Nanotecnología aplicada a la medicina
La nanotecnología comprende la creación, manipulación y utilización de materiales en el orden nanométrico (10-9 m) y su aplicación en los campos de la química, la biología, la física, la ingeniería y en especial la medicina, se ha traducido en trascendentales avances en la ciencia. La hoy denominada nanomedicina ha comportado significativos adelantos en la monitorización, la reparación de tejidos, el diagnostico, el tratamiento y la prevención de enfermedades mediante el uso de herramientas como nanosensores, nanopartículas y nanotubos, y en el futuro cercano nanorrobots capaces de penetrar en el organismo humano para repararlo. La nanotecnología se encuentra en una fase de crecimiento y ha capturado el interés de empresas, gobiernos e inversionistas que buscan desarrollar productos útiles para aplicarlos en diferentes esferas, especialmente en medicina
Annonaceae squamosa una fuente de acetogeninas en la actualidad
Chemical studies on plants of the Annonaceae family have intensified in recent decades due to the discovery of molecules with medicinal potential. Moreover, until 2004, 593 acetogenins (ACG), 51 species in 13 genera were identified. This suggests that plants of this family allocate significant resources for the biosynthesis of these compounds. Despite the diversity of these molecules, their biological functions, including their physiological and ecological functions are not well understood. This paper presents a review of their knowledge, in particular on the biosynthetic pathway and biogenetic acetogenins, which will improve our understanding of secondary metabolism of these pharmacologically important molecules.Estudios químicos en plantas de la familia Annonaceae se han intensificado en las últimas décadas, debido al descubrimiento de moléculas con potencial medicinal. Además, hasta el 2004, se identificaron 593 acetogeninas (ACG), de 51 especies en 13 géneros. Esto sugiere que las plantas de esta familia asignan recursos importantes para la biosíntesis de estos compuestos. A pesar de la diversidad de estas moléculas, sus funciones biológicas, incluyendo sus funciones fisiológicas o ecológicos, no son bien entendidos. Este trabajo presenta una revisión sobre su conocimiento, en particular sobre la ruta biosintética y biogenética de las acetogeninas, lo cual mejorará nuestra comprensión del metabolismo secundario de estas moléculas farmacológicamente importantes
Review of Occupational Implications Due to Exposure to Lead
El plomo, considerado uno de los metales tóxicos más comunes en el medio
ambiente, es frecuentemente utilizado en el mundo para la fabricación de armas,
baterías y aditivos para la gasolina, entre otras aplicaciones. La exposición crónica
a este contaminante es un problema de salud pública que afecta sistemas
importantes como el nervioso, el reproductivo y el circulatorio. En varios países
la búsqueda de materiales que reemplacen el plomo ha sido implementada con
buenos resultados. En Colombia no existen regulaciones nacionales concretas
que garanticen el desempeño ocupacional en ambientes libres de plomo, por
lo que se hace necesaria la creación de regulaciones que permitan minimizar el
riesgo de exposición a este metal tóxico.Lead, considered one of the most common toxic metals in the environment,
is often used in the world for the manufacture of weapons, batteries,
and gasoline additives, among other applications. Chronic exposure to
this contaminant is a health problem, affecting the nervous, reproductive,
and circulatory systems. The search for materials to replace lead has
been implemented in several countries with good results. In Colombia,
no regulations exist to ensure occupational performance
in lead-free environments; thereby, it is necessary to create regulations

Tribolium castaneum HERBST (Coleoptera: Tenebrionidae) AS a toxicity model for the study of volatile chemicals
Humans are exposed to thousands of chemical substances, therefore, the
responses of these toxic substances and their interactions with
environmental factors can occur at various time scales and at different
levels of biological complexity. Currently, there are more than five
million man-made chemicals and the number of new products that
appear annually has increased enormously, generating a growing gap
between the evaluation capacity and the need of new toxicity detection
tools. In vivo studies are used to determine different levels of toxicity;
however, these tests can be very expensive, consume more time and use
a large number of animals. Therefore, it is necessary to develop new
models for the evaluation of toxicity based on the principles 3R - Reduce,
Replace, Refine. Invertebrate organisms have an easy and economical
maintenance, a short life cycle, small size, simple anatomy and are
extremely useful to provide an early and sensitive detection of toxicity,
as well as a better understanding of the biochemical mechanisms
involved, allowing a more predictable of the toxicity of unknown
compounds. In addition, a large number of invertebrates can be studied
in a single experiment within a short period of time, with fewer ethical
problems and its maintenance cost is lower compared to other animals.
Tribolium castaneum, is a model organism for agricultural and medical
research, some authors consider it a better representative of the Insecta
class than Drosophila melanogaster. In view of the need for new toxicity
models, this work aimed to propose the T. castaneum insect as a toxicity
model for the study of volatile chemicals of natural and synthetic origin,
evaluating the molecular mechanisms involved.
