Studies on anti-inflammatory and vasoactive effects of mPGES-1 inhibition
Inflammation is the basis for various serious illnesses such as rheumatic diseases,
cardiovascular diseases, and cancer. Prostaglandin E2 (PGE2) is a pro-inflammatory lipid
mediator produced by cyclooxygenases (COX1/2) and the microsomal prostaglandin E
synthase 1 (mPGES-1). Nonsteroidal anti-inflammatory drugs (NSAIDS) targeting COX
successfully reduce pain and inflammation. However, gastrointestinal and cardiovascular side
effects associated with blockade of all prostaglandins limit their use. Selective inhibition of
mPGES-1 is an alternative therapeutic strategy to impede PGE2 production while sparing or
even upregulating other lipid mediators. When and where the shunting of PGH2 to other
prostanoids occurs and whether it interferes with or contributes to the therapeutic effects of
mPGES-1 inhibition is not fully understood. The overall aim of this thesis was to study the
anti-inflammatory and vasoactive effects of mPGES-1 inhibition in models of inflammation
and cardiovascular disease and to investigate the possible shunting of PGH2 to PGD2 and PGI2.
The methodological approach was principally based on liquid chromatography-tandem mass
spectrometry, biochemical assays as well as wire-myography.
In Paper I five new mPGES-1 inhibitors were characterized. The inhibitors selectively
suppressed PGE2 formation in in-vitro and in-vivo assays, reduced acute paw swelling in rats,
and reduced adrenergic vasoconstriction. The results of this study serve as a basis for the
application of these inhibitors in pre-clinical research. Depletion of mPGES-1 may lead to the
re-direction of PGH2 into the PGD2/15-deoxy-Î12,14-PGJ2 (15dPGJ2) pathway, which has been
described to be anti-inflammatory and pro-resolving. In Paper II, we studied the biosynthesis
and metabolism of 15dPGJ2 via conjugation to glutathione in immune cells and upon inhibition
of mPGES-1. The results of this study demonstrate the formation of 15dPGJ2-glutathione and
15dPGJ2-cysteine conjugates in immune cells, the involvement of MGST3 in this pathway and
the preservation of the PGD2/15dPGJ2 pathway upon inhibition of mPGES-1. Another
important aspect of mPGES-1 inhibition is the redirection of excess PGH2 into the PGI2
pathway. A decrease in PGI2 levels as a result of COX-2 inhibition has been associated with
increased cardiovascular risk in patients treated with selective NSAIDs. In Paper III we aimed
to study the effects of mPGES-1 inhibition on human resistance arteries. Inhibition of mPGES-
1 significantly reduced adrenergic vasoconstriction and enhanced relaxation. Our results
suggest that multiple pathways in addition to shunting to PGI2 may be involved in the
vasoactive effects of mPGES-1 inhibition in human microcirculation. In Paper IV we studied
the effects of mPGES-1 inhibition in a mouse model of MI. The results of this study indicate
that pharmacological inhibition of mPGES-1 could improve cardiac function after MI and
increase the PGI2/PGE2 metabolite ratio in urine compared with controls.
The results from this thesis contribute to a better understanding of the mechanisms underlying
the effects seen after inhibition of mPGES-1 in models of inflammation and cardiovascular
disease
Schwingungsbeeinflussung geometrisch komplexer Strukturen mittels piezoelektrischer FlÀchenwandler am Beispiel des Getriebeheulens
Das vor allem im Automobilbereich bekannte Getriebeheulen ist ein tonales StörgerÀusch, welches unter bestimmten Betriebsbedingungen durch die periodische Belastung der einzelnen ZÀhne der im Eingriff befindlichen ZahnrÀder angeregt wird. Die Folge sind resonante GehÀuseschwingungen, die als akustische KomfortbeeintrÀchtigung wahrgenommen werden.
Bislang werden nur passive MaĂnahmen eingesetzt, um diesem Problem zu begegnen. In der vorliegenden Arbeit werden passive und aktive AnsĂ€tze numerisch untersucht, um bei verschiedener Verwendung piezoelektrischer FlĂ€chenwandler eine Schwingungsminderung zu realisieren. FĂŒr die Simulation dieser AnsĂ€tze wird ein vom Getriebehersteller zur VerfĂŒgung gestelltes, rĂ€umlich hoch aufgelöstes Finite-Elemente-Modell verwendet. Dieses wird zunĂ€chst an die dynamischen Eigenschaften des realen Systems, die mittels einer experimentellen Modalanalyse identifizierten werden, abgeglichen.
