    Positive selection in the adhesion domain of Mus sperm Adam genes through gene duplications and function-driven gene complex formations

    Background: Sperm and testes-expressed Adam genes have been shown to undergo bouts of positive selection in mammals. Despite the pervasiveness of positive selection signals, it is unclear what has driven such selective bouts. The fact that only sperm surface Adam genes show signals of positive selection within their adhesion domain has led to speculation that selection might be driven by species-specific adaptations to fertilization or sperm competition. Alternatively, duplications and neofunctionalization of Adam sperm surface genes, particularly as it is now understood in rodents, might have contributed to an acceleration of evolutionary rates and possibly adaptive diversification. Results: Here we sequenced and conducted tests of selection within the adhesion domain of sixteen known sperm-surface Adam genes among five species of the Mus genus. We find evidence of positive selection associated with all six Adam genes known to interact to form functional complexes on Mus sperm. A subset of these complex-forming sperm genes also displayed accelerated branch evolution with Adam5 evolving under positive selection. In contrast to our previous findings in primates, selective bouts within Mus sperm Adams showed no associations to proxies of sperm competition. Expanded phylogenetic analysis including sequence data from other placental mammals allowed us to uncover ancient and recent episodes of adaptive evolution. Conclusions: The prevailing signals of rapid divergence and positive selection detected within the adhesion domain of interacting sperm Adams is driven by duplications and potential neofunctionalizations that are in some cases ancient (Adams 2, 3 and 5) or more recent (Adams 1b, 4b and 6)

    The Deep Plumbing System of Ischia: a Physico-chemical Window on the Fluid-saturated and CO2-sustained Neapolitan Volcanism (Southern Italy)

    Ischia, a volcanic island located 18 miles SW of Naples (Southern Italy), is a densely populated active caldera that last erupted in AD 1302. Melt inclusions in phenocrysts of the Vateliero and Cava Nocelle shoshonite-latite eruptive products (6th to 4th centuries BC) constrain the structure and nature of the Ischia deep magmatic feeding system. Their geochemical characteristics make Ischia a natural borehole for probing the physico-chemical conditions of magma generation in mantle contaminated by slab-derived fluids or melts, largely dominated by CO2. Volatile concentrations in olivine-hosted melt inclusions require gas-melt equilibria at between 3 and 18 km depth. In agreement with what has already been demonstrated at the other neighboring Neapolitan volcanoes (Procida, Campi Flegrei caldera and Somma-Vesuvius volcanic complex), a major crystallization depth at 8-10 km has been identified.The analyzed melt inclusions provide clear evidence for CO2-dominated gas fluxing and consequent dehydration of magma batches stagnating at crustal discontinuities. Gas fluxing is further supported by selective enrichment in K owing to fluid-transfer during magma differentiation.This takes place under oxidized conditions (Fe3+/EFe>=0·3) that can be fixed by an equimolar proportion of divalent and trivalent iron in the melt if post-entrapment crystallization of the host olivine is discarded. The melt inclusion data, together with data from the literature for other Neapolitan volcanoes, show that magmatism and volcanism in the Neapolitan area, despite differences in composition and eruption dynamics, are closely linked to supercritical CO2-rich fluids.These fluids are produced by devolatilization of subducting terrigenous-pelagic metasediments and infiltrate the overlying mantle wedge, generate magmas and control their ascent up to eruption. Geochemical characteristics of Ischia and the other Neapolitan volcanoes reveal that the extent of fluid or melt contamination of the pre-subduction asthenospheric mantle wedge was similar among these volcanoes. However, differences in the isotopic compositions of the erupted magmas (more enriched in radiogenic Sr at Ischia, Campi Flegrei and Somma-Vesuvius with respect to Procida) and the amount of H2O in the plumbing system of these volcanoes (almost double at Ischia, Campi Flegrei and Somma-Vesuvius than at Procida) reflect the different flow-rates of deep slab-derived fluids or melts through the mantle wedge, which, in turn, control the amount of generated magma.The high bulk permeability of the lithosphere below Ischia, Campi Flegrei and Somma-Vesuvius, determined by the occurrence of intersecting NW-SE and NE-SW regional fault systems, favours fluid ascent and accumulation at crustal levels, with consequent larger magma production and storage than at Procida, located along the NE-SW system

    Third chromosome candidate genes for conspecific sperm precedence between D. simulans and D. mauritiana

