    Two Dimensional Ultrasonic Vibration Assisted Chemical Mechanical Grinding and Experiment on Si Wafer

    为了提高材料去除率和加工通用性,本文提出了工具施加二维超声波振动辅助的化学机械磨削(CMg)复合加工方式,开发具有伸缩和弯曲两种模态的二维超声波振动子及实验装置.以单晶硅片为加工对象,进行单点切削加工轨迹特性分析,并比较不同加工模式以及加工参数对表面粗糙度和材料去除率的影响.实验结果表明,二维超声辅助下的单点切削轨迹存在更多延性加工趋势.在同样普通机床精度条件下,随着时间的增加,二维超声辅助CMg表面粗糙度明显改善,达到纳米级.较无超声情况下二维超声波辅助CMg复合加工材料去除率提高约1倍,可获得最优表面粗糙度5 nM,一维径向超声辅助加工结果次之.To improve material removal rate and machining popularity,two dimensional ultrasonic vibration assisted chemical mechanical grinding(CMG) was introduced in this paper as the combined grinding method.And two dimensional vibrators with longitudinal and bending modes and integrated experimental installation were developed.Si wafer was taken as machining material,and the experiments were carried out to examine the characteristic of machining locus using single point cutting,and the influence of different machining modes and parameters on surface roughness and material removal rate.The results show that two dimensional ultrasonic assisted single point cutting locus makes more ductile removal.When using the general-purpose machine,two dimensional ultrasonic assisted CMG can obtain better surface roughness at nanometer level as the machining time increases.Compared with conventional CMG without ultrasonic vibration the material removal rate improves by 100% and the best surface roughness is 5 nm.Two dimensional ultrasonic assisted CMG yields the best surface roughness and the largest material removal rate,and one dimensional radial ultrasonic assisted CMG is relatively backward.国家高技术研究发展计划(863计划)资助项目(2008AA042501);国家自然科学基金资助项目(50905150

    基于知识图谱的药品联用建模及风险评价

    目的:根据临床用药实践中的药品联用模式构建知识图谱模型,从而自动化地评价药品联用风险。方法:基于海量处方用药大数据,采用知识图谱建模方法,构建概率图模型,并利用子图搜索算法进行药品联用风险评价。结果:在某三甲医院门诊妇科的59703条真实用药处方中,随机挑选30%数据作为测试集,得到药品联用风险预测准确率达98.3%,能够有效识别合理药品联用模式。结论:基于处方大数据构建的药品联用知识图谱模型能准确地对药品联用风险进行评估,可降低人工成本,减轻药师负担,为临床药事管理提供决策依据,提高联合用药的安全性

