    Thesis with the title "Zam - Zam kebab UMKM Development Strategy in Ploso Baru 104F". The Zam - Zam Kebab business is a home business that sells various kinds of produced kebabs. This zam – zam kebab business has been running for approximately 8 years. The agribusiness system in managing this business starts from production to marketing. The problem faced by the zam - zam kebab business is how the development strategy of this MSME is experiencing a decline due to market competition, limited time, limited circulation of money and consumer tastes which influence demand. The production of frozen Zamzam kebab is carried out independently because they have different responsibilities and there is a limited number of human resources, which is only 3 people. The aim of this research is to find out the Zam-Zam Kebab strategy in Ploso Baru 104F so that it can compete with the market out there and find out the factors that hinder the Zam-Zam Kebab business itself and find a solution. The method this research uses is qualitative with a phenomenological type of research. The subject determination technique uses purposive sampling. This research was conducted at Ploso Baru 104F which is located in Tambak Sari District, Surabaya City. In the data collection method the author uses observation, interviews and documentation. To analyze using Talcott Parsons' structural functionalism theory. The results of this research are that the zam - zam kebab UMKM development strategy should run optimally, but the limited manpower (HR) in production is hampering the development process and also the level of promotion of this UMKM is also lacking. It would be nice for the owner of Zam-Zam Kebab to add more workers so that the development process of the Zam-Zam Kebab UMKM can be better known out there

The Quraan has stated in verse 21:30 ‘and we have made of water everything living, will they not then believe’. It is because of its religious character that Zamzam has a unique identification: the most sacred and miraculous water well in the history of Islam if not the entire world.     The Effect of Zam-zam Water to the Lipid Profile (HDL & LDL) at White Mouse (Rattus novergicus)

    The estimates from the experts of the World Health Organization (WHO) indicate that 50% of the 12 million world population die from coronary heart disease blood vessel disease. Low-Density Lipid (LDL) and High-Density Lipid (HDL) ratios are often calculated to estimate the risk of coronary heart disease. Zam-zam water is thought to be efficacious in reducing blood lipid profile levels. This study aims to know the effect of Zam-zam Water on the lipid profile (HDL LDL) on a white mouse (Rattus Novergicus). The type of this research is an experimental laboratory design with a pre-post control group conducted for 28 days. The subjects of this study were 25 Rattus norvegicus wistar strains, divided into five groups namely control (aquades) group and four treatment groups (concentrations of 25%, 50%, 75% and 100%). The data were obtained by measuring the level of LDL and HDL before and after receiving the treatment of Cholesterol Oxidase Para Aminophenazone (CHODPAP). Also, the data were analyzed using ANOVA test and paired t-test. The Anova analysis showed that the administration of Zam-zam water affected the level of HDL and LDL as proven by the significance of differences among study groups (p 0.05). It is obtained that zam-zam water is 100% proven to have the most influence on HDL and LDL levels. The analysis of paired T-test showed that there were significant differences before and after treatment (p 0.05). Therefore, it can be concluded that the administration of zam-zam water affected the level of HDL and LDL of Rattus norvegicus blood lipid profiles

    Commercially, and fulfills the requirements set by the government. One of the components of the sharia tourism industry in the Mandailing Natal district is the Zam-Zam Syariah Hotel. Hotels that are in accordance with sharia principles are hotels which, in providing lodging, eating and drinking services and other services for the public, are managed and do not conflict with sharia principles. In fact, not all of these efforts have implemented sharia principles as a whole in accordance with the Fatwa of the DSN MUI No. 108/DSN-MUI/X/2016 concerning Guidelines for the Implementation of Tourism Based on Sharia Principles. The type of research used is field research (Field Research) with a qualitative descriptive approach. Data were collected through observation, interviews, and documentation. Inductive analysis technique is to draw things that are specific to things that are general. The results of the study state that, First, the prohibition of checking in for non-Muslims by implementing a screening process for hotel guests, Hotel Zam-Zam Syariah provides adequate facilities, equipment, and facilities in the implementation of worship, Hotel Zam-Zam Syariah does not provide entertainment facilities that leads to disobedience such as cafes, karoeke, swimming pools, and therapeutic facilities and only provides entertainment facilities for children. Hotel Zam-Zam Syariah has implemented and used the Ijarah contract with hotel guests. Second, the Sharia Zam Zam Hotel has not complied with the guidelines for establishing sharia tourism as stated in the Fatwa of the DSN MUI No. 108/DSN-MUI/X/2016 concerning Guidelines for the Implementation of Tourism Based on Sharia Principles because it has not met the criteria set by the Indonesian Ulema Council

