    Combining Electrochemical Nitrate Reduction and Anammox for Treatment of Nitrate-Rich Wastewater: A Short Review

    Treatment of nitrate-rich wastewater is important but challenging for the conventional biological denitrification process. Here, we propose combining the electrochemical reduction and anaerobic ammonium oxidation (anammox) processes together for treatment of nitrate-rich wastewater. This article reviews the mechanism and current research status of electrochemical reduction of nitrate to ammonium as well as the mechanism and applicability of the anammox process. This article discusses the principles, superiorities, and challenges of this combined process. The feasibility of the combined process depends on the efficiency of electrochemical nitrate reduction to ammonium and the conditions in the anammox process to use the reduced ammonium as the substrate to achieve deep nitrogen removal. The article provides a feasible strategy for using the electrochemical reduction and anammox combined process to treat nitrate-rich wastewater

    Ultrasound hyperthermia induces apoptosis in head and neck squamous cell carcinoma : an in vitro study

    Hyperthermia is considered an efficient complement in the treatment of head and neck squamous cell carcinoma (HNSCC). Hyperthermia induces cell apoptosis in a temperature- and time-dependent manner. However, the molecular mechanism of hyperthermia remains unclear. The aim of this study was to investigate the mechanism of apoptosis induced by ultrasound hyperthermia in HNSCC cell lines HN-30 and HN-13. We examined the dynamic changes of early apoptosis and secondary necrosis in HN-30 and HN-13 cells treated by hyperthermia at 42°C for 10 min. We further examined mitochondrial membrane potential in vitro by ultrasound hyperthermia for different heating temperatures (38-44°C, 10 min) and heating times (42°C, 10-50 min). After heating by ultrasound at 42°C for 10 min, the apoptosis index achieved its highest level at 8 h after treatment, decreased rapidly and remained constant at a reduced level at 12 h. The level of secondary necrosis increased with the level of early apoptosis but remained at a higher level until 14 h. The level of secondary necrosis correlated with the level of early apoptosis (HN-13: r=0.7523, P=0.0030; HN-30: r=0.6510, P=0.016). The fractions of cells with low mitochondrial membrane potential (??) in the heating-temperature grads group and heating-time grads group decreased significantly over time. Therefore, HN-30 and HN-13 cells developed apoptosis after ultrasound hyperthermia treatment with decreases in the mitochondrial transmembrane potential level.     Loss of NLRP3 reduces oxidative stress and polarizes intratumor macrophages to attenuate immune attack on endometrial cancer

    IntroductionThe interaction between endometrial cancer (EMC) cells and intratumoral macrophages plays a significant role in the development of the disease. PYD domains-containing protein 3 (NLRP3) inflammasome formation triggers caspase-1/IL-1β signaling pathways and produces reactive oxygen species (ROS) in macrophages. However, the role of NLRP3-regulated ROS production in macrophage polarization and the subsequent growth and metastasis of EMC remains unknown.MethodsWe conducted bioinformatic analysis to compare NLRP3 levels in intratumoral macrophages from EMC and normal endometrium. In vitro experiments involved knocking out NLRP3 in macrophages to shift the polarization from an anti-inflammatory M1-like phenotype to a proinflammatory M2-like phenotype and reduce ROS production. The impact of NLRP3 depletion on the growth, invasion, and metastasis of co-cultured EMC cells was assessed. We also evaluated the effect of NLRP3 depletion in macrophages on the growth and metastasis of implanted EMC cells in mice.ResultsOur bioinformatic analysis showed significantly lower NLRP3 levels in intratumoral macrophages from EMC than those from normal endometrium. Knocking out NLRP3 in macrophages shifted their polarization to a proinflammatory M2-like phenotype and significantly reduced ROS production. NLRP3 depletion in M2-polarized macrophages increased the growth, invasion, and metastasis of co-cultured EMC cells. NLRP3 depletion in M1-polarized macrophages reduced phagocytic potential, which resulted in weakened immune defense against EMC. Additionally, NLRP3 depletion in macrophages significantly increased the growth and metastasis of implanted EMC cells in mice, likely due to compromised phagocytosis by macrophages and a reduction in cytotoxic CD8+ T cells.DiscussionOur results suggest that NLRP3 plays a significant role in regulating macrophage polarization, oxidative stress, and immune response against EMC. NLRP3 depletion alters the polarization of intratumoral macrophages, leading to weakened immune defense against EMC cells. The reduction in ROS production by the loss of NLRP3 may have implications for the development of novel treatment strategies for EMC