    间歇式轴向压应力对组织工程骨种子细胞的黏附增殖与成骨分化促进作用的研究

    目的探究间歇式轴向压应力对组织工程骨种子细胞黏附、增殖与成骨分化能力的影响。方法构建表达绿色荧光蛋白的兔骨髓间充质干细胞(rBMSCs)作为示踪种子细胞,运用旋转细胞培养仪将松质骨支架和种子细胞共培养7 d获得组织工程骨(TEB),实验组在第7~14天施加大小10 N、频率1 Hz、4 h/d的间歇式轴向压应力刺激,对照组常规培养,14 d后胰酶消化法获取两组种子细胞并比较其黏附、增殖和成骨分化能力。采用两组独立样本t检验进行统计学分析。结果 (1)流式细胞术显示rBMSCs被成功提取分离。(2)倒置荧光显微镜及扫描电镜显示TEB中种子细胞与支架相容性良好。(3)活体荧光成像系统及扫描电镜显示应力刺激组种子细胞的生长状况要优于非应力刺激组,前者平均荧光密度及细胞数/500倍视野均大于后者,差异均具有统计学意义(平均荧光密度:(3.75±0.34)×108 vs(2.91±0.22)×108,t=2.90,P=0.04;细胞数/500倍视野:30.50±4.43 vs 21.00±5.13,t=3.14,P=0.01)。(4)细胞黏附实验显示,应力刺激组种子细胞的75﹪细胞贴壁时间短于非应力刺激组,两组时间分别为(3.00±0.41) h、(13.33±1.70)h,差异具有统计学意义(t=8.20,P <0.01),前者的最终细胞贴壁率高于后者(99.97﹪±0.34﹪vs 85.83﹪±1.18﹪),差异具有统计学意义(t=11.31,P <0.01)。(5) CCK-8检测显示,在培养第48~96 h,应力刺激组种子细胞的增殖能力优于非应力刺激组,将两者的450 nm吸光度值在第48小时(0.49±0.02、0.40±0.02)、72 h(0.76±0.07、0.64±0.04)和96 h(1.58±0.07、1.34±0.13)分别进行比较,差异均具有统计学意义(t=5.15、2.57、2.86,P均<0.01)。(6)在成骨诱导14 d后,应力刺激组种子细胞的ALP和Ca结节染色阳性率要强于非应力刺激组:两组ALP染色阳性率分别为26.73﹪±4.56﹪、16.68﹪±3.89﹪,差异具有统计学意义(t=3.33,P=0.03);两组Ca结节染色阳性率分别为41.81﹪±3.56﹪、27.40﹪±2.35﹪,差异具有统计学意义(t=3.68,P=0.02)。结论间歇性轴向压应力可促进组织工程骨种子细胞的黏附、增殖与成骨分化

    [中文文摘]用PCR扩增从激活的人中性粒细胞中得到的编码可溶性Fas配体胞外区中第14 1个到第2 81个氨基酸的cDNA ,将其与hCG_β信号肽片段融合到质粒MB12neo中,随后导入到盘基网柄菌AX3细胞中,得到分泌性表达hFasL的重组菌AX3_H3。为提高shFasL的表达量,对质粒pMB12neo作了改造,得到衍生质粒pMB74。利用质粒pMB74克隆表达shFasL ,得到高通量表达shFasL的重组菌AX3_pLu8。在复杂培养基HL_5C中,重组菌的细胞密度可达(1 5~2 )×10 7 mL ,AX3_H3及AX3_pLu8分泌的shFasL浓度分别为2 3 5 μg L及2 0 6 μg L。利用合成培养基SIH培养重组菌AX3_H3及AX3_pLu8,细胞密度均达到(4~5 )×10 7 mL ,shFasL浓度则分别达到111μg L和4 2 0 μg L。[英文文摘]An expression system is described for high-yield production of recombinant soluble human FasL (shFasL) in Dictyostelium discoideum cells. DNA encoding amino acids 141~281 of hFasL was PCR amplified from cDNA derived from activated human neutrophils. The resulting product was fused with a DNA fragment encoding hCG-β signal peptide and cloned in the expression vector pMB12neo. Dictyostelium strain AX3 was transfected with this plasmid, yielding a recombinant strain called AX3-pCESFL95-H3. In order to mprove the shFasL expression level , pMB12neo was optimized by replacing its transcriptional terminatorPpolyadenylation segment of the 2H3 gene with an actin8 terminatorPpolyadenylation segment , yielding derived expression vector pMB741 The recombinant Dictyostelium strain called AX3-pLu8 was generated with this new plasmid. When the recombinant cells were cultivated in a complex HL-5C medium, a cell density of (115~2) ×107PmL was reached , and the shFasL level expressed by strains AX3-pCESFL95-H3 and AX3-pLu8 was 2315 μgPL and 206μgPL , respectively. By using a newly developed synthetic medium called SIH as culture medium, higher cell density of (4~5) ×107PmL was achieved. Correspondently, 111 μgPL and 420 μgPL shFasL were secreted by recombinant strains AX3-pCESFL95-H3 and AX3-pLu8 ,respectively.国家自然科学基金资助项目(No.20306025,30370039
