    FOXD3 Regulates VISTA Expression in Melanoma.

    Immune checkpoint inhibitors have improved patient survival in melanoma, but the innate resistance of many patients necessitates the investigation of alternative immune targets. Many immune checkpoint proteins lack proper characterization, including V-domain Ig suppressor of T cell activation (VISTA). VISTA expression on immune cells can suppress T cell activity; however, few studies have investigated its expression and regulation in cancer cells. In this study, we observe that VISTA is expressed in melanoma patient samples and cell lines. Tumor cell-specific expression of VISTA promotes tumor onset in vivo, associated with increased intratumoral T regulatory cells, and enhanced PDL-1 expression on tumor-infiltrating macrophages. VISTA transcript levels are regulated by the stemness factor Forkhead box D3 (FOXD3). BRAF inhibition upregulates FOXD3 and reduces VISTA expression. Overall, this study demonstrates melanoma cell expression of VISTA and its regulation by FOXD3, contributing to the rationale for therapeutic strategies that combine targeted inhibitors with immune checkpoint blockade

    Male great tits assort by personality during the breeding season

    Animal personalities can influence social interactions among individuals, and thus have major implications for population processes and structure. Few studies have investigated the significance of the social context of animal personalities, and such research has largely focused on the social organization of nonterritorial populations. Here we address the question of whether exploratory behaviour, a well-studied personality trait, is related to the social structure of a wild great tit, Parus major, population during the breeding season. We assayed the exploration behaviour of wild-caught great tits and then established the phenotypic spatial structure of the population over six consecutive breeding seasons. Network analyses of breeding proximity revealed that males, but not females, show positive assortment by behavioural phenotype, with males breeding closer to those of similar personalities. This assortment was detected when we used networks based on nearest neighbours, but not when we used the Thiessen polygon method where neighbours were defined from inferred territory boundaries. Further analysis found no relationship between personality assortment and local environmental conditions, suggesting that social processes may be more important than environmental variation in influencing male territory choice. This social organization during the breeding season has implications for the strength and direction of both natural and sexual selection on personality in wild animal populations

    Response and Resistance to Paradox-Breaking BRAF Inhibitor in Melanomas

    FDA-approved BRAF inhibitors produce high response rates and improve overall survival in patients with BRAF V600E/K-mutant melanoma, but are linked to pathologies associated with paradoxical ERK1/2 activation in wild-type BRAF cells. To overcome this limitation, a next-generation paradox-breaking RAF inhibitor (PLX8394) has been designed. Here, we show that by using a quantitative reporter assay, PLX8394 rapidly suppressed ERK1/2 reporter activity and growth of mutant BRAF melanoma xenografts. Ex vivo treatment of xenografts and use of a patient-derived explant system (PDeX) revealed that PLX8394 suppressed ERK1/2 signaling and elicited apoptosis more effectively than the FDA-approved BRAF inhibitor, vemurafenib. Furthermore, PLX8394 was efficacious against vemurafenibresistant BRAF splice variant-expressing tumors and reduced splice variant homodimerization. Importantly, PLX8394 did not induce paradoxical activation of ERK1/2 in wild-type BRAF cell lines or PDeX. Continued in vivo dosing of xenografts with PLX8394 led to the development of acquired resistance via ERK1/2 reactivation through heterogeneous mechanisms; however, resistant cells were found to have differential sensitivity to ERK1/2 inhibitor. These findings highlight the efficacy of a paradox-breaking selective BRAF inhibitor and the use of PDeX system to test the efficacy of therapeutic agents. © 2017 American Association for Cancer Research

    Wherever I may roam: social viscosity and kin affiliation in a wild population despite natal dispersal

    Dispersal affects the social contexts individuals experience by redistributing individuals in space, and the nature of social interactions can have important fitness consequences. During the vagrancy stage of natal dispersal, after an individual has left its natal site and before it has settled to breed, social affiliations might be predicted by opportunities to associate (e.g., distance in space and time between natal points of origin) or kin preferences. We investigated the social structure of a population of juvenile great tits (Parus major) and asked whether social affiliations during vagrancy were predicted by 1) the distance between natal nest-boxes, 2) synchrony in fledge dates, and 3) accounting for spatial and temporal predictors, whether siblings tended to stay together. We show that association strength was affected predominantly by spatial proximity at fledging and, to a lesser extent, temporal proximity in birth dates. Independently of spatial and temporal effects, sibling pairs associated more often than expected by chance. Our results suggest that the structure of the winter population is shaped primarily by limits to dispersal through incomplete population mixing. In addition, our results reveal kin structure, and hence the scope for fitness-related interactions between particular classes of kin. Both spatial-mediated and socially mediated population structuring can have implications for our understanding of the evolution of sociality

    Neolithic dispersal implications of murids from late Holocene archaeological and modern natural deposits in the Talaud Islands, northern Sulawesi

    The Sangihe-Talaud Archipelago represents a group of 77 remote islands located between the Philippines and North Sulawesi, in the northern sector of Wallacea. The Talaud Islands have a rich and significant archaeological record going back to the Pleistocene and are instrumental in understanding Pleistocene colonisation of small islands and later models of Austronesian language dispersal. Here we report on vertebrate material excavated from Leang Mande'et, a late Holocene rockshelter on Karakelang, the main island in the Talaud group. The site represents a periodically occupied shelter used for gardening. Fauna recovered predominately comprises murid elements, with at least four taxa (Rattus rattus, Rattus exulans and two Melomys species) found. The rodents show clear signs of being deposited by raptors rather than humans, unlike the small number of fish remains also recovered. DNA sequences from several Rattus rattus specimens are referrable to Rattus rattus complex lineage IV, a lineage with a southern Indochinese origins and broad modern representation through Indonesia and the Philippines. The lack of any Rattus rattus complex lineage II from Leang Mande’et suggests that the first agricultural inhabitants of this island did not come from Taiwan or northern Indochina, but rather that they either originated from the south or that, once established in the Talauds, opened up significant trade networks to the south and in the process acquired a significant pest

    Molecular Brightness Approach for FRET Analysis of Donor-Linker-Acceptor Constructs at the Single Molecule Level: A Concept

    In this report, we have developed a simple approach using single-detector fluorescence autocorrelation spectroscopy (FCS) to investigate the Förster resonance energy transfer (FRET) of genetically encoded, freely diffusing crTC2.1 (mTurquoise2.1-linker-mCitrine) at the single molecule level. We hypothesize that the molecular brightness of the freely diffusing donor (mTurquoise2.1) in the presence of the acceptor (mCitrine) is lower than that of the donor alone due to FRET. To test this hypothesis, the fluorescence fluctuation signal and number of molecules of freely diffusing construct were measured using FCS to calculate the molecular brightness of the donor, excited at 405 nm and detected at 475/50 nm, in the presence and absence of the acceptor. Our results indicate that the molecular brightness of cleaved crTC2.1 in a buffer is larger than that of the intact counterpart under 405-nm excitation. The energy transfer efficiency at the single molecule level is larger and more spread in values as compared with the ensemble-averaging time-resolved fluorescence measurements. In contrast, the molecular brightness of the intact crTC2.1, under 488 nm excitation of the acceptor (531/40 nm detection), is the same or slightly larger than that of the cleaved counterpart. These FCS-FRET measurements on freely diffusing donor-acceptor pairs are independent of the precise time constants associated with autocorrelation curves due to the presence of potential photophysical processes. Ultimately, when used in living cells, the proposed approach would only require a low expression level of these genetically encoded constructs, helping to limit potential interference with the cell machinery
