    Water transfer and crack regimes in nano-colloidal gels

    International audienceDirect observations of the surface and shape of model nano-colloidal gels associated with measurements of the spatial distribution of water content during drying show that air starts to significantly penetrate the sample when the material stops shrinking. We show that whether the material fractures or not during desiccation, as air penetrates the porous body, the water saturation decreases but remains almost homogeneous throughout the sample. This air-invasion is at the origin of another type of fracture due to capillary effects; these results provide a new insight in the liquid dynamics at the nano-scale.     Grape marc extract acts as elicitor of plant defence responses

    International audiencePlant protection based on novel alternative strategies is a major concern in agriculture to sustain pest management. The marc extract of red grape cultivars reveals plant defence inducer properties. Treatment with grape marc extract efficiently induced hypersensitive reaction-like lesions with cell death evidenced by Evans Blue staining of tobacco leaves. Examination of the infiltration zone and the surrounding areas under UV light revealed the accumulation of autofluorescent compounds. Both leaf infiltration and a foliar spray of the red grape extract on tobacco leaves induced defence gene expression. The PR1 and PR2 target genes were upregulated locally and systemically in tobacco plants following grape marc extract treatment. The grape extract elicited an array of plant defence responses making this natural compound a potential phytosanitary product with a challenging issue and a rather attractive option for sustainable agriculture and environmentally friendly practices

    Un nouvel indicateur intégré d’évaluation des dégâts occasionnés aux grappes par des bioagresseurs majeurs au vignoble

    Communication faite au cours du colloque DinABio2013, 13 et 14 novembre 2013; Tours, FranceAn original and integrative evaluation indicator has been developed to quantify the cumulated damage from major pests and diseases affecting grape bunches: downy mildew, powdery mildew, gray mould and tortricid moths. It made it possible to estimate the associated crop losses and to relate them to the plant protection strategy in different modes of production (organic farming, in-transition, conventional). Thus, overall plant losses were higher in 2012 than in 2011. The in-transition growers’ strategy, with reduced copper doses but increased numbers of sprays, led to a 20% increase in average severity on bunches (essentially due to Downy mildew). The more pragmatic approach of experienced organic growers and conventional ones (higher doses and fewer sprays) reduced the yield losses. The proposed indicator is used for two purposes, i) evaluating the quantitative losses due to pest attacksand ii) differentiating them from other non-pest ones. A more detailed analysis including the impact on performance will be achieved and published soon.Un indicateur d’évaluation, l’IEDG (Indicateur d’Evaluation des Dégâts sur Grappes), a été mis au point pour quantifier les dégâts cumulés dus aux principaux bioagresseurs affectant les grappes de raisin : mildiou, oïdium, pourriture grise et tordeuses. Il permet d’estimer la perte de récolte imputable au cortège parasitaire et de faire le lien avec la stratégie phytosanitaire adoptée (caractérisée ici par l’IFT) et le mode de production (AB, conversion, conventionnel). Ainsi, les pertes sanitaires ont été supérieures en 2012 par rapport à 2011. La stratégie phytosanitaire des viticulteurs en conversion, basée sur des réductions de dose de cuivre de près de 80% et des passages plus nombreux dans les parcelles, n’a pas été efficiente en 2012 avec des sévérités proches de 20% sur grappe, essentiellement dues au mildiou. L’utilisation de doses d’applications supérieures et moins de passages dans les parcelles limite les dommages chez les autres viticulteurs. L’indicateur proposé permet d’évaluer les pertes quantitatives générées par les attaques de bioagresseurs et de les différencier des autres pertes non parasitaires. Une analyse plus fine incluant l’effet région et l’impact sur le rendement devra être réalisée

    2-Substituted vs 4-substituted-9,9′-spirobifluorene host materials for green and blue phosphorescent OLEDs: a structure-property relationship study

    Nanotek for organic synthesis, and organic synthesis for nanotekInternational audienceWe report a structure-property relationship study of four 9,9′-spirobifluorene (SBF) derivatives (4-5Pm-SBF, 2-5Pm-SBF, 4-Ph-SBF and 2-Ph-SBF), substituted with either phenyl or pyrimidine at the \C2\ or \C4\ position of the \SBF\ core. Structural, thermal, electrochemical and photophysical properties have been examined and correlated to theoretical calculations in order to study the influence of the nature and the position of the substituent. The emission properties of 4- versus 2-substituted \SBFs\ are noticeably different highlighting, in the excited state, the remarkable effect of substitution in ortho position of SBF. All compounds have been used as host material for green dopant in PhOLEDs with very high performances (2-5Pm-SBF: CE>58 cd/A, PE>35 lm/W, EQE>14%). More importantly, the two 4-substituted \SBFs\ have been used as host materials in blue PhOLEDs, displaying high performance and a decrease of \VTH\ for 4-5Pm-SBF due to the incorporation of the electron-withdrawing pyrimidine

