it susceptible to proteolysis by FheCL1. This process is enhanced by glutathione (GSH), the main reducing agent contained in red blood cells. Using mass spectrometry, we show that FheCL1 can degrade Hb to small peptides, predominantly of 4–14 residues, but cannot release free amino acids. Therefore, we suggest that Hb degradation is not completed in the gut lumen but that the resulting peptides are absorbed by the gut epithelial cells for further processing by intracellular di- and amino-peptidases to free amino acids that are distributed through the parasite tissue for protein anabolism

    Dietary habits and supplementation practices of young women during pregnancy: an online cross-sectional survey of young mothers and health care professionals

    Background: Nutrition is a modifiable factor affecting birth outcomes, particularly in adolescent pregnancies. This study explores diet and supplementation practices, information and advice before, during and after pregnancy from the perspectives of pregnant or new young mothers and healthcare professionals. Methods: Two cross-sectional surveys used online questionnaires for young women who were currently pregnant or who had recently given birth, and health care professionals providing antenatal care. The surveys utilised a combination of question types including free text and multiple choice. Recruitment was conducted via the Tommy's website, online forums for young mothers and professional networks. Results: A total of 205 young women and 146 health care professionals were included in the study. Most young women reported taking supplements at some stage of pregnancy (93.2%), with 54.6% taking it on a daily basis. Those who reported taking supplements less than 7 days a week stated it was mainly due to forgetting. Health care professionals however reported that some young women had difficulties accessing healthy start supplements. Young women reported positive dietary changes; however a significant proportion of participants indicated that they avoided some foods unnecessarily. Avoiding or reducing foods such as red meat (22.7%), eggs (40.6%), oily fish (60.4%) and soft cheese (36.2%) is of concern. Midwife/family nurse (38.0%) was young women's current favourite information source; smartphone applications (apps) and recipe booklets were suggested by over 50% of participants as a new addition to existing services. Health care professionals reported they included nutritional information and support as part of their role; however they felt there were some gaps in knowledge and confidence. Midwives in particular suggested a lack of sufficient time and resources as a main challenge in providing adequate support
