    Fire and Ice in Central Idaho: Modern and Holocene Fires, Debris Flows, and Climate in the Payette River Basin, and Quaternary and Glacial Geology in the Sawtooth Mountains

    This 2-day trip will highlight recent fire and storm-related debris flows in the Payette River region, Holocene records of fires and fire-related sedimentation events preserved in alluvial fan stratigraphic sequences, and geomorphology and geology of alpine glaciations in the spectacular Sawtooth Mountains and Stanley Basin of central Idaho. Storm events and associated scour following recent fires in the South Fork Payette basin have exposed Holocene fire-related debris-flow deposits, flood sediments, and other alluvial fan-building deposits that yield insights into Holocene environmental change. Moraine characteristics and sediment cores from the southeastern Sawtooth Mountains and Stanley Basin provide evidence of late Pleistocene alpine glaciation. A combination of these glacial records with reconstructions of regional equilibrium line elevations produces late-glacial paleoclimatic inferences for the area. Day one of the trip will examine recent and Holocene fire-related deposits along the South Fork Payette River; day two will focus on alpine glaciation in the Sawtooth Mountains (fig. 1). A description of the scope, methods, results and interpretation of the South Fork Payette fire study is given below. Background information on late Pleistocene alpine glaciation in the eastern Sawtooth Mountains is presented with the material for day 2 of the trip. The road log for day 1 of the trip begins at Banks, Idaho, and ends in Stanley, Idaho. Stop locations are shown on figure 2. At Stop 1, we will provide an introduction to interpretation of alluvial fan stratigraphic sections, and discuss the Boise Ridge fault. At Stops 2–4 (Hopkins Creek, Deadwood River, and Jughead creek), we will examine recent debrisflow deposits and Holocene alluvial fan stratigraphic sections. At Stop 5 (Helende Campground), we will look at a series of well-preserved Holocene and Pleistocene terraces and at Stop 6 (Canyon Creek), we will briefly inspect fire-related deposits in higher-elevation alluvial fan stratigraphic sections. The road log for day 2 begins at Stanley, Idaho, and ends in Sun Valley, Idaho. Stop locations are shown on figure 2. Stop 1, at Redfish Lake, will focus on regional equilibrium line altitude reconstructions and on the general pattern of late Pleistocene glaciation on the eastern flank of the Sawtooth Mountains. Stop 2 will be at Pettit Lake, where we will examine the moraine sequence and discuss relative weathering criteria and moraine groupings. At Stop 3, near Alturas Lake, we will discuss lake sediment coring, moraine chronology, and implications for latest Pleistocene paleoclimatic inferences. Stop 4 will be a brief stop at Galena Summit for an overview of the Sawtooth Mountains and a discussion of ice accumulation patterns. The trip will end at a set of moraines in the Trail Creek valley, near Sun Valley, where we will examine moraine morphology and weathering rind data that constrain the moraine ages

    Testcrosses are an efficient strategy for identifying cis-regulatory variation: Bayesian analysis of allele-specific expression (BayesASE)

    Allelic imbalance (AI) occurs when alleles in a diploid individual are differentially expressed and indicates cis acting regulatory variation. What is the distribution of allelic effects in a natural population? Are all alleles the same? Are all alleles distinct? The approach described applies to any technology generating allele-specific sequence counts, for example for chromatin accessibility and can be applied generally including to comparisons between tissues or environments for the same genotype. Tests of allelic effect are generally performed by crossing individuals and comparing expression between alleles directly in the F1. However, a crossing scheme that compares alleles pairwise is a prohibitive cost for more than a handful of alleles as the number of crosses is at least (n2-n)/2 where n is the number of alleles. We show here that a testcross design followed by a hypothesis test of AI between testcrosses can be used to infer differences between nontester alleles, allowing n alleles to be compared with n crosses. Using a mouse data set where both testcrosses and direct comparisons have been performed, we show that the predicted differences between nontester alleles are validated at levels of over 90% when a parent-of-origin effect is present and of 60%-80% overall. Power considerations for a testcross, are similar to those in a reciprocal cross. In all applications, the testing for AI involves several complex bioinformatics steps. BayesASE is a complete bioinformatics pipeline that incorporates state-of-the-art error reduction techniques and a flexible Bayesian approach to estimating AI and formally comparing levels of AI between conditions. The modular structure of BayesASE has been packaged in Galaxy, made available in Nextflow and as a collection of scripts for the SLURM workload manager on github (https://github.com/McIntyre-Lab/BayesASE)

    Softsensors: key component of property control in forming technology

    The constantly increasing challenges of production technology for the economic and resource-saving production of metallic workpieces require, among other things, the optimisation of existing processes. Forming technology, which is confronted with new challenges regarding the quality of the workpieces, must also organise the individual processes more efficiently and at the same time more reliably in order to be able to guarantee good workpiece quality and at the same time to be able to produce economically. One way to meet these challenges is to carry out the forming processes in closed-loop control systems using softsensors. Despite the many potential applications of softsensors in the field of forming technology, there is still no definition of the term softsensor. This publication therefore proposes a definition of the softsensor based on the definition of a sensor and the distinction from the observer, which on the one hand is intended to stimulate scientific discourse and on the other hand is also intended to form the basis for further scientific work. Based on this definition, a wide variety of highly topical application examples of various softsensors in the field of forming technology are given

