Technologies for Climate Change Mitigation - Agriculture Sector
This guidebook describes crop and livestock management technologies and practices that contribute to climate change mitigation while improving crop productivity, reducing reliance on synthetic fertilizers, and lowering water consumption. It is co-authored by internationally recognised experts in the areas of crops, livestock, emissions, and economics, and we are grateful for their efforts in producing this cross disciplinary work.This publication is part of a technical guidebook series produced by the UNEP Risø Centre on Energy, Climate and Sustainable Development (URC) as part of the Technology Needs Assessment (TNA) project(http://tech-action.org) that is assisting developing countries in identifying and analysing the priority technology needs for mitigating and adapting to climate change. The TNA process involves differentstakeholders in a consultative process, enabling all stakeholders to understand their technology needs in a cohesive manner, and prepare Technology Action Plans (TAPs) accordingly.The TNA project is funded by the Global Environment Facility (GEF) and is being implemented by UNEP and the URC in 36 developing countries
7‑hydroxymitragynine is an active metabolite of mitragynine and a key mediator of its analgesic effects
Mitragynina speciosa, more commonly known as kratom, is a
plant native to Southeast Asia, the leaves of which have been used
traditionally as a stimulant, analgesic, and treatment for opioid addiction. Recently,
growing use of the plant in the United States and concerns that kratom
represents an uncontrolled drug with potential abuse liability, have
highlighted the need for more careful study of its pharmacological activity. The
major active alkaloid found in kratom, mitragynine, has been reported to have
opioid agonist and analgesic activity in vitro
and in animal models, consistent with the purported effects of kratom leaf in
humans. However, preliminary research has provided some evidence that
mitragynine and related compounds may act as atypical opioid agonists, inducing
therapeutic effects such as analgesia, while limiting the negative side effects
typical of classical opioids. Here we report evidence that an active metabolite
plays an important role in mediating the analgesic effects of mitragynine. We
find that mitragynine is converted in
vitro in both mouse and human liver preparations to the much more potent
mu-opioid receptor agonist 7-hydroxymitragynine, and that this conversion is
mediated by cytochrome P450 3A isoforms. Further, we show that 7-hydroxymitragynine
is formed from mitragynine in mice and that brain concentrations of this
metabolite are sufficient to explain most or all of the opioid-receptor-mediated
analgesic activity of mitragynine. At the same time, mitragynine is found in the
brains of mice at very high concentrations relative to its opioid receptor
binding affinity, suggesting that it does not directly activate opioid
receptors. The results presented here provide a metabolism-dependent mechanism
for the analgesic effects of mitragynine and clarify the importance of route of
administration for determining the activity of this compound. Further, they
raise important questions about the interpretation of existing data on
mitragynine and highlight critical areas for further research in animals and
humans.</p
The effectiveness of interventions on nutrition social behaviour change communication in improving child nutritional status within the first 1000 days : evidence from a systematic review and meta-analysis
This systematic review and meta-analysis aimed to assess the robustness of designs and tools used in nutrition social behaviour change communication (NSBCC) interventions and establish their effectiveness. EBSCOhost as an umbrella database including Medline (Ovid) and CINAHL, EMBASE, and ProQUEST databases were searched for peer-reviewed articles from January 1960 to October 2018. Additional sources were searched to identify all relevant studies including grey literature. Studies' biases were assessed according to Cochrane handbook. Pooled estimate of effectiveness of interventions on infant and young child feeding (IYCF) practices and child nutritional status with 95% confidence intervals were measured using random-effects models. Eighty studies were included in this review: Fifty-one (64%) were cluster randomised controlled trials (RCTs), 13 (16%) were RCTs and 16 (20%) quasi-experimental. Of the included studies, 22 (27%) measured early initiation of breastfeeding, 38 (47%) measured exclusive breastfeeding, 29 (36%) measured minimum dietary diversity, 21 (26%) measured minimum meal frequency, 26 (32%) measured height for age z-scores (HAZ), 23 (29%) measured weight for height z-scores (WHZ), 27 (34%) measured weight for age z-scores (WAZ), 20 (25%) measured stunting, 14 (17%) measured wasting, and 11 (14%) measured underweight. The overall intervention's effect was significant for exclusive breastfeeding (EBF) (odds ratio = 1.73; 95% confidence interval [CI]: 1.35–2.11, p < 0.001), HAZ (standardized mean differences [SMD] = 0.19; 95% CI: 0.17–0.21; p < 0.001), WHZ (SMD = 0.02; 95% CI: 0.004–0.04; p < 0.001), and WAZ (SMD = 0.04; 95% CI: 0.02–0.06; p < 0.001). Evidence shows the effectiveness of NSBCC in improving EBF and child anthropometric outcomes. Further research should test the impact on child nutritional status with clearly specified and detailed NSBCC interventions
Evaluation of adenovirus capsid labeling versus transgene expression
Adenoviral vectors have been utilized for a variety of gene therapy applications. Our group has incorporated bioluminescent, fluorographic reporters, and/or suicide genes within the adenovirus genome for analytical and/or therapeutic purposes. These molecules have also been incorporated as capsid components. Recognizing that incorporations at either locale yield potential advantages and disadvantages, our report evaluates the benefits of transgene incorporation versus capsid incorporation. To this end, we have genetically incorporated firefly luciferase within the early region 3 or at minor capsid protein IX and compared vector functionality by means of reporter readout