    Background: The retroviral Integrase protein catalyzes the insertion of linear viral DNA into host cell DNA. Although different retroviruses have been shown to target distinctive chromosomal regions, few of them display a site-specific integration. ZAM, a retroelement from Drosophila melanogaster very similar in structure and replication cycle to mammalian retroviruses is highly site-specific. Indeed, ZAM copies target the genomic 59-CGCGCg-39 consensus-sequences. To enlighten the determinants of this high integration specificity, we investigated the functional properties of its integrase protein denoted ZAM-IN. Principal Findings: Here we show that ZAM-IN displays the property to nick DNA molecules in vitro. This endonuclease activity targets specific sequences that are present in a 388 bp fragment taken from the white locus and known to be a genomic ZAM integration site in vivo. Furthermore, ZAM-IN displays the unusual property to directly bind specific genomic DNA sequences. Two specific and independent sites are recognized within the 388 bp fragment of the white locus: the CGCGCg sequence and a closely apposed site different in sequence. Conclusion: This study strongly argues that the intrinsic properties of ZAM-IN, ie its binding properties and its endonuclease activity, play an important part in ZAM integration specificity.     Selective inhibition of hepatitis C virus replication by alpha-zam, a Nigella sativa seed formulation

    Background: Hepatitis C virus (HCV) infection became curable because of the development of direct acting antivirals (DAAs). However, the high cost of DAAs has greatly impeded their potential impact on the treatment of HCV infection. As a result, hepatitis C will continue to cause substantial morbidity, and mortality among chronically infected individuals in low and middle income countries. Thus, urgent need exists for developing cheaper drugs available to hepatitis C patients in these countries.Materials and Methods: Alpha-zam, an indigenous herbal formulation from Nigella sativa seed, was examined for its anti-HCV activity and cytotoxicity in genotype 1b HCV replicon cells. The antiviral activity was determined by luciferase expression and viral RNA synthesis, while the cytotoxicity was assessed by viable cell number and glyceraldehyde-3-phosphate dehydrogenase RNA synthesis in the replicon cells.Results: Alpha-zam was found to be a selective inhibitor of HCV replication. The 50% effective dilution and 50% cytotoxic dilution of Alpha-zam were 761- and < 100-fold, respectively, in the subgenomic replicon cells LucNeo#2. Its selective inhibition of HCV was also confirmed by HCV RNA levels in LucNeo#2 and in the full-genome HCV replicon cells NNC#2 using real-time reverse transcriptase polymerase chain reaction. Furthermore, the anti-HCV activity of Alpha-zam was not due to the induction of interferon.Conclusion: Alpha-zam selectively inhibits HCV replication and therefore has potential for a novel antiviral agent against HCV infection.Keywords: Alpha-zam, chronic hepatitis, hepatitis C virus, antiviral assay, Nigella sativ

    Viral particles of the endogenous retrovirus ZAM from Drosophila melanogaster use a pre-existing endosome/exosome pathway for transfer to the oocyte

    BACKGROUND: Retroviruses have evolved various mechanisms to optimize their transfer to new target cells via late endosomes. Here, we analyzed the transfer of ZAM, a retroelement from Drosophila melanogaster, from ovarian follicle cells to the oocyte at stage 9–10 of oogenesis, when an active yolk transfer is occurring between these two cell types. RESULTS: Combining genetic and microscopic approaches, we show that a functional secretory apparatus is required to tether ZAM to endosomal vesicles and to direct its transport to the apical side of follicle cells. There, ZAM egress requires an intact follicular epithelium communicating with the oocyte. When gap junctions are inhibited or yolk receptors mutated, ZAM particles fail to sort out the follicle cells. CONCLUSION: Overall, our results indicate that retrotransposons do not exclusively perform intracellular replication cycles but may usurp exosomal/endosomal traffic to be routed from one cell to another