    Remarkable changes have taken in social operation mode and consumers’ behavior mode because of the foot ban during the pandemic spread of COVID-19. Digital technologies such as Blockchain have shown potential in gaining competitive advantages for enterprises in such situations. This study aims to provide an insight into how to gain consumer loyalty through the use of modern and efficient Blockchain technology. In contrast to the current literature, this study combined the technology acceptance model of planned behavior theory, social exchange theory to explain the loyalty of an online travel agency (OTA) consumer. A self-administered questionnaire was used to collect data from citizens in Hangzhou, a city full of technological innovation atmosphere. Using structural equation modeling with SmartPLS©, responses from 1403 citizens were analyzed. Social norm was discovered to have a positive and significant association with the consumers’ attitude toward technology and thus enhancing the perceived usability and hedonism of OTA application, which can increase consumer loyalty. The findings suggested OTAs may gain consumers’ loyalty through adopting Blockchain technology, and local governments have played a key role in creating such an environment.     Transmission Risk Prediction and Evaluation of Mountain-Type Zoonotic Visceral Leishmaniasis in China Based on Climatic and Environmental Variables

    With global warming and socioeconomic developments, there is a tendency toward the emergence and spread of mountain-type zoonotic visceral leishmaniasis (MT-ZVL) in China. Timely identification of the transmission risk and spread of MT-ZVL is, therefore, of great significance for effectively interrupting the spread of MT-ZVL and eliminating the disease. In this study, 26 environmental variables—namely, climatic, geographical, and 2 socioeconomic indicators were collected from regions where MT-ZVL patients were detected during the period from 2019 to 2021, to create 10 ecological niche models. The performance of these ecological niche models was evaluated using the area under the receiver-operating characteristic curve (AUC) and true skill statistic (TSS), and ensemble models were created to predict the transmission risk of MT-ZVL in China. All ten ecological niche models were effective at predicting the transmission risk of MT-ZVL in China, and there were significant differences in the mean AUC (H = 33.311, p p 0.95, TSS > 0.85). Ensemble models predicted a transmission risk of MT-ZVL in the provinces of Shanxi, Shaanxi, Henan, Gansu, Sichuan, and Hebei, which was centered in Shanxi Province and presented high spatial clustering characteristics. Multiple ensemble ecological niche models created based on climatic and environmental variables are effective at predicting the transmission risk of MT-ZVL in China. This risk is centered in Shanxi Province and tends towards gradual radiation dispersion to surrounding regions. Our results provide insights into MT-ZVL surveillance in regions at high risk of MT-ZVL

    Primary undifferentiated pleomorphic sarcoma in oral-maxillary area: retrospective study and molecular analysis

    y. Undifferentiated pleomorphic sarcoma (UPS) in oral-maxillary area is rarely reported. Herein, we aimed to investigate the clinical characteristics, treatment strategies, prognosis, and molecular features of the oral-maxillary UPS. In total, 10 cases with primary oral-maxillary UPS were included. The rapidly progressive UPS can easily develop to an advanced and life-threatening stage, especially concerning the complex anatomical structures and spaces in the oral-maxillary area. The final diagnosis for UPS greatly depended on histological findings and immunohistochemistry staining after the exclusion of all possible differential diagnoses. Retrospectively, the treatment strategies for the included cases still referred to those of oral squamous cell carcinoma (OSCC). Statistically, the median overall survival (OS) for all the included cases was 7.75 months (range: 5-17 months). Comparatively, 3 cases had improved OS (median survival: 17 months, range: 17-18 months) and experienced PR/SD with neoadjuvant chemotherapy (anlotinib). The molecular features were demonstrated by using whole exonic sequencing for 1 included case. Cancer driver gene detection revealed GBP4 as a candidate driver gene for the primary oralmaxillary UPS. Additionally, a missense mutation in gene PIK3CA (p.E545K) was also identified. Our findings could greatly expand the knowledge about primary oral-maxillary UPS, and provide molecular evidences to improve the therapeutic options for primary oral-maxillary UPS
