    Effect of pectin feeding on obesity development and duodenal alkaline phosphatase activity in Sprague-Dawley rats fed with high-fat/high-energy diet

    Purpose The objective of this study was to evaluate whether pectin feeding would affect the small intestinal function and whether these changes would lead to obesity prevention in rats fed with high-fat diet. Three groups of weaned male rats (ad lib. fed; rats fed with diet containing 15% w/w of citrus pectin; restrictedly pair-fed rats) were fed with either a standard diet (9.5% fat) or a high-fat diet (30% fat) for 10 days. Results Our results revealed that pectin feeding led to significant decreases in body weight, energy intake and fat pad weight in rats fed with the standard as well as high-fat diet. Moreover, compared to the restrictedly pair-fed rats, in both groups of rats fed with the diet containing pectin, significant decrease in duodenal alkaline phosphatase (AP) activity was observed in histochemically stained cryostat sections. In contrast, despite their lower energy intake, restrictedly pair-fed rats showed similar fat pad deposition accompanied by unchanged values of AP activity in comparison to the controls. Conclusions Our findings indicate that daily pectin consumption could be beneficial in suppressing body weight gain and reducing probability of obesity risk in rats fed with a high-fat diet

    Effect of neonatal β3-adrenoceptor agonist CL 316,243 treatment on body fat accumulation and intestinal alkaline phosphatase activity in rats from reduced nests

    Introduction. The aim of this study was to evaluate the effects of early life administration of β3-adrenoceptor agonist (CL 316,243) on somatic and feeding parameters, obesity development as well as small intestinal enzyme activity in 21- and 40-day-old overfed male Sprague-Dawley rats. Material and methods. To induce postnatal overnutrition, litter size was reduced to 4 pups/litter (small litters, SL); while in normally-nourished groups (NL) the litter size was adjusted to 10 pups/litter. From days 5 to 15, half of the suckling pups from NL and SL groups received CL 316,243 subcutaneously. From 21st to 40th day, in the post-weaning period, both control (NL, SL) and CL 316,243-treated (NL-CL, SL-CL) rats had free access to standard diet and water. Body composition was determined by magnetic resonance imaging, blood pressure was measured using non-invasive tail-cuff, and intestinal alkaline phosphatase (AP) activity was assessed by histochemistry. Results. At 21 and 40 days of age the SL rats showed higher body mass, displayed higher adiposity and had significantly increased duodenal and jejunal AP activity compared with NL animals. On day 21, NL and SL rats treated with CL 316,243 showed significantly less fat deposition and jejunal AP activity than the non-treated controls. In contrast, treatment-related changes in adiposity and AP activity were not observed in 40-day-old NL-CL, SL-CL rats. Conclusions. These results indicate that early pharmacological intervention with CL did not permanently influence physiological processes involved in body weight/fat regulation, which resulted in the development of early-programmed overweight status in SL rats after weaning

    Cloud Native Data Platform for Network Telemetry and Analytics

    In this manuscript, we present a prototype of a modular data platform that is able to continuously ingest, process, retain, and analyse large amounts of network telemetry data in a scalable and straightforward manner. It follows a recently proposed Data Lakehouse architectural pattern, which is an evolution of two well-known approaches used in this area -- data warehouses and data lakes. The platform is based on open standards and open-source components, and it follows cloud native principles in order to be able to run in modern computing environments such as public, private, and hybrid clouds. The primary focus of the prototype is network telemetry and analytics over traffic flows and infrastructure logs for the purposes of cyber-security digital forensics and incident response. During the demonstration part, we will further describe internal workings of the presented data platform and showcase its capabilities and possible applications on a public dataset

    Práce se zabývá návrhem a implementací frameworku částečné dynamické rekonfigurace pro FPGA architekturu Virtex-5.  Framework má usnadnit tvorbu aplikací s hardwarovými akcelerátory využívajících částečnou dynamickou rekonfiguraci. S využitím frameworku byla vytvořena demonstrační aplikace pro pattern-matching nad příchozími síťovými pakety. Řízení procesu částečné dynamické rekonfigurace obstarává systém typu GNU/Linux, který běží na procesoru MicroBlaze. To navíc umožňuje běh méně náročných aplikací a zpracování paketů pomocí softwaru.The thesis is focused on design and implementiation of a framework for Dynamic Partial Reconfiguration for FPGA architecture Virtex-5. The aim of the framework is to simplify creating applications with hardware accelerators using  Dynamic Partial Reconfiguration. Using this framework, a demonstration application was created for pattern-matching incoming network packets. The process of Dynamic Partial Reconfiguration is controlled by GNU/Linux type operating system, which runs on MicroBlaze processor. This also allows to run less demanding applications and the processing of packets using software.

    High-quality and universal empirical atomic charges for chemoinformatics applications.

    BackgroundPartial atomic charges describe the distribution of electron density in a molecule and therefore provide clues to the chemical behaviour of molecules. Recently, these charges have become popular in chemoinformatics, as they are informative descriptors that can be utilised in pharmacophore design, virtual screening, similarity searches etc. Especially conformationally-dependent charges perform very successfully. In particular, their fast and accurate calculation via the Electronegativity Equalization Method (EEM) seems very promising for chemoinformatics applications. Unfortunately, published EEM parameter sets include only parameters for basic atom types and they often miss parameters for halogens, phosphorus, sulphur, triple bonded carbon etc. Therefore their applicability for drug-like molecules is limited.ResultsWe have prepared six EEM parameter sets which enable the user to calculate EEM charges in a quality comparable to quantum mechanics (QM) charges based on the most common charge calculation schemes (i.e., MPA, NPA and AIM) and a robust QM approach (HF/6-311G, B3LYP/6-311G). The calculated EEM parameters exhibited very good quality on a training set ([Formula: see text]) and also on a test set ([Formula: see text]). They are applicable for at least 95 % of molecules in key drug databases (DrugBank, ChEMBL, Pubchem and ZINC) compared to less than 60 % of the molecules from these databases for which currently used EEM parameters are applicable.ConclusionsWe developed EEM parameters enabling the fast calculation of high-quality partial atomic charges for almost all drug-like molecules. In parallel, we provide a software solution for their easy computation (http://ncbr.muni.cz/eem_parameters). It enables the direct application of EEM in chemoinformatics

Within the experimental program, tests of compressive strength, transverse tensile strength and flexural tensile strength were performed. The selected type of wires is dosed in 4 series from 0 to 110 kg / m3. Konkrétne sa budú skúšať mechanické vlastnosti na dvoch železobetónových nosníkoch, prvý s rozmermi 4500 x 400 x 150 mm a druhý s rozmermi 6000 x 400 x 165 mm. V teoretickej časti diplomovej práce sú stručne uvedené základné informácie o železobetóne a výstuži spolu s možnosťami ich testovania. V rámci experimentálnej časti boli prevedené skúšky pevnosti v tlaku na jadrových vývrtoch, identifikácia výstuže, ťahová pevnosť výstuže, sledovanie vývoju trhlín a odolnosť voči CHRL. Tieto skúšky sú ďalej doplnené o nedeštruktívne testovanie nosníkov Schmidtovým tvrdomerom na zistenie pevnosti a ultrazvukovou impulzívnou metódou na zistenie dynamického modulu pružnosti.Raček, M.