The use of phylogeny to interpret cross-cultural patterns in plant use and guide medicinal plant discovery: an example from Pterocarpus (Leguminosae)
The study of traditional knowledge of medicinal plants has led to discoveries that have helped combat diseases and improve healthcare. However, the development of quantitative measures that can assist our quest for new medicinal plants has not greatly advanced in recent years. Phylogenetic tools have entered many scientific fields in the last two decades to provide explanatory power, but have been overlooked in ethnomedicinal studies. Several studies show that medicinal properties are not randomly distributed in plant phylogenies, suggesting that phylogeny shapes ethnobotanical use. Nevertheless, empirical studies that explicitly combine ethnobotanical and phylogenetic information are scarce.In this study, we borrowed tools from community ecology phylogenetics to quantify significance of phylogenetic signal in medicinal properties in plants and identify nodes on phylogenies with high bioscreening potential. To do this, we produced an ethnomedicinal review from extensive literature research and a multi-locus phylogenetic hypothesis for the pantropical genus Pterocarpus (Leguminosae: Papilionoideae). We demonstrate that species used to treat a certain conditions, such as malaria, are significantly phylogenetically clumped and we highlight nodes in the phylogeny that are significantly overabundant in species used to treat certain conditions. These cross-cultural patterns in ethnomedicinal usage in Pterocarpus are interpreted in the light of phylogenetic relationships.This study provides techniques that enable the application of phylogenies in bioscreening, but also sheds light on the processes that shape cross-cultural ethnomedicinal patterns. This community phylogenetic approach demonstrates that similar ethnobotanical uses can arise in parallel in different areas where related plants are available. With a vast amount of ethnomedicinal and phylogenetic information available, we predict that this field, after further refinement of the techniques, will expand into similar research areas, such as pest management or the search for bioactive plant-based compounds
Invited Article: First Flight in Space of a Wide-Field-of-View Soft X-Ray Imager Using Lobster-Eye Optics: Instrument Description and Initial Flight Results
We describe the development, launch into space, and initial results from a prototype wide eld-of-view (FOV) soft X-ray imager that employs Lobster-eye optics and targets heliophysics, planetary, and astrophysics science. The Sheath Transport Observer for the Redistribution of Mass (STORM) is the rst instrument using this type of optics launched into space and provides proof-of-concept for future ight instruments capable of imaging structures such as the terrestrial cusp, the entire dayside magnetosheath from outside the magnetosphere, comets, the moon, and the solar wind interaction with planetary bodies like Venus and Mars
Pharmacological characterization of novel synthetic opioids (NSO) found in the recreational drug marketplace
Novel synthetic opioids (NSO) are increasingly encountered in illicit heroin and counterfeit pain pills. Many NSO are resurrected from older biomedical literature or patent applications, so limited information is available about their biological effects. Here we examined the pharmacology of three structurally-distinct NSO found in the recreational drug market: N-(1-(2-phenylethyl)-4-piperidinyl)-N-phenylbutyramide (butyrylfentanyl), 3,4-dichloro-N-[(1R,2R)-2-(dimethylamino)cyclohexyl]-N-methylbenzamide (U-47700) and 1-cyclohexyl-4-(1,2-diphenylethyl)piperazine (MT-45). Radioligand binding and GTPγS functional assays were carried out in cells transfected with murine mu- (MOR-1), delta- (DOR-1) or kappa-opioid receptors (KOR-1). Antinociceptive effects were determined using the radiant heat tail flick technique in mice, and opioid specificity was assessed with the mu-opioid antagonist naloxone. Butyrylfentanyl, U-47700 and MT-45 displayed nM affinities at MOR-1, but were less potent than morphine, and had much weaker effects at DOR-1 and KOR-1. All NSO exhibited agonist actions at MOR-1 in the GTPγS assay. Butyrylfentanyl and U-47700 were 31- and 12-fold more potent than morphine in the tail flick assay, whereas MT-45 was equipotent with morphine. Analgesic effects were reversed by naloxone and absent in genetically-engineered mice lacking MOR-1. Our findings confirm that butyrylfentanyl, U-47700 and MT-45 are selective MOR-1 agonists with in vitro affinities less than morphine. However, analgesic potencies vary more than 30-fold across the compounds, and in vitro binding affinity does not predict in vivo potency. Taken together, our findings highlight the risks to humans who may unknowingly be exposed to these and other NSO when taking adulterated heroin or counterfeit pain medications. Connecting Warning with Decision and Action: A Partnership of Communicators and Users
In this chapter, we explore the challenges of achieving a level of awareness of disaster risk, by each person or organisation receiving a warning, which allows them to take actions to reduce potential impacts while being consistent with the warning producer’s capabilities and cost-effectiveness considerations. Firstly we show how people respond to warnings and how the nature and delivery of the warning affects their response. We look at the aims of the person providing the warning, the constraints within which they must act and the judgement process behind the issue of a warning. Then we address the delivery of the warning, noting that warning messages need to be tailored to different groups of receivers, and see how a partnership between warner and warned can produce a more effective result. We include illustrative examples of co-design of warning systems in Argentina and Nepal, experience in communicating uncertainty in Germany and the Weather-Ready Nation initiative in the USA. We conclude with a summary of aspects of the warning that need to be considered between warner and decision-maker when designing or upgrading a warning system
