    Genome-wide association studies in schizophrenia: Recent advances, challenges and future perspective

    Genome-wide association studies (GWAS) have proved to be a powerful approach for gene discovery in schizophrenia; their findings have important implications not just for our understanding of the genetic architecture of the disorder, but for the potential applications of personalised medicine through improved classification and targeted interventions. In this article we review the current status of the GWAS literature in schizophrenia including functional annotation methods and polygenic risk scoring, as well as the directions and challenges of future research. We consider recent findings in East Asian populations and the advancements from trans-ancestry analysis, as well as the insights gained from research looking across psychiatric disorders

    A farinha de mandioca é o principal produto em importância alimentar servindo como fonte de energia e geração de renda para os agricultores familiares da Amazônia. A castanha-do-brasil é uma das principais fontes de proteína vegetal, geração de emprego e renda para os extrativistas da Amazônia. Este trabalho teve como objetivo analisar o arranjo produtivo local da farinha mista de mandioca com castanha-do-brasil e a produtividade de mandioca em área de agricultores familiares na Reserva Extrativista Cazumbá-Iracema, Sena Madureira-AC. Foram avaliados os critérios de dimensões econômicas, agronômicas, ambientais e sociais destes arranjos produtivos locais. O potencial produtivo da mandioca e da castanha foram avaliados através de ensaios de campo e entrevistas técnicas junto às famílias dos agricultores e extrativistas das comunidades Cuidado, Núcleo Cazumbá e Alto Caeté. O resultado desta análise indica que existe grande potencial de desenvolvimento social e econômico de toda a RESEX, com geração e distribuição de renda

    A transcriptome-wide association study implicates specific pre- and post-synaptic abnormalities in schizophrenia

    chizophrenia is a complex highly heritable disorder. Genome-wide association studies (GWAS) have identified multiple loci that influence the risk of developing schizophrenia, although the causal variants driving these associations and their impacts on specific genes are largely unknown. We identify a significant correlation between schizophrenia risk and expression at 89 genes in dorsolateral prefrontal cortex (P ≤ 9.43x10−6), including 20 novel genes. Genes whose expression correlate with schizophrenia were enriched for those involved in abnormal CNS synaptic transmission (PFDR = 0.02) and antigen processing and presentation of peptide antigen via MHC class I (PFDR = 0.02). Within the CNS synaptic transmission set, we identify individual significant candidate genes to which we assign direction of expression changes in schizophrenia. The findings provide strong candidates for experimentally probing the molecular basis of synaptic pathology in schizophrenia

    Cerliponase Alfa for the Treatment of Atypical Phenotypes of CLN2 Disease: A Retrospective Case Series

    Background: The classic phenotype of CLN2 disease (neuronal ceroid lipofuscinosis type 2) typically manifests between ages 2 and 4 years with a predictable clinical course marked by epilepsy, language developmental delay, and rapid psychomotor decline. Atypical phenotypes exhibit variable time of onset, symptomatology, and/or progression. Intracerebroventricular-administered cerliponase alfa (rhTPP1 enzyme) has been shown to stabilize motor and language function loss in patients with classic CLN2 disease, but its impact on individuals with atypical phenotypes has not been described. Methods: A chart review was conducted of 14 patients (8 male, 6 female) with atypical CLN2 phenotypes who received cerliponase alfa. Pre- and posttreatment CLN2 Clinical Rating Scale Motor and Language (ML) domain scores were compared. Results: Median age at first presenting symptom was 5.9 years. First reported symptoms were language abnormalities (6 [43%] patients), seizures (4 [29%]), ataxia/language abnormalities (3 [21%]), and ataxia alone (1 [7%]). Median age at diagnosis was 10.8 years. ML score declined before treatment in 13 (93%) patients. Median age at treatment initiation was 11.7 years; treatment duration ranged from 11 to 58 months. From treatment start, ML score remained stable in 11 patients (treatment duration 11-43 months), improved 1 point in 1 patient after 13 months, and declined 1 point in 2 patients after 15 and 58 months, respectively. There were 13 device-related infections in 8 patients (57%) and 10 hypersensitivity reactions in 6 (43%). Conclusions: Cerliponase alfa is well tolerated and has the potential to stabilize motor and language function in patients with atypical phenotypes of CLN2 disease

