The MSW and the MPA: Confrontation of Two Professions in Public Welfare
From its inception in the 1930\u27s, public welfare has emerged as a major service industry commanding a sizeable portion of the public tax dollar. Concomitant with growth and size and expenditure has come the federalization of the program. In the face of a series of changes--the growth of welfare, added administrative complexity, and the emergence of new economic function--public welfare\u27s identification with social work was weakened. Correspondingly, and perhaps logically, its identification with public administration has been strengthened. In this paper, there is exploration of the changes leading to the confrontation between social work and public administration within public welfare. A comparative analysis of this confrontation is undertaken, and the methods of conflict resolution between the professions are explored. The principal purpose of the paper, however, is to draw attention to the argument, stimulate further debate about this course of events, and encourage some public policy direction in public welfare

Interactions of Lipids with Aquaporin-0 and Other Membrane Proteins
The structure of aquaporin-0 (AQP0) has recently been determined by electron crystallography of two-dimensional (2D) crystals and by x-ray crystallography of three-dimensional (3D) crystals. The electron crystallographic structure revealed nine lipids per AQP0 monomer, which form an almost complete bilayer. The lipids adopt a wide variety of conformations and tightly fill the space between adjacent AQP0 tetramers. The conformations of the lipid acyl chains appear to be determined not only by the protein surface but also by the acyl chains of adjacent lipid molecules. In the x-ray structure, the hydrophobic region of the protein is surrounded by a detergent micelle, with two ordered detergent molecules per AQP0 monomer. Despite the different environments, the electron crystallographic and x-ray structures of AQP0 are virtually identical, but they differ in the temperature factors of the atoms that either contact the lipids in the 2D crystals or are exposed to
detergents in the 3D crystals. The temperature factors are higher in the x-ray structure, suggesting that the detergent-exposed AQP0 residues are less ordered than the corresponding ones contacting lipids in the 2D crystals. An examination of ordered detergent molecules in crystal structures of other aquaporins and of lipid molecules in 2D and 3D crystals of bacteriorhodopsin suggests that the increased conformational variability of detergent-exposed residues compared to lipid-contacting residues is a general feature.Chemistry and Chemical BiologyMolecular and Cellular Biolog
Principles of membrane protein interactions with annular lipids deduced from aquaporin-0 2D crystals
We have previously described the interactions of aquaporin-0 (AQP0) with dimyristoyl phosphatidylcholine (DMPC) lipids. We have now determined the 2.5 Å structure of AQP0 in two-dimensional (2D) crystals formed with Escherichia coli polar lipids (EPLs), which differ from DMPC both in headgroups and acyl chains. Comparison of the two structures shows that AQP0 does not adapt to the different length of the acyl chains in EPLs and that the distance between the phosphodiester groups in the two leaflets of the DMPC and EPL bilayers is almost identical. The EPL headgroups interact differently with AQP0 than do those of DMPC, but the acyl chains in the EPL and DMPC bilayers occupy similar positions. The interactions of annular lipids with membrane proteins seem to be driven by the propensity of the acyl chains to fill gaps in the protein surface. Interactions of the lipid headgroups may be responsible for the specific interactions found in tightly bound lipids but seem to have a negligible effect on interactions of generic annular lipids with membrane proteins
Migrations- und Integrationsforschung: Jahresbericht 2013 des Forschungszentrums Migration, Integration und Asyl im Bundesamt für Migration und Flüchtlinge
Der Jahresbericht gibt einen Überblick über die Tätigkeitsschwerpunkte des Forschungszentrums im Jahr 2013. Neben der Darstellung der laufenden und abgeschlossenen Forschungsprojekte werden Informationen zu Publikationen, Veranstaltungen, Vorträgen und der Vernetzung des Forschungszentrums gegeben. Aufgabe des Forschungszentrums des Bundesamts ist die Beobachtung des Migrationsgeschehens nach und von Deutschland, die Analyse von Auswirkungen von Migrationsprozessen und die Gewinnung von Erkenntnissen für die Migrationssteuerung. Daneben wird auch der Prozess der Integration von Menschen mit Migrationshintergrund sowie die Weiterentwicklung von Integrationsmaßnahmen auf Bundesebene begleitet. Zu diesem Zweck führt das Forschungszentrum zahlreiche Forschungsprojekte durch. Für 