Anhand dieses Modells wird die fĂŒr das Getriebeheulen verantwortliche Eigenform identifiziert und geeignete Aktorpositionen bestimmt, auf die im FE-Modell piezoelektrische FlĂ€chenwandler integriert werden. Das so entstandene elektromechanisch gekoppelte Gesamtsystem, bestehend aus Getriebe und FlĂ€chenwandler, wird reduziert und fĂŒr zeiteffiziente Simulationen der verschiedenen AnsĂ€tze verwendet.
Beim passiven Shunt-Damping werden dabei mehrere FlĂ€chenwandler jeweils mit einem Widerstand und einer InduktivitĂ€t zu elektrischen Schwingkreisen kombiniert, die eine zu mechanischen Tilgern Ă€quivalente Wirkung aufweisen. Ăber einen klassischen und einen neu entwickelten Ansatz zur Auslegung solcher Multi-Shunt-Systeme wird eine geringe Schwingungsamplitudenreduktion des GetriebgehĂ€uses erreicht, wobei der neu entwickelte Ansatz dem klassischen ĂŒberlegen ist. FĂŒr eine praktische Umsetzung sind die erreichbaren Reduktionen jedoch nicht ausreichend.
Das Potential aktiver AnsĂ€tze wird ĂŒber eine untersuchte optimale StörgröĂenaufschaltung abgeschĂ€tzt. Hierauf aufbauend wird eine realitĂ€tsnahe Simulation unter BerĂŒcksichtigung der dynamischen Eigenschaften des LeistungsverstĂ€rkers und der digitalen Echtzeitdatenverarbeitung eines einzelnen ĂŒber den FxLMS-Algorithmus angesteuerten FlĂ€chenwandlers durchgefĂŒhrt. Mit dieser Methode werden bereits bei der Verwendung eines einzelnen Aktors deutliche Reduktionen erreicht. Die kombinierte Anwendung mehrerer ĂŒber diesen Algorithmus angesteuerten FlĂ€chenwandler ermöglicht deutliche Amplitudenreduktionen, erfordern andererseits jedoch erheblichen Aufwand insbesondere hinsichtlich der benötigten LeistungsverstĂ€rker. AbschlieĂend wird somit eine praktische Umsetzung dieses Ansatzes nur eingeschrĂ€nkt empfohlen

Um PlasmainstabilitĂ€ten zu begegnen, werden in vier der oberen Ports im ITER VakuumgefÀà Electron Cyclotron Launcher installiert. Diese bestehen im Wesentlichen aus einer trapezförmigen Stahlkonstruktion, welche die Mikrowellenkomponenten (im Wesentlichen Spiegel und Wellenleiter) beherbergt. Bei der Konstruktion eines solchen Launchers mĂŒssen als wesentliche Vorgaben die mechanische Festigkeit, die ausreichende KĂŒhlung des Systems und wirksame Abschirmung gegen Neutronen berĂŒcksichtigt werden

Melting Domain Size and Recrystallization Dynamics of Ice Revealed by Time-Resolved X-ray Scattering
The phase transition between water and ice is ubiquitous and one of the most
important phenomena in nature. Here, we performed time-resolved x-ray
scattering experiments capturing the melting and recrystallization dynamics of
ice. The ultrafast heating of ice I is induced by an IR laser pulse and probed
with an intense x-ray pulse, which provided us with direct structural
information on different length scales. From the wide-angle x-ray scattering
(WAXS) patterns, the molten fraction, as well as the corresponding temperature
at each delay, were determined. The small-angle x-ray scattering (SAXS)
patterns, together with the information extracted from the WAXS analysis,
provided the time-dependent change of the size and the number of the liquid
domains. The results show partial melting (~13 %) and superheating of ice
occurring at around 20 ns. After 100 ns, the average size of the liquid domains
grows from about 2.5 nm to 4.5 nm by the coalescence of approximately six
adjacent domains. Subsequently, we capture the recrystallization of the liquid
domains, which occurs on microsecond timescales due to the cooling by heat
dissipation and results to a decrease of the average liquid domain size
