Reproduction affects immune defenses in the guinea pig even under ad libitum food
Reproduction is one of the costliest processes in the life of an animal. Life history theory assumes that when resources are limiting allocation to reproduction will reduce allocation to other essential processes thereby inducing costs of reproduction. The immune system is vital for survival. If reproduction reduces investment in immune function, this could increase the risk of disease, morbidity and mortality. We here test in the guinea pig, if even under ad libitum food conditions, pregnancy and lactation reduce the activity of the adaptive and innate immune system compared to the reaction of non-reproducing animals. In response to a challenge with keyhole limpet haemocyanin the antibody-mediated adaptive immunity during (pregnancy and) lactation was reduced. Pregnant and lactating females showed higher levels of bacterial killing activity, an integrated measure of innate immunity, than non-reproducing females. However, two major effectors of the innate immunity, the natural antibody and the complement of pregnant and lactating females showed lower levels than in non-reproducing females. Pregnant and lactating females did not differ significantly in the expressed levels of innate immunity. Our results indicate that changes in the immune response during reproduction are physiological adjustments to predictable allocation problems, because they happen even under ad libitum food availability
Suitability of short-period sensors for retrieving reliable H/V peaks for frequencies less than 1 Hz
Using three different short-period electromagnetic sensors with resonant frequencies of 1 Hz (Mark L4C-3D), 2 Hz (Mark L-22D), and 4.5 Hz (I/O SM-6), coupled with three digital acquisition system, the PDAS Teledyne Geotech, the REFTEK 72A, and the Earth Data Logger PR6-24 (EDL), the effect of the seismic instruments on the horizontal-to-vertical spectral ratio (H/V) using seismic noise for frequencies less than 1 Hz has been evaluated. For all possible sensors - acquisition system pairs, the background seismic signal and instrumental self-noise power spectral densities have been calculated and compared. The results obtained when coupling the short-period sensors with different acquisition systems show that the performance of the considered instruments at frequencies < 1 Hz strongly depends upon the sensor-acquisition system combination and the gain used, with the best performance obtained for sensors with the lowest resonance frequency. For all acquisition systems, it was possible to retrieve correctly the H/V peak down to 0.1-0.2 Hz by using a high gain and a 1 Hz sensor. In contrast, biased H/V spectral ratios were retrieved when low-gain values were considered. Analytische Unterstützung zur Vorbereitung des Ausdampf-Versuchs QUENCH-11
Im QUENCH-Vorhaben des Forschungszentrums Karlsruhe soll das Fluten eines teilweise zerstörten Kerns untersucht werden. Der zweite LACOMERA Versuch Q-L2 (QUENCH-11) beginnt mit einer Ausdampfphase des Bündels, bis der Wasserspiegel das untere Bündel¬ende erreicht hat. Ein derartiger Versuch wurde bislang noch nicht in der QUENCH-Anlage durchgeführt, so dass mit SCDAP/RELAP5 mod3.2.irs eine Machbarkeitsstudie erforderlich war. Die Ergebnisse zeigen, dass eine Zusatzheizung im unteren Plenum notwendig ist, um den Wasserstand und die Verdampfungsrate (Dampfmassenstrom in der Ausdampfphase) unabhängig von der angestrebten Maximaltemperatur im Bündel zu regeln. Für eine verläss¬liche Versuchsplanung sowie zur Erstellung der Energiebilanz muss die Zusatzheizung in¬nerhalb des unteren Plenums unterhalb der Wasseroberfläche installiert werden, damit die Heizleistung vollständig in das Wasser eingekoppelt wird. Um die Verdampfungsrate über längere Zeit aufrecht zu erhalten, muss zusätzlich Wasser in das untere Plenum eingespeist werden.
Anhand dieser Rechnungen wird der Testablauf im Detail diskutiert. Eine entsprechende Studie zeigte die Durchführbarkeit eines solchen Ausdampftests und war die Grundlage für die oben erwähnten Änderungen in der Anlage und der Versuchs-Durchführung gegenüber früheren Tests. Eine Reihe von Vorversuche wurde durchgeführt, um die Brauchbarkeit der Änderungen an der Anlage und der geplanten Versuchsführung zu prüfen und um Daten für das thermohydraulische Verhalten der Anlage zu bekommen, an denen die Code-Modelle für die Voraus- und Nachrechnungen von QUENCH-11 getestet werden können. Im Anschluss an die Vorversuche wurden wie bei früheren QUENCH-Tests detaillierte Vorausrechnungen mit verschiedenen Codes zu Versuchsablauf und -steuerung durchgeführt. Drei Forschungs¬einrichtungen in der EU waren beteiligt. Die berechneten Ergebnisse reagieren empfindlich auf Änderungen der Versuchsparameter wie das anfängliche axiale Temperaturprofil und die eingespeiste elektrische Leistung, wie es auch für die untersuchten physikalischen Bedin¬gungen im Versuch erwartet werden kann

