    Fas antigen is a member of the tumor necrosis factor receptor family and transduces a lethal signal in Fas-sensitive cells. We previously demonstrated that the sensitivity of Fasmediated apoptosis was modulated by changing the intracellular gluthatione (GSH) level in human T lymphocytes. When the Fas-sensitive cells were incubated with N-acetylcysteine (NAC), intracellular GSH was increased and Fas-mediated apoptosis was blocked. In this report, we examined whether lymphokine-activated-killer (LAK) activity is increased by NAC treatment via inhibition of Fas-mediated activation-induced cell death (AICD). We previously established the Fas-resistant variant cell line JKT2D1R from the parental Fas-sensitive cell line Jurkat. AICD in JKT2D1R was markedly reduced. Induction of Fas antigen on the cell surface was confirmed after incubation of normal PBL with PHA and IL-2 for 7 days. After incubation with NAC, the intracellular GSH level was increased. In this context, surface expression of Fas, CD3 and HLA class I and intracellular expression of TIA-1 remained unchanged. However, production of TNF was markedly increased by NAC treatment. Therefore, the results suggest that LAK activity may be increased in terms of TNF production, most likely due to the inhibition of AICD. These results suggest that the increase of intracellular GSH induced by the administration of NAC may increase LAK activity and have clinical significance in tumor immunology

    We investigated the effects of different concentrations (0, 1, 2 and 4 mM) of putrescine on chilling injury, fruit quality, ethylene production rate, fatty acid composition and the antioxidant system of cold-stored kiwifruit (Actinidia chinensis Planch. var. chinensis 'Hongyang'). We achieved a significant decrease in ethylene production, maintained fruit quality and alleviated chilling injury during storage via treatment with 2 mM putrescine. Furthermore, putrescine treatment inhibited increases in superoxide anion production rate and H2O2 concentration, while maintaining higher membrane lipid unsaturation as well as increased activities of superoxide dismutase and catalase. In addition, putrescine treatment enhanced the activities of antioxidant enzymes related to the ascorbate-glutathione cycle while causing higher levels of ascorbic acid and reduced glutathione. Our results suggest that induced tolerance against chilling injury via putrescine treatment in cold-stored kiwifruit may be due to enhanced antioxidant activity, increased unsaturation of membrane lipids, and inhibited ethylene production

    微柱离心-HPLC法测定六甲蜜胺脂质体包封率

    目的:对六甲蜜胺脂质体进行质量评价,建立六甲蜜胺脂质体包封率的测定方法。方法:采用(凝胶)微柱离心法分离游离药物与脂质体;利用流动相:甲醇-乙腈-水(50∶35∶15);柱温:30℃;流速:0.8 mL·min-1;紫外检测波长:228 nm的HPLC条件测定六甲蜜胺的含量,最终计算得包封率。结果:微柱离心法能够将100%的空白脂质体和小于3.5%的游离药物洗脱下来,游离药物与脂质体可被分离,游离药物基本上被截留于微柱中。在本法选择的色谱条件下,六甲蜜胺得到良好的分离,辅料不干扰测定,六甲蜜胺在2.0~40.0 mg·L-1内线性关系良好(r=0.9996),日内和日间RSD均<2.0%(n=5),加样回收率在97.9%~98.5%之间(RSD<2.0%,n=5)。用该方法测得3份六甲蜜胺脂质体的包封率在82.49%~86.56%之间(RSD均<2.0%,n=3)。结论:本方法可用于测定六甲蜜胺脂质体包封率

    For better understanding of real estate in a multi-asset portfolio, this research focuses on time-series relation of real estate with its other asset classes in total return. VECM confirmed a lead-lag relationship between REITs, equities and investment real estate, that JREITs, large-cap equities and mid-cap equities lead real estate. It was also found that JREITs have a significant impact on forecast errors of real estate at almost 40%, and that large-cap equities have large impacts on both JREITs and real estate in forecast errors