Based on the above, the work contemplates four phases, described
below. In the first, the development of the insect in laboratory conditions
and its life cycle was evaluated. In the second, the effects of benzene,
naphthalene, toluene, xylene, and thinner are studied in several genetic
markers of toxicity on the red flour beetle, using the polymerase chain
reaction (PCR) in real time, and the lethal concentration 50 (LC50) of each
chemical was determined. In addition, gas chromatography coupled to
mass spectrometry was used to identify the compounds present in the
thinner and the naphthalin balls. The third phase, evaluates three
toxicological endpoints (development, growth, and reproduction) in the
Coleoptera exposed to eight toxic chemicals. Finally, the expression of
genes related to neurotransmission in T. castaneum exposed to linalool
and β-pinene was studied, using real-time PCR, in addition the repellent
concentration 50 (RC50) was determined and in silico interaction with
important proteins for the insect was evaluated.
The results obtained show that the development cycle of the immature
stages of the insect is in the range of 46-81 days. The egg stage has a
duration of 6-7 days, and a size of 0.6 ± 0.0 x 0.3 ± 0.0 mm, the larvae can
reach up to eight instars and can go to the pupal stage from the fourth
instar. The pupa stage lasts 7 ± 1.7 days and a size of 3 ± 0.1 x 1.1 ± 0.0
mm. Adults have a size of 3.6 ± 0.0 x 1.1 ± 0.0? mm.
The LC50 for naphthalene, naphthalin and benzene was 63.6, 20.0 and
115.9 μL/L in air, respectively. Naphthalin balls mainly contained
naphthalene and in a lesser proportion benzo[b]thiophene. Real-time
PCR analysis revealed changes in the expression of genes related to
oxidative stress, metabolism, reproduction, metamorphosis, and
neurotransmission in naphthalene-exposed insects. Adults exposed to
benzene overexpressed genes related to neurotransmission,
reproduction, metamorphosis and development.
The main compounds identified in the thinner with peak area > 2% were
p-xylene (6.00%), toluene (3.98%), 2,4-dimethylheptane (2.99%),
methylcyclohexane, (2.75%), 2-methylheptane, (2.04%), cyclohexanone
(2.58%) and nonane (2.10%). Xylene was highly toxic and most animals
did not survive 4 hours of exposure. The LC50 values for toluene, xylene
and thiner at 48 h of exposure were 97.7, <40.0 and 99.8 μL/L of air,
respectively. The gene expression of glutathione-S-transferase increased
after exposure to thiner and toluene. Expression of the superoxide
dismutase gene increased after exposure to toluene.
On the other hand, the effect observed at the level of reproduction and
development after exposure to eight chemical compounds was
abnormalities in the stages of larva and pupa induced by bisphenol A and
mercury (II) chloride, while phenol, toluene and metronidazole, showed
an effect on the pupa stage. Hydroquinone and hydrazine hydrochloride
affected the weight of the larva and pupa, respectively. The main
anomalies observed were necrosis in appendages of larvae, in pupae the
absence of papillae for sexual differentiation and abnormality in the
formation of head, extremities, wings and appendages.
Studies with two components of natural origin, indicated that β-pinene
was more potent than linalool to induce insect repellency, with RC50
values of 0.03 and 0.11 μL / cm2, respectively. Both compounds induced
overexpression of the Hiscl2 gene in adult insects, and β-pinene
promoted overexpression of the Grd and Ace1 gene. However, these
monoterpenes had little potential for coupling in computer-generated
models for several important proteins in the neurotransmission of T.
castaneum since their respective binding affinities were marginal, and
therefore the repellent action probably involved mechanisms other than
the interaction direct with these protein targets.
In summary, with the obtained results it is demonstrated that the T.
