    Integrated and Sustainable Management of Post-industrial Coasts

    The sustainable management of post-industrial coasts is a major emerging issue globally. Along such coasts, there may be a significant legacy of both contaminated land (including historic landfills and non-managed waste disposal) and contaminated sediments in and around urban and industrial areas, which require new strategies for cost-effective and integrated risk management under future sea-level rise and climate change scenarios. Here, we review current approaches to managing contamination in post-industrial coastlines, discuss emerging integrated management strategies (building on low input approaches to sustainable brownfields regeneration) and present an approach and framework for assessing and comparing different scenarios for coastal brownfield regeneration to soft re-use and other end-points. This framework can be applied to explore the opportunities for synergy and realization of wider environmental, economic and societal benefits between coastal protection, dredged material re-use and the management of brownfield land. As such, the approach we propose supports planning and options appraisal to realize maximum benefit and value from integrated coastal management strategies

    Bound states of spin-half particles in a static gravitational field close to the black hole field

    We consider the bound-state energy levels of a spin-1/2 fermion in the gravitational field of a near-black hole object. In the limit that the metric of the body becomes singular, all binding energies tend to the rest-mass energy (i.e. total energy approaches zero). We present calculations of the ground state energy for three specific interior metrics (Florides, Soffel and Schwarzschild) for which the spectrum collapses and becomes quasi-continuous in the singular metric limit. The lack of zero or negative energy states prior to this limit being reached prevents particle pair production occurring. Therefore, in contrast to the Coulomb case, no pairs are produced in the non-singular static metric. For the Florides and Soffel metrics the singularity occurs in the black hole limit, while for the Schwarzschild interior metric it corresponds to infinite pressure at the centre. The behaviour of the energy level spectrum is discussed in the context of the semi-classical approximation and using general properties of the metric.Comment: 16 pages, 6 Figures.     Spherical perspective

    We survey the present state of spherical perspective, regarding both mathematical structure and drawing practice, with a view to applications in the visual arts. We define a spherical perspective as the entailment of a conical anamorphosis with a compact flattening of the visual sphere. We examine a general framework for solving spherical perspectives, exemplified with the azimuthal equidistant (“fisheye”) and equirectangular cases. We consider the relation between spherical and curvilinear perspectives. We briefly discuss computer renderings but focus on methods adapted to freehand sketching or technical drawing with simple instruments such as ruler and compass. We discuss how handmade spherical perspective drawings can generate immersive anamorphoses, which can be rendered as virtual reality panoramas, leading to hybrid visual creations that bridge the gap between traditional drawing and digital environments.info:eu-repo/semantics/publishedVersio

    Sialylation on O-linked glycans protects von Willebrand factor from macrophage galactose lectin mediated clearance

    Terminal sialylation determines the plasma half-life of von Willebrand factor (VWF). A role for macrophage galactose lectin (MGL) in regulating hyposialylated VWF clearance has recently been proposed. In this study, we showed that MGL influences physiological plasma VWF clearance. MGL inhibition was associated with a significantly extended mean residence time and 3-fold increase in endogenous plasma VWF antigen levels (P<0.05). Using a series of VWF truncations, we further demonstrated that the A1 domain of VWF is predominantly responsible for enabling the MGL interaction. Binding of both full-length and VWF-A1-A2-A3 to MGL was significantly enhanced in the presence of ristocetin (P<0.05), suggesting that the MGL-binding site in A1 is not fully accessible in globular VWF. Additional studies using different VWF glycoforms demonstrated that VWF O-linked glycans, clustered at either end of the A1 domain, play a key role in protecting VWF against MGLmediated clearance. Reduced sialylation has been associated with pathological, increased clearance of VWF in patients with von Willebrand disease. Herein, we demonstrate that specific loss of α2-3 linked sialylation from O-glycans results in markedly increased MGL-binding in vitro, and markedly enhanced MGL-mediated clearance of VWF in vivo. Our data further show that the asialoglycoprotein receptor (ASGPR) does not have a significant role in mediating the increased clearance of VWF following loss of O-sialylation. Conversely however, we observed that loss of N-linked sialylation from VWF drives enhanced circulatory clearance predominantly via the ASGPR. Collectively, our data support the hypothesis that in addition to regulating physiological VWF clearance, the MGL receptor works in tandem with ASGPR to modulate enhanced clearance of aberrantly sialylated VWF in the pathogenesis of von Willebrand disease

    First trimester diagnosis and screening for fetal aneuploidy

    Maternal serum screening for neural tube defects and fetal aneuploidy in the second trimester has been incorporated into obstetrical practice over the past two decades. Now, as a result of several multicenter trials, first trimester screening between 11 and 14 weeks has been shown to be an effective and reliable screening test for Down syndrome and trisomy 18. Benefits of first trimester screening include earlier identification of the pregnancy at risk for fetal aneuploidy and anatomic defects, in particular, cardiac anomalies, and the option of earlier diagnosis by chorionic villus sampling, if available. This policy updates the American College of Medical Genetics policy statement entitled Second Trimester Maternal Serum Screening for Fetal Open Neural Tube Defects and Aneuploidy (2004) and complements the sections of American College of Medical Genetic’s Standards and Guidelines for Clinical Genetics Laboratories entitled “Prenatal screening for Down syndrome that includes first trimester biochemistry and/or ultrasound measurements.