    Potentiality of coal mining waste to obtain geopolymers

    Abstract Geopolymers are chemically bonded ceramics formed at room temperature and obtained using aluminosilicates and alkali activators. Geopolymers have found applications beyond construction, such as in wastewater treatment and ceramics. This research aims to develop geopolymers using coal mining waste (CMW) for various purposes. The waste used in this study was obtained from the Barro Branco layer in companies located in southern Santa Catarina/Brazil. The waste was thermally treated at 900 °C for 160 min, followed by grinding and characterization using X-ray fluorescence spectroscopy, X-ray diffraction, and particle size analysis. Geopolymeric samples were produced using 60 wt% waste, 25 wt% sodium silicate, and 15 wt% 10 M NaOH. The specimens were divided into three groups and cured using different methods. The results showed that thermal curing enhanced waste reactivity, reduced curing time, and improved compressive strength. However, long-term submersion curing decreased compressive strength due to sodium leaching, reducing reactivity

    A path forward in the debate over health impacts of endocrine disrupting chemicals

    Several recent publications reflect debate on the issue of “endocrine disrupting chemicals” (EDCs), indicating that two seemingly mutually exclusive perspectives are being articulated separately and independently. Considering this, a group of scientists with expertise in basic science, medicine and risk assessment reviewed the various aspects of the debate to identify the most significant areas of dispute and to propose a path forward. We identified four areas of debate. The first is about the definitions for terms such as “endocrine disrupting chemical”, “adverse effects”, and “endocrine system”. The second is focused on elements of hormone action including “potency”, “endpoints”, “timing”, “dose” and “thresholds”. The third addresses the information needed to establish sufficient evidence of harm. Finally, the fourth focuses on the need to develop and the characteristics of transparent, systematic methods to review the EDC literature. Herein we identify areas of general consensus and propose resolutions for these four areas that would allow the field to move beyond the current and, in our opinion, ineffective debate

    Os métodos multicritérios agregam um valor significativo à informação contábil, na medida em que não somente permitem a abordagem de problemas considerados complexos e, por isto mesmo, não tratáveis pelos procedimentos intuitivo-empíricos usuais, mas também conferem, ao processo de tomada de decisão uma clareza e conseqüente transparência não disponíveis quando esses procedimentos, ou outros métodos de natureza monocritério, são utilizados. Discorre-se acerca dos métodos multicriteriais de apoio à tomada de decisão, citando conceitos e referências históricas, abordando medidas subjetivas e seu uso pela contabilidade gerencial, apresentando os principais métodos multi-atributos de apoio à tomada de decisão e alguns casos de aplicação na área contábil.Multicriterion methods aggregate a significant value to accounting information, since they do not only allow for the treatment of problems that are considered complex and that, therefore, cannot be treated by the usual intuitive-empirical procedures, but also provide the decision-making process with a level of clearness and, consequently, transparency that is not available when these procedures or other monocriterion methods are used. This article deals with the multicriterion methods that support decisionmaking, quoting concepts and historical references, discussing subjective measures and their use in management accounting and presenting the main multi-attribute methods used in support of decision-making and some examples of their application in the accounting area

    Modelling distributions of Aedes aegypti and Aedes albopictus using climate, host density and interspecies competition.

    Florida faces the challenge of repeated introduction and autochthonous transmission of arboviruses transmitted by Aedes aegypti and Aedes albopictus. Empirically-based predictive models of the spatial distribution of these species would aid surveillance and vector control efforts. To predict the occurrence and abundance of these species, we fit a mixed-effects zero-inflated negative binomial regression to a mosquito surveillance dataset with records from more than 200,000 trap days, representative of 53% of the land area and ranging from 2004 to 2018 in Florida. We found an asymmetrical competitive interaction between adult populations of Aedes aegypti and Aedes albopictus for the sampled sites. Wind speed was negatively associated with the occurrence and abundance of both vectors. Our model predictions show high accuracy (72.9% to 94.5%) in validation tests leaving out a random 10% subset of sites and data since 2017, suggesting a potential for predicting the distribution of the two Aedes vectors

    An Effective Assessment of Simvastatin-Induced Toxicity with NMR-Based Metabonomics Approach

    BACKGROUND: Simvastatin, which is used to control elevated cholesterol levels, is one of the most widely prescribed drugs. However, a daily excessive dose can induce drug-toxicity, especially in muscle and liver. Current markers for toxicity reflect mostly the late stages of tissue damage; thus, more efficient methods of toxicity evaluation are desired. METHODOLOGY/PRINCIPAL FINDINGS: As a new way to evaluate toxicity, we performed NMR-based metabonomics analysis of urine samples. Compared to conventional markers, such as AST, ALT, and CK, the urine metabolic profile provided clearer distinction between the pre- and post-treatment groups treated with toxic levels of simvastatin. Through multivariate statistical analysis, we identified marker metabolites associated with the toxicity. Importantly, we observed that the treatment group could be further categorized into two subgroups based on the NMR profiles: weak toxicity (WT) and high toxicity (HT). The distinction between these two groups was confirmed by the enzyme values and histopathological exams. Time-dependent studies showed that the toxicity at 10 days could be reliably predicted from the metabolic profiles at 6 days. CONCLUSIONS/SIGNIFICANCE: This metabonomics approach may provide a non-invasive and effective way to evaluate the simvastatin-induced toxicity in a manner that can complement current measures. The approach is expected to find broader application in other drug-induced toxicity assessments