    Observation of an Excited Bc+ State

    Using pp collision data corresponding to an integrated luminosity of 8.5 fb-1 recorded by the LHCb experiment at center-of-mass energies of s=7, 8, and 13 TeV, the observation of an excited Bc+ state in the Bc+π+π- invariant-mass spectrum is reported. The observed peak has a mass of 6841.2±0.6(stat)±0.1(syst)±0.8(Bc+) MeV/c2, where the last uncertainty is due to the limited knowledge of the Bc+ mass. It is consistent with expectations of the Bc∗(2S31)+ state reconstructed without the low-energy photon from the Bc∗(1S31)+→Bc+γ decay following Bc∗(2S31)+→Bc∗(1S31)+π+π-. A second state is seen with a global (local) statistical significance of 2.2σ (3.2σ) and a mass of 6872.1±1.3(stat)±0.1(syst)±0.8(Bc+) MeV/c2, and is consistent with the Bc(2S10)+ state.     Observation of J/ψpJ/\psi p resonances consistent with pentaquark states in Λb0J/ψKp{\Lambda_b^0\to J/\psi K^-p} decays

    Observations of exotic structures in the J/ψpJ/\psi p channel, that we refer to as pentaquark-charmonium states, in Λb0J/ψKp\Lambda_b^0\to J/\psi K^- p decays are presented. The data sample corresponds to an integrated luminosity of 3/fb acquired with the LHCb detector from 7 and 8 TeV pp collisions. An amplitude analysis is performed on the three-body final-state that reproduces the two-body mass and angular distributions. To obtain a satisfactory fit of the structures seen in the J/ψpJ/\psi p mass spectrum, it is necessary to include two Breit-Wigner amplitudes that each describe a resonant state. The significance of each of these resonances is more than 9 standard deviations. One has a mass of 4380±8±294380\pm 8\pm 29 MeV and a width of 205±18±86205\pm 18\pm 86 MeV, while the second is narrower, with a mass of 4449.8±1.7±2.54449.8\pm 1.7\pm 2.5 MeV and a width of 39±5±1939\pm 5\pm 19 MeV. The preferred JPJ^P assignments are of opposite parity, with one state having spin 3/2 and the other 5/2.Comment: 48 pages, 18 figures including the supplementary material, v2 after referee's comments, now 19 figure

    Observation of the Bs0J/ψϕϕB_s^0 \rightarrow J/\psi \phi \phi decay

    The Bs0J/ψϕϕB_s^0 \rightarrow J/\psi \phi \phi decay is observed in pppp collision data corresponding to an integrated luminosity of 3 fb1^{-1} recorded by the LHCb detector at centre-of-mass energies of 7 TeV and 8 TeV. This is the first observation of this decay channel, with a statistical significance of 15 standard deviations. The mass of the Bs0B_s^0 meson is measured to be 5367.08±0.38±0.155367.08\,\pm \,0.38\,\pm\, 0.15 MeV/c2^2. The branching fraction ratio B(Bs0J/ψϕϕ)/B(Bs0J/ψϕ)\mathcal{B}(B_s^0 \rightarrow J/\psi \phi \phi)/\mathcal{B}(B_s^0 \rightarrow J/\psi \phi) is measured to be 0.0115\,\pm\, 0.0012\, ^{+0.0005}_{-0.0009}. In both cases, the first uncertainty is statistical and the second is systematic. No evidence for non-resonant Bs0J/ψϕK+KB_s^0 \rightarrow J/\psi \phi K^+ K^- or Bs0J/ψK+KK+KB_s^0 \rightarrow J/\psi K^+ K^- K^+ K^- decays is found.Comment: All figures and tables, along with any supplementary material and additional information, are available at https://lhcbproject.web.cern.ch/lhcbproject/Publications/LHCbProjectPublic/LHCb-PAPER-2015-033.htm

    Amplitude analysis of B+J/ψϕK+B^+\to J/\psi \phi K^+ decays

    The first full amplitude analysis of B+J/ψϕK+B^+\to J/\psi \phi K^+ with J/ψμ+μJ/\psi\to\mu^+\mu^-, ϕK+K\phi\to K^+K^- decays is performed with a data sample of 3 fb1^{-1} of pppp collision data collected at s=7\sqrt{s}=7 and 88 TeV with the LHCb detector. The data cannot be described by a model that contains only excited kaon states decaying into ϕK+\phi K^+, and four J/ψϕJ/\psi\phi structures are observed, each with significance over 55 standard deviations. The quantum numbers of these structures are determined with significance of at least 44 standard deviations. The lightest has mass consistent with, but width much larger than, previous measurements of the claimed X(4140)X(4140) state. The model includes significant contributions from a number of expected kaon excitations, including the first observation of the K(1680)+ϕK+K^{*}(1680)^+\to\phi K^+ transition.Comment: 62 pages 26 figure