    <p>Abstract</p> <p>Background</p> <p>Male - female incompatibilities can be critical in keeping species as separate and discrete units. Premating incompatibilities and postzygotic hybrid sterility/inviability have been widely studied as isolating barriers between species. In recent years, a number of studies have brought attention to postmating prezygotic barriers arising from male - male competition and male - female interactions. Yet little is known about the genetic basis of postmating prezygotic isolation barriers between species.</p> <p>Results</p> <p>Using <it>D. simulans </it>lines with mapped introgressions of <it>D. mauritiana </it>into their third chromosome, we find at least two <it>D. mauritiana </it>introgressions causing male breakdown in competitive paternity success. Eighty one genes within the mapped introgressed regions were identified as broad-sense candidates on the basis of male reproductive tract expression and male-related function. The list of candidates was narrowed down to five genes based on differences in male reproductive tract expression between <it>D. simulans </it>and <it>D. mauritiana</it>. Another ten genes were confirmed as candidates using evidence of adaptive gene coding sequence diversification in the <it>D. simulans </it>and/or <it>D. mauritiana </it>lineage. Our results show a complex genetic basis for conspecific sperm precedence, with evidence of gene interactions between at least two third chromosome loci. Pleiotropy is also evident from correlation between conspecific sperm precedence and female induced fecundity and the identification of candidate genes that might exert an effect through genetic conflict and immunity.</p> <p>Conclusions</p> <p>We identified at least two loci responsible for conspecific sperm precedence. A third of candidate genes within these two loci are located in the 89B cytogenetic position, highlighting a possible major role for this chromosome position during the evolution of species specific adaptations to postmating prezygotic reproductive challenges.</p

    Chronology of the 2007 eruption of Stromboli and the activity 1 of the Scientific Synthesis Group

    On 27 February 2007, at 12.49 GMT, a new eruption of Stromboli took place with the effusion of a 15 lava flow from a fracture cutting the flank of the NE cone, which rapidly reached the sea. The eruption 16 had been heralded by an increase in the amplitude of tremor and flank movement since at least the 14th 17 of February. Short-term precursors were an increase in the rate of occurrence of small landslides within 18 the “Sciara del Fuoco” scar on the North-western flank of the volcano. A new effusive vent opened at 19 18.30 GMT on the Sciara del Fuoco at an height of 400 m asl. The new lava emission caused the 20 sudden termination of the summit flow and initiated a period of non-stationary lava outpouring which 21 ended on 2 April, 2007. The eruption has been characterized by a rapid decrease in the eruption rate after the first days and subsequently by episodic pulse increases. On the 15th 22 of March, the increase in 23 lava outpouring, monitored by a thermal camera, heralded by 9 minutes the occurrence of a violent 24 paroxysmal explosion with the formation of an impulsive eruption column and the emission of small 25 pumices mingled with black scoriae. The pumice had a bulk composition similar to that of the lava and 26 of the black scoriae, but with a distinct lower content of phenocrysts. A similar feature has been 27 repeateadly observed during the major explosive paroxysms of Stromboli. Short term precursors of the 28 paroxysm were recorded by strainmeter and tiltmeter stations. The volcano monitoring activity has 29 been made by a joint team of researchers from the INGV sections of Catania, Napoli, Palermo and 30 Rome, along with researchers from the University of Florence, Pisa, Roma Tre, and Palermo. The 31 scientific activity was coordinated by a Synthesis Group made up by the responsibles of the different 32 monitoring techniques of INGV and Universities and by the volcanic experts of Commissione Nazionale Grandi Rischi of the Prime Minister Office (Civil Protection Department). 33 The group made a 34 daily evaluation of the state of the volcano and transmitted its recommendations to the Civil Protection 35 Department (DPC). Several prevention measures were adopted by DPC, the main of which were the 36 evacuation of the coast zone when strong acceleration of the Sciara del Fuoco slope motion (occurred 37 twice) could led to a dangerous tsunami by flank collapse (as last occurred on 30 December 2002) and 38 four days before the 15 March paroxysm when access was prohibited to the part of the volcano above 39 290m as

    Mating with more than one male is the norm for females of many species. In addition to generating competition between the ejaculates of different males, multiple mating may allow females to bias sperm use. In Drosophila melanogaster, the last male to inseminate a female sires approximately 80% of subsequent progeny. Both sperm displacement, where resident sperm are removed from storage by the incoming ejaculate of the copulating male, and sperm incapacitation, where incoming seminal fluids supposedly interfere with resident sperm, have been implicated in this pattern of sperm use. But the idea of incapacitation is problematic because there are no known mechanisms by which an individual could damage rival sperm and not their own. Females also influence the process of sperm use, but exactly how is unclear. Here we show that seminal fluids do not kill rival sperm and that any 'incapacitation' is probably due to sperm ageing during sperm storage. We also show that females release stored sperm from the reproductive tract (sperm dumping) after copulation with a second male and that this requires neither incoming sperm nor seminal fluids. Instead, males may cause stored sperm to be dumped or females may differentially eject sperm from the previous mating
