    The Kazhdan-Lusztig conjecture for W-algebras

    The main result in this paper is the character formula for arbitrary irreducible highest weight modules of W algebras. The key ingredient is the functor provided by quantum Hamiltonian reduction, that constructs the W algebras from affine Kac-Moody algebras and in a similar fashion W modules from KM modules. Assuming certain properties of this functor, the W characters are subsequently derived from the Kazhdan-Lusztig conjecture for KM algebras. The result can be formulated in terms of a double coset of the Weyl group of the KM algebra: the Hasse diagrams give the embedding diagrams of the Verma modules and the Kazhdan-Lusztig polynomials give the multiplicities in the characters.Comment: uuencoded file, 29 pages latex, 5 figure

    Identification of Lactobacillus plantarum genes modulating the cytokine response of human peripheral blood mononuclear cells

    <p>Abstract</p> <p>Background</p> <p>Modulation of the immune system is one of the most plausible mechanisms underlying the beneficial effects of probiotic bacteria on human health. Presently, the specific probiotic cell products responsible for immunomodulation are largely unknown. In this study, the genetic and phenotypic diversity of strains of the <it>Lactobacillus plantarum </it>species were investigated to identify genes of <it>L. plantarum </it>with the potential to influence the amounts of cytokines interleukin 10 (IL-10) and IL-12 and the ratio of IL-10/IL-12 produced by peripheral blood mononuclear cells (PBMCs).</p> <p>Results</p> <p>A total of 42 <it>Lactobacillus plantarum </it>strains isolated from diverse environmental and human sources were evaluated for their capacity to stimulate cytokine production in PBMCs. The <it>L. plantarum </it>strains induced the secretion of the anti-inflammatory cytokine IL-10 over an average 14-fold range and secretion of the pro-inflammatory cytokine IL-12 over an average 16-fold range. Comparisons of the strain-specific cytokine responses of PBMCs to comparative genome hybridization profiles obtained with <it>L. plantarum </it>WCFS1 DNA microarrays (also termed gene-trait matching) resulted in the identification of 6 candidate genetic loci with immunomodulatory capacities. These loci included genes encoding an <it>N</it>-acetyl-glucosamine/galactosamine phosphotransferase system, the LamBDCA quorum sensing system, and components of the plantaricin (bacteriocin) biosynthesis and transport pathway. Deletion of these genes in <it>L. plantarum </it>WCFS1 resulted in growth phase-dependent changes in the PBMC IL-10 and IL-12 cytokine profiles compared with wild-type cells.</p> <p>Conclusions</p> <p>The altered PBMC cytokine profiles obtained with the <it>L. plantarum </it>WCFS1 mutants were in good agreement with the predictions made by gene-trait matching for the 42 <it>L. plantarum </it>strains. This study therefore resulted in the identification of genes present in certain strains of <it>L. plantarum </it>which might be responsible for the stimulation of anti- or pro-inflammatory immune responses in the gut.</p

    Cystic Ovarian Follicles in Cattle

    The objective of this study was to determine the effects of different intramuscular dosages of human chorionic gonadotropin (hCG) on ovarian follicular development of dairy cows diagnosed with refractory cystic ovarian follicles (COFs). Cows diagnosed with COFs (≥25mm in diameter) were allocated to four treatment groups: hCG-1 (n = 3), a single dose of 4,500 IU on day 1; hCG-2 (n = 3), 2,250 IU on days 1 and 3; hCG-3 (n = 3), 1,500 IU on days 1, 3, and 5; and hCG-C (n = 3) received saline on day 1. Blood sampling and ovarian ultrasonographic (US) examinations were performed on days 1, 3, 5, 7, and 14. A progesterone (P4) value 1 ng/ml was 100% (3/3) and 100% (3/3) in group hCG-1; 100% (3/3) and 67% (2/3) in group hCG-2; 67%(2/3) and 100%(3/3) in group hCG-3; and 33%(1/3) and 33%(1/3) in group hCG-C, respectively. Strong tendencies of P4 increases in group hCG-1 (P = 0.054) and hCG-2 (P = 0.051) were measured after hCG administration. Additionally, P4 values tended to be higher (P = 0.07) for group hCG-1 compared to group hCG-C on day 5. The preliminary findings of this study suggest that multiple smaller doses of hCG might be equally effective as a single large dose of hCG in modulating ovarian follicular development in dairy cows with COFs

