    Effects of Diosmetin from Carbonized Cirsium japonicum on Apoptosis in Human MCF-7 Breast Cancer Cells and its Mechanisms

    研究大蓟炭中香叶木素诱导人乳腺癌MCF-7细胞凋亡及其作用机制。采用硅胶和Sephadex; LH-20柱层析从大蓟炭中分离鉴定了三个黄酮类化合物,经NMR和MS鉴定他们的结构分别为香叶木素(1)、刺槐素(2)和柳川鱼黄素(3)。采用MT; S方法检测不同浓度的香叶木素对MCF-7的细胞活力的影响;流式细胞术检测不同浓度的香叶木素处理对MCF-7细胞凋亡的作用;Western; blot法检测香叶木素处理对细胞PARP、P-JNK等细胞凋亡相关蛋白表达的影响。MTS及流式细胞术结果显示香叶木素能显著抑制MCF-7的增殖并; 且诱导细胞凋亡;香叶木素可上调P-JNK促进细胞凋亡。结果表明香叶木素在体外实验能通过激活JNK细胞凋亡通路抑制MCF-7的增殖及促进细胞凋亡。To study the effects of diosmetin from carbonized Cirsium japonicum on; apoptosis in human MCF-7 breast cancer cells and its mechanisms. Three; flavonoids were isolated and purified from carbonized C. japonicum by; silica gel and Sephadex LH-20 chromatography methods. Their structures; were elucidated by NMR and MS spectroscopic data and identified as; diosmetin(1),acacetin(2) and pectolinarigenin(3). MTS assay was; performed to detect the viability of MCF-7 cells treated by different; concentrations of diosmetin. The cell apoptosis rate was further; analyzed by flow cytometry(FCM). Western blot assay was applied to; measure the apoptosis related protein expression levels of PARP,P-JNK.; Diosmetin treatment on MCF-7 cells significantly inhibited cell; proliferation and induced cell apoptosis. Diosmetin significantly; downregulated P-JNK and upregulated cleaved-PARP protein expression.; Diosmetin inhibited MCF-7 cell proliferation and induced cell apoptosis; by activation of JNK pathway in vitro.江西中医药大学校级科研项目; 国家自然科学基金; 江西省自然科学基

    Study on the Thermal Degradation Behavior of Tanshinones during the Process of Reflux Extraction

    用甲醇作提取溶剂,在回流条件下考察了从丹参药材中提取丹参酮类有效成分的过程中,隐丹参酮、丹参酮I及丹参酮IIA等三种丹参酮的热降解行为。结果表明,回流提取过程中所考察的三种丹参酮均发生严重的热降解,降解速率:丹参酮IIA>丹参酮I>隐丹参酮,其热降解均具有零级反应动力学特征;同时,回流提取过程中丹参酮的热降解是在丹参酮共萃物存在下发生的。Thermal degradation behavior of cryptotanshinone,tanshinone I and tanshinone IIA has been studied during the process of reflux extraction of tanshinones from Salvia miltiorrhiza Bge.with methanol as extracting solvent.The obtained results showed that the thermal degradation of cryptotanshinone,tanshinone I and tanshinone IIA occurred obviously during reflux extraction in the order of degradation rate:tanshinone IIA>tanshinone I>cryptotanshinone,and their thermal degradation was featured with zero order reaction;meanwhile,it was under the existence of other components coextracted with tanshinones that the thermal degradation of cryptotanshinone,tanshinone I and tanshinone IIA occurred.国家自然科学基金重点项目(20235020

    利用电感耦合等离子体质谱 (ICP -MS) ,对福建建瓯产大叶、小叶泽泻中 17种元素的含量进行了测定 ,在此基础上 ,运用SAS软件的DISCRIM过程对两类泽泻样品进行了判别分析 ,取得了良好的分类判别结果 ,利用上述方法得到的泽泻元素含量分布图谱有可能为中药材指纹图谱的建立及其规范化种植提供新的途径。element contents of Rhizoma alismatics in Jianou Fujian were determined by ICP-MS. On the basis of the results, the classification discrimination of the two sorts of Rhizome Alismatics, which were Alismao rientalis (Sam.) Juzep and Alisma canaliculatum A. Braun et Bouche, respectively. was obtained by the software of the SAS.     用环氧树脂包埋的半薄切片经PAS反应和苏丹黑染色,研究阳春砂花药发育中的多糖和脂类物质分布特征。结果发现小孢子母细胞和四分体小孢子中积累了一些脂滴,但没有淀粉。阳春砂小孢子母细胞和四分体没有胼胝质壁。晚期小孢子中除了仍有很多脂滴外,细胞核周围开始出现淀粉粒;成熟花粉粒贮存丰富的淀粉粒和脂滴,且花粉壁由多糖物质构成。阳春砂花药壁结构比较特殊:花药壁由10余层细胞组成;最内层的绒毡层细胞在小孢子时期开始解体,细胞质转变为脂滴,供花粉吸收。开花时,花药壁由表皮和几层薄壁细胞以及径向壁纤维加厚的变形细胞组成。Anther development of Amomum villosum Lour.were studied using epoxy resin semi-thin sections with periodic acid-Schiff's technique and Sudan black staining method.The results showed that microspore mother cells and tetrad microspores accumulated numerous lipids but no starches.The late microspore began appear starches around the nucleus beside lipids.Mature pollen grains accumulated abundant polysaccharide starches and lipids.Pollen wall was stained red,suggesting its polysaccharide feature.The anther wall of A.villosum consists of more than ten lay cells.The tapetal cells degenerated at microspore stage and its cytoplasm transform into lipids for the microspore absorbing.At anthesis,the mature anther wall included epidermis and some layers of parenchymal cells and several layers of endothecium cells with an evidently thickened radial wall.福建省教育厅科技项目(JA12437); 漳州市科技计划项目(Z2011060

    Preparative Separation of Salvianolic Acid B from Salvia miltiorrhiza by High Speed Counter-Current Chromatorgraphy

    采用高速逆流色谱法分离纯化丹参水溶性成分丹酚酸类物质,制备丹酚酸B化学对照品。分离采用的溶剂系统为正己烷-乙酸乙酯-水-甲醇(1.5:5:5:1.5),上相做固定相,下相做流动相,流速为1.7 mL/min,仪器转速850 rpm,进样量80 mg,纯度用HPLC方法测定。结果表明:一次分离可制备63.4 mg丹酚酸B,其纯度为98.6%。该方法操作简单,可作为高纯度丹酚酸B化学对照品的制备分离方法。Salvianolic acid B,a major bioactive component in Danshen(Salvia miltiorrhiza Bge.),was isolated and purified by high-speed counter-current chromatography(HSCCC) in semi-preparative scale from the crude extract of Danshen.Separation was performed with a two-phase solvent system composed of hexane-ethyl acetate-water-methanol(1.5:5:5:1.5,v/v) by eluting the lower aqueous phase at a flow-rate of 1.7 mL/min and a revolution speed of 850 rpm with injection volume of 80 mg.After the proceeding of separation,63.4 mg salvianolic acid B was obtained,and analyzed by normalization method according to HPLC areas giving a purity of 98.6%.HSCCC is a recommendable method to prepare and purify the active constituents from Salvia miltiorrhiza.国家自然科学基金重点项目(20235020);; 厦门市科技创新重点项目(3502Z20031096
