    VALIDATION OF A 3-DIMENSIONAL VIDEO MOTION CAPTURE SYSTEM FOR DETERMINING BARBELL POWER AND VELOCITY DURING THE BENCH PRESS

PURPOSE: To determine the validity of a 3-dimensional video markerless motion capture system for determining barbell kinetics and kinematics. METHODS: Two 3-D video cameras sampling at 30 Hz and mounted on the top of a power rack were interfaced with a self-contained computer and software system, and operated with a touch screen (EliteForm, Lincoln, NE). For laboratory comparison purposes, a ceiling–mounted linear position transducer (Unimeasure, Corvallis, OR) was attached via a tether to the barbell. Data from the position transducer was sampled at 1000 Hz using a BioPac data acquisition system (Goleta, CA). Velocity (m.s-1) and power (W) were derived using LabView software (National Instruments, Austin, TX). One weight-trained male subject (age = 25 yrs, hgt = 1.75 m, BW = 82.6 kg, 1 RM = 161.0 kg) performed the barbell bench press exercise for 10 sets x 1 repetition at 30, 40, 50, 60, 70 and 80% 1 RM loads using maximal acceleration during the concentric phase. Dependent variables included peak (PV) and X̅ velocity (MV) and peak (PP) and X̅ power (MP). Linear regressions between lab-derived and 3-D video-derived data provided correlation coefficients, and regression slopes (b). Bland-Altman plots were used to determine X̅ differences, from which effect sizes (Cohen’s D) and % error for the 3-D camera system was determined. RESULTS: Lab-derived mean values for all loads ranged as follows; MV = 0.36 – 1.00 m.s-1, PV = 0.47 – 1.60 m.s-1, MP = 460.9 – 621.6 W, and PP = 619.9 – 1055.6 W. CONCLUSION: The 3-D video markerless motion capture system provided accurate and valid barbell velocity and power data for the bench press exercise. Supported in part by Nebraska Global LL

    Abstract-In considering the reliability of numerical programs, it is normal to "limit our study to the semantics dealing with numerical precision" (Martel, 2005). On the other hand, there is a great deal of work on the reliability of programs that essentially ignores the numerics. The thesis of this paper is that there is a class of problems that fall between these two, which could be described as "does the lowlevel arithmetic implement the high-level mathematics". Many of these problems arise because mathematics, particularly the mathematics of the complex numbers, is more dif¿cult than expected: for example the complex function log is not continuous, writing down a program to compute an inverse function is more complicated than just solving an equation, and many algebraic simpli¿cation rules are not universally valid. The good news is that these problems are theoretically capable of being solved, and are practically close to being solved, but not yet solved, in several real-world examples. However, there is still a long way to go before implementations match the theoretical possibilities

    Animating the Carbon Cycle

The role of living organisms has been accounted for, but the focus has traditionally been on contributions of plants and microbes. We develop the case that fully “animating” the carbon cycle requires broader consideration of the functional role of animals in mediating biogeochemical processes and quantification of their effects on carbon storage and exchange among terrestrial and aquatic reservoirs and the atmosphere. To encourage more hypothesis-driven experimental research that quantifies animal effects we discuss the mechanisms by which animals may affect carbon exchanges and storage within and among ecosystems and the atmosphere. We illustrate how those mechanisms lead to multiplier effects whose magnitudes may rival those of more traditional carbon storage and exchange rate estimates currently used in the carbon budget. Many animal species are already directly managed. Thus improved quantitative understanding of their influence on carbon budgets may create opportunity for management and policy to identify and implement new options for mitigating CO2 release at regional scales.US National Science FoundationNERCBBSRCNippon Foundatio

    A “piano movers” problem reformulated

    Abstract-It has long been known that cylindrical algebraic decompositions (CADs) can in theory be used for robot motion planning. However, in practice even the simplest examples can be too complicated to tackle. We consider in detail a "Piano Mover's Problem" which considers moving an infinitesimally thin piano (or ladder) through a right-angled corridor. Producing a CAD for the original formulation of this problem is still infeasible after 25 years of improvements in both CAD theory and computer hardware. We review some alternative formulations in the literature which use differing levels of geometric analysis before input to a CAD algorithm. Simpler formulations allow CAD to easily address the question of the existence of a path. We provide a new formulation for which both a CAD can be constructed and from which an actual path could be determined if one exists, and analyse the CADs produced using this approach for variations of the problem. This emphasises the importance of the precise formulation of such problems for CAD. We analyse the formulations and their CADs considering a variety of heuristics and general criteria, leading to conclusions about tackling other problems of this form