    Dynamic beam based calibration of orbit monitors at LEP

    The offsets of the beam position monitors (BOM) with respect to the centre of the quadrupoles were determined for 16 wide band electronic and 18 narrow band electronic monitors. The first are located near to the IPs and the latter are in all other parts. The distribution of the wide band monitor offsets at the low beta focusing quadrupole magnets has a mean value of 1000 µm and a width of s = 600 mm. The offsets are almost identical with the beam position of the orbit which allows to obtain highest luminosity. The distribution of the narrow band monitor offsets has a mean value of 19 mm and a width of s = 245 mm

    Relationship between quantum decoherence times and solvation dynamics in condensed phase chemical systems

    A relationship between the time scales of quantum coherence loss and short-time solvent response for a solute/bath system is derived for a Gaussian wave packet approximation for the bath. Decoherence and solvent response times are shown to be directly proportional to each other, with the proportionality coefficient given by the ratio of the thermal energy fluctuations to the fluctuations in the system-bath coupling. The relationship allows the prediction of decoherence times for condensed phase chemical systems from well developed experimental methods.Comment: 10 pages, no figures, late

    Graphical genotyping as a method to map Ny ((o,n)sto) and Gpa5 using a reference panel of tetraploid potato cultivars

    The method of graphical genotyping is applied to a panel of tetraploid potato cultivars to visualize haplotype sharing. The method allowed to map genes involved in virus and nematode resistance. The physical coordinates of the amount of linkage drag surrounding these genes are easily interpretable. Graphical genotyping is a visually attractive and easily interpretable method to represent genetic marker data. In this paper, the method is extended from diploids to a panel of tetraploid potato cultivars. Application of filters to select a subset of SNPs allows one to visualize haplotype sharing between individuals that also share a specific locus. The method is illustrated with cultivars resistant to Potato virus Y (PVY), while simultaneously selecting for the absence of the SNPs in susceptible clones. SNP data will then merge into an image which displays the coordinates of a distal genomic region on the northern arm of chromosome 11 where a specific haplotype is introgressed from the wild potato species S. stoloniferum (CPC 2093) carrying a gene (Ny ((o,n)sto) ) conferring resistance to two PVY strains, PVYO and PVYNTN. Graphical genotyping was also successful in showing the haplotypes on chromosome 12 carrying Ry-f (sto) , another resistance gene derived from S. stoloniferum conferring broad-spectrum resistance to PVY, as well as chromosome 5 haplotypes from S. vernei, with the Gpa5 locus involved in resistance against Globodera pallida cyst nematodes. The image also shows shortening of linkage drag by meiotic recombination of the introgression segment in more recent breeding material. Identity-by-descent was found to be a requirement for using graphical genotyping, which is proposed as a non-statistical alternative method for gene discovery, as compared with genome-wide association studies.     Molecular dynamics simulations reveal that AEDANS is an inert fluorescent probe for the study of membrane proteins

    Computer simulations were carried out of a number of AEDANS-labeled single cysteine mutants of a small reference membrane protein, M13 major coat protein, covering 60% of its primary sequence. M13 major coat protein is a single membrane-spanning, α-helical membrane protein with a relatively large water-exposed region in the N-terminus. In 10-ns molecular dynamics simulations, we analyze the behavior of the AEDANS label and the native tryptophan, which were used as acceptor and donor in previous FRET experiments. The results indicate that AEDANS is a relatively inert environmental probe that can move unhindered through the lipid membrane when attached to a membrane protein