Reduction of Campylobacter jejuni in Broiler Chicken by Successive Application of Group II and Group III Phages
Background Bacteriophage treatment is a promising tool to reduce Campylobacter
in chickens. Several studies have been published where group II or group III
phages were successfully applied. However, these two groups of phages are
different regarding their host ranges and host cell receptors. Therefore, a
concerted activity of group II and group III phages might enhance the efficacy
of a treatment and decrease the number of resistant bacteria. Results In this
study we have compared the lytic properties of some group II and group III
phages and analysed the suitability of various phages for a reduction of C.
jejuni in broiler chickens. We show that group II and group III phages exhibit
different kinetics of infection. Two group III and one group II phage were
selected for animal experiments and administered in different combinations to
three groups of chickens, each containing ten birds. While group III phage
CP14 alone reduced Campylobacter counts by more than 1 log10 unit, the
concomitant administration of a second group III phage (CP81) did not yield
any reduction, probably due to the development of resistance induced by this
phage. One group of chickens received phage CP14 and, 24 hours later, group II
phage CP68. In this group of animals, Campylobacter counts were reduced by
more than 3 log10 units. Conclusion The experiments illustrated that
Campylobacter phage cocktails have to be carefully composed to achieve the
best results

This feature article discusses the optical trapping and manipulation of plasmonic nanoparticles, an area of current interest with potential applications in nanofabrication, sensing, analytics, biology and medicine. We give an overview over the basic theoretical concepts relating to optical forces, plasmon resonances and plasmonic heating. We discuss fundamental studies of plasmonic particles in optical traps and the temperature profiles around them. We place a particular emphasis on our own work employing optically trapped plasmonic nanoparticles towards nanofabrication, manipulation of biomimetic objects and sensing
Harmonic Generation from Relativistic Plasma Surfaces in Ultra-Steep Plasma Density Gradients
Harmonic generation in the limit of ultra-steep density gradients is studied
experimentally. Observations demonstrate that while the efficient generation of
high order harmonics from relativistic surfaces requires steep plasma density
scale-lengths () the absolute efficiency of the harmonics
declines for the steepest plasma density scale-length , thus
demonstrating that near-steplike density gradients can be achieved for
interactions using high-contrast high-intensity laser pulses. Absolute photon
yields are obtained using a calibrated detection system. The efficiency of
harmonics reflected from the laser driven plasma surface via the Relativistic
Oscillating Mirror (ROM) was estimated to be in the range of 10^{-4} - 10^{-6}
of the laser pulse energy for photon energies ranging from 20-40 eV, with the
best results being obtained for an intermediate density scale-length
Chronic "Candidatus Mycoplasma turicensis" infection
"Candidatus Mycoplasma turicensis" infects felids. The pathogenesis of "Candidatus M. turicensis" chronic infection is poorly understood. The goals of the present study were to (1) induce reactivation of the infection in chronic carrier cats by attempted immunosuppression, (2) identify potential tissue sequestration using real-time TaqMan® PCR and (3) monitor the humoral immune response by DnaK enzyme-linked immunosorbent assay (ELISA). Ten specified pathogen-free cats that had ostensibly recovered from experimental "Candidatus M. turicensis" infection were used: five cats (group 1) received high dose methylprednisolone (attempted immunosuppression), while five cats served as untreated controls (group 2). Besides weekly blood samples, tissue samples were collected from bone marrow, kidney, liver and salivary glands at selected time points. The cats in group 1 had significantly lower lymphocyte counts and higher blood glucose levels after methylprednisolone administration than the controls. After methylprednisolone administration one blood and three tissue samples from cats in group 1 tested PCR-positive; before the administration, only one sample was positive. All other samples tested PCR-negative. All cats stayed seropositive; the antibody levels of the cats in group 1 showed a significant transient decrease after methylprednisolone administration. This is the first study to report the presence of "Candidatus M. turicensis" in tissues of chronically infected cats and the persistence of anti-feline hemoplasma antibodies in the absence of detectable bacteremia. Methylprednisolone administration did not lead to a significant reactivation of the infection. Our results enhance the knowledge of "Candidatus M. turicensis" infection pathogenesis and are clinically relevant to the prognosis of hemoplasma-infected cats

Ground-based off-line aerosol measurements at Praia, Cape Verde, during the Saharan Mineral Dust Experiment: Microphysical properties and mineralogy
A large field experiment of the Saharan Mineral Dust Experiment (SAMUM) was performed in Praia, Cape Verde, in
January and February 2008. This work reports on the aerosol mass concentrations, size distributions and mineralogical
composition of the aerosol arriving at Praia. Three dust periods were recorded during the measurements, divided by
transitional periods and embedded in maritime-influenced situations. The total suspended particle mass/PM10/PM2.5
were 250/180/74μg/m3 on average for the first dust period (17–21 January) and 250/230/83μg/m3 for the second (24–26
January). The third period (28 January to 2 February) was the most intensive with 410/340/130 μg/m3. Four modes were
identified in the size distribution. The first mode (50–70 nm) and partly the second (700–1100 nm) can be regarded as
of marine origin, but some dust contributes to the latter. The third mode (2–4 μm) is dominated by advected dust, while
the intermittently occurring fourth mode (15–70 μm) may have a local contribution. The dust consisted of kaolinite
(dust/maritime period: 35%wt./25%wt.),K-feldspar (20%wt./25%wt.), illite (14%wt./10%wt.), quartz (11%wt./8%wt.),
smectites (6%wt./4%wt.), plagioclase (6%wt./1%wt.), gypsum (4%wt./7%wt.), halite (2%wt./17%wt.) and calcite
(2%wt./3%wt.)