    Expression of UDP-glucuronosyltransferase 1A4 in human placenta at term

    The placenta contains a large variety of metabolizing enzymes, among them UDP-glucuronosyltransferase (UGT). Several UGT2B isozymes have so far been detected in human placenta, but little is known on placental expression of UGT1A isozymes. The antiepileptic drug lamotrigine (LTG) is a UGT1A4-substrate, and its serum concentration falls by over 50% during pregnancy, leading to impaired seizure control. The placenta may be involved in this. Microsomes from term placentas of 4 LTG-users and 10 healthy control subjects were prepared. Western blot analysis detected UGT1A proteins in all placentas. The presence of UGT1A4 in placenta from LTG users was confirmed with UGT1A4 commercial standard and a specific UGT1A4 primary antibody. Since LTG is primarily metabolized by UGT1A4 and this isozyme is shown to be present in placenta at term, it may be hypothesized that the placenta is involved in the fall of LTG serum concentrations during pregnancy

    Selected reactive oxygen species and antioxidant enzymes in common bean after Pseudomonas syringae pv. phaseolicola and Botrytis cinerea infection

    Phaseolus vulgaris cv. Korona plants were inoculated with the bacteria Pseudomonas syringae pv. phaseolicola (Psp), necrotrophic fungus Botrytis cinerea (Bc) or with both pathogens sequentially. The aim of the experiment was to determine how plants cope with multiple infection with pathogens having different attack strategy. Possible suppression of the non-specific infection with the necrotrophic fungus Bc by earlier Psp inoculation was examined. Concentration of reactive oxygen species (ROS), such as superoxide anion (O2 -) and H2O2 and activities of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) were determined 6, 12, 24 and 48 h after inoculation. The measurements were done for ROS cytosolic fraction and enzymatic cytosolic or apoplastic fraction. Infection with Psp caused significant increase in ROS levels since the beginning of experiment. Activity of the apoplastic enzymes also increased remarkably at the beginning of experiment in contrast to the cytosolic ones. Cytosolic SOD and guaiacol peroxidase (GPOD) activities achieved the maximum values 48 h after treatment. Additional forms of the examined enzymes after specific Psp infection were identified; however, they were not present after single Bc inoculation. Subsequent Bc infection resulted only in changes of H2O2 and SOD that occurred to be especially important during plant–pathogen interaction. Cultivar Korona of common bean is considered to be resistant to Psp and mobilises its system upon infection with these bacteria. We put forward a hypothesis that the extent of defence reaction was so great that subsequent infection did not trigger significant additional response

    Predictors of linkage to care following community-based HIV counseling and testing in rural Kenya

    Despite innovations in HIV counseling and testing (HCT), important gaps remain in understanding linkage to care. We followed a cohort diagnosed with HIV through a community-based HCT campaign that trained persons living with HIV/AIDS (PLHA) as navigators. Individual, interpersonal, and institutional predictors of linkage were assessed using survival analysis of self-reported time to enrollment. Of 483 persons consenting to follow-up, 305 (63.2%) enrolled in HIV care within 3 months. Proportions linking to care were similar across sexes, barring a sub-sample of men aged 18–25 years who were highly unlikely to enroll. Men were more likely to enroll if they had disclosed to their spouse, and women if they had disclosed to family. Women who anticipated violence or relationship breakup were less likely to link to care. Enrolment rates were significantly higher among participants receiving a PLHA visit, suggesting that a navigator approach may improve linkage from community-based HCT campaigns.Vestergaard Frandse

    FAK acts as a suppressor of RTK-MAP kinase signalling in Drosophila melanogaster epithelia and human cancer cells

    Receptor Tyrosine Kinases (RTKs) and Focal Adhesion Kinase (FAK) regulate multiple signalling pathways, including mitogen-activated protein (MAP) kinase pathway. FAK interacts with several RTKs but little is known about how FAK regulates their downstream signalling. Here we investigated how FAK regulates signalling resulting from the overexpression of the RTKs RET and EGFR. FAK suppressed RTKs signalling in Drosophila melanogaster epithelia by impairing MAPK pathway. This regulation was also observed in MDA-MB-231 human breast cancer cells, suggesting it is a conserved phenomenon in humans. Mechanistically, FAK reduced receptor recycling into the plasma membrane, which resulted in lower MAPK activation. Conversely, increasing the membrane pool of the receptor increased MAPK pathway signalling. FAK is widely considered as a therapeutic target in cancer biology; however, it also has tumour suppressor properties in some contexts. Therefore, the FAK-mediated negative regulation of RTK/MAPK signalling described here may have potential implications in the designing of therapy strategies for RTK-driven tumours