2013 sind dabei insbesondere die Forschungsprojekte "Integration von zugewanderten Ehepartnerinnen und Ehepartnern in Deutschland (BAMF-Heiratsmigrationsstudie 2013)", "Beschäftigungssituation von ausländischen Studienabsolventen deutscher Hochschulen", "Migrationspotenziale", "(Spät-)Aussiedler in Deutschland: Eine Analyse aktueller Daten und Forschungsergebnisse" und "Geschlechterrollen bei Deutschen und Zuwanderern christlicher und muslimischer Religionszugehörigkeit" zu nennen. Ein weiteres herausragendes Projekt ist der Migrationsbericht, der jährlich im Auftrag der Bundesregierung durch das Bundesamt erstellt wird und u.a. eine differenzierte Darstellung der einzelnen Zuwanderergruppen liefert. Neben einem Gesamtüberblick über alle Forschungsprojekte geht der Jahresbericht auch auf die vielfältigen Aktivitäten der Mitarbeiterinnen und Mitarbeiter des Forschungszentrums in Bezug auf Wissenstransfer, Öffentlichkeitsarbeit und Vernetzung ein, gibt Informationen zu den zahlreichen externen Publikationen und stellt eine Auswahl von Vorträgen dar

Allosteric β\u3csub\u3e1\u3c/sub\u3e integrin antibodies that stabilize the low affinity state by preventing the swing-out of the hybrid domain
The ligand binding function of integrins can be modulated by various monoclonal antibodies by both direct and indirect mechanisms. We have characterized an anti-β1 antibody, SG/19, that had been reported to inhibit the function of the β1 integrin on the cell surface. SG/19 recognized the wild type β1 subunit that exists in a conformational equilibrium between the high and low affinity states but bound poorly to a mutant β1 integrin that had been locked in a high affinity state. Epitope mapping of SG/19 revealed that Thr82 in the β1 subunit, located at the outer face of the boundary between the I-like and hybrid domains, was the key binding determinant for this antibody. Direct visualization of the α5β1 headpiece fragment in complex with SG/19 Fab with electron microscopy confirmed the location of the binding surface and showed that the ligand binding site is not occluded by the bound Fab. Surface plasmon resonance showed that α5β1 integrin bound by SG/19 maintained a low affinity toward its physiological ligand fibronectin (Fn) whereas binding by function-blocking anti-α5 antibodies resulted in a complete loss of fibronectin binding. Thus a class of the anti-β antibodies represented by SG/19 attenuate the ligand binding function by restricting the conformational shift to the high affinity state involving the swing-out of the hybrid domain without directly interfering with ligand docking
Novel perspectives for the application of total internal reflection microscopy
Total Internal Reflection Microscopy (TIRM) is a sensitive non-invasive
technique to measure the interaction potentials between a colloidal particle
and a wall with femtonewton resolution. The equilibrium distribution of the
particle-wall separation distance z is sampled monitoring the intensity I
scattered by the Brownian particle under evanescent illumination. Central to
the data analysis is the knowledge of the relation between I and the
corresponding z, which typically must be known a priori. This poses
considerable constraints to the experimental conditions where TIRM can be
applied (short penetration depth of the evanescent wave, transparent surfaces).
Here, we introduce a method to experimentally determine I(z) by relying only on
the distance-dependent particle-wall hydrodynamic interactions. We demonstrate
that this method largely extends the range of conditions accessible with TIRM,
and even allows measurements on highly reflecting gold surfaces where multiple
reflections lead to a complex I(z).Comment: 11 pages, 7 figure
Assembly of the SIR Complex and Its Regulation by O-Acetyl-ADP-Ribose, a Product of NAD-Dependent Histone Deacetylation
SummaryAssembly of silent chromatin domains in budding yeast involves the deacetylation of histone tails by Sir2 and the association of the Sir3 and Sir4 proteins with hypoacetylated histone tails. Sir2 couples deacetylation to NAD hydrolysis and the synthesis of a metabolite, O-acetyl-ADP-ribose (AAR), but the functional significance of NAD hydrolysis or AAR, if any, is unknown. Here we examine the association of the Sir2, Sir3, and Sir4 proteins with each other and histone tails. Our analysis reveals that deacetylation of histone H4-lysine 16 (K16), which is critical for silencing in vivo, is also critical for the binding of Sir3 and Sir4 to histone H4 peptides in vitro. Moreover, AAR itself promotes the association of multiple copies of Sir3 with Sir2/Sir4 and induces a dramatic structural rearrangement in the SIR complex. These results suggest that Sir2 activity modulates the assembly of the SIR complex through both histone deacetylation and AAR synthesis
