    The effect of autonomy, training opportunities, age and salaries on job satisfaction in the South East Asian retail petroleum industry

    South East Asian petroleum retailers are under considerable pressure to improve service quality by reducing turnover. An empirical methodology from this industry determined the extent to which job characteristics, training opportunities, age and salary influenced the level of job satisfaction, an indicator of turnover. Responses are reported on a random sample of 165 site employees (a 68% response rate) of a Singaporean retail petroleum firm. A restricted multivariate regression model of autonomy and training opportunities explained the majority (35.4%) of the variability of job satisfaction. Age did not moderate these relationships, except for employees >21 years of age, who reported enhanced job satisfaction with additional salary. Human Capital theory, Life Cycle theory and Job Enrichment theory are invoked and explored in the context of these findings in the South East Asian retail petroleum industry. In the South East Asian retail petroleum industry, jobs providing employees with the opportunity to undertake a variety of tasks that enhanced the experienced meaningfulness of work are likely to promote job satisfaction, reduce turnover and increase the quality of service

    Measurement of the ratio of the ppˉWp\bar{p}\to W+cc-jet cross section to the inclusive ppˉWp\bar{p}\to W+jets cross section

    We present a measurement of the fraction of inclusive WW+jets events produced with net charm quantum number ±1\pm1, denoted WW+cc-jet, in ppˉp\bar{p} collisions at s=1.96\sqrt{s}=1.96 TeV using approximately 1~fb1^{-1} of data collected by the D0 detector at the Fermilab Tevatron Collider. We identify the WW+jets events via the leptonic WW boson decays. Candidate WW+cc-jet events are selected by requiring a jet containing a muon in association with a reconstructed WW boson and exploiting the charge correlation between this muon and WW boson decay lepton to perform a nearly model-independent background subtraction. We measure the fraction of WW+cc-jet events in the inclusive WW+jets sample for jet pT>20p_{T}>20 GeV and pseudorapidity η<2.5|\eta|<2.5 to be 0.074±0.019\pm0.019(stat.)±0.0140.012\pm^{0.012}_{0.014}(syst.), in agreement with theoretical predictions. The probability that background fluctuations could produce the observed fraction of WW+cc-jet events is estimated to be 2.5×1042.5\times 10^{-4}, which corresponds to a 3.5 σ\sigma statistical significance.Comment: submitted to Physics Letters

    Cancer cell growth requires fatty acids to replicate cellular membranes. The kinase Akt is known to up-regulate fatty acid synthesis and desaturation, which is carried out by the oxygen-consuming enzyme stearoyl-CoA desaturase (SCD)1. We used 13C tracers and lipidomics to probe fatty acid metabolism, including desaturation, as a function of oncogene expression and oxygen availability. During hypoxia, flux from glucose to acetyl-CoA decreases, and the fractional contribution of glutamine to fatty acid synthesis increases. In addition, we find that hypoxic cells bypass de novo lipogenesis, and thus, both the need for acetyl-CoA and the oxygen-dependent SCD1-reaction, by scavenging serum fatty acids. The preferred substrates for scavenging are phospholipids with one fatty acid tail (lysophospholipids). Hypoxic reprogramming of de novo lipogenesis can be reproduced in normoxic cells by Ras activation. This renders Ras-driven cells, both in culture and in allografts, resistant to SCD1 inhibition. Thus, a mechanism by which oncogenic Ras confers metabolic robustness is through lipid scavenging

    Genome-wide association studies have identified a signal at the SLC22A1 locus for serum acylcarnitines, intermediate metabolites of mitochondrial oxidation whose plasma levels associate with metabolic diseases. Here, we refined the association signal, performed conditional analyses, and examined the linkage structure to find coding variants of SLC22A1 that mediate independent association signals at the locus. We also employed allele-specific expression analysis to find potential regulatory variants of SLC22A1 and demonstrated the effect of one variant on the splicing of SLC22A1. SLC22A1 encodes a hepatic plasma membrane transporter whose role in acylcarnitine physiology has not been described. By targeted metabolomics and isotope tracing experiments in loss- and gain-of-function cell and mouse models of Slc22a1, we uncovered a role of SLC22A1 in the efflux of acylcarnitines from the liver to the circulation. We further validated the impacts of human variants on SLC22A1-mediated acylcarnitine efflux in&nbsp;vitro, explaining their&nbsp;association with serum acylcarnitine levels. Our findings provide the detailed molecular mechanisms of the GWAS association for serum acylcarnitines at the SLC22A1 locus by functionally validating the impact of SLC22A1 and its variants on acylcarnitine transport