    Universal growth scheme for quantum dots with low fine-Structure splitting at various emission wavelengths

    Efficient sources of individual pairs of entangled photons are required for quantum networks to operate using fibre optic infrastructure. Entangled light can be generated by quantum dots (QDs) with naturally small fine-structure-splitting (FSS) between exciton eigenstates. Moreover, QDs can be engineered to emit at standard telecom wavelengths. To achieve sufficient signal intensity for applications, QDs have been incorporated into 1D optical microcavities. However, combining these properties in a single device has so far proved elusive. Here, we introduce a growth strategy to realise QDs with small FSS in the conventional telecom band, and within an optical cavity. Our approach employs droplet-epitaxy of InAs quantum dots on (001) substrates. We show the scheme improves the symmetry of the dots by 72%. Furthermore, our technique is universal, and produces low FSS QDs by molecular beam epitaxy on GaAs emitting at ~900nm, and metal-organic vapour phase epitaxy on InP emitting at 1550 nm, with mean FSS 4x smaller than for Stranski-Krastanow QDs

    TOM40 Mediates Mitochondrial Dysfunction Induced by α-Synuclein Accumulation in Parkinson's Disease.

    Alpha-synuclein (α-Syn) accumulation/aggregation and mitochondrial dysfunction play prominent roles in the pathology of Parkinson's disease. We have previously shown that postmortem human dopaminergic neurons from PD brains accumulate high levels of mitochondrial DNA (mtDNA) deletions. We now addressed the question, whether alterations in a component of the mitochondrial import machinery -TOM40- might contribute to the mitochondrial dysfunction and damage in PD. For this purpose, we studied levels of TOM40, mtDNA deletions, oxidative damage, energy production, and complexes of the respiratory chain in brain homogenates as well as in single neurons, using laser-capture-microdissection in transgenic mice overexpressing human wildtype α-Syn. Additionally, we used lentivirus-mediated stereotactic delivery of a component of this import machinery into mouse brain as a novel therapeutic strategy. We report here that TOM40 is significantly reduced in the brain of PD patients and in α-Syn transgenic mice. TOM40 deficits were associated with increased mtDNA deletions and oxidative DNA damage, and with decreased energy production and altered levels of complex I proteins in α-Syn transgenic mice. Lentiviral-mediated overexpression of Tom40 in α-Syn-transgenic mice brains ameliorated energy deficits as well as oxidative burden. Our results suggest that alterations in the mitochondrial protein transport machinery might contribute to mitochondrial impairment in α-Synucleinopathies

    Transcriptome analysis of the central nervous system of the mollusc Lymnaea stagnalis

    Background: The freshwater snail Lymnaea stagnalis (L. stagnalis) has served as a successful model for studies in the field of Neuroscience. However, a serious drawback in the molecular analysis of the nervous system of L. stagnalis has been the lack of large-scale genomic or neuronal transcriptome information, thereby limiting the use of this unique model. Results: In this study, we report 7,712 distinct EST sequences (median length: 847 nucleotides) of a normalized L. stagnalis central nervous system (CNS) cDNA library, resulting in the largest collection of L. stagnalis neuronal transcriptome data currently available. Approximately 42% of the cDNAs can be translated into more than 100 consecutive amino acids, indicating the high quality of the library. The annotated sequences contribute 12% of the predicted transcriptome size of 20,000. Surprisingly, approximately 37% of the L. stagnalis sequences only have a tBLASTx hit in the EST library of another snail species Aplysia californica (A. californica) even using a low stringency e-value cutoff at 0.01. Using the same cutoff, approximately 67% of the cDNAs have a BLAST hit in the NCBI non-redundant protein and nucleotide sequence databases (nr and nt), suggesting that one third of the sequences may be unique to L. stagnalis. Finally, using the same cutoff (0.01), more than half of the cDNA sequences (54%) do not have a hit in nematode, fruitfly or human genome data, suggesting that the L. stagnalis transcriptome is significantly different from these species as well. The cDNA sequences are enriched in the following gene ontology functional categories: protein binding, hydrolase, transferase, and catalytic enzymes. Conclusion: This study provides novel molecular insights into the transcriptome of an important molluscan model organism. Our findings will contribute to functional analyses in neurobiology, and comparative evolutionary biology.     This chapter consists of a systematic introduction to the nature and function of habitus in Latin medieval philosophy. Over the course of this introduction, several topics are treated: the theoretical necessity to posit habitus; their nature; their causal contribution to the production of internal and external acts; how and why habitus can grow and decay; what makes their unity when they can have multiple objects and work in clusters. Finally we examine two specific questions: why intellectual habitus represent a special case that triggered considerable debate; how human beings can be said to be free if their actions are determined by moral habitus