castaneum insect can be used as a toxicological model for the study of
volatile compounds, the effects are quantitatively measurable at the level of gene expression, development, growth, reproduction, and mortalityDoctoradoDoctor(a) en Toxicología Ambienta
Dominio fonológico como predictor del aprendizaje lector en niños y niñas de transición y primer año de educación básica primaria
Introduction: It is common to find among students who have learning difficulties, that this is accompanied by a deficit at the level of the reading processes, which makes it necessary to evaluate this complex and very important skill. Objectives: The study aimed to determine the phonological awareness skills as predictors of reading problems in transition children and first year of basic education, evaluated with the format: "Evaluation of Phonological Awareness (ECOFON)". Method: The sample consisted of 136 students from a Public Educational Institution of Sincelejo (Colombia). The research is descriptive of each of the components of phonological awareness. Results: obtaining as a result transition students obtained lower scores in the initial phoneme detection and suppression components. While the first-year students obtained the highest scores in segmentation and count of syllables, in the other components the scores were low. Conclusion: The results show that the phonological difficulties for rhyme at an early age are related to the reading domain; as well as the ignorance of the grapheme-phoneme relationship.Introducción: Es común encontrar entre los estudiantes que presentan dificultades del aprendizaje, que ésta, venga acompañada de déficit a nivel de los procesos lectores, lo que hace necesario evaluar esta habilidad compleja y muy importante. Objetivos: Se buscó determinar las habilidades de conciencia fonológica como predictor de problemas de lectura en niños de transición y primer año de educación básica, evaluados con el formato: “Evaluación de la Conciencia Fonológica (ECOFON)”. Método: La muestra la conformaron 136 alumnos de una Institución Educativa. de corte descriptivo. Resultados: Se encontró que estudiantes de transición obtuvieron puntuaciones más bajas en los componentes detección del fonema inicial y supresión. Mientras que los estudiantes de primero obtuvieron las puntuaciones más altas en segmentación y conteo de silabas, en los demás componentes las puntuaciones fueron bajas. Conclusión: Los resultados muestran que las dificultades fonológicas para la rima a temprana edad se relacionan con el dominio lector; al igual que el desconocimiento de la relación grafema-fonema
Química verde: Un nuevo reto
Currently there is a significant environmental degradation that has generated the need to search for alternatives leading to environmental sustainability. One of these tools is "green chemistry", a concept that involves the design of products and processes that reduce the generation of hazardous substances and maximize the efficiency in the utilization of material and energetic resources. The use of less pollutant technologies will allow chemical companies to mitigate the environmental effects associated with their business, reducing material consumption and increasing the participation of renewable resources. To achieve these goals, 12 basic principles of green chemistry have been proposed, applicable to different fields, such as medicine, agriculture, and chemical and pharmaceutical industries. This review outlines the principles and main uses of green chemistry, and their application as a philosophy of work to move towards a truly sustainable development.En la actualidad, existe un enorme deterioro del medio ambiente que ha generado la necesidad de buscar alternativas que conduzcan a la sostenibilidad ambiental. Una de estas herramientas es la "química verde", concepto que contempla el diseño de productos y procesos que reduzcan la generación de sustancias peligrosas y maximicen la eficiencia en la utilización de recursos materiales y energéticos. El empleo de tecnologías menos contaminantes, permitirá a las empresas químicas mitigar los efectos ambientales asociados a su actividad, reduciendo el consumo de materiales e incrementando la participación de recursos renovables. Para alcanzar estas metas, se han propuesto 12 principios básicos de química verde, aplicables en diferentes campos, tales como la medicina, la agricultura, la industria química y farmacéutica. Identificación in silico de moléculas potencialmente inhibidoras de CDK5, proteína relacionada con la enfermedad de Alzheimer
RESUMEN Introducción: La enfermedad de Alzheimer exhibe un compromiso neurodegenerativo e irreversible. Hoy, numerosas investigaciones promueven la inhibición de algunas quinasas para su tratamiento, de especial mención la CDK5. Objetivo: Identificación de nuevas moléculas con posibilidad de interactuar con la proteína quinasa dependiente de ciclina 5, CDK5, inhibiendo su función. Material y Métodos: Se realizó un estudio in silico, para lo cual se extrajeron 911 moléculas de pubchem, y mediante AutoDock Vina se hicieron acoplamientos moleculares con la proteína CDK5 extraída de Protein Data Bank y con un inhibidor conocido para la proteína. Además se realizó un acoplamiento inverso para la identificación de otros posibles blancos moleculares con los mejores ligandos seleccionados. Resultados: Con los resultados obtenidos fueron identificadas cinco moléculas con valores de afinidad entre -11,6 hasta -17,7 Kcal/mol que se unen en el sitio activo de la proteína, de igual forma que lo hace el inhibidor conocido de la misma, e interactúan con los residuos cisteína 83 y glutamina 81. Conclusiones: Las moléculas identificadas pueden interactuar con la CDK5 a nivel de su sitio activo, por lo que podrían actuar como inhibidores de esta quinasa. Esto abre una futura ventana terapéutica en el tratamiento de la enfermedad de Alzheimer

Identificación in silico de moléculas potencialmente inhibidoras de CDK5, proteína relacionada con la enfermedad de Alzheimer
Introduction: The illness of Alzheimer exhibits a neurodegenerative and irreversible commitment. Today, numerous investigations promote the inhibition of some kinases to the treatment, of special mention the CDK5. Objective: Identification of new molecules witch are able to interact with the cicline dependent kinase protein 5, CDK5, inhibiting their function. Material and Methods: it was carried out a study in silico, for that 911 pubchem molecules were extracted, and by means of AutoDock Vina molecular joining were made with the protein CDK5 extracted from the Protein Data Bank and with a well-known inhibitor for the protein. It was also carried out an inverse joining for the identification of other possible molecular targets with the best selected ligands. Results: With the obtained results five molecules were identified with values of likeness among -11,6 until -17,7 Kcal/mol that joins in the active site of the protein, in the same form that makes it the well-known inhibitor of the CDK5, and interact with the residuals cysteine 83 and glutamine 81. Conclusions: The identified molecules can interact with the CDK5 at level of their active place, for what you/they could act as inhibitors of this quinasa. This opens a future therapeutic window in the treatment of the illness of Alzheimer.