    Putting hornets on the genomic map

    Hornets are the largest of the social wasps, and are important regulators of insect populations in their native ranges. Hornets are also very successful as invasive species, with often devastating economic, ecological and societal effects. Understanding why these wasps are such successful invaders is critical to managing future introductions and minimising impact on native biodiversity. Critical to the management toolkit is a comprehensive genomic resource for these insects. Here we provide the annotated genomes for two hornets, Vespa crabro and Vespa velutina. We compare their genomes with those of other social Hymenoptera, including the northern giant hornet Vespa mandarinia. The three hornet genomes show evidence of selection pressure on genes associated with reproduction, which might facilitate the transition into invasive ranges. Vespa crabro has experienced positive selection on the highest number of genes, including those putatively associated with molecular binding and olfactory systems. Caste-specific brain transcriptomic analysis also revealed 133 differentially expressed genes, some of which are associated with olfactory functions. This report provides a spring-board for advancing our understanding of the evolution and ecology of hornets, and opens up opportunities for using molecular methods in the future management of both native and invasive populations of these over-looked insects

    Scaphoideus titanus up-to-the-minute: biology, ecology, and role as a vector

    Native to the Nearctic region, Scaphoideus titanus Ball (Hemiptera: Cicadellidae) has become a major threat for grapevine production after being unintentionally introduced into Europe, where it became the main vector of flavescence dorée phytoplasma, being mainly associated with the genus Vitis. Scaphoideus titanus is a highly efficient vector of the most important phytoplasma affecting grapevine. For this reason, compulsory insecticide treatments have been introduced against this pest in many European countries. Moreover, the continuous expansion of its geographical distribution makes this leafhopper a serious threat for several non-European Countries. In this article, we review the current knowledge about its taxonomy, morphology, biology, ecology, and its role as a vector. Finally, we point out the main challenges for research aimed at reducing S. titanus and flavescence dorée expansion across Europe and avoiding spread of the disease outside the Old World

    Scaphoideus titanus forecasting and management: quo vadis?

    Scaphoideus titanus Ball (Hemiptera: Cicadellidae) is a major pest for European viticulture due to its high efficiency in the transmission of one of the most destructive pathogens for grapevine, namely flavescence dorée phytoplasmas. Although it plays a major role in spreading this disease, S. titanus is part of a complex epidemiological cycle involving several alternative vectors with variable relevance for phytoplasma spread. Here we provide an updated review on S. titanus monitoring and modelling, as well as the available tools for management of this pest and for limiting phytoplasma transmission and, thus, also spread. Insecticide-based control is examined; additional emphasis is placed on innovative and low-impact control approaches, such as vibrational mating disruption, biocontrol, and methods to reduce vector competence. We also discuss the main emerging challenges to the implementation of effective and sustainable control programs against S. titanus

    miR-200 Enhances Mouse Breast Cancer Cell Colonization to Form Distant Metastases

    BACKGROUND: The development of metastases involves the dissociation of cells from the primary tumor to penetrate the basement membrane, invade and then exit the vasculature to seed, and colonize distant tissues. The last step, establishment of macroscopic tumors at distant sites, is the least well understood. Four isogenic mouse breast cancer cell lines (67NR, 168FARN, 4TO7, and 4T1) that differ in their ability to metastasize when implanted into the mammary fat pad are used to model the steps of metastasis. Only 4T1 forms macroscopic lung and liver metastases. Because some miRNAs are dysregulated in cancer and affect cellular transformation, tumor formation, and metastasis, we examined whether changes in miRNA expression might explain the differences in metastasis of these cells. METHODOLOGY/PRINCIPAL FINDINGS: miRNA expression was analyzed by miRNA microarray and quantitative RT-PCR in isogenic mouse breast cancer cells with distinct metastatic capabilities. 4T1 cells that form macroscopic metastases had elevated expression of miR-200 family miRNAs compared to related cells that invade distant tissues, but are unable to colonize. Moreover, over-expressing miR-200 in 4TO7 cells enabled them to metastasize to lung and liver. These findings are surprising since the miR-200 family was previously shown to promote epithelial characteristics by inhibiting the transcriptional repressor Zeb2 and thereby enhancing E-cadherin expression. We confirmed these findings in these cells. The most metastatic 4T1 cells acquired epithelial properties (high expression of E-cadherin and cytokeratin-18) compared to the less metastatic cells. CONCLUSIONS/SIGNIFICANCE: Expression of miR-200, which promotes a mesenchymal to epithelial cell transition (MET) by inhibiting Zeb2 expression, unexpectedly enhances macroscopic metastases in mouse breast cancer cell lines. These results suggest that for some tumors, tumor colonization at metastatic sites might be enhanced by MET. Therefore the epithelial nature of a tumor does not predict metastatic outcome